| 1 | Development of oligonucleotide probes for FISH karyotyping in Haynaldia villosa,a wild relative of common wheat显示文摘Haynaldia villosa is a wild relative of wheat and a valuable gene resource for wheat improvement.Owing to the limited number of probes available for fluorescence in situ hybridization(FISH),the resolution at which the karyotype of H.villosa can be characterized is poor,hampering accurate characterization of small segmental alien introgressions.We designed ten oligonucleotide probes using tandem repeats in DNA sequences derived from the short arm of H.villosa chromosome 6 V(6 VS).FISH with seven of them resulted in clear signals on H.villosa chromosomes.Using these,we constructed FISH karyotypes for H.villosa using oligo-6 VS-1 and oligo-6 VS-35 oligonucleotides and characterized the distribution of the two probes in five different H.villosa accessions.The new FISH probes can efficiently characterize H.villosa introgressions into wheat. | Jia Lei Jiawen Zhou Haojie Sun Wentao Wan Jin Xiao Chunxia Yuan Miroslava Karafiátová Jaroslav Dolezel Haiyan Wang Xiue Wang | 2020 | The Crop Journal2020,8,4: | 3 |
| 2 | Diagnosis and treatment of diverticulum of the uterus in pregnancy through hysteroscopy and methotrexate: a case report显示文摘A 27-year-old patient was diagnosed by hysteroscopy with uterine diverticulum in pregnancy and admitted to the hospital. Under the guidance of hysteroscopy combined with methotrexate, the scraping of the uterus was done and operation successfully completed. | 5 | Hyperbaric oxygen protects against PC12 and H9C2 cell damage caused by oxygen-glucose deprivation/reperfusion via the inhibition of cell apoptosis and autophagy显示文摘In this study,we investigated the protective effect of hyperbaric oxygen(HBO)on PC12 and H9C2 cell damage caused by oxygen-glucose deprivation/reperfusion and its possible mechanism.PC12 and H9C2 cell oxygen-glucose deprivation/reperfusion model were established.Cells were divided into a control group,model group,hyperbaric air(HBA)group and HBO group.The cell viability was detected by the CCK8 assay.Hoechst 33342 and PI staining assays and mitochondrial membrane potential(MMP)assays were used to detect cell apoptosis.The ultrastructure of cells,including autophagosomes,lysosomes,and apoptosis,were examined using a transmission electron microscope.The expression of autophagy-related proteins was detected by cellular immunofluorescence and immunocytochemistry.Our results showed that HBO can significantly improve the vitality of damaged PC12 and H9C2 cells caused by oxygen–glucose deprivation/reperfusion.HBO can significantly inhibit apoptosis of PC12 and H9C2 cells caused by oxygenglucose deprivation/reperfusion.Importantly,we found that the protective mechanism of PC12 and H9C2 cell damage caused by oxygen-glucose deprivation/reperfusion may be related to the inhibition of the autophagy pathway.In this study,the results of cellular immunofluorescence and immunocytochemistry experiments showed that the 4E-BP1,p-AKt and mTOR levels of PC12 and H9C2 cells in the model group decreased,while the levels of LC3B,Atg5 and p53 increased.However,after HBO treatment,these autophagy-related indexes were reversed.In addition,observation of the cell ultrastructure with transmission electron microscopy found that in the model group,a significant increase in the number of autophagic vesicles was observed.In the HBO group,a decrease in autophagic vesicles was observed.The study demonstrated that hyperbaric oxygen protects against PC12 and H9C2 cell damage caused by oxygen-glucose deprivation/reperfusion via the inhibition of cell apoptosis and autophagy. | JIANRONG YANG WAN CHEN XING ZHOU YAOXUAN LI ZHIHUANG NONG LIYUAN ZHOU XUAN WEI XIAORONG PAN CHUNXIA CHEN WENSHENG LU | 2022 | BIOCELL2022,46,1: | 0 |
