1The Reference Genome of Tea Plant and Resequencing of 81 Diverse Accessions Provide Insights into Its Genome Evolution and Adaptation显示文摘Tea plant is an important economic crop,which is used to produce the world's oldest and most widely consumed tea beverages.Here,we present a high-quality reference genome assembly of the tea plant(Camellia sinensis var.sinensis)consisting of 15 pseudo-chromosomes.LTR retrotransposons(LTR-RTs)account for 70.38%of the genome,and we present evidence that LTR-RTS play critical roles in genome size expansion and the transcriptional diversification of tea plant genes through preferential insertion in promoter regions and introns.Genes,particularly those coding for terpene biosynthesis pro-teins,associated with tea aroma and stress resistance were significantly amplified through recent tandem duplications and exist as gene clusters in tea plant genome.Phylogenetic analysis of the sequences of 81 tea plant accessions with diverse origins revealed three well-differentiated tea plant populations,support-ing the proposition for the southwest origin of the Chinese cultivated tea plant and its later spread to western Asia through introduction.Domestication and modern breeding left significant signatures on hundreds of genes in the tea plant genome,particularly those associated with tea quality and stress resis-tance.The genomic sequences of the reported reference and resequenced tea plant accessions provide valuable resources for future functional genomics study and molecular breeding of improved cul-tivars of tea plants.Enhua Xia Wei Tong Yan Hou Yanlin An Linbo Chen Qiong Wu Yunlong Liu Jie Yu Fangdong Li Ruopei Li Penghui Li Huijuan Zhao Ruoheng Ge Jin Huang Ali Inayat Mallano Yanrui Zhang Shengrui Liu Weiwei Deng Chuankui Song Zhaoliang Zhang Jian Zhao Shu Wei Zhengzhu Zhang Tao Xia Chaoling Wei Xiaochun Wan 2020Molecular Plant2020,13,7:51
2Relationships between vegetation and stomata,and between vegetation and pollen surface soil in Yunnan,Southwest China显示文摘Surface pollen and stomata of 61 samples collected in a study area ranging from tropical seasonal rainforest to oak forest(Quercus spinosa) in the Yulong Snow Mountain region in Yunnan,China,are used to distinguish vegetation communities.The results show that tropical seasonal rainforest(and mountain rainforest),south subtropical evergreen broad-leaved forest,and Quercus shrub are distinguished effectively from other vegetation types by analysis of surface pollen.The south subtropical evergreen broad-leaved forest,Pinus kesiya forest and evergreen broadleaf forest are distinguished effectively from other types of vegetation by pollen analysis.However,P.yunnanensis forest is not distinguished from other vegetation types,and P.armandii,P.densata forest and temperate deciduous conifer mixed forest are not distinguished.The over-representation of Pinus pollen is the main reason that these vegetation communities are not distinguished from each other.Conifer stomata analysis is an effective tool for identifying and distinguishing different types of coniferous forest,and this method performs well even with a small number of sampling points.SHEN HuaDong LI ChunHai WAN HeWen TONG GuoBang LIU JinSong DAN Johnson 2013Chinese Science Bulletin2013,58,15:12
3Influence on Cellular Signal Transduction Pathway in Dairy Cow Mammary Gland Epithelial Cells by Galactopoietic Compound Isolated from Vaccariae segetalis显示文摘The galactopoietic mechanism of Vaccaria segetalis is still unknown. Understanding dibutyl phthalate (DBP) separated from Vaccaria segetalis on the expression of lactation signal transduction genes of mammary gland epithelial cells, including prlr, erα, akt1, socs2, pparγ and elf5, will be helpful to reveal the molecular mechanism. Western blot and qRT- PCR were used to study the change of prlr, erα, akt, socs2, pparγ, and elf5 expression at mRNA and protein level. Co- localization expression of prolactin receptor (PRLR) and estrogen receptor α (ERα) was observed by immunofluorescence; the expression changes of miRNAs (21, 125b, 143, and 195) and the secretion of β-casein and lactose were detected by qRT-PCR and RP-HPLC. The results showed that Vaccaria segetalis active compound had similar fuctions as estrogen and/or prolactin (PRL) in dairy cow mammary gland epithelial cells (DCMECs), increased the expressions of prlr, erα, akt1, and elf5 genes, while repressed pparγ expressions. DBP promoted socs2 mRNA expression, but its protein expressions were repressed. Furthermore, both DBP and PRL could repress the expressions of miRNA-125b, miRNA-143 and miRNA- 195 in DCMECs. DBP could repress the expression of miRNA-21, while the influence of PRL on miRNA-21 was not certain. 