1Effect of viscoelasticity on skin pain sensation显示文摘Pain sensation may appear under long-lasting mechanical stimulation. Although people have the experience that pain sensation generally decreases with time while the stimulation remains, the underlying mechanism remains elusive. We experimentally studied the thermal and strain ratedependent viscoelastic behavior of skin in uniaxial stretch and numerically investigated the effects of temperature and strain rate on pain sensation. The results indicate that the viscosity of skin tissue decreases with increasing temperature and reducing strain rate, which subsequently decreases the discharge frequency of skin nociceptor and thus relieves the pain sensation. 9Enzymatic synthesis of indigo derivatives by tuning P450 BM3 peroxygenases显示文摘Indigoids,a class of bis-indoles,have long been applied in dyeing,food,and pharmaceutical industries.Recently,interest in these‘old’molecules has been renewed in the field of organic semiconductors as functional building blocks for organic electronics due to their excellent chemical and physical properties.However,these indigo derivatives are difficult to access through chemical synthesis.In this study,we engineer cytochrome P450 BM3 from an NADPH-dependent monooxygenase to peroxygenases through directed evolution.A select number of P450 BM3 variants are used for the selective oxidation of indole derivatives to form different indigoid pigments with a spectrum of colors.Among the prepared indigoid organic photocatalysts,a majority of indigoids demonstrate a reduced band gap than indigo due to the increased light capture and improved charge separation,making them promising candidates for the development of new organic electronic devices.Thus,we present a useful enzymatic approach with broad substrate scope and cost-effectiveness by using low-cost H2O2 as a cofactor for the preparation of diversified indigoids,offering versatility in designing and manufacturing new dyestuff and electronic/sensor components.Li Ma Tianjian Sun Yunjie Liu Yue Zhao Xiaohui Liu Yuxuan Li Xinwei Chen Lin Cao Qianqian Kang Jiawei Guo Lei Du Wei Wang Shengying Li 2023Synthetic and Systems Biotechnology2023,8,3:0
10Nondestructive monitoring of annealing and chemical-mechanical planarization behavior using ellipsometry and deep learning显示文摘The Cu-flling process in through-silicon via(TSV-Cu)is a key technology for chip stacking and three-dimensional vertical packaging.During this process,defects resulting from chemical-mechanical planarization(CMP)and annealing severely affect the reliability of the chips.Traditional methods of defect characterization are destructive and cumbersome.In this study,a new defect inspection method was developed using Mueller matrix spectroscopic ellipsometry.TSV-Cu with a 3-μm-diameter and 8-μm-deep Cu filling showed three typical types of characteristics:overdishing(defect-OD),protrusion(defect-P),and defect-free.The process dimension for each defect was 13 nm.First,the three typical defects caused by CMP and annealing were investigated.With single-channel deep learning and a Mueller matrix element(MME),the TSV-Cu defect types could be distinguished with an accuracy rate of 99.94%.Next,seven effective MMEs were used as independent channels in the artificial neural network to quantify the height variation in the Cu flling in the z-direction.The accuracy rate was 98.92%after training,and the recognition accuracy reached 1 nm.The proposed approach rapidly and nondestructively evaluates the annealing bonding performance of CMP processes,which can improve the reliability of high-density integration.Qimeng Sun Dekun Yang Tianjian Liu Jianhong Liu Shizhao Wang Sizhou Hu Sheng Liu Yi Song 2023Microsystems & Nanoengineering2023,9,2:0
11Application of eDNA Metabarcoding for Detecting Anura in North China显示文摘eDNA metabarcoding is an advanced method formonitoring biodiversityproposed in recent years.By analyzing DNA in water,soil and sediment samples,the technology obtains species distribution and population quantity information.It was found that macrobarcode technology is more accurate than the traditional method in measuring the species richness of some groups.In Europe,America and South America,the relia bility of this technology in monitoring amphibian diversity in the wild was studied,and it was found to be better than traditional biodiversity monitoring methods in detecting species diversity.At present,amphibian monitoring mainly depends on various traditional methods,such as