| 1 | Stable expression of Arabidopsis vacuolar Na^+/H^+ antiporter gene AtNHX1,and salt tolerance in transgenic soybean for over six generations显示文摘The Arabidopsis vacuolar Na+/H+ antiporter gene,AtNHX1,was introduced into soybean by Agrobacterium-mediated transformation.Four independent kanamycin resistant lines were obtained.The result of PCR,Southern blotting and Northern blotting analyses demonstrated that the AtNHX1 gene was successfully inserted into the soybean genome and stably expressed in these kanamycin resistant lines.The stability of AtNHX1 expression and salt resistance were evaluated in the soybean transformants for over 6 generations.Two independently derived transgenic lines with high expression level of AtNHX1 were selected,and propagated to generation T5 in the absence of selection pressure.PCR and RT-PCR examinations revealed that AtNHX1 was highly expressed in all investigated transgenic T5 progenies.Furthermore,all transgenic T5 plants showed resistant to salt stress,same as those of homozygous T2 plants.Taken together,our results indicated that constitutive expression of AtNHX1 enhanced salt tolerance in soybean for over 6 generations,suggesting a great potential use of AtNHX1 for improving salt tolerance in plants by genetic engineering. | LI TianXing ZHANG Yue LIU Hua WU YuTing LI WenBin ZHANG HongXia | 2010 | Chinese Science Bulletin2010,55,12: | 8 |
| 2 | A Virulent PEDV Strain FJzz1 with Genomic Mutations and Deletions at the High Passage Level Was Attenuated in Piglets via Serial Passage In Vitro显示文摘Highly virulent porcine epidemic diarrhea virus(PEDV)strains re-emerged and circulated in China at the end of 2010,causing significant economic losses in the pork industry worldwide.To understand the genetic dynamics of PEDV during its passage in vitro,the PEDV G2 strain FJzzl was serially propagated in Vero cells for up to 200 passages.The susceptibility and adaptability of the FJzzl strain increased gradually as it was serially passaged in vitro.Sequence analysis revealed that amino acid(aa)changes were mainly concentrated in the S glycoprotein,which accounted for 72.22%-85.71%of all aa changes.A continuous aa deletion(^(55)I^(56)G^(57)E→^(55)K^(56)Δ^(57)Δ)occurred in the N-terminal domain of S1(Sl-NTD).To examine how the aa changes affected its virulence,FJzzl-F20 and FJzzl-F200 were selected to simultaneously evaluate their pathogenicity in suckling piglets.All the piglets in the FJzzl-F20-infected group showed typical diarrhea at 24 h postinfection,and the piglets died successively by 48 h postinfection.However,the clinical signs of the piglets in the FJzzl-F200-infected group were significantly weaker,and no deaths occurred.The FJzzl-F200-infected group also showed a lower level of fecal viral shedding and lower viral loads in the intestinal tissues,and no obvious histopathological lesions.TypeⅠandⅢinterferon were induced in the FJzzl-F200 infection group,together with pro-inflammatory cytokines,such as TNF-α,IL-1βand IL-8.These results indicate that the identified genetic changes may contribute to the attenuation of FJzzl strain,and the attenuated FJzzl-F200 may have the potential for developing PEDV live-attenuated vaccines. | Pengfei Chen Xiongwei Zhao Shuting Zhou Tianxing Zhou Xiangmei Tan Xia Wu Wu Tong Fei Gao Lingxue Yu Yifeng Jiang Hai Yu Zhibiao Yang Guangzhi Tong Yanjun Zhou | 2021 | Virologica Sinica2021,36,5: | 4 |
| 3 | Novel metastable Bi:Co and Bi:Fe alloys nanodots@carbon as anodes for high rate K-ion batteries显示文摘Bi is a promising anode material for potassium-ion batteries(PIBs)due to its high theoretical capacity.However,severe pulverization upon cycling limits its practical applications.In this work,we propose a new approach of using metastable alloys with Bi elements.Metastable Bi:Co and Bi:Fe alloys nanodots@carbon anode materials(Bi:Co and Bi:Fe@C)are synthesized for the first time via simple annealing of their metal-organic frameworks(MOF)precursors.These prepared materials are demonstrated as ideal hosts for high-rate K-ion