| 6 | CsGDH2.1 negatively regulates theanine accumulation in late-spring tea plants (Camellia sinensis var. sinensis)显示文摘Theanine,a unique and the most abundant non-proteinogenic amino acid in tea plants,endows tea infusion with the umami taste and anti-stress effects.Its content in tea correlates highly with green tea quality.Theanine content in new shoots of tea plants is high in mid-spring and greatly decreases in late spring.However,how the decrease is regulated is largely unknown.In a genetic screening,we observed that a yeast mutant,glutamate dehydrolase 2(gdh2),was hypersensitive to 40 mM theanine and accumulated more theanine.This result implied a role of CsGDH2s in theanine accumulation in tea plants.Therefore,we identified the two homologs of GDH2,CsGDH2.1 and CsGDH2.2,in tea plants.Yeast complementation assay showed that the expression of CsGDH2.1 in yeast gdh2 mutant rescued the theanine hypersensitivity and hyperaccumulation of this mutant.Subcellular localization and tissue-specific expression showed CsGDH2.1 localized in the mitochondria and highly expressed in young tissues.Importantly,CsGDH2.1 expression was low in early spring,and increased significantly in late spring,in the new shoots of tea plants.These results all support the idea that CsGDH2.1 regulates theanine accumulation in the new shoots.Moreover,the in vitro enzyme assay showed that CsGDH2.1 had glutamate catabolic activity,and knockdown of CsGDH2.1 expression increased glutamate and theanine accumulation in the new shoots of tea plants.These findings suggested that CsGDH2.1-mediated glutamate catabolism negatively regulates theanine accumulation in the new shoots in late spring,and provides a functional gene for improving late-spring green tea quality. | Tingting Chen Jingzhen Ma Huiping Li Shijia Lin Chunxia Dong Yunxia Xie Xiaomei Yan Shupei Zhang Tianyuan Yang Xiaochun Wan Zhaoliang Zhang | 2023 | Horticulture Research2023,10,1: | 0 |
| 7 | A novel on-tissue cycloaddition reagent for mass spectrometry imaging of lipid C=C position isomers in biological tissues显示文摘Application of matrix-assisted laser desorption/ionization mass spectrometry imaging(MALDI-MSI) to investigate the spatiotemporal alterations of lipids in biological tissues has brought many significant results.However, the presence of structural isomers varying in C=C double bond(DB) locations makes isomerresolved MSI an urgent need. Herein, we introduce a new type of light-driven on-tissue [2 + 2] cycloaddition reaction coupled with MALDI-MS/MS imaging to identify lipid DB position isomers and their spatial signatures in biological tissues. 3-Benzoylpyridine was introduced as a novel derivatization reagent, and it exhibited great reactivity toward lipid C=C bond to form oxetanes under both ultraviolet light and visible light irradiation. | 8 | Ensemble Based Diagnosis of the Track Errors of Super Typhoon Mangkhut(2018)显示文摘Super Typhoon Mangkhut(2018)was the most high-impact typhoon in 2018 because of its long lifespan and significant intensity.The operational track forecasts in the short-to-medium range(deterministic and probabilistic forecast)showed a great uncertainty and the forecast landing points varied with different lead times.This study applied ensembles of high-resolution ECMWF forecasts to investigate the major factors and mechanisms of the bias production of the Mangkhut forecast track.The ensembles with the largest track bias were analyzed to examine the possible bias associated factors.The results suggested that environmental steering flows were the main cause for the erroneous southward track error with a variance contribution of 72%.The tropical cyclone(TC)size difference and the interaction of the TC with the subtropical high(SH)were other two key factors that contributed to the track error.Particularly,larger TCs may have led to a stronger erosion of the southern part of the SH,and thus induced significant changes in the large-scale environment and eventually resulted in an additional northward movement of TC. | Ling HUANG Qilin WAN Chunxia LIU Huijun HUANG | 2020 | Journal of Meteorological Research2020,34,2: | 0 |
