| 4 | Manipulation of SlMXl for enhanced carotenoids accumulation and drought resistance in tomato显示文摘Plants are the ultimate source of nutrients in the human diet. To ensure adequate availability of high quality food for an increasing world population, traits including improved tolerance of stresses and nutrient levels need to be selected in crops, both individually and in combination. Here we report the identification of Sl MX1 encoding a MIXTA-like MYB transcription factor in tomato that simultaneously modulates drought resistance and metabolic processes through regulating key structural and regulatory genes of the corresponding pathways. Over-expression of Sl MX1 results in substantially increased drought tolerance and improved fruit quality, while knocking down Sl MX1 resulted in the opposite phenotypes. | 5 | Comprehensive profiling and natural variation of flavonoids in rice显示文摘Flavonoids constitute a major group of plant phenolic compounds.While extensively studied in Arabidopsis,profiling and naturally occurring variation of these compounds in rice(Oryza sativa),the monocot model plant,are less reported.Using a collection of rice germplasm,comprehensive profiling and natural variation of flavonoids were presented in this report.Application of a widely targeted metabolomics method facilitated the simultaneous identification and quantification of more than 90 flavonoids using liquid chromatography tandem mass spectrometry(LC-MS/MS).Comparing flavonoid contents in various tissues during different developmental stages revealed tissue-specific accumulation of most flavonoids.Further investigation indicated that flavone mono-C-glycosides,malonylated flavonoid O-hexosides,and some flavonoid O-glycosides accumulated at significantly higher levels in indica than in japonica,while the opposite was observed for aromatic acylated flavone C-hexosyl-O-hexosides.In contrast to the highly differential accumulation between the two subspecies,relatively small variations within subspecies were detected for most flavonoids.Besides,an association analysis between flavonoid accumulation and its biosynthetic gene sequence polymorphisms disclosed that natural variation of flavonoids was probably caused by sequence polymorphisms in the coding region of flavonoid biosynthetic genes.Our work paves the way for future dissection of biosynthesis and regulation of flavonoid pathway in rice. | Xuekui Dong Wei Chen Wensheng Wang Hongyan Zhang Xianqing Liu Jie Luo | 2014 | Journal of Integrative Plant Biology2014,56,9: | 8 |

| 7 | A UV-B-responsive glycosyltransferase,OsUGT706C2,modulates flavonoid metabolism in rice显示文摘Although natural variations in rice flavonoids exist, and biochemical characterization of a few flavonoid glycosyltransferases has been reported, few studies focused on natural variations in tricin-lignan-glycosides and their underlying genetic basis. In this study, we carried out metabolic profiling of tricin-lignan-glycosides and identified a major quantitative gene annotated as a UDPdependent glycosyltransferase OsUGT706C2 by metabolite-based genome-wide association analysis. The putative flavonoid glycosyltransferase OsUGT706C2 was characterized as a flavonoid 7-O-glycosyltransferas in vitro and in vivo. Although the in vitro enzyme activity of OsUGT706C2 was similar to that of OsUGT706D1, the expression pattern and induced expression profile of OsUGT706C2 were very different from those of OsUGT706D1. Besides, OsUGT706C2 was specifically induced by UV-B. Constitutive expression of OsUGT706C2 in rice may modulate phenylpropanoid metabolism at both the transcript and metabolite levels. Furthermore, overexpressing OsUGT706C2 can enhance UV-B tolerance by promoting ROS scavenging in rice. | 11 | Lipid Oxidation and Fatty Acid Composition in Salt-Dried Yellow Croaker(Pseudosciaena polyactis) During Processing显示文摘Lipid oxidation in salt-dried yellow croaker(Pseudosciaena polyactis) was evaluated during processing with commonly used analytical indices, such as the peroxide value(POV), the thiobarbituric acid reactive substances(TBARS) value, and