1Drug resistance mechanisms and novel drug targets for tuberculosis therapy显示文摘Drug-resistant tuberculosis(TB) poses a significant challenge to the successful treatment and control of TB worldwide.Resistance to anti-TB drugs has existed since the beginning of the chemotherapy era.New insights into the resistant mechanisms of anti-TB drugs have been provided.Better understanding of drug resistance mechanisms helps in the development of new tools for the rapid diagnosis of drugresistant TB.There is also a pressing need in the development of new drugs with novel targets to improve the current treatment of TB and to prevent the emergence of drug resistance in Mycobacterium tuberculosis.This review summarizes the anti-TB drug resistance mechanisms,furnishes some possible novel drug targets in the development of new agents for TB therapy and discusses the usefulness using known targets to develop new anti-TB drugs.Whole genome sequencing is currently an advanced technology to uncover drug resistance mechanisms in M.tuberculosis.However,further research is required to unravel the significance of some newly discovered gene mutations in their contribution to drug resistance.Md Mahmudul Islam H.M. 2Effects of root morphology and anatomy on cadmium uptake and translocation in rice(Oryza sativa L.)显示文摘A clear description of the certain mechanisms of cadmium(Cd) uptake and translocation in rice(Oryza sativa L.) may help to reduce Cd accumulation in rice grain. Hydroponic experiments were carried out to determine the effects of cultivation conditions(aerated and stagnant) on the uptake, translocation and subcellular distribution of Cd in relation to the morphology and anatomy of roots in two rice genotypes with different Cd accumulations in grains. Marked differences in morphology and anatomy were observed between these two genotypes under different cultivation conditions. Genotypes with low Cd accumulation in grains tended to develop fewer root tips per root surface area, larger root porosity and more mature apoplastic barriers. The stagnant cultivation condition decreased the number of root tips per root surface area but increased root porosity and accelerated apoplastic barrier formation in root tissues.Correlative Cd uptake studies revealed that rice plants with fewer number of root tips per root surface area reduced root Cd uptake ability, while mature apoplastic barriers increased root Cd retention in cell walls and the symplast. Thus, the fewer number of root tips per root surface area and the earlier formation of mature apoplastic barriers led to lower Cd uptake and translocation. The results indicated that the morphology and anatomy of roots could play important roles in Cd uptake and translocation in rice, and could be influenced by both genotype and cultivation conditions. 3Ce and Er Co-doped TiO2 for rapid bacteria- killing using visible light显示文摘Bacterial infection and related diseases are threatening the health of human beings.Photocatalytic disinfection as a simple and low-cost disinfection strategy is attracting more and more attention.In this work,TiO2 nanoparticles(NPs)were modified by co-doping of Ce and Er using the sol-gel method,which endowed TiO2 NPs with enhanced visible light photocatalytic performance but not pure ultraviolet photocatalytic properties compared the untreated TiO2.Our results disclosed that as the doping content of Er increased,the photocatalytic activity of modified TiO2 NPs initially increased and subsequently decreased.The same trend occurred for Ce doping.When the doping dose of Er and Ce is 0.5 mol%and 0.2 mol%,the 0.5Ce0.2Ti-O calcined at 800℃ presented the best antibacterial properties,with the antibacterial efficiency of 91.23%and 92.8%for Staphylococcus aureus and Escherichia coli,respectively.The existence of Er ions is thought to successfully turn the near-infrared radiation into visible region,which is easier to be absorbed by TiO2 NPs.Meanwhile,the addition of Ce ions can effectively extend spectral response range and inhibit the recombination of electrons and holes,enhancing the photocatalytic disinfection activity of co-doped TiO2.Yawei Ren Yajing Han Zhaoyang Li Xiangmei Liu Shengli Zhu Yanqin Liang Kelvin Wai Kwok Yeung Shuilin Wu 2020Bioactive Materials2020,5,2:7

