CXCL5/CXCR2 axis in tumor microenvironment as potential diagnostic biomarker and therapeutic target显示文摘The components of the tumor microenvironment(TME)in solid tumors, especially chemokines, are currently attracting much attention from scientists.C-X-C motif chemokine ligand 5(CXCL5)is one of the important chemokines in TME.Over-expression of CXCL5 is closely related to the survival time, recurrence and metasta-sis of cancer patients. In TME,CXCL5 binds to its receptors, such as C-X-C motif chemokine receptor 2(CXCR2),to participate in the recruitment of immune cells and promote angiogenesis, tumor growth, and metastasis. The CXCL5/CXCR2 axis can act as a bridge between tumor cells and host cells in TME.Blocking the trans-mission of CXCL5/CXCR2 signals can increase the sensitivity and effectiveness of immunotherapy and slow down tumor progression.CXCL5 and CXCR2 are also regarded as biomarkers for predicting prognosis and molecular targets for customiz-ing the treatment. In this review,we summarized the current literature regarding the biological functions and clinical significance of CXCL5/CXCR2 axis in TME.The possibility to use CXCL5 and CXCR2 as potential prognostic biomarkers and thera-peutic targets in cancer is also discussed.Wen Zhang Huishan Wang Mingyang Sun Xueting Deng Xueru Wu Yilan Ma Mengjing Li Said Maisam Shuoa Qiang You Lin Miao 2020Cancer Communications2020,40,2:19
The wonders of BMP9:From mesenchymal stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism to regenerative medicine显示文摘Although bone morphogenetic proteins(BMPs)initially showed effective induction of ectopic bone growth in muscle,it has since been determined that these proteins,as members of the TGF-b superfamily, play a diverse and critical array of biological roles. These roles include regulating skeletal and bone formation, angiogenesis, and development and homeostasis of multiple organ systems.Disruptions of the members of the TGF-b/BMP superfamily result in severe skeletal and extra-skeletal irregularities, suggesting high therapeutic potential from understanding this family of BMP proteins.Although it was once one of the least characterized BMPs,BMP9 has revealed itself to have the highest osteogenic potential across numerous experiments both in vitro and in vivo, with recent studies suggesting that the exceptional potency of BMP9 may result from unique signaling pathways that differentiate it from other BMPs.The effectiveness of BMP9 in inducing bone formation was recently revealed in promising experiments that demonstrated efficacy in the repair of critical sized cranial defects as well as compatibility with bone-inducing bio-implants,revealing the great translational promise of BMP9.Furthermore,emerging evidence indicates that, besides its osteogenic activity,BMP9 exerts a broad range of biological functions, including stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism. This review aims to summarize our current understanding of BMP9 across biology and the body.Sami Mostafa Mikhail Pakvasa Elam Coalson Allen Zhu Alex Alverdy Hector Castillo Jiaming Fan Alex Li Yixiao Feng Di Wu Elliott Bishop Scott Du Mia Spezia Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Sherwin S·Ho Aravind Athiviraham Michael J·Lee Jennifer Moriatis Wolf Guillermo A·Ameer Hue H·Luu Rex C·Haydon Jason Strelzow Kelly Hynes Tong-Chuan He Russell R·Reid 2019Genes & Diseases2019,6,3:15
Side Effects during Treatment of Advanced Gastric Carcinoma by Chemotherapy Combined with CIK-cell Transfusion in Elderly People显示文摘OBJECTIVE To study the side effects and therapeutic results of autologous cytokine-induced killer(CIK)cell treatment in elderly patients with advanced gastric cancer. METHODS CIK cells were induced and cultured using biotechnics in vitro, and then the cells were infused back into the patients.Sixty elderly gastric cancer patients treated by chemotherapy(FOLFOX4 protocol)were followed-up.Among them,29 patients were treated with CIK cells during application of chemotherapy.Short-term curative effects and adverse events from the CIK transfusion and chemotherapy were observed. RESULTS Eight cases developed partial remission(PR),9 cases moderate remission(MR),7 cases stable disease(SD)and 5 cases progressive disease(PD).Out of a total of 29 patients who received chemotherapy combined with autologous CIK therapy, the total remission rate(PR+MR)was 58.6%.The total remission rate following chemotherapy alone was 45.2%,including 5 PR cases,9 MR cases,7 SD cases, and 10 PD cases. There was a