oxidative-relative lipoxygenase(LOX) activity. Additionally, fatty acids were analyzed using gas chromatography-mass spectrometry. Both POV and TBARS increased significantly(P < 0.05) at the rinsing stage. POV reached its peak value of 3.63 meq O_2 per kg sample at the drying stage, whereas TBARS constantly increased from 0.05 to 0.20 mg MDA per kg sample. Processing of salt-dried yellow croaker had an extremely significant(P < 0.01) effect on LOX activity. Twenty-six fatty acids were identified. Combined eicosapentaenoic acid(EPA; C20:5n3) and docosahexaenoic acid(DHA; C22:6n3) content varied between(19.20 ±0.37) mg g^(-1) and(23.45 ± 1.05) mg g^(-1). The polyunsaturated fatty acid/saturated fatty acid(PUFA/SFA) ratio in yellow croaker was 0.73–1.10, and the n-6/n-3 PUFA ratio was approximately 0.13–0.20. The contents of most fatty acids varied significantly(P <0.05) during the different processing stages, and these differences were caused by lipid oxidation. C18:0, C16:1n7, C19:0, and C22:6n3 showed clear changes in principle component one of a principle components analysis. While the tectonic characteristics is different obviously between the east and the west because of the special tectonic position and tectonic evolution process. In terms of submarine geomorphology,the eastern shelf-slope structure in Pearl River Mouth Basin is characterized by having wide sub-basins and narrow intervening highs, whereas the western(Qiongdongnan Basin) structure is characterized by narrow subbasins and wide uplift. As to the structural features, the deep-water sags in the east are all structurally halfgrabens, controlled by a series of south-dipping normal faults. While the west sags are mainly characterised by graben structures with faulting in both the south and north. With regards to the tectonic evolution, the east began neotectonic activity when the post-rifting stage had completed at the end of the Middle Miocene. In the Baiyun Sag, tectonic activity became strong and was characterised by rapid subsidence and obvious faulting. | 14 | A targeted sequencing approach to find novel pathogenic genes associated with sporadic aortic dissection显示文摘Aortic dissection(AD) is a heterogeneous genetic disease of the aorta with high mortality and poor prognosis. However, only few genetic causes of AD have been explored till date. After conducting a broad literature review focused on identifying potential pathogenic pathways, we designed a panel containing 152 AD-associated genes to conduct massively parallel targeted nextgeneration sequencing of 702 sporadic aortic dissection patients and 163 matched healthy controls. After validation by Sanger sequencing, we identified 21 definitely pathogenic and 635 likely pathogenic variants in 61.25%(430/702) of patients. In these patients, 34.88%(150/430) harbored more than one variant that was either definitely or likely to be pathogenic. Among the candidate genes, we identified 546 likely pathogenic variants in 47.72%(335/702) of patients. Importantly, we identified 94 lossof-function(LOF) variants in 45 genes in AD patients, but only five LOF variants in the controls(P=1.34×10^(-4)). With a burden test, we highlighted RNF213 as an important new gene for AD pathogenesis. We also performed transcriptome sequencing of human aorta tissues to evaluate the expression levels of these newly identified genes. Our study has compiled a comprehensive genetic map of sporadic AD in the Han Chinese population. | 16 | Variants of genes encoding collagens and matrix metalloproteinase system increased the risk of aortic dissection显示文摘Aortic dissection(AD) is a devastating,heterogeneous condition of aorta.The homeostasis between collagens and matrix metalloproteases(MMPs)/tissue inhibitors of MMPs(TIMPs) system in the extracellular matrix plays an important role for structure and functions of aorta.However,our knowledge on association between variants of genes in this system and pathogenesis of AD is very limited.We analyzed all yet known coding human genes of collagens(45 genes),MMPs/TIMPs(27genes) in 702 sporadic AD