| 1 | Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota. | Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng | 2019 | Cell Research2019,29,10: | 192 |

| 3 | Liver transplant recipients with portal vein thrombosis:a single center retrospective study显示文摘BACKGROUND:Portal vein thrombosis(PVT)used to be a contraindication for liver transplantation(LT).This obstacle has been delt with following the improvement of LT-related techniques and therapeutic approaches to thrombosis. But the effect of PVT on LT outcomes is still controversial. We reviewed retrospectively the outcome of LT patients with PVT as well as risk factors and surgical management according to PVT grades. METHODS:A total of 465 adult LTs were performed from December 2002 through December 2006.Operative findings and the result of preoperative ultrasonography and imaging were reviewed for PVT grading(Yerdel grading).Comparison of risk factors,variables associated with perioperative period and prognosis between recipients with and without PVT is based on the grades. RESULTS:In the 465 LTs,42 were operatively confirmed to have PVT(9.0%)(19 recipients with grade 1,14 with grade 2,7 with grade 3,and 2 with grade 4).Increased age and treatment of portal hypertension were associated with PVT.Grade 1 or 2 PVT was treated by direct anastomosis or single thrombectomy.In grade 3 PVT patients,the donor PV was directly anastomosed to the dilated branch of the recipient portal venous system or to the distal open superior mesenteric vein through an interposition vein graft if needed.Grade 4 PVT was managed by our modified cavoportal hemitransposition technique.The comparison between PVT patients and controls showed no significant difference in operative duration and blood transfusion(P>0.05).The flow rate of the PV was lower in the PVT patients(48.881±12.788 cm/s)than in the controls(57.172±21.715 cm/s,P<0.05).The PVT patients had such postoperative complications as renal failure and PV rethrombosis(P<0.05).The 1-year survival rates in PVT and non-PVT patients were 78.6%and 76.4%respectively (P>0.05);the 3-year survival rates were 58.8%and 56.4% respectively(P>0.05). | 4 | Isolation and culture of adult Sertoli cells and their effects on the function of co-cultured allogeneic islets in vitro显示文摘Background Globally, 180 million people suffer from diabetes mellitus. Islet transplantation is believed to be an almost ideal therapy for insulin-dependent patients. How to maintain the viability and the function of isolated human islets is a challenge in clinical practice. Sertoli cells are considered ‘nurse cells’in the seminiferous tubules and have been used in cell graft protocols for neurodegenerative diseases and diabetes in many studies. Many researchers have used immature murine testes as the primarily source of Sertoli cells in islet transplantation because they are easily purified. Mature human Sertoli cells have been seldom investigated. In the present study, we developed a method for the isolation and culture of Sertoli cells derived from adult human testes, and investigated their effects on the function of allogeneic islets when they were cultured together in vitro. Methods Adult Sertoli cells were prepared successfully by two-step enzyme digestion with trypsin, collagenase and hyaluronidase. They were identified by morphological characteristics and their activity was determined by MTT colorimetry over a 28-day culture time in vitro. A glucose-stimulated insulin secretion test was performed to detect the effects of Sertoli cells on allogeneic islets’ function when they were co-cultured for 21 days in vitro. Results In cultured cells, mature human Sertoli cells accounted for more than 90% of total cells. The activity of Sertoli cells reached 95% and they remained highly cytoactive for a long time in vitro (P>0.05). Compared with the islets cultured alone, the co-cultured islets with allogeneic Sertoli cells maintained higher sensitivity to glucose stimulation for the duration of the experiment (P<0.01). Conclusions A method of isolation and culture of Sertoli cells from adult testes has been established. Sertoli cells could enhance allogeneic islets’ function when they were co-cultured in vitro. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Real-time signal and information processor is an important module for real-time positioning, imaging, detection and recognition of targets. With the development of ultra-wideband technology, synthetic aperture technology, signal and information processing technology, the radar coverage, detection accuracy and resolution have been greatly improved, especially in terms of one-dimensional(1D) high-resolution radar detection, tracking, recognition, and two-dimensional(2D) synthetic aperture radar imaging technology. Meanwhile, for the application of radar detection and remote sensing with high resolution and wide swath, the amount of data has been greatly increased. Therefore, the radar is required to have low-latency and real-time processing capability under the constraints of size, weight and power consumption. This paper systematically introduces the new technology of high resolution radar and real-time signal and information processing. The key problems and solutions are discussed, including the detection and tracking of 1D high-resolution radar, the accurate signal modeling and wide-swath imaging for geosynchronous orbit synthetic aperture radar, and real-time signal and information processing architecture and efficient algorithms. | 15 | Two case reports and literature review for hepatic epithelioid angiomyolipoma: Pitfall of misdiagnosis显示文摘BACKGROUND Hepatic epithelioid angiomyolipoma(HEAML) is a rare liver disease and is easily misdiagnosed. Enhanced recognition of HEAML is beneficial to the differential diagnosis of rare liver diseases.CASE SUMMARY We presented two cases of HEAML in Changzheng Hospital, Naval Medical University, and then collected and analyzed all reports about HEAML recorded in PubMed, MEDLINE, China Science Periodical Database, and VIP database from January 2000 to March 2018. A total of 409 cases of HEAML in 97 reports were collected, with a ratio of men to women of 1:4.84 and an age range from 12 years to 80 years(median 44 years). Among the patients with clinical symptoms