relatively lower rate of severe chemotherapic toxicities in the CIK- cell transfusion group.Side e?ects of autologous CIK transfusion included chills(13 cases),fever(9 cases),nausea and vomiting (1 case)and general malaise(3 cases).Side effects were treated with conventional therapy resulting in their amelioration. No patients developed shock, blood capillary leakage syndrome,or abnormalities in routine blood, urine, liver and renal function tests. CONCLUSION Adoptive immunotherapy with autologous CIK cells may decrease the clinical signs and symptoms of elderly patients who suffer from advanced gastric cancer.Adverse reactions of patients can be alleviated by conventional therapy. Autologous CIK-cell transfusion may improve endurance to chemotherapy.Jingting Jiang Changping Wu Liangrong Shi Ning Xu Haifeng Deng Mingyang Lu Mei Ji Hbei Zhu Xueguang Zhang 2008Chinese Journal of Clinical Oncology2008,5,2:10

Integrated transcriptome and proteome analysis provides insight into chilling-induced dormancy breaking in Chimonanthus praecox显示文摘Chilling has a critical role in the growth and development of perennial plants. The chilling requirement(CR)for dormancy breaking largely depends on the species. However, global warming is expected to negatively affect chilling accumulation and dormancy release in a wide range of perennial plants. Here,we used Chimonanthus praecox as a model to investigate the CR for dormancy breaking under natural and artificial conditions. We determined the minimum CR(570 chill units,CU)needed for chilling-induced dormancy breaking and analyzed the transcriptomes and proteomes of flowering and non-flowering flower buds(FBs,anther and ovary differentiation completed)with different CRs.The concentrations of ABA and GA3 in the FBs were also determined using HPLC.The results indicate that chilling induced an upregulation of ABA levels and significant downregulation of SHORT VEGETATIVE PHASE(SVP)and FLOWERING LOCUS T(FT)homologs at the transcript level in FBs when the accumulated CR reached 570 CU(IB570)compared to FBs in November(FB.Nov,CK)and nF16(non-flowering FBs after treatment at 16℃for−300 CU),which suggested that dormancy breaking of FBs could be regulated by the ABA-mediated SVP-FT module.Overexpression in Arabidopsis was used to confirm the function of candidate genes, and early flowering was induced in 35S::CpFT1 transgenic lines. Our data provide insight into the minimum CR(570 CU)needed for chilling-induced dormancy breaking and its underlying regulatory mechanism in C.praecox, which provides a new tool for the artificial regulation of flowering time and a rich gene resource for controlling chilling-induced blooming.Zhineng Li Ning Liu Wei Zhang Chunyu Wu Yingjie Jiang Jing Ma Mingyang Li Shunzhao Sui 2020Horticulture Research2020,7,1:6
Stem cell therapy for chronic skin wounds in the era of personalized medicine:From bench to bedside显示文摘With the significant financial burden of chronic cutaneous wounds on the healthcare system, not to the personal burden mention on those individuals afflicted,it has become increasingly essential to improve our clinical treatments. This requires the translation of the most recent benchtop approaches to clinical wound repair as our current treatment modalities have proven insufficient. The most promising potential treatment options rely on stem cellbased therapies.Stem cell proliferation and signaling play crucial roles in every phase of the wound healing process and chronic wounds are often associated with impaired stem cell function.Clinical approaches involving stem cells could thus be utilized in some cases to improve a body’s inhibited healing capacity. We aim to present the laboratory research behind the mechanisms and effects of this technology as well as current clinical trials which showcase their therapeutic potential.Given the current problems and complications presented by chronic wounds,we hope to show that developing the clinical applications of stem cell therapies is the rational next step in improving wound care.Elam Coalson Elliot Bishop Wei Liu Yixiao Feng Mia Spezia Bo Liu Yi Shen Di Wu Scott Du Alexander J.Li Zhenyu Ye Ling Zhao Daigui Cao Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Rex C.Haydon Lewis Shi Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Guillermo A.Ameer Tong-Chuan He Russell R.Reid 2019Genes & Diseases2019,6,4:6
PDCD5 regulates iNKT cell terminal maturation and iNKT1 fate decision显示文摘Invariant natural killer T1(iNKT1)cells are characterized by the preferential expression of T-box transcription factor T-bet(encoded by Tbx21)and the production of cytokine IFN-γ,but the relationship between the developmental process and iNKT1 lineage diversification in the thymus remains elusive. We report in the present study a crucial role of programmed cell death 5(PDCD5)in iNKT cell terminal maturation and iNKT1 fate determination.Mice with T cell-specific deletion of PDCD5 had decreased numbers of thymic and peripheral iNKT cells with a predominantly immature phenotype and defects in response toα-galactosylceramide.Loss of PDCD5 also selectively abolished the iNKT1 lineage by reducing T-bet expression in iNKT cells at an early thymic developmental stage(before CD44 upregulation). We further demonstrated that TOX2,one of the high mobility group proteins that was highly expressed in iNKT cells at stage 1 and could be stabilized by PDCD5,promoted the permissive histone H3K4me3 modification in the promoter region of Tbx21.These data indicate a pivotal and unique role of PDCD5/TOX2 in iNKT1 lineage determination. They also suggest that the fate of iNKT1 may be programmed at the developmental stage of iNKT cells in the thymus.Ke Wang Xinwei Zhang Yifan Wang Gaowen Jin Mingyang Li Shusong Zhang Jie Hao Rong Jin Xiaojun Huang Hounan Wu Jun Zhang Yingyu Chen Qing Ge 2019Cellular & Molecular Immunology2019,16,9:4
Selective depression action of taurine in flotation separation of specularite and chlorite显示文摘Chlorite,as the most representative gangue mineral associated with specularite,of which the separation of these two minerals is difficult. This paper investigated the depression effect of taurine on specularite/chlorite separation via flotation experiments, adsorption tests, contact angle measurements,Zeta potential detection,FT-IR measurements, and XPS analyses. The results of single mineral flotation indicated that chlorite could be depressed selectively by taurine with the recovery of less than 30%,but the floatability of specularite remains high with recovery rate of 81.77%at pH 10.The artificial mixed mineral flotation results confirmed the effectiveness of taurine as a depressant.Surface adsorption, contact angle, and Zeta potential detection revealed taurine primarily adsorbs on the chlorite surface, which hampered the DDA’s subsequent adsorption and results in the chlorite’s poor floatability. The FT-IR spectra and XPS analyses provided further proof that taurine adsorbed on chlorite surface as an electron donor, and part of the electrons transferred from the sulfonic acid group of taurine to metal ions during the adsorption process. In addition, the hydrogen bond between amino-group of taurine and O ions in chlorite surface was also formed in the adsorption process. Finally, optimized adsorption configurations of taurine on chlorite surfaces were proposed.Mingyang Li Cheng Yang Zhaoyang Wu Xiangpeng Gao Xiong Tong Xiankun Yu Hongming Long 2022International Journal of Mining Science and Technology2022,32,3:4
A wave of Foxp3^(+) regulatory T cell accumulation in the neonatal liver plays unique roles in maintaining self-tolerance显示文摘Newborn animals require tightly regulated local and systemic immune environments to govern the development and maturation of multiple organs/tissues even though the immune system itself is far from mature during the neonatal period.Regulatory T cells(Tregs)are essential for maintaining immune tolerance/homeostasis and modulating inflammatory responses. The features of Tregs in the neonatal liver under steady-state conditions are not well understood. The present study aimed to investigate the phenotype, functions, and significance of neonatal Tregs in the liver. We found a wave of thymus-derived Treg influx into the liver during 1–2 weeks of age.Depletion of these Tregs between days 7 and 11 after birth rapidly resulted in Th1-type liver inflammation and metabolic disorder.More Tregs in the neonatal liver than in the spleen underwent MHC II-dependent activation and proliferation, and the liver Tregs acquired stronger suppressive functions. The transcriptomic profile of these neonatal liver Tregs showed elevated expression of PPARγand T-bet and features of Tregs that utilize lipid metabolic machinery and are capable of regulating Th1 responses. The accumulation of Tregs with unique features in the neonatal liver is critical to ensure self-tolerance and liver maturation.Mingyang Li Weijia Zhao Yifan Wang Lixue Jin Gaowen Jin Xiuyuan Sun Wei Wang Ke Wang Xi Xu Jie Hao Rong Jin Wenxian Fu Ying Sun Yingjun Chang Xiaojun Huang Xuyu Zhou Hounan Wu Kunshan Zhang Qing Ge 2020Cellular & Molecular Immunology2020,17,5:4