6CMOS-compatible high-index doped silica waveguide with an embedded silicon-nanocrystal strip for all-optical analog-to-digital conversion显示文摘Passive all-optical signal processors that overcome the electronic bottleneck can potentially be the enabling components for the next-generation high-speed and lower power consumption systems. Here, we propose and experimentally demonstrate a CMOS-compatible waveguide and its application to the all-optical analog-to-digital converter(ADC) under the nonlinear spectral splitting and filtering scheme. As the key component of the proposed ADC, a 50 cm long high-index doped silica glass spiral waveguide is composed of a thin silicon-nanocrystal(Si-nc) layer embedded in the core center for enhanced nonlinearity. The device simultaneously possesses low loss(0.16 dB/cm at 1550 nm), large nonlinearity(305 W^-1∕km at 1550 nm), and negligible nonlinear absorption.A 2-bit ADC basic unit is achieved when pumped by the proposed waveguide structure at the telecom band and without any additional amplification. Simulation results that are consistent with the experimental ones are also demonstrated, which further confirm the feasibility of the proposed scheme for larger quantization resolution.This demonstrated approach enables a fully monolithic solution for all-optical ADC in the future, which can digitize broadband optical signals directly at low power consumption. 8Monosialoganglioside 1 may alleviate neurotoxicity induced by propofol combined with remifentanil in neural stem cells显示文摘Monosialoganglioside 1(GM1) is the main ganglioside subtype and has neuroprotective properties in the central nervous system. In this study, we aimed to determine whether GM1 alleviates neurotoxicity induced by moderate and high concentrations of propofol combined with remifentanil in the immature central nervous system. Hippocampal neural stem cells were isolated from newborn Sprague-Dawley rats and treated with remifentanil(5, 10, 20 ng/m L) and propofol(1.0, 2.5, 5.0 μg/m L), and/or GM1(12.5, 25, 50 μg/m L). GM1 reversed combined propofol and remifentanil-induced decreases in the percentage of 5-bromodeoxyuridine(+) cells and also reversed the increase in apoptotic cell percentage during neural stem cell proliferation and differentiation. However, GM1 with combined propofol and remifentanil did not affect β-tubulin(+) or glial fibrillary acidic protein(+) cell percentage during neural stem cell differentiation. 10Gigantic electric-field-induced second harmonic generation from an organic conjugated polymer enhanced by a band-edge effect显示文摘Electric-field-induced second harmonic generation(EFISH),a third-order nonlinear process,arises from the interaction between the electric field of an external bias and that of two incident photons.EFISH can be used to dynamically control the nonlinear optical response of materials and is therefore promising for active nonlinear devices.However,it has been challenging to achieve a strong modulation with EFISH in conventional nonlinear materials.Here,we report a large tunability of an EFISH signal from a subwavelength-thick polymer film sandwiched between a transparent electrode and a metallic mirror.By exploiting the band-edge-enhanced third-order nonlinear susceptibility of the organic conjugated polymer,we successfully demonstrate a gigantic EFISH effect with a modulation ratio up to 422%V^(−1) at a pumping wavelength of 840 nm.The band-edge-enhanced EFISH opens new avenues for modulating the intensity of SHG signals and for controlling nonlinear electro-optic interactions in nanophotonic devices.Shumei Chen King Fai Li Guixin Li Kok Wai Cheah Shuang Zhang 2019Light(Science & Applications)2019,8,1:3

13Whether Syndrome Differentiation Affects Treatment Result:Study Protocol of MaZiRenWan(麻子仁丸)for Functional Constipation in A Randomized Controlled Trial显示文摘Background: Syndrome is one of the most important concepts in Chinese medicine(CM) theory. However, it was not well accounted in most of randomized controlled trials(RCTs). Objective: To determine whether CM syndrome differentiation affects the treatment results, functional constipation(FC) was selected as a target disease, and MaZiRenWan(麻子仁丸, MZRW), a classic CM formula commonly used for constipation with excessive heat syndrome, was selected for study. Methods: It is an 18-week prospective double-blinded, doubledummy RCT, including 2-week run-in, 8-week treatment and 8-week post treatment follow-up. A total of 120 FC patients diagnosed as excessive heat syndrome will be recruited from the First Teaching Hospital of Tianjin University of