1Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders.Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao 2020Cell Research2020,30,9:7
2Single-cell RNA sequencing profiling of the effects of aging on alveolar stem cells显示文摘The aging of alveolar stem cells has been linked to many chronic lung diseases, including pulmonary fibrosis. However, the effects of aging on alveolar stem cells during homeostasis and post-injury alveolar repair have not been well characterized. Here we conducted a single-cell RNA sequencing(scRNA-seq) analysis of alveolar stem cells of 3-month-old and 12-month-old mice to characterize the aging effect on alveolar stem cells. Our results have shown that the transcriptomes of alveolar stem cells of 3-month-old and 12-month-old mice are not significantly different under the steady condition. However, after a bleomycin-induced lung injury, the alveolar stem cells of 12-month-old mice show enhanced inflammatory responses and decreased lipid metabolism. 4Sodium Danshensu promotes the healing of stage 2 pressure injury wounds in ischemia/reperfusion injury rat models:possible regulation of apoptosis and inflammatory response显示文摘OBJECTIVE:To investigate the protective effect and possible mechanism of sodium Danshensu(SDSS)against pressure injury caused by ischemia/reperfusion(I/R)injury.METHODS:Sprague-Dawley rats were randomly divided into five groups of eight rats each:control group,model group,10 mg/kg SDSS-treated group,20 mg/kg SDSS-treated group,and 40 mg/kg SDSS-treated group.We used two round ferrite magnetic plates of 15 mm diameter and 3 mm thickness to establish stage 2 pressure injury model rats.Each rat was subjected to five cycles of ischemia and reperfusion to induce pressure injury.One cycle consisted of 2 h of ischemia and 0.5 h of reperfusion,which meant that each cycle included2 h of pressure and 0.5 h of pressure relief.The outline of the wound was delineated by butter paper and marker pen,and histopathological changes were observed by hematoxylin and eosin staining.In addition,the number of apoptotic cells and the activity of caspase-3 were assessed by terminal deoxynucleotidyl transferase-mediated nick end labeling and caspase-3 assay kits,respectively.The expression of apoptosis-regulatory proteins and inflammatory mediators was investigated by enzyme-linked immunosorbent assay.RESULTS:Results showed that treatment with SDSS for 7 d after establishing the pressure injury model remarkably improved the healing rate of the wound.SDSS also inhibited the levels of tumor necrosis factor-α,myeloperoxidase,and intercellular cell adhesion molecule-1;decreased the number of apoptotic cells;increased the ratio of B-cell lymphoma-2(Bcl-2)/Bcl-2-associated X(Bax);and regulated the expression and activity of caspase-3.CONCLUSION:Our results suggest that SDSS exhibits a treatment efficacy for pressure injury caused by I/R injury possibly by inhibiting apoptosis and inflammatory response.ZHANG Qiongzhi FU Tingting DAI Jianing ZHOU Zhinan SHEN Cuizhen 2021Journal of Traditional Chinese Medicine2021,41,4:5
5Estimating antiwear properties of esters as potential lubricantbased oils using QSTR models with CoMFA and CoMSIA显示文摘Comparative molecular field analysis and comparative molecular similarity indices analysis were employed to analyze the antiwear properties of a series of 57 esters as potential lubricant-based oils. Predictive 3D-quantitative structure tribo-ability relationship models were established using the SYBYL multifit molecular alignment rule with a training set and a test set. 6Ecotoxicological effects of waterborne PFOS exposure on swimming performance and energy expenditure in juvenile goldfish (Carassius auratus)显示文摘The potential risks of perfluorooctane sulfonate (PFOS) are of increasing ecological concern. Swimming performance is linked to the fitness and health of fish. However, the impacts of PFOS on swimming performance remain largely unknown. We investigated the ecotoxicological effects of acute exposure to PFOS on the swimming performance and energy expenditure of juvenile goldfish (Carassius auratus). The fish were exposed to a range of PFOS concentrations (0, 0.5, 2, 8 and 32 mg/L) for 48 hr. The spontaneous swimming activity, fast-start swimming performance, critical swimming speed (Ucrit) and active metabolic rate (AMR) of the goldfish were examined after exposure to PFOS. PFOS exposure resulted in remarkable effects on spontaneous activity. Motion distance was reduced, and the proportion of motionless time increased with increasing concentrations of PFOS. However, no significant alterations in the fast-start performance-related kinematic parameters, such as latency time, maximum linear velocity, maximum linear acceleration or escape distance during the first 120 msec after stimulus, were observed after PFOS exposure. Unexpectedly, although PFOS exposure had marked influences on the swimming oxygen consumption rates and AMR of goldfish, the U crit of the goldfish was not significantly affected by PFOS. This may result in a noteworthy increase in the energetic cost of transport. 