| 1 | Effect of Jiaweiwumei decoction on regulatory T cells and interleukin-10 in a rat model of ulcerative colitis显示文摘OBJECTIVE:To investigate the effect of a decoction made from the Traditional Chinese Medicine Wumei pill,on regulatory T cells and interleukin-10(IL-10) in a rat model of ulcerative colitis induced by2,4,6-trinitrobenzene sulfonic acid(TNBS).METHODS:Rat ulcerative colitis was induced with TNBS.All modeled rats were randomly divided into six groups:normal control group;model group;sulfasalazine suppositories treatment group;and high,moderate,and low dosage of Jiaweiwumei decoction groups(12 rats each).Colon injury index was evaluated after 14 days.After peripheral blood lymphocyte separation,CD4 + T cells and CD4+/CD25+ T cell percentage was detected by flow cytometry.The content of IL-10 in serum and intestinal mucosa tissue was detected by sandwich enzyme-linked immunosorbent assay.RESULTS:Colon injury indices in the decoction groups were effectively reduced,compared with the model group(P < 0.05).Compared with that of the control group,the CD4+/CD25+ to CD4+ T lymphocyte ratio of the model group was significantly lower,while the decoction treatment improved the CD4 +/CD25 + to CD4 + T lymphocyte ratio(P <0.05).The serum and mucosal IL-10 content of the model group was significantly lower(P < 0.05) than that in the control group,while the decoction group had significantly higher serum and intestinal mucosal IL-10 content than that in the model group(P < 0.05).The regulatory T cell content was negatively correlated with the colonic injury index(r = 0.68,P < 0.05),and positively correlated with the content of serum IL-10(r = 0.87,P < 0.05) and intestinal mucosal IL-10(r= 0.79,P < 0.05).CONCLUSION:Jiaweiwumei decoction had significant effects on regulatory T cells and IL-10 in rats with TNBS-induced ulcerative colitis. | Tao Mei Wang Xuyan Wang Aiqin Gao Ninghan Pan Jie Liu Xiaoquan Wang Xiaoye | 2015 | Journal of Traditional Chinese Medicine2015,35,3: | 14 |
| 2 | Erosion effect on the productivity of black soil in Northeast China显示文摘Continuous soil erosion has caused serious land degradation in the black soil area of Northeast China. The primary objective of this study was to determine the effects of accelerated erosion on soil productivity, as measured by soybean (Glycine max L. Merr.) yields. Eight erosion levels, 0, 10, 20, 30, 40, 50, 60, and 70 cm, were simulated by imitating the integrated process of natural erosion and tillage activity. Each erosion level had two sub-treatments: conventional fertilization and no fertilization. Soil erosion was found to affect survival probability and to cause remarkable reductions in the Leaf Area Index (LAI), plant height, pod number, biomass, and yield. Soybean yield was exponentially decreased with the increase of soil erosion depth. Compared to erosion depth of 0 cm, erosion levels of 10, 20, 30, 40, 50, 60, and 70 cm experienced reductions in soybean yield by 28.8%, 37.8%, 43.5%, 52.6%, 53.1%, 52.9%, and 64.1% respectively when fertilized whereas the reductions at those levels were 32.6%, 42.2%, 53.0%, 54.0%, 65.8%, 69.7%, and 72.6%, respectively, when unfertilized. At the erosion depths of 10, 20, 30, 40, 50, 60, and 70 cm, the yield reductions per 10 cm of soil eroded when fertilized were 28.8%, 18.9%, 14.5%, 13.2%, 10.6%, 8.8%, and 9.2%, averaged 14.9%, but when unfertilized they were 32.6%, 21.1%, 17.7%, 13.5%, 13.2%, 11.6%, and 10.4%, averaged 17.1%. The results also showed that chemical fertili zers could enhance the yields of eroded soil, but could not recover the yields to the pre-erosion level. Additionally, the results indicated that the primary reason for the decrease in soybean yield with increasing erosion depth was the loss of soil organic matter, soil N and P. | 3 | The zinc transporter Slc39a5 controls glucose sensing and insulin secretion in pancreatic β-cells via Sirt1- and Pgc-1α-mediated regulation of Glut2显示文摘Zinc levels are high in pancreatic