Traditional Chinese Medicine and the Baokang Affiliated Hospital of Tianjin University of Traditional Chinese Medicine. Patients will be randomly allocated into fixed MZRW(f_MZRW) granule group, modified MZRW(m_MZRW) granule group or bisacodyl group. For m_MZRW group, no more than two herbal granules can be added according to the syndrome differentiation for individual participants. The primary end point is the mean of complete spontaneous bowel movements(CSBMs) per week during the treatment period. Secondary end points include mean of CSBMs per week during follow-up, stool form, global symptom improvement, constipation and constipation-related symptoms assessment, CM syndrome change, and reported adverse events. Discussion: This trial is designed to evaluate the effectiveness of these three interventions for FC patients with the CM syndrome of excessive heat, and to determine the change of CM syndrome and the progress of disease during the treatment course. 14Inhibition of the CDK9-cyclin T1 protein-protein interaction as a new approach against triple-negative breast cancer显示文摘Cyclin-dependent kinase 9(CDK9) activity is correlated with worse outcomes of triplenegative breast cancer(TNBC) patients. The heterodimer between CDK9 with cyclin T1 is essential for maintaining the active state of the kinase and targeting this protein-protein interaction(PPI) may offer promising avenues for selective CDK9 inhibition. Herein, we designed and generated a library of metal complexes bearing the 7-chloro-2-phenylquinoline CN ligand and tested their activity against the CDK9cyclin T1 PPI. Complex 1 bound to CDK9 via an enthalpically-driven binding mode, leading to disruption of the CDK9-cyclin T1 interaction in vitro and in cellulo. Importantly, complex 1 showed promising anti-metastatic activity against TNBC allografts in mice and was comparably active compared to cisplatin. To our knowledge, 1 is the first CDK9-cyclin T1 PPI inhibitor with anti-metastatic activity against TNBC. 15A novel glasses-free virtual reality rehabilitation system on improving upper limb motor function among patients with stroke:A feasibility pilot study显示文摘Background:Virtual reality(VR)technology is increasingly used in stroke rehabilitation.This study aimed to investigate the effectiveness of using the glasses-free VR training to improve motor function of upper limb in patients with stroke.Methods:Twelve patients with stroke were recruited to participate in the intervention of 3 weeks.At the baseline and post intervention,two times of evaluation including Fugl-Meyer upper-extremity scale(FMS-UE),transcranial magnetic stimulation(TMS)measurement and motion evaluation were performed.Results:No significant difference was observed between two groups at baseline evaluation.After the intervention,the FMS-UE scores presented a greater improvement in the VR group compared with the control group.TMS measurement showed that there was significant difference in cortex latency and central motor conduction time between two groups after the intervention,but no significant difference in the amplitude of motor event potential was observed.In addition,there was a significant correlation between game scores and FMS-UE scores.Conclusions:The novel glasses-free VR training was at least as effective as conventional occupational therapy in upper limb motor function,improving nerve conduction time and corticospinal excitability in patient with stroke.Haoyu Xie Hantao Zhang Haowen Liang Hang Fan Jianying Zhou Wai Leung Ambrose Lo Le Li 2021Medicine in Novel Technology and Devices2021,,3:2
16A Z-scheme heterojunction of ZnO/CDots/C_(3)N^(4) for strengthened photoresponsive bacteria-killing and acceleration of wound healing显示文摘The abuse of antibiotics is leading to the emergence of resistant bacteria.In this work,a drug-free composite of ZnO/CDots/g-C3N4 with Z-scheme heterojunction was developed,which was employed to kill bacteria effectively within a very short time under the irradiation of visible light due to the enhanced photocatalytic and photothermal effects.In this system,the CDots acted as a bridge between g-C3N4 and ZnO,effectively inhibiting the recombination of photo-generated electrons with holes and consequently enhancing the photocatalytic properties.In addition,CDots endowed the system with excellent photothermal effects.As a result,the antibacterial efficacy of ZnO/CDots/g-C3N4 composite against S.aureus and E.coli reached up to 99.97%and 99.99%respectively,after 15 min of visible light irradiation,due to the synergistic action of photo-inspired radical oxygen species and hyperthermia.The Zn ions released from the composite promoted the growth of fibroblasts,which accelerated the wound healing process.Yiming Xiang Qilin Zhou Zhaoyang Li Zhenduo Cui Xiangmei Liu Yanqin Liang Shengli Zhu Yufeng Zheng Kelvin Wai Kwok Yeung Shuilin Wu 2020Journal of Materials Science & Technology2020,54,22:2