Rational design of palladium single-atoms and clusters supported on silicoaluminophosphate-31 by a photochemical route for chemoselective hydrodeoxygenation of vanillin显示文摘Chemoselective hydrodeoxygenation of vanillin is of great importance in converting biomass into high value-added chemicals.Herein,we describe a facile photochemical route to access palladium single atoms and clusters supported on silicoaluminophosphate-31(SAPO-31)as a highly active, chemoselective, and reusable catalyst for hydrodeoxygenation of vanillin.Characterizations by aberration-corrected high-angle annular dark-field scanning transmission electron microscopy, extended X-ray absorption fine structure measurement, and CO-absorbed diffuse reflectance infrared Fourier transform spectroscopy reveal the atomically dispersed palladium single atoms and clusters are loosely bonded and randomly dispersed, without forming strong palladium-palladium metallic bonding, over the SAPO-31 support. This catalyst, with a full metal availability to the reactants, exhibits exceptional catalytic activity(TOF:3,000 h^(−1),Yield:>99%)in the hydrodeoxygenation of vanillin toward 2-methoxy-4-methylphenol(MMP)under mild conditions(1 atm,80°C,30 min),along with excellent stability, scalability(up to 100-fold),and wide substrate scope. The superior catalytic performance can be attributed to the synergistic effect of the positively charged palladium single atoms and fully exposed clusters,as well as the strong metal-support interactions. This work may offer a new avenue for the design and synthesis of fully exposed metal catalysts with targeted functionalities.Xiaowen Lu Chunmu Guo Mingyang Zhang Leipeng Leng J.Hugh Horton Wei Wu Zhijun Li 2021Nano Research2021,14,11:3
Memantine combined with environmental enrichment improves spatial memory and alleviates Alzheimer's disease-like pathology in senescence-accelerated prone-8(SAMP8) mice显示文摘Memantine is a N-methyl-D-aspartate(NMDA) receptor antagonist approved for the treatment of moderate to severe Alzheimer's disease(AD).Environmental enrichment(EE) has shown significant beneficial effects on functional improvement in AD.In this study,we sought to determine whether combining these two distinct therapies would yield greater benefit than either drug used alone. We investigated the effect of memantine combined with EE on spatial learning and memory and AD-like pathology in a widely used AD model, the senescence-accelerated prone mice(SAMP8).The SAMP8 mice were randomly assigned to enriched housing(EH) or standard housing(SH),where either memantine(20 mg/kg) or saline was given by gastric lavage once daily continuously for eight weeks. Our results showed that, when provided separately, memantine and EE significantly improved spatial learning and memory by shortening escape latencies and increasing the frequency of entrance into the target quadrant. When combined, memantine and EE showed additive effect on learning and memory as evidenced by significant shorter escape latencies and higher frequency of target entrance than either drug alone.Consistent with the behavior results, pathological studies showed that both memantine and EE significantly reduced hippocampal CA1 neurofibrilliary tangles(NFTs) as well as amyloid beta precursor protein(APP) levels.Combining both therapies synergistically lessened NFTs and APP expression compared to either drug alone in SAMP8 mice, indicating that the combination of memantine with EE could offer a novel and efficient therapeutic strategy for the treatment of AD.Jingde Dong Mi Zhou Xiaoqiang Wu Mingyang Du Xiaoshan Wang 2012The Journal of Biomedical Research2012,26,6:2
Polarization microwave-induced thermoacoustic imaging for quantitative characterization of deep biological tissue microstructures显示文摘Polarization optical imaging can be used to characterize anisotropy in biological tissue microstructures and has been demonstrated to be a powerful tool for clinical diagnosis. However, the approach is limited by an inability to image targets deeper than ~1 mm due to strong optical scattering in biological tissues. As such, we propose a novel polarization microwave-induced thermoacoustic imaging(P-MTAI) technique to noninvasively detect variations in deep tissue by exploiting the thermoacoustic signals induced by four pulsed microwaves of varying polarization orientations. The proposed P-MTAI method overcomes the penetration limits of conventional polarization optical imaging and provides submillimeter resolution over depths of several centimeters. As part of the paper, the structural characteristics of tissues were quantified using a new parameter, the degree of microwave absorption anisotropy. P-MTAI was also applied to the noninvasive detection of morphological changes in cardiomyocytes as they transitioned from ordered to disordered states, providing a potential indication of myocardial infarction.YUJING LI SHANXIANG ZHANG LINGHUA WU ZHONGWEN CHENG ZHENHUI ZHANG HAOHAO WANG SHUXIANG ZHAO MINGYANG REN SIHUA YANG DA XING HUAN QIN 2022Photonics Research2022,10,5:2