patients and in 163 matched healthy controls,by using massively targeted next-generation and Sanger sequencing.To define the pathogenesis of potential disease-causing candidate genes,we performed transcriptome sequencing and pedigree co-segregation analysis in some genes and generated Col5a2 knockout rats.We identified 257 pathogenic or likely pathogenic variants which involved 88.89%(64/72) genes in collagens-MMPs/TIMPs system and accounted for 31.05%(218/702) sporadic AD patients.In them,84.86%patients(185/218) carried one variant,12.84%two variants and 2.30%more than two variants.Importantly,we identified 52 novel probably pathogenic loss-of-function(LOF) variants(20 nonsense,16 frameshift,14 splice sites,one stop-loss,one initiation codon) in 11.06%(50/452) AD patients,which were absent in 163controls(P=2.5×10^(-5)).Transcriptome sequencing revealed that identified variants induced dyshomeostasis in expression of collagens-TIMPs/MMPs systems.The Col5α2^(-/-) rats manifested growth retardation and aortic dysplasia.Our study provides a first comprehensive map of genetic alterations in collagens-MMPs/TIMPs system in sporadic AD patients and suggests that variants of these genes contribute largely to AD pathogenesis. | Zongzhe Li Chengming Zhou Lun Tan Peng Chen Yanyan Cao Chenze Li Xianqing Li Jiangtao Yan Hesong Zeng Dao-Wu Wang Dao-Wen Wang | 2017 | Science China(Life Sciences)2017,60,1: | 4 |

| 18 | Raising the production of phloretin by alleviation of by-product of chalcone synthase in the engineered yeast显示文摘Phloretin is an important skin-lightening and depigmenting agent from the peel of apples. Although de novo production of phloretin has been realized in microbes using the natural pathway from plants, the efficiency of phloretin production is still not enough for industrial application. Here, we established an artificial pathway in the yeast to produce phloretin via assembling two genes of p-coumaroyl-CoA ligase(4CL) and chalcone synthase(CHS). CHS is a key enzyme which conventionally condenses a CoA-tethered starter with three molecules of malonyl-CoA to form the backbone of flavonoids. However, there was 33% of byproduct generated via CHS by condensing two molecules of malonyl-CoA during the fermentation process. Hence, we introduced a more efficient CHS and improved the supply of malonyl-CoA through two pathways;the by-product ratio was decreased from 33% to 17% and the production of phloretin was improved from 48 to 83.2 mg L^(-1). Finally, a fed-batch fermentation process was optimized and the production of phloretin reached 619.5 mg L^(-1), which was 14-fold higher than that of the previous studies. | 19 | Geochemistry, Geochronology and Lu-Hf Isotopes of Peraluminous Granitic Porphyry from the Walegen Au Deposit, West Qinling Terrane显示文摘Walegen Au deposit is closely correlated with granitic intrusions of Triassic age, which are composed of granite and quartz porphyries. Both granite porphyry and quartz porphyry consist of quartz, feldspar and muscovite as primary minerals. Weakly peraluminous granite porphyry(A/CNK=1.10–1.15) is enriched in LREE, depleted in HREE with Nb-Ta-Ti anomalies, and displays subduction-related geochemistry. Quartz porphyry is strongly peraluminous(A/CNK=1.64–2.81) with highly evolved components, characterized by lower TiO_2, REE contents, Mg~#, K/Rb, Nb/Ta, Zr/Hf ratios and higher Rb/Sr ratios than the granite porphyry. REE patterns of quartz porphyry exhibit lanthanide tetrad effect, resulting from mineral fractionation or participation of fluids with enriched F and Cl. LAICP-MS zircon U-Pb dating indicates quartz porphyry formed at 233±3 Ma. The ages of relict zircons from Triassic magmatic rocks match well with the detrital zircons from regional area. In addition, ε_(Hf)(t) values of Triassic magmatic zircons from the granite and quartz porphyries are -14.2 to -9.1(with an exception of +4.1) and -10.8 to -8.6 respectively, indicating a crustal-dominant source. Regionally, numerous Middle Triassic granitoids were previously reported to be formed under the consumption of Paleotethyan Ocean.
