| 1 | Construction of a genetic map with SRAP markers and localization of the gene responsible for the first-flower-node trait in cucumber (Cucumis sativus L.)显示文摘With an F 2 population from the cross of two cucumber inbred lines, S06 and S52, sequence-related amplified polymorphism (SRAP) was used to construct a genetic linkage map in cucumber (Cucumis sativus L.). Sixty-four SRAP primer combinations generated 108 polymorphic bands in the F 2 population analysis. The average of polymorphic bands produced by one primer pair was 1.5, and the maximum was 5. Using Mapmaker 3.0, a linkage map was constructed, which consisted of 77 SRAP markers distributed in nine linkage groups (LOD≥3.0) and spanned 1114.2 cM with an average interval of 14.5 cM between markers. | 2 | Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. | 4 | Localization of genes for lateral branch and female sex expression and construction of a molecular linkage map in cucumber (Cucumis sativus L.) with RAPD markers显示文摘A cucumber (Cucumis sativus L.) molecular linkage map, including 79 random-amplified polymorphic DNAs (RAPD) and two genes , lb for lateral branch and f for female sex expression, is constructed from a cross between a line, S52, with weak lateral growing ability and staminate from Dabieshan Mountains area in China and another line, S06, with strong lateral growing ability and gynoecious from Europe. The map contains nine linkage groups and spans 1110.0 cM with an average distance of 13.7 cM between loci. The lb locus is located in a longer linkage group LG-2 and flanked by two markers, OP-Q5-1 and OP-M-2-2, at 9.3 cM and 15.9 cM, respectively. In the meantime, the RAPD loci, OP-Q5-2 and BC151, in a short linkage group were found to flank f at 13.7 cM and 13.4 cM, respectively. | 6 | Capacity of soil to protect organic carbon and biochemical characteristics of density fractions in Ziwulin Haplic Greyxems soil显示文摘Physical protection is one of the important ways to stabilize organic carbon in soils. In order to under- stand the role of soils as a carbon sink or source in global climatic change and carbon cycles and properly manage soils as a carbon sink, we ought to know how many organic car- bon (OC) in a given soil could be protected. By a density fractionation approach and ultrasonic technique, each soil sample was divided into three fractions: free light fraction (free-LF), occluded fraction (occluded-LF) and heavy frac- tion (HF). The obtained fractions were analyzed for total OC content, carbohydrate content and recalcitrant OC content. The results showed: (ⅰ) In the whole soil profile, dominance of OC consistently decreased in the following order: HF, free-LF, occluded-LF. This suggested that OC in soils were mostly protected. From 0—10 to 60—80 cm horizons, the OC in free-LF decreased from 25.27% to 3.72%, while OC in HF they were increased from 72.57% to 95.39%. The OC in oc- cluded-LF was between 2.16% and 0.89%. (ⅱ) Organic carbon recalcitrance in free-LF was similar to that in HF, and was even higher than that in HF below the surface hori- zon. This suggested that free-LF was not always the most fresh and non-decomposed fraction. | 7 | Ecophysiological Evidence for the Competition Strategy of Two Psammophytes Artemisia halodendron and A. frigida in Horqin Sandy Land, Nei Mongol显示文摘Gas exchange, water relations and leaf chemical characteristics were examined of twodominant psammophytes: Artemisia frigida Willd and A halodendron Turcz. ex Bess in Horqin sandy land, NeiMongol, China under different water regimes. The measurements were conducted by submitting the plantsto five different irrigation levels. A. fTigida was characterized by lower photosynthetic rate (Pn), lowertranspiration rate ( TR and lower shoot water potential (ψuw) relative to A. halodendron. Foliage of A frigdahad higher values of relative water deficit (RWD), bound water content (BWC), ratio of bound water contentto free water content (BWC/FWC) and integrated drought-resistant index (DI than that of A. halodendron.Water relations differed significantly between two species in response to soil water availability. ψw, BWCand BWC/FWC ratio of A halodendron exhibited large variation with gradual decrease of soil moisture.However, in terms of these parameters, A. fRIGIDA was characterized by higher capacity of water holding anddrought tolerance relative to A halodendron. Proline and total soluble sugar contents of A frigida and Ahalodendron tended to increase with decrease of soil moisture and the former had a larger increaseamplitude than the latter. This shows that A frigida has a higher osmotic regulation ability than A halodendron.Under the extreme drought conditions, ψw, RWD, BWC and BWC/FWC of two species were approximate,but soluble proteins degraded largely. A large amount of accumulation of organic matter, proline and totalsoluble sugars were observed in both A halodendron and A frigida. The increase in proline and total solublesugar contents and soluble protein degradation of A frigida far exceeded those of A. halodendron. Webelieve that the accumulated materials at this moment are mostly of nutrient substances