β-cells, and zinc is involved in the synthesis, processing and secretion of insulin in these cells. However, precisely how cellular zinc homeostasis is regulated in pancreatic β-cells is poorly understood. By screening the expression of 14 Slc39a metal importer family member genes, we found that the zinc transporter Slc39a5 is significantly downregulated in pancreatic β-cells in diabetic db/db mice, obese ob/ob mice and high-fat diet-fed mice. Moreover,β-cell-specific Slc39a5 knockout mice have impaired insulin secretion. In addition, Slc39a5-deficient pancreatic islets have reduced glucose tolerance accompanied by reduced expression of Pgc-1α and its downstream target gene Glut2. The down-regulation of Glut2 in Slc39a5-deficient islets was rescued using agonists of Sirt1, Pgc-1α and Ppar-γ. At the mechanistic level, we found that Slc39a5-mediated zinc influx induces Glut2 expression via Sirt1-mediated Pgc-1α activation. | 7 | E3 ligase FBXW7 aggravates TMPD-induced systemic lupus erythematosus by promoting cell apoptosis显示文摘Systemic lupus erythematosus(SLE)is a systemic autoimmune disease,and the pathogenesis of SLE has not been fully elucidated.The E3 ubiquitin ligase FBXW7 has been well characterized in cancer as a tumor suppressor that can promote the ubiquitination and subsequent degradation of various oncoproteins;however,the potential role of FBXW7 in autoimmune diseases is unclear.In the present study,we identified that FBXW7 is a crucial exacerbating factor for SLE development and progression in a mouse model induced by 2,6,10,14-tetramethylpentadecane(TMPD).Myeloid cell-specific FBXW7-deficient(Lysm+FBXW7f/f)C57BL/6 mice showed decreased immune complex accumulation,glomerulonephritis,glomerular mesangial cell proliferation,and basemembrane thickness in the kidney.Lysm+FBXW7f/f mice produced fewer anti-Sm/RNP and anti-ANA autoantibodies and showed a decreased MHC II expression in B cells.In Lysm+FBXW7f/f mice,we observed that cell apoptosis was reduced and that fewer CD11b+Ly6Chi inflammatory monocytes were recruited to the peritoneal cavity.Consistently,diffuse pulmonary hemorrhage(DPH)was also decreased in Lysm+FBXW7f/f mice.Mechanistically,we clarified that FBXW7 promoted TMPD-induced cell apoptosis by catalyzing MCL1 degradation through K48-linked ubiquitination.Our work revealed that FBXW7 expression in myeloid cells played a crucial role in TMPD-induced SLE progression in mice,which may provide novel ideas and theoretical support for understanding the pathogenesis of SLE. | Zhenlu Chong Chunjing Bao Jia He Tianxiao Chen Lijia Zhong Gaopeng Li Huanle Li Lutong Fang Yinjing Song Guoxiang Fu Xuyan Yang Lihua Lai Yang Liu Qingqing Wang | 2018 | Cellular & Molecular Immunology2018,15,12: | 2 |

| 11 | Molecular cloning, expression and biochemical property analysis of AtKP1, a kinesin gene from Arabidopsis thaliana显示文摘Kinesins are common in a variety of eukaryotic cells with diverse functions. A cDNA encoding a member of the Kinesin-14B subfamily is obtained using 3′-RACE technology and named AtKP1 (for Arabidopsis kinesin protein 1). This cDNA has a maximum open reading frame of 3.3 kb encoding a polypeptide of 1087 aa. Protein domain analysis shows that AtKP1 contains the motor domain and the calponin homology domain in the central and amino-terminal regions, respectively. The carboxyl-terminal region with 202 aa residues is diverse from other known kinesins. Northern blot analysis shows that AtKP1 is widely expressed at a higher level in seedlings than in mature plants. 2808 bp of the AtKP1 promoter region is cloned and fused to GUS. GUS expression driven by the AtKP1 promoter region shows that AtKP1 is mainly expressed in vasculature of young organs and young leaf trichomes, indicating that AtKP1 may participate in the differentiation or development of Arabidopsis thaliana vascular bundles and trichomes. A truncated AtKP1 protein containing the putative motor domain is expressed in E. coli and affinity-purified.
