2SP1 governs primordial folliculogenesis by regulating pregranulosa cell development in mice显示文摘Establishment of the primordial follicle(PF)pool is pivotal for the female reproductive lifespan;however,the mechanism of primordial folliculogenesis is poorly understood.Here,the transcription factor SP1 was shown to be essential for PF formation in mice.Our results showed that SP1 is present in both oocytes and somatic cells during PF formation in the ovary.Knockdown of Sp1 expression,especially in pregranulosa cells,significantly suppressed nest breakdown,oocyte apoptosis,and PF formation,suggesting that SP1 expressed by somatic cells functions in the process of primordial folliculogenesis.We further demonstrated that SP1 governs the recruitment and maintenance of Forkhead box L2-positive(FOXL2^+)pregranulosa cells using an Lgr5-EGFP-IRES-CreER^T2(Lgr5-KI)reporter mouse model and a FOXL2^+ cell-specific knockdown model.At the molecular level,SP1 functioned mainly through manipulation of NOTCH2 expression by binding directly to the promoter of the Notch2 gene.Finally,consistent with the critical role of granulosa cells in follicle survival in vitro,massive loss of oocytes in SP1 knockdown ovaries was evidenced before puberty after the ovaries were tranSP1anted under the renal capsules.Conclusively,our results reveal that SP1 controls the establishment of the ovarian reserve by regulating pregranulosa cell development in the mammalian ovary.Han Cai Bingying Liu Huarong Wang Guanghong Sun Lizhao Feng Ziqi Chen Jiaqi Zhou Jiawei Zhang Tuo Zhang Meina He Tingting Yang Qirui Guo Zhen Teng Qiliang Xin Bo Zhou Hua Zhang Guoliang Xia Chao Wang 2020Journal of Molecular Cell Biology2020,12,3:3

4Geochemistry, Sr-Nd-Pb Isotopic Compositions and Zircon U-Pb Geochronology of Neoproterozoic Mafic Dyke in the Douling Complex, South Qinling Belt, China显示文摘Mafic dyke swarm is widely developed in Proterozoic continental lithosphere.The Gangou diabase dyke from the Xixia area,intruded into the Douling complex of the South Qinling belt,yields zircon UPb age of 731 Ma.It has low SiO2 content(49.02 wt.%-49.37 wt.%)and Mg^#(34.0–37.7)and shows characteristics of subalkaline tholeiite series.They show highΣREE(155.5×10^-6–184.7×10^-6),weak negative Eu anomaly(δEu=0.88-0.93),slight depletion of Nb and Ta,and enrichment of LILE.Their major and trace element,Sr-Nd-Pb isotope and clinopyroxene compositions indicate that it originated from the partial melting of the asthenospheric mantle within-plate extension setting and was slightly contaminated by crust compositions.The similarities of formation age,petrogenesis,source characteristic and tectonic setting for Gangou diabase with mafic dykes in Wudang Block and mafic volcanic rocks in Yaolinghe Group indicate that the South Qinling belt underwent strong continental extension in Neoproterozoic during 796–685 Ma most likely correspondence to the breaking-up of the Rodinia supercontinent.Tong Li Liang Liu Xiao-Ying Liao Yong-Sheng Gai Tuo Ma Chao Wang 2020Journal of Earth Science2020,31,2:2
5Three-dimensional characterization of micro-fractures in shale reservoir rocks显示文摘Fractures are crucial for unconventional hydrocarbon exploitation,but it is difficult to accurately observe the 3D spatial distribution characteristics of fractures.Microtomography(micro-CT)technology makes it possible to observe the 3D structures of fractures at micro-scale.In this study,micron-CT scanning is conducted on multiple mud-shale samples of source rocks in the Permian Lucaogou Formation,Junggar Basin.The Avizo^(®) software is applied to process and segment the micron-CT images,so as to obtain the 3D fracture structure model inside rock core.Therefore,the independently-developed CTSTA program is adopted to quantitatively describe the micro-fractures inside rock core,including fracture dimension,extension direction and extension scale.Meanwhile,this study summarizes the classification characteristics of fractures and their anisotropy.On this basis,the fractal dimensions of fractures can also be extracted.Previous studies show that the geometric features of fractures have self-similarity at large and small scales,which can be described by exponential laws;and the fractal dimension is a typical exponent.Through the quantitative description or characterization of 3D fractures at micro-scale,the distribution characteristics of fractures in large scales could be known.Chao Qi Xiaoqi Wang Wei Wang Jie Liu Jincai Tuo Keyu Liu 2018Petroleum Research2018,3,3:1