transects,drift fence traps,artificial shelters and mark-recapture.These monitoring techniques have many shortcomings,such as low accuracy and strong subjectivity of study results.These technologies have poor effects on rare,invasive and endangered species with strong concealment ability,low density and strong seasonality and are difficult to implement in sites inaccessible to people.Traditional monitoring technology also requires considerable investment of human and material resources,and the economic cost is relatively high,while eDNA metabarcoding ismore efficient and less costly,so it is important to use eDNA meta barcoding in amphibian monitoring in China.In this study,the eDNA meta barcoding and traditional line transect method(TLTM)were used to study the characteristics of the two methods in the Beijing-Tianjin-Hebeiregion.Repeated samplingwas conducted on 58 waterbodies in July 2019 and June 2020.After sequencing the samples using highthroughput sequencing technology,the differences between metabarcoding and commonly used TLTM surveys in detecting the diversity of four amphibians in North China were assessed.Our results showed that eDNA meta barcoding is more sensitive to the detection of the four amphibian species in the sampling area,and the combined use of eDNA metabarcoding and TLTM can improve the survey results of amphibians in the survey area to the greatest extent.In addition,in the process of species classification and identification of metabarcoding results,7 species of reptiles were detected,indicating that eDNA metabarcoding is also useful to detect reptiles.The results of this study indicate that metabarcoding in combination with TLTM can accurately estimate the diversityof amphibians in a short-term survey in North China and is also useful in reptile species detection.Wenhao LI Mingshuo QIN Xianglei HOU Jiaqi ZHANG Siqi WANG Yu LI Zexu LUO Teng DENG Tianjian SONG Chunxia XU Xuan LIU Xuyu WANG Yiming LI 2022Asian Herpetological Research2022,13,4:0
12Development of a micro-indentation device for measuring the mechanical properties of soft materials显示文摘Indentation is a simple and nondestructive method to measure the mechanical properties of soft materials,such as hydrogels,elastomers and soft tissues.In this work,we have developed a micro-indentation system with high-precision to measure the mechanical properties of soft materials,where the shear modulus and Poisson’s ratio of the materials can be obtained by analyzing the load–relaxation curve.We have validated the accuracy and stability of the system by comparing the measured mechanical properties of a polyethylene glycol sample with that obtained from a commercial instrument.The mechanical properties of another typical polydimethylsiloxane sample submerged in heptane are measured by using conical and spherical indenters,respectively.The measured values of shear modulus and Poisson’s ratio are within a reasonable range.Xiang He Pengfei Wang Guoyou Huang Shaobao Liu Minglong Xu Feng Xu TianJian Lu 2013Theoretical & Applied Mechanics Letters2013,3,5:0
13Development of MEMS directed evolution strategy for multiplied throughput and convergent evolution of cytochrome P450 enzymes显示文摘Directed evolution(DE)inspired by natural evolution(NE)has been achieving tremendous successes in protein/enzyme engineering.However,the conventional'one-protein-for-one-task'DE cannot match the'multi-proteins-for-multi-tasks'NE in terms of screening throughput and efficiency,thus often failing to meet the fast-growing demands for biocatalysts with desired properties.In this study,we design a novel'multi-enzymes-for-multi-substrates'(MEMS)DE model and establish the proof-ofconcept by running a NE-mimicking and higher-throughput screening on the basis of'two-P450 s-against-seven-substrates'(2P×7S)in one pot.With the multiplied throughput and improved hit rate,we witness a series of convergent evolution events of the two archetypal cytochrome P450 enzymes(P450 BM3 and P450 cam)in laboratory.It is anticipated that the new strategy of MEMS DE will find broader application for a larger repertoire of enzymes in the future.Furthermore,structural and substrate docking analysis of the two functionally convergent P450 variants provide important insights into how distinct P450 active-sites can reach a common catalytic goal.Li Ma Fengwei Li Xingwang Zhang Hui Chen Qian Huang Jing Su Xiaohui Liu Tianjian Sun Bo Fang Kun Liu Dandan Tang Dalei Wu Wei Zhang Lei Du Shengying Li 2022Science China(Life Sciences)2022,65,3:0
