| 1 | High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat. | Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu | 2020 | Molecular Plant2020,13,9: | 8 |

| 3 | Single copper sites dispersed on hierarchically porous carbon for improving oxygen reduction reaction towards zinc-air battery显示文摘The demand for high-performance non-precious-metal electrocatalysts to replace the noble metal-based catalysts for oxygen reduction reaction(ORR)is intensively increasing.Herein,single-atomic copper sites supported on N-doped three-dimensional hierarchically porous carbon catalyst(Cu_(1)/NC)was prepared by coordination pyrolysis strategy.Remarkably,the Cu_(1)/NC-900 catalyst not only exhibits excellent ORR performance with a half-wave potential of 0.894 V(vs.RHE)in alkaline media,outperforming those of commercial Pt/C(0.851 V)and Cu nanoparticles anchored on N-doped porous carbon(CuNPs/NC-900),but also demonstrates high stability and methanol tolerance.Moreover,the Cu_(1)/NC-900 based Zn-air battery exhibits higher power density,rechargeability and cyclic stability than the one based on Pt/C.Both experimental and theoretical investigations demonstrated that the excellent performance of the as-obtained Cu_(1)/NC-900 could be attributed to the synergistic effect between copper coordinated by three N atoms active sites and the neighbouring carbon defect,resulting in elevated Cu d-band centers of Cu atoms and facilitating intermediate desorption for ORR process.This study may lead towards the development of highly efficient non-noble metal catalysts for applications in electrochemical energy conversion. | Wenjie Wu Yan Liu Dong Liu Wenxing Chen Zhaoyi Song Ximin Wang Yamin Zheng Ning Lu Chunxia Wang Junjie Mao Yadong Li | 2021 | Nano Research2021,14,4: | 4 |
| 4 | The tea plant CsLHT1 and CsLHT6 transporters take up amino acids, as a nitrogen source, from the soil of organic tea plantations显示文摘Organic tea is more popular than conventional tea that originates from fertilized plants.Amino acids inorganic soils constitute a substantial pool nitrogen(N)available for plants.However,the amino-acid contents in soils of tea plantations and how tea plants take up these amino acids remain largely unknown.In this study,we show that the amino-acid content in the soil of an organic tea plantation is significantly higher than that of a conventional tea plantation.Glutamate,alanine,valine,and leucine were the most abundant amino acids in the soil of this tea plantation.When 15 N-glutamate was fed to tea plants,it was efficiently absorbed and significantly increased the contents of other amino acids in the roots.We cloned seven CsLHT genes encoding amino-acid transporters and found that the expression of CsLHT1,CsLHT2,and CsLHT6 in the roots significantly increased upon glutamate feeding.Moreover,the expression of CsLHT1 or CsLHT6 in a yeast amino-acid uptake-defective mutant,22Δ10α,enabled growth on media with amino acids constituting the sole N source.Amino-acid uptake assays indicated that CsLHT1 and CsLHT6 are H^(+)-dependent high-and low-affinity amino-acid transporters,respectively.We further demonstrated that CsLHT1 and CsLHT6 are highly expressed in the roots and are localized to the plasma membrane.Moreover,overexpression of CsLHT1 and CsLHT6 in Arabidopsis significantly improved the uptake of exogenously supplied 15 N-glutamate and 15 N-glutamine.Taken together,our findings are consistent with the involvement of CsLHT1 and CsLHT6 in amino-acid uptake from the soil,which is particularly important for tea plants grown inorganic tea plantations. | Fang Li Chunxia Dong Tianyuan Yang Shilai Bao Wanping Fang William J.Lucas Zhaoliang Zhang | 2021 | Horticulture Research2021,8,1: | 3 |

| 9 | Beneficial Effects of Celastrol on Immune Balance by Modulating Gut Microbiota in Experimental Ulcerative Colitis Mice显示文摘Ulcerative colitis(UC)is a chronic inflammatory bowel disease caused by many factors including colonic inflammation and microbiota dysbiosis.Previous studies have indicated that celastrol(CSR)has strong anti-inflammatory and immune-inhibitory effects.Here,we investigated the effects of CSR on colonic inflammation and mucosal immunity in an experimental colitis model,and addressed the mechanism by which CSR exerts the protective effects.We characterized the therapeutic effects and the potential mechanism of CSR on treating UC using histological staining,intestinal permeability assay,cytokine assay,flow cytometry,fecal microbiota transplantation(FMT),16S rRNA sequencing,untargeted metabolomics,and cell differentiation.CSR administration significantly ameliorated the dextran sodium sulfate(DSS)-induced colitis in mice,which was evidenced by the recovered body weight and colon length as well as the decreased disease activity index(DAI)score and intestinal permeability.Meanwhile,CSR down-regulated the production of pro-inflammatory cytokines and up-regulated the amount of anti-inflammatory mediators at both mRNA and protein levels,and improved the balances of Treg/Th1 and Treg/Th17 to maintain the colonic immune homeostasis.Notably,all the therapeutic effects were exerted in a gut microbiota-dependent manner.Furthermore,CSR treatment increased the gut microbiota diversity and changed the compositions of the gut microbiota and metabolites,which is probably associated with the gut microbiota-mediated protective effects.In conclusion,this study provides the strong evidence that CSR may be a promising therapeutic drug for UC. | Mingyue Li Weina Guo Yalan Dong Wenzhu Wang Chunxia Tian Zili Zhang Ting Yu Haifeng Zhou Yang Gui Kaming Xue Junyi Li Feng Jiang Alexey Sarapultsev Huafang Wang Ge Zhang Shanshan Luo Heng Fan Desheng Hu | 2022 | Genomics, Proteomics & Bioinformatics2022,20,2: | 1 |