9Comparison of percutaneous endoscopic cervical discectomy versus anterior cervical discectomy and fusion for cervical disc herniation:a meta-analysis显示文摘Purpose:The purpose of the study is to perform a systematic review and meta-analysis to evaluate the clinical results of percutaneous endoscopic cervical discectomy(PECD)and anterior cervical discectomy and fusion(ACDF)for the treatment of cervical disc herniation(CDH).Methods:Randomized controlled trials or non-randomized controlled trials published from the time when databases were built to October 2020 that compared the clinical effectiveness of PECD and ACDF surgical approaches for the treatment of CDH were acquired hy a comprehensive search in four electronic databases(Pub Med,EMBASE,and Web of Science).A total of 5 studies(559 patients)were included in this systematic review and meta-analysis.Pooled mean differences(MD)and odds ratios(OR)and with 95% CIs were calculated for the outcomes.Results:The results showed that there were no statistically between the PECD group and ACDF group in terms of preoperative VASN score(WMD=0.22;95%CI:-0.52 to 0.97;P=0.56),preoperative VASA score(WMD=-0.13;95%CI:-0.42 to 0.16;P=0.37),postoperative VASN score(WMD=0.14;95%CI:-0.12 to 0.40;P=0.28),postoperative VASA score(WMD=-0.08;95%CI:-0.28 to 0.11;P=0.40),preoperative NDI(WMD=-0.75;95%CI:-5.60 to 4.11;P=0.76),postoperative NDI(WMD=0.22;95%CI:-1.44 to 1.88;P=0.80),,reoperation rate(OR=1.35;95%CI:0.49 to 3.72;P=0.57)or operation time(WMD=-11.76;95%CI:-71.23 to 47.72;P=0.70).PECD group was associated with lower complication rate(OR=0.34;95%CI:0.15 to 0.78;P=0.01)and shorter hospital stay(WMD=-2.59;95%CI:-3.67 to -1.52;P<0.00001).Conclusion:The existing evidence indicates that no superiority exists between the two surgical approaches for the treatment of CDH in terms of functional outcome,operation lime,and reoperation rate,despite that PECD surgical group can achieve lower complication rates and shorter hospital stay than ACDF surgical group.李庆达 章雪芳 杨俊松 Chen Chienmin Liu Tuanjiang He Baorong Jiang Chao Tian Xin Yang Mingyi Chai Xin Tuo Yuan Hao Dingjun 2021美中国际创伤杂志2021,20,2:0
10Unveiling clinical significance and tumor immune landscape of CXCL12 in bladder cancer: Insights from multiple omics analysis显示文摘Objective: The interplay between chemokine C-X-C motif ligand 12(CXCL12) and its specific receptors is known to trigger various signaling pathways, contributing to tumor proliferation and metastasis. Consequently,targeting this signaling axis has emerged as a potential strategy in cancer therapy. However, the precise role of CXCL12 in clinical therapy, especially in immunotherapy for bladder cancer(BCa), remains poorly elucidated.Methods: We gathered multiple omics data from public databases to unveil the clinical relevance and tumor immune landscape associated with CXCL12 in BCa patients. Univariate and multivariate Cox regression analyses were employed to assess the independent prognostic significance of CXCL12 expression and formulate a nomogram. The expression of CXCL12 in BCa cell lines and clinical tissue samples was validated using enzymelinked immunosorbent assays(ELISA) and immunohistochemistry(IHC).Results: While transcriptional expression of CXCL12 exhibited a decrease in nearly all tumor tissues, CXCL12 methylation expression was notably increased in BCa tissues. Single-cell RNA analysis highlighted tissue stem cells and endothelial cells as the primary sources expressing CXCL12. Abnormal CXCL12 expression, based on transcriptional and methylation levels, correlated with various clinical characteristics in BCa patients. Functional analysis indicated enrichment of CXCL12 and its co-expression genes in immune regulation and cell adhesion. The immune landscape analysis unveiled a significant association between CXCL12 expression and M2 macrophages(CD163~+ cells) in BCa tissues.