4Metabolic Regulation of Mammary Gland Epithelial Cells of Dairy Cow by Galactopoietic Compound Isolated from Vaccariae segetalis显示文摘In previous experiment, we isolated a compound dibutyl phthalate (DBP) from Vaccaria segetalis which had galactopoietic function on mammary gland epithelial cells of dairy cow (DCMECs). In this experiment, we ascertained the metabolic regulation function of DBP on DCMECs. Many genes related to lactation including Stat5, AMPK, b-casein, Glut1, SREBP-1, PEPCK, and ACC were detected by real-time PCR. Furthermore, Stat5 and AMPK were detected by Western blot and immunofluorescence co-localization, respectively. The results showed that DBP stimulates the expression of Stat5 and p-Stat5, thus activates Stat5 cell signal transduction pathway and stimulates b-casein synthesis. DBP also raises the activities of Glut1 and AMPK to stimulate glucose uptake and glycometabolism and activates the expression of AMPK downstream target genes PEPCK and ACC and expression of SREBP-1 to stimulate milk fat synthesis. In addition, the activities of HK, G-6-PDH, ICDH, ATPase, and energy charges were stimulated by DBP to increase the energy metabolism level of DCMECs. 6Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. 7Inhibition of cyclooxygenase-2 activity in subchondral bone modifies a subtype of osteoarthritis显示文摘Osteoarthritis(OA) causes the destruction of joints. Its pathogenesis is still under investigation, and there is no effective diseasemodifying therapy. Here, we report that elevated cyclooxygenase-2(COX-2) expression in the osteocytes of subchondral bone causes both spontaneous OA and rheumatoid arthritis(RA). The knockout of COX-2 in osteocytes or treatment with a COX-2 inhibitor effectively rescues the structure of subchondral bone and attenuates cartilage degeneration in spontaneous OA(STR/Ort)mice and tumor necrosis factor-α transgenic RA mice. Thus, elevated COX-2 expression in subchondral bone induces both OAassociated and RA-associated joint cartilage degeneration. 8Inhibition of miR-135a-5p attenuates vascular smooth muscle cell proliferation and vascular remodeling in hypertensive rats显示文摘Proliferation of vascular smooth muscle cells(VSMCs)greatly contributes to vascular remodeling in hypertension.This study is to determine the roles and mechanisms of miR-135a-5p intervention in attenuating VSMC proliferation and vascular remodeling in spontaneously hypertensive rats(SHRs).MiR-135a-5p level was raised,while fibronectin type III domain-containing 5(FNDC5)mRNA and protein expressions were reduced in VSMCs of SHRs compared with those of Wistar–Kyoto rats(WKYs).Enhanced VSMC proliferation in SHRs was inhibited by miR-135a-5p knockdown or miR-135a-5p inhibitor,but exacerbated by miR-135a-5p mimic.VSMCs of SHRs showed reduced myofilaments,increased or even damaged mitochondria,increased and dilated endoplasmic reticulum,which were attenuated by miR-135a-5p inhibitor.Dual-luciferase reporter assay shows that FNDC5 was a target gene of miR-135a-5p.Knockdown or inhibition of miR-135a-5p prevented the FNDC5 downregulation in VSMCs of SHRs,while miR-135a-5p mimic inhibited FNDC5 expressions in VSMCs of both WKYs and SHRs.FNDC5 knockdown had no significant effects on VSMC proliferation of WKYs,but aggravated VSMC proliferation of SHRs.Exogenous FNDC5 or FNDC5 overexpression attenuated VSMC proliferation of SHRs,and prevented miR-135a-5p mimic-induced enhancement of VSMC proliferation of SHR.MiR-135a-5p knockdown in SHRs attenuated hypertension,normalized FNDC5 expressions and inhibited vascular smooth muscle proliferation,and alleviated vascular remodeling.These results indicate that miR-135a-5p promotes while FNDC5 inhibits VSMC proliferation in SHRs.Silencing of miR-135a-5p attenuates VSMC proliferation and vascular remodeling in SHRs via disinