| 1 | IAE-adaptive Kalman filter for INS/GPS integrated navigation system显示文摘A marine INS/GPS adaptive navigation system is presented in this paper. GPS with two antenna providing vessel’s altitude is selected as the auxiliary system fusing with INS to improve the performance of the hybrid system. The Kalman filter is the most frequently used algorithm in the integrated navigation system, which is capable of estimating INS errors online based on the measured errors between INS and GPS. The standard Kalman filter (SKF) assumes that the statistics of the noise on each sensor are given. As long as the noise distributions do not change, the Kalman filter will give the optimal estimation. However GPS receiver will be disturbed easily and thus temporally changing measurement noise will join into the outputs of GPS, which will lead to performance degradation of the Kalman filter. Many researchers introduce fuzzy logic control method into innovation-based adaptive estimation adaptive Kalman filtering (IAE-AKF) algorithm, and accordingly propose various adaptive Kalman filters. However how to design the fuzzy logic controller is a very complicated problem still without a convincing solution. A novel IAE-AKF is proposed herein, which is based on the maximum likelihood criterion for the proper computation of the filter innovation covariance and hence of the filter gain. The approach is direct and simple without having to establish fuzzy inference rules. After having deduced the proposed IAE-AKF algorithm theoretically in detail, the approach is tested by the simulation based on the system error model of the developed INS/GPS integrated marine navigation system. Simulation results show that the adaptive Kalman filter outperforms the SKF with higher accuracy, robustness and less computation. | 2 | Propofol differentially induces unconsciousness and respiratory depression through distinct interactions between GABAa receptor and GABAergic neuron in corresponding nuclei显示文摘Propofol is the most commonly used intravenous anesthetic worldwide.It can induce loss of consciousness prior to the occurrence of severe respiratory suppression,which is also a pharmacodynamic feature of all general anesthetics.However,the neural mechanisms underlying this natural phenomenon are controversial and highly related to patient safety.In the present study,we demonstrated that the pharmacodynamic effects of propofol(50 and 100 μM)on suppression of consciousness-related excitatory postsynaptic currents in the medial prefrontal cortex(mPFC)and centromedian nucleus of the thalamus(CMT)were lower than those in the kernel respiratory rhythmogenesis nucleus pre-Botzinger complex(PrBo).Furthermore,we unexpectedly found that the GABAa receptor β3 subunit is the key target for propofol's action and that it is mutually and exclusively expressed in GABAergic neurons.It is also more abundant in the mPFC and CMT,but mainly co-localized with GABAergic neurons in the PrBo.As a result,the differentiated expression pattern should mediate more neuron suppression through the activation of GABAergic neurons in the mPFC and CMT at low doses of propofol(50 μM).However,PrBo GABAergic neurons were only activated by propofol at a high dose(100 μM).These results highlight the detailed pharmacodynamic effects of propofol on consciousness-related and respiration-related nuclei and provide the distinct interaction mechanism between the β3 subunit and GABAergic neurons in mediating the suppression of consciousness compared to the inhibition of respiration. | Junli Jiang Yingfu Jiao Po Gao Wen Yin Wei Zhou Yunchun Zhang Yanjun Liu Daxiang Wen Yuan Wang Liang Zhou Tian Yu Weifeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,8: | 11 |

| 4 | De-Noising Stochastic Noise in FOG Based on Second-Generation DB4 Wavelet and SURE-Threshold显示文摘An effective de-noising method for fiber optic gyroscopes (FOGs) is proposed. This method is based on second-generation Daubechies D4 (DB4) wavelet transform (WT) and level-dependent threshold estimator called Stein's unbiased risk estimator (SURE). The whole approach consists of three critical parts: wavelet decomposition module, parameters estimation module and SURE de-noising module. First, DB4 wavelet is selected as lifting base of the second-generation wavelet in the decomposition module. Second, in the parameters estimation module, maximum likelihood estimation (MLE) is used for stochastic noise parameters estimation. Third, combined with soft threshold de-noising technique, the SURE de-noising module is designed. For comparison, both the traditional universal threshold wavelet and the second-generation Harr wavelet method are also investigated. The experiment results show that the computation cost is 40% less than that of the traditional wavelet method. | 5 | Negative selection of murine medullary-type single positive thymocytes显示文摘Negative selection depletes self-reactive T cells, thus ensuring self-tolerance. It is usually considered that negative selection imposed on double-positive (DP) thymocytes that reside at the cortico-medullary junction. Negative selection model was set up by injecting mice with anti-T cell receptor (TCR) monoclonal antibody (mAb) intraperitoneally in this work. As shown in phenotypic analysis of thymocytes, negative selection destroys not only cortical-type DP thymocytes, but also medullary-type CD3+TCRαβ+CD4SP and CD3+TCRαβ+CD8SP thymocytes. Negative selection of medullary-type single positive (SP) are more susceptible to apoptosis, while with development of the cells, their resistance to apoptosis increases. Therefore, negative selection does not operate on functionally mature thymocytes at the late stage. This result is a supplement to the traditional theory of negative selection. | 16 | Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology.