available for therecovery of plants after the drought. This is one of the reasons why A halodendron plants died while Afrigida plants survived under extremely drought condition. | 8 | APPROACH TO IMPROVEMENT OF ROBOT TRAJECTORY ACCURACY BY DYNAMIC COMPENSATION显示文摘Some dynamic factors, such as inertial forces and friction, may affect the robot trajectory accuracy. But these effects are not taken into account in robot motion control schemes. Dynamic control methods, on the other hand, require the dynamic model of robot and the implementation of new type controller. A method to improve robot trajectory accuracy by dynamic compensation in robot motion control system is proposed. The dynamic compensation is applied as an additional velocity feedforward and a multilayer neural network is employed to realize the robot inverse dynamics. The complicated dynamic parameter identification problem becomes a learning process of neural network connecting weights under supervision. The finite Fourier series is used to activate each actuator of robot joints for obtaining training samples. Robot control system, consisting of an industrial computer and a digital motion controller, is implemented. The system is of open architecture with velocity feedforward function. The proposed method is not model-based and combines the advantages of close-loop position control and computed torque control. | 10 | Preparation of monoclonal antibody against human KIAA0100 protein and Northern blot analysis of human KIAA0100 gene显示文摘Monoclonal antibodies(MAbs) are important tools for the study of proteins′ function and structure. But there has been no report on the preparation of MAbs against human KIAA0100 protein up to date. Here, first, we generated the mouse MAb against human KIAA0100 protein using purified recombinant 6×Histidinc(6×His)-tagged human KIAA0100 protein segment(1557–2234) as an antigen; then, the m RNA expression of human KIAA0100 gene was detected in U937 cells using Northern blot analysis. The results showed that the mouse MAb against human KIAA0100 protein could sensitively recognize the human KIAA0100 protein using Western blot analysis and immunocytochemistry analysis. Besides, Western blot analysis revealed that human KIAA0100 gene possibly encoded two different protein products(254 k Da and < 250 k Da) in U937 cells. Moreover,Northern blot analysis confirmed that human KIAA0100 gene might produced two different m RNA products(6000–10000 bp and 5000–6000 bp) in U937 cells. | 11 | An effective rolling process of magnesium alloys for suppressing edge cracks: Width-limited rolling显示文摘To suppress the edge crack of the magnesium alloy sheet during the ordinary rolling process, a new rolling process named width-limited rolling was proposed in this paper. Width-limited rolling is a rolling method in which the width of the alloy sheet is limited by modifying the shape of the rollers, allowing a compressive stress field to form at the edge portion of the alloy sheet during rolling, resulting in the reduction of edge cracks. At present work, magnesium alloy sheets were separately subjected to ordinary rolling and width-limited rolling. The microstructure evolution and mechanical properties of the rolled sheets were investigated by EBSD, TEM, and XRD. The results exhibited that under the same rolling conditions, the sheet after ordinary rolling exhibited obvious edge cracks while no crack was found at the edge of the sheet after width-limited rolling. The edge crack suppressing effect was attributed to the reduction of the tensile stress along rolling direction during WLR, promoting the synchronous extension of the edge and center regions to suppress edge crack tendency. Microstructure observation showed that the compressive twins formed in the sheet after ordinary rolling usually exhibited as thin plates and cannot continue to fully develop due to the premature generation of the edge cracks. However, the compressive twins developed maturely in some of which double twins formed and various slip systems with different dislocation Burgers vectors occurred in the rolled sheet after WLR. More twin intersections and shear bands, providing more potential recrystallization nucleation sites, which are beneficial to weaken basal texture. | 19 | Effects of Annealing Temperature on the Properties of Copper Films Prepared by Magnetron Sputtering显示文摘Copper oxide thin films were prepared by a direct-current magnetron sputtering method followed by a thermal annealing treatment at 100-500 °C. The obtained films were characterized by X-ray diffraction, UV–vis absorption spectroscopy, scanning electron microscopy, Raman spectroscopy, and X-ray photoelectron spectroscopy. With the increase of the annealing temperature, it was found that the films transformed sequentially from amorphous to single-phase Cu(100 ℃), mixed-phase of Cu and Cu2O(150 ℃), single-phase Cu2O(200 ℃), then to mixed-phase of Cu2 O and Cu O(300 ℃), and finally to single-phase Cu O(400- 500 ℃). Further analyses indicated that the Cu/Cu2 O thin films and the Cu2 O thin films presented no further oxidation even on the surface in air atmosphere.