storage.Bi_(0.85)Co_(0.15)@C and Bi_(0.83)Fe_(0.17)@C electrodes respectively deliver superior 178 and 253 mAh·g^(−1)at 20 A·g^(−1),as well as stable cycling performance at 2 A·g^(−1).Ex situ scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD),and transmission electron microscopy(TEM)studies on Bi:Co@C indicate that the elemental Co separates out during the initial potassiation and stands during the following discharge/charge cycles.In situ formed Co precipitates can act as(1)“conductive binders”as well as(2)“separators”to prevent the severe aggregation of adjacent active elemental Bi nanoparticles and(3)accelerate the potassiation/de-potassiation kinetics in elemental Bi precipitates after initial discharge/charge cycles.This work could inspire the development of metal-type anodes. | Zhongqiu Tong Tianxing Kang Yan Wu Fan Zhang Yongbing Tang Chun-Sing Lee | 2022 | Nano Research2022,15,8: | 2 |

| 15 | Characteristics of alveolar macrophages in bronchioalveolar lavage fluids from active tuberculosis patients identified by single-cell RNA sequencing显示文摘Tuberculosis(TB),is an infectious disease caused by Mycobacterium tuberculosis(M.tuberculosis),and presents with high morbidity and mortality.Alveolar macrophages play an important role in TB pathogenesis although there is heterogeneity and functional plasticity.This study aimed to show the characteristics of alveolar macrophages from bronchioalveolar lavage fluid(BALF)in active TB patients.Single-cell RNA sequencing(scRNA-seq)was performed on BALF cells from three patients with active TB and additional scRNA-seq data from three healthy adults were established as controls.Transcriptional profiles were analyzed and compared by differential gene expression and functional enrichment analysis.We applied pseudo-temporal trajectory analysis to investigate correlations and heterogeneity within alveolar macrophage subclusters.Alveolar macrophages from active TB patients at the single-cell resolution are described.We found that TB patients have higher cellular percentages in five macrophage subclusters.Alveolar macrophage subclusters with increased percentages were involved in inflammatory signaling pathways as well as the basic macrophage functions.The TB-increased alveolar macrophage subclusters might be derived from M1-like polarization state,before switching to an M2-like polarization state with the development of M.tuberculosis infection.Cell-cell communications of alveolar macrophages also increased and enhanced in active TB patients.Overall,our study demonstrated the characteristics of alveolar macrophages from BALF in active TB patients by using scRNA-seq. | Qianqian Chen Chunmei Hu Wei Lu Tianxing Hang Yan Shao Cheng Chen Yanli Wang Nan Li Linling Jin Wei Wu Hong Wang Xiaoning Zeng Weiping Xie | 2022 | The Journal of Biomedical Research2022,36,3: | 1 |

| 17 | Evaluation of antimicrobial and anticancer activities of three peptides identified from the skin secretion of Hylarana latouchii显示文摘The skins of frogs of the family Ranidae are particularly rich sources of biologically active peptides, among which antimicrobial peptides (AMPs) constitute the major portion. Some of these have attracted the interest of researchers because they possess both antimicrobial and anticancer activities. In this study, with ‘shotgun’ cloning & MS/MS fragmentation, three AMPs, homologues of family brevinin-1 (brevinin-1HL), & temporin (temporin-HLa and temporin-HLb), were discovered from the skin secretion of the broad-folded frog, Hylarana latouchii. They exhibited various degrees of antimicrobial & antibiofilm activities against test microorganisms & hemolysis on horse erythrocytes. It was found that they could induce bacteria death through disrupting cell membranes & binding to bacterial DNA. In addition, they also showed different potencies towards human cancer cell lines. The secondary structure & physicochemical properties of each peptide were investigated to preliminarily reveal their structure–activity relationships. Circular dichroism spectrometry showed