7Nanostructured N-doped carbon materials derived from expandable biomass with superior electrocatalytic performance towards V^(2+)/V^(3+) redox reaction for vanadium redox flow battery显示文摘Vanadium redox flow battery(VRFB)is one of the most promising large-scale energy storage systems,which ranges from kilowatt to megawatt.Nevertheless,poor electrochemical activity of electrode for two redox couples still restricts the extensive applications of VRFB.Compared with V^(2+)/V^(3+)redox reaction,V^(2+)/V^(3+)reaction plays a more significant role in voltage loss of VRFB owing to slow heterogeneous electron transfer rate.Herein,N-doped carbon materials derived from scaphium scaphigerum have been developed as negative electrocatalyst by hydrothermal carbonization and high-temperature nitridation treatments.The undoped carbon material hardly has electrocatalytic ability for V^(2+)/V^(3+)reaction.Based on this,N-doped carbon materials with urea as nitrogen source exhibit excellent electrocatalytic properties.And the material nitrided at 850°C(SSC/N-850)exhibits the best performance among those from700 to 1000℃.SSC/N-850 can accelerate the electrode process including V^(2+)/V^(3+)reaction and mass transfer of active ions due to the large reaction place,more active sites,and good hydrophilicity.The effect of catalyst on comprehensive performance of cell was evaluated.SSC/N-850 can improve the charge-discharge performance greatly.Utilization of SSC/N-850 can lessen the electrochemical polarization of cell,further resulting in increased discharge capacity and energy efficiency.Discharge capacity and energy efficiency increase by 81.5%and 9.8%by using SSC/N-850 as negative catalyst at 150 m A cm^(-2),respectively.Our study reveals that the developed biomass-derived carbon materials are the low-cost and efficient negative electrocatalyst for VRFB system.Yingqiao Jiang Mengchen Du Gang Cheng Peng Gao Tingting Dong Jing Zhou Xiaojian Feng Zhangxing He Yuehua Li Lei Dai Wei Meng Ling Wang 2021Journal of Energy Chemistry2021,30,8:3
8Influence of N doping and the functional groups of graphene on a RGO/TiO2 composite photocatalyst显示文摘Reduced graphene oxide(RGO)assisted TiO2 composite photocatalysts have attracted increased attention because of their high photocatalytic performance under both UV-and visible-light irradiation.However,the corresponding properties under the long wavelength irradiation is far from expected.In this study,a series of RGO samples with well-designed N impurity and optimized surface functional groups are prepared and employed during hybridization with TiO2(named N-RGO/TiO2).The coupling between the doped N atoms and surface functional groups is analyzed based on the resulting photocatalytic performances,and the specific mechanism is revealed by the EPR,IR and lifetime of the photoinduced electrons.After comparing the corresponding performances of the RGO/TiO2 and RGO/N-TiO2 specimens,the results show that the surface functional groups of the RGO serve as a precondition to achieveπ-d coupling between the graphene basal plane and TiO2 nanoparticles,while N atoms in the RGO enhance the resulting photoactivity under a long wavelength range(>510 nm).The findings provide a potential approach to promote the photocatalytic properties of graphene-based composite photocatalysts with the low-energy incident photons.TANG Bo HE YanFeng ZHANG ZhenYu WANG ZhengWei JI Li MA TingTing LI Sen DAI YuanZhe ZHANG GuoLiang 2020Science China(Technological Sciences)2020,63,6:3
9Fiber-Optic Raman Spectrum Sensor for Fast Diagnosis of Esophageal Cancer显示文摘A fiber-optic Raman spectrum sensor system is used for the fast diagnosis of esophageal cancer during clinical endoscopic examination.The system contains a 785nm exciting laser,a Raman fiber-optic probe with 7 large core fibers and a focus lens,and a highly sensitive spectrum meter.The Raman spectrum of the tissue could be obtained within 1 second by using such a system. 10Item-level Forecasting for E-commerce Demand with High-dimensional Data Using a Two-stage Feature Selection Algorithm显示文摘With the rapid development of information technology and fast growth of Internet users,e-commerce nowadays is facing complex business environment and accumulating large-volume and highdimensional data.This brings two challenges for demand forecasting.First,e-merchants need to find appropriate approaches to leverage the large amount of data and extract forecast features to capture various factors affecting the demand.Second,they need to efficiently identify the most important features to improve the forecast accuracy and better understand the key drivers for demand changes.To solve these challenges,this study conducts a multi-dimensional feature engineering by constructing five feature categories including historical demand,price,page view,reviews,and competition for e-commerce demand forecasting on item-level.We then propose a two-stage random forest-based feature selection algorithm to effectively identify the important features from the high-dimensional feature