| 13 | CsGDH2.1 negatively regulates theanine accumulation in late-spring tea plants (Camellia sinensis var. sinensis)显示文摘Theanine,a unique and the most abundant non-proteinogenic amino acid in tea plants,endows tea infusion with the umami taste and anti-stress effects.Its content in tea correlates highly with green tea quality.Theanine content in new shoots of tea plants is high in mid-spring and greatly decreases in late spring.However,how the decrease is regulated is largely unknown.In a genetic screening,we observed that a yeast mutant,glutamate dehydrolase 2(gdh2),was hypersensitive to 40 mM theanine and accumulated more theanine.This result implied a role of CsGDH2s in theanine accumulation in tea plants.Therefore,we identified the two homologs of GDH2,CsGDH2.1 and CsGDH2.2,in tea plants.Yeast complementation assay showed that the expression of CsGDH2.1 in yeast gdh2 mutant rescued the theanine hypersensitivity and hyperaccumulation of this mutant.Subcellular localization and tissue-specific expression showed CsGDH2.1 localized in the mitochondria and highly expressed in young tissues.Importantly,CsGDH2.1 expression was low in early spring,and increased significantly in late spring,in the new shoots of tea plants.These results all support the idea that CsGDH2.1 regulates theanine accumulation in the new shoots.Moreover,the in vitro enzyme assay showed that CsGDH2.1 had glutamate catabolic activity,and knockdown of CsGDH2.1 expression increased glutamate and theanine accumulation in the new shoots of tea plants.These findings suggested that CsGDH2.1-mediated glutamate catabolism negatively regulates theanine accumulation in the new shoots in late spring,and provides a functional gene for improving late-spring green tea quality. | Tingting Chen Jingzhen Ma Huiping Li Shijia Lin Chunxia Dong Yunxia Xie Xiaomei Yan Shupei Zhang Tianyuan Yang Xiaochun Wan Zhaoliang Zhang | 2023 | Horticulture Research2023,10,1: | 0 |

| 17 | High-performance fiber strain sensor of carbon nanotube/thermoplastic polyurethane@styrene butadiene styrene with a double percolated structure显示文摘In this work,a high-performance fiber strain sensor is fabricated by constructing a double percolated structure,consisting of carbon nanotube(CNT)/thermoplastic polyurethane(TPU)continuous phase and styrene butadiene styrene(SBS)phase,incompatible with TPU(CNT/TPU@SBS).Compared with other similar fiber strain sensor systems without double percolated structure,the CNT/TPU@SBS sensor achieves a lower percolation threshold(0.38 wt.%)and higher electrical conductivity.The conductivity of 1%-CNT/TPU@SBS(4.12×10^(-3) S·m^(-1))is two orders of magnitude higher than that of 1%-CNT/TPU(3.17×10^(-5) S·m^(-1))at the same CNT loading of 1 wt.%.Due to double percolated structure,the 1%-CNT/TPU@SBS sensor exhibits a wide strain detection range(0.2%-100%)and an ultra-high sensitivity(maximum gauge factor(GF)is 32411 at 100%strain).Besides,the 1%-CNT/TPU@SBS sensor shows a high linearity(R^(2)=0.97)at 0%-20%strain,relatively fast response time(214 ms),and stability(500 loading/unloading cycles).The designed sensor can efficiently monitor physiological signals and movements and identify load distribution after being woven into a sensor array,showing broad application prospects in wearable electronics. | Dong XIANG Libing LIU Xiaoyu CHEN Yuanpeng WU Menghan WANG Jie ZHANG Chunxia ZHAO Hui LI Zhenyu LI Ping WANG Yuntao LI | 2022 | Frontiers of Materials Science2022,16,1: | 0 |