that they all adopted a canonical α-helical conformation in membrane-mimetic solvents. Notably, the prepropeptide of brevinin-1HL from H. latouchii was highly identical to that of brevinin-1GHd from Hylarana guentheri, indicating a close relationship between these two species. | 18 | A biosensing system employing nanowell microelectrode arrays to record the intracellular potential of a single cardiomyocyte显示文摘Electrophysiological recording is a widely used method to investigate cardiovascular pathology,pharmacology and developmental biology.Microelectrode arrays record the electrical potential of cells in a minimally invasive and highthroughput way.However,commonly used microelectrode arays primarily employ planar microelectrodes and cannot work in applications that require a recording of the intracellular action potential of a single cell.In this study,we proposed a novel measuring method that is able to record the intracellular action potential of a single cardiomyocyte by using a nanowell patterned microelectrode array(NWMEA).The NWMEA consists of five nanoscale wells at the center of each circular planar microelectrode.Biphasic pulse electroporation was applied to the NWMEA to penetrate the cardiomyocyte membrane,and the intracellular action potential was continuously recorded.The intracellular potential recording of cardiomyocytes by the NWMEA measured a potential signal with a higher quality(213.76±25.85%),reduced noise root-mean-square(~33%),and higher signal-to-noise ratio(254.36±12.61%)when compared to those of the extracellular recording.Compared to previously reported nanopillar microelectrodes,the NWMEA could ensure single cell electroporation and acquire high-quality action potential of cardiomyocytes with reduced fabrication processes.This NWMEA-based biosensing system is a promising tool to record the intracellular action potential of a single cell to broaden the usage of microelectrode arrays in electrophysiological investigation. | Yuting Xiang Haitao Liu Wenjian Yang Zhongyuan xu Yue Wu Zhajian Tang Zhjing Zhu Zhiyong Zeng Depeng Wang Tianxing Wang Ning Hu Diming Zhang | 2022 | Microsystems & Nanoengineering2022,8,3: | 0 |
| 19 | Numerical Investigation of the Influence of Fuel Concentration Gradient on Propane/Oxygen Detonation Propagation显示文摘In this paper,one-and two-dimensional numerical simulations are carried out to study the effects of fuel concentration gradients(such as steep,intermediate and shallow)on the detonation wave behavior.The equivalent ratio range of detonation propagation,the quenching mechanism and the change of cell size are discussed in detail.The simulation results show,as the fuel concentration gradient increases,the detonation wavefront decays faster and decouples into a leading shock and a following flame at equivalence ratios of 0.68,0.64 and 0.62,respectively.Moreover,there are two modes of the quenching mechanism.One occurs in the steep gradient that the detonation wave fails rapidly.The O_(2)in front of the detonation wave passes through the detonation wave and forms some unburned O_(2)pockets.The unburned pockets are affected by the marginal walls and reduce the heat release.The other occurs in the intermediate and shallow gradients that more triple points will survive in the flow field,which leads to a difference in the propagation speed of the detonation wavefront.This makes the detonation wavefront bent and deformed.The unburned O_(2)pockets are affected by the strong instability near the triple points and show different distribution characteristics compared with the steep gradient,which may be helpful to the detonation propagation.In addition,as the fuel concentration gradient increases,the triple points moving toward the wall gradually disappear while the triple points that move toward the center can continue to survive,which leads to the gradual increase in cell size and irregularity of the cell structure. | SHAO Xiaofeng WU Tianxing MENG Qingyang ZHAO Ningbo QI Lei ZHENG Hongtao | 2023 | Journal of Thermal Science2023,32,6: | 0 |