17Bacteroides ovatus-mediated CD27^(−)MAIT cell activation is associated with obesity-related T2D progression显示文摘Type 2 diabetes(T2D)is highly associated with obesity.However,the factors that drive the transition from excessive weight gain to glucose metabolism disruption are still uncertain and seem to revolve around systemic immune disorder.Mucosal-associated invariant T(MAIT)cells,which are innate-like T cells that recognize bacterial metabolites,have been reported to be altered in obese people and to lead to metabolic dysfunction during obesity.By studying the immunophenotypes of blood MAIT cells from a cross-sectional cohort of obese participants with/without T2D,we found an elevation in CD27^(-)negative(CD27−)MAIT cells producing a high level of IL-17 under T2D obese conditions,which could be positively correlated with impaired glucose metabolism in obese people.We further explored microbial translocation caused by gut barrier dysfunction in obese people as a triggering factor of MAIT cell abnormalities.Specifically,accumulation of the bacterial strain Bacteroides ovatus in the peripheral blood drove IL-17^(-)producing CD27−MAIT cell expansion and could be associated with T2D risk in obese individuals.Overall,these results suggest that an aberrant gut microbiota–immune axis in obese people may drive or exacerbate T2D.Importantly,CD27−MAIT cell subsets and Bacteroides ovatus could represent targets for novel interventional strategies.Our findings extend current knowledge regarding the clinical relevance of body mass index(BMI)-associated variation in circulating MAIT cells to reveal the role of these cells in obesity-related T2D progression and the underlying cellular mechanisms.Yue Li Yi Yang Jin Wang Peihong Cai Mei Li Xixiang Tang Ying Tan Yuchan Wang Fan Zhang Xiaofeng Wen Qiaoxing Liang Yuanpeng Nie Tufeng Chen Xiang Peng Xuemin He Yanhua Zhu Guojun Shi Wai WCheung Lai Wei Yanming Chen Yan Lu 2022Cellular & Molecular Immunology2022,19,7:2

19Ag_(3)PO_(4) decorated black urchin-like defective TiO_(2) for rapid and long-term bacteria-killing under visible light显示文摘Both phototherapy via photocatalysts and physical puncture by artificial nanostructures are promising substitutes for antibiotics when treating drug-resistant bacterial infectious diseases.However,the photodynamic therapeutic efficacy of photocatalysts is seriously restricted by the rapid recombination of photogenerated electron-hole pairs.Meanwhile,the nanostructures of physical puncture are limited to two-dimensional(2D)platforms,and they cannot be fully used yet.Thus,this research developed a synergistic system of Ag_(3)PO_(4) nanoparticles(NPs),decorated with black urchin-like defective TiO_(2)(BU-TiO_(2-X)/Ag_(3)PO_(4)).These NPs had a decreased bandgap compared to BU-TiO_(2-X),and BU-TiO_(2)-X/Ag_(3)PO_(4)(3:1)exhibited the lowest bandgap and the highest separation efficiency for photogenerated electron-hole pairs.After combination with BU-TiO_(2-X),the photostability of Ag_(3)PO_(4) improved because the oxygen vacancy of BU-TiO_(2-X) retards the reduction of Ag^(+) in Ag_(3)PO_(4) into Ag^(0),thus reducing its toxicity.In addition,the nanospikes on the surface of BU-TiO_(2-X) can,from all directions,physically puncture bacterial cells,thus assisting the hybrid’s photodynamic therapeutic effects,alongside the small amount of Ag^(+) released from Ag_(3)PO_(4).This achieves synergy,endowing the hybrid with high antibacterial efficacy of 99.76±0.15%and 99.85±0.09%against Escherichia coli and Staphylococcus aureus,respectively,after light irradiation for 20 min followed by darkness for 12 h.It is anticipated that these findings may bring new insight for developing synergistic treatment strategies against bacterial infectious diseases or pathogenic bacterial polluted environments.Yingde Xu Xiangmei Liu Yufeng Zheng Changyi Li Kelvin Wai Kwok Yeung Zhenduo Cui Yanqin Liang Zhaoyang Li Shengli Zhu Shuilin Wu 2021Bioactive Materials2021,6,6:2