set and avoid overfittlng.We test our proposed algorithm with a large-scale dataset from the largest e-commerce platform in China.The numerical results from 21,111 items and 109 million sales observations show that our proposed random forest-based forecasting framework with a two-stage feature selection algorithm delivers 11.58%,5.81%and 3.68%forecast accuracy improvement,compared with the Autoregressive Integrated Moving Average(ARIMA),Random Forecast,and Random Forecast with one-stage feature selection approach,respectively,which are widely used in literature and industry.This study provides a useful tool for the practitioners to forecast demands and sheds lights on the B2C e-commerce operations management.Hongyan Dai Qin Xiao Nina Yan Xun Xu Tingting Tong 2022Journal of Systems Science and Systems Engineering2022,31,2:1

14Determination of trophic levels of marine fish in the Yellow Sea and northern East China Sea using nitrogen stable isotope (δ^(15)N) analysis of otoliths显示文摘Fish otolithδ^(15) N(δ^(15) N_(oto))is a demonstrated source of information of dietary history for marine fi sh as it is available iN_(oto)lith archives and sedimentary deposits unlike white muscle tissue(WMT).WMT and stomach content data are insufficient for trophic level(TL)data of past fi shes which is important for the changes of marine fi shery resources over long time scales.To determine the correlation betweenδ^(15) N_(oto) and fi sh WMTδ^(15) N(δ^(15) N_(wmt))and the feasibility of usingδ^(15) N_(oto) in characterizing the TLs of marine fi shes,we conducted nitrogen stable isotope analysis(SIA)in the otolith and WMT of 36 marine fi sh species sampled from the Yellow Sea and northern East China Sea in 2011-2014.Bothδ^(15) N_(oto) andδ^(15) N_(wmt) were analyzed using an elemental analyzer coupled with an isotope ratio mass spectrometer(EA-IRMS).Multiple otoliths were combined to make each otolith measurement and were analyzed as-is without a carbonate dissolution pre-processing step.δ^(15) N_(oto) andδ^(15) N_(wmt) comparisons for species in the Yellow Sea and northern East China Sea are currently lacking and would be helpful for both regional studies and for increasing the number of species for whichδ^(15) N_(oto) andδ^(15) N_(wmt) have been compared.Additionally,to determine the relative accuracy of trophic level calculated usingδ^(15) N_(oto),we compared TL calculated fromδ^(15) N_(oto) to traditional trophic level metrics calculated usingδ^(15) N_(wmt).The results showed a positive and highly signifi cant correlation(R=0.780,P<0.001)betweenδ^(15) N_(oto) andδ^(15) N_(wmt).Trophic level estimation using WMT(TL wmt)and otolith(TL oto)showed congruence in our study,which is not entirely surprising given thatδ^(15) N_(oto) was regressed againstδ^(15) N_(wmt) and the resulting regression coefficient was used to convertδ^(15) N_(oto) toδ^(15) N_(wmt) prior to calculating TL oto.This conversion was required in order to be consistent with previousδ^(15) N_(wmt)-based calculations of TL for comparison.TL oto calculations resulted in TL values that were largely within 5%-10%of TL values calculated withδ^(15) N_(wmt).Our fi ndings show thatδ^(15) N_(oto) is a feasible technique for characterizing the TLs of marine fi sh and can also assist in food web and marine ecosystem studies.Huaiyu BAI Yukun WANG Tingting ZHANG Fangqun DAI Lingfeng HUANG Yao SUN 2022Journal of Oceanology and Limnology2022,40,2:1

16Postoperative infection caused by Acinetobacter baumannii misdiagnosed as a free-living amoeba species in a humeral head hemiarthroplasty patient:a case report显示文摘Background:Acinetobacter baumannii is ubiquitous,facultative intracellular,and opportunistic bacterial pathogen.Its unique abilities allow it to survive in a diverse range of environments,including health care settings,leading to nosocomial infections.And its exceptional ability to develop resistance to multiple antibiotics leaves few drug options for treatment.It has been recognized as a leading cause of nosocomial pneumonia and bacteremia over the world.Case presentation:In this case,a 73-year-old woman presented with a Neer Group VI proximal humeral fracture.Six hours after a successfully performed hemiarthroplasty,she developed continuous fever.Clinical examination revealed that the vitals were regular.Laboratory and radiographic examinations revealed only elevated procalcitonin levels.Blood culture revealed no bacterial or fungal growth.Cooling treatment and empirical broad-spectrum antibiotic therapy showed no apparent effect.Conclusions:We report a postoperative infection caused by Acinetobacter baumannii.The infectious pathogen was identified via molecular DNA sequencing and was initially misidentified as a free-living amoeba species upon microscopic examinations.The patient was mistreated with antiamebic combination therapy.Her symptoms persisted for over 4 months and were eventually followed by her death.Jiaxin Tang Huaimin Zhu Li Cai Tingting Tang Jian Tang Yuehua Sun Ming Liu Kerong Dai Zhiguang Qiao Chao Yu 2018Infectious Diseases of Poverty2018,7,1:1