Precise robust motion control of cell puncture mechanism driven by piezoelectric actuators with fractional-order nonsingular terminal slidingmode control显示文摘A novel robust controller is proposed in this study to realize the precise motion control of a cell puncture mechanism(CPM)driven by piezoelectric ceramics(PEAs).The entire dynamic model of CPM is constructed based on the Bouc–Wen model, and the nonlinear part of the dynamic model is optimized locally to facilitate the construction of a robust controller.A model-based,nonlinear robust controller is constructed using time-delay estimation(TDE)and fractional-order nonsingular terminal sliding mode(FONTSM).The proposed controller does not require prior knowledge of unknown disturbances due to its real-time online estimation and compensation of unknown terms by using the TDE technology. The controller also has finite-time convergence and high-precision trajectory tracking capabilities due to FONTSM manifold and fast terminal sliding mode-type reaching law.The stability of the closed-loop system is proved by Lyapunov stability theory.Computer simulation and hardware-in-loop simulation experiments of CPM verify that the proposed controller outperforms traditional terminal sliding mode controllers, such as the integer-order or model-free controller. The proposed controller can also continuously output without chattering and has high control accuracy.Zebrafish embryo is used as a verification target to complete the cell puncture experiment. From the engineering application perspective, the proposed control strategy can be effectively applied in a PEA-driven CPM.Shengdong Yu Hongtao Wu Mingyang Xie Haiping Lin Jinyu Ma 2020Bio-Design and Manufacturing2020,3,4:2
Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however, whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied. Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously, including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception, cigarette smoking, alcohol drinking, and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites, which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition, the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring, infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers, indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
Liang Kang Ziwei Zeng Shuangling Luo Hong Zhang Quan Wang Mingyang Ren Miao Wu Weidong Tong Qing Xu Yi Xiao Aiwen Wu Yuan-Guang Chen Bo Feng Zhanlong Shen Liang Huang Xingwei Zhang Minhua Zheng Jian-Ping Wang 2021Gastroenterology Report2021,9,1:2
MOF-derived Co_(9)S_(8) nano-flower cluster array modified separator towards superior lithium sulfur battery显示文摘Lithium sulfur batteries with high energy density are thought to be the most potential energy storage technology that can be commercialized. However, the shuttle effect of polysulfides deteriorates its electrochemical performance.Herein,a novel Co_(9)S_(8)nanostructure derived from metal organic framework material(MOF)was explored by simple liquid phase reaction and heat vulcanization of2-methylimidazole and Co(NO_(3))_(2)·6 H_(2)O on the surface of the original PP separator. The Co_(9)S_(8)nano-flower cluster array wall was vertically and closely arranged with the thickness of 200 nm,and the polysulfide can be adsorbed by its physical and chemical action to slow down the'shuttle effect'.It is found that the cell with the modified separator can achieve an ideal discharge capacity of about 600 mAh/g at 1 C.The specific capacity is maintained at 500 mAh/g after 200 cycles, with only 0.11%of capacity decay per cycle. It provides a new way for the utilization of MOF material derivatives to modify the separator in order to improve the electrochemical performance of lithium-sulfur batteries.Qi Wang Hanqing Zhao Boya Li Chenhui Yang Mingyang Li Yanan Li Peng Han Miaomiao Wu Tengyu Li Ruiping Liu 2021Chinese Chemical Letters2021,32,3:2