mentioned, 61.93%(205/331) were asymptomatic, 34.74%(115/331) showed upper or right upper quadrant abdomen discomfort, while a few of them showed abdominal mass, gastrointestinal symptoms, low fever, or weight loss. The misdiagnosis rate of HEAML was as high as 40.34%(165/409) due to its nonspecific imaging findings. Most of the tumors were solitary and round in morphology, with clear boundaries. Ultrasound scan indicated low echo with internal nonuniformity and rich blood supply in most cases. Computer tomography/magnetic resonance imaging enhanced scan showed varied characteristics. The ratio of fast wash-in and fast wash-out, fast wash-in and slow wash-out, and delayed enhancement was roughly 4:5:1. A definite diagnosis of HEAML depended on the pathological findings of the epithelioid cells in lesions and the expression of human melanoma black 45, smooth muscle actin,melanoma antigen, and actin by immunohistochemical staining. HEAML had a relatively low malignant rate of 3.91%. | 16 | An optimization method for metamorphic mechanisms based on multidisciplinary design optimization显示文摘The optimization of metamorphic mechanisms is different from that of the conventional mechanisms for its characteristics of multi-configuration. There exist complex coupled design variables and constraints in its multiple different configuration optimization models. To achieve the compatible optimized results of these coupled design variables, an optimization method for metamorphic mechanisms is developed in the paper based on the principle of multidisciplinary design optimization(MDO). Firstly, the optimization characteristics of the metamorphic mechanism are summarized distinctly by proposing the classification of design variables and constraints as well as coupling interactions among its different configuration optimization models. Further, collaborative optimization technique which is used in MDO is adopted for achieving the overall optimization performance. The whole optimization process is then proposed by constructing a two-level hierarchical scheme with global optimizer and configuration optimizer loops. | 17 | Eogenetic Karst in Interbedded Carbonates and Evaporites and Its Impact on Hydrocarbon Reservoir: A New Case from Middle Triassic Leikoupo Formation in Sichuan Basin, Southwest China显示文摘Karst in interbedded carbonates and evaporites has been reported to have important and complex impacts on reservoir. It is significant for exploration and karst geology. Here, we report such a new case from Middle Triassic Leikoupo Formation of Sichuan Basin, Southwest China. Stratigraphic incompleteness and the occurrence of unconformity provide evidence for the presence of eogenetic karst. Under the impact of this eogenetic karst, residual weathered and solution-collapse breccia, solution pores and silicification and dedolomitization have been observed. Classic stratigraphic zonation of karst is not readily distinguishable, which is ascribed to the stratigraphic collapse of carbonate rocks resulting from the dissolution of evaporites by lateral subsurface fluid flow. In terms of impact on reservoir quality, karst can generally improve the initial physical property of the porous layers in theory. However, subsurface fluid flow dissolved the evarporitic beds and facilitated the collapse of overlying strata. As a consequence, the lateral continuity of the reservoirs would be destroyed, and relatively high-quality reservoirs can only be developed with little collapse of overlying strata, reflecting reservoir heterogeneities. | 18 | Electrical and optical properties of ZnO:Al films with different hydrogen contents in sputtering gas显示文摘Aluminum-doped zinc oxide(Zn O:Al) films were deposited by direct current magnetron sputtering in incorporating hydrogen in sputtering gas at room temperature. The influences of hydrogen content in sputtering gas on the structural, optical, and electrical properties of Zn O:Al films were systematically investigated. It is found that hydrogen incorporated into Zn O lattice forms shallow donors in Zn O:Al films and plays an important role in the properties of Zn O:Al films. The electrical conductivity and infrared(IR) reflectance are improved due to the increase of electron carrier concentration, and the average transmittance decreases, which is ascribed to the strong scattering from the hydrogen incorporated and oxygen vacancies in Zn O:Al films. In this study,the resistivity of 5.5×10^(-4) Ω·cm is obtained, the average transmittance of the wavelength in the range of 400–900 nm is almost 86%, and the IR reflectance reaches 75% at 2,500 nm,which is higher than that of reported TCO films. The band gap determined by optical absorption is a result of competition between Burstein–Moss effect and many-body perturbation effect. | 20 | Clonality and allelotype analyses of focal nodular hyperplasia compared with hepatocellular adenoma and carcinoma显示文摘AIM: To identify clonality and genetic alterations in focal nodular hyperplasia (FNH) and the nodules derived from it. METHODS: Twelve FNH lesions were examined. Twelve hepatocellular adenomas (HCAs) and 22 hepatocellular carcinomas (HCCs) were used as references. Nodules of different types were identified and isolated from FNH by microdissection. An X-chromosome inactivation assay was employed to describe their clonality status. Loss of heterozygosity (LOH) was detected, using 57 markers, for genetic alterations.RESULTS: Nodules of altered hepatocytes (NAH), the putative precursors of HCA and HCC, were found in all the FNH lesions. Polyclonality was revealed in 10 FNH lesions from female patients, and LOH was not detected in any of the six FNH lesions examined, the results apparently showing their polyclonal nature. In contrast, monoclonality was demonstrated in all the eight HCAs and in four of the HCCs from females, and allelic imbalances were found in the HCAs (9/9) and HCCs (15/18), with chromosomal arms 11p, 13q and 17p affected in the former, and 6q, 8p, 11p, 16q and 17p affected in the latter lesions in high frequencies (≥ 30%). Monoclonality was revealed in 21 (40%) of the 52 microdissected NAH, but was not found in any of the f ive ordinary nodules. LOH was found in all of the 13 NAH tested, being highly frequent at six loci on 8p, 11p, 13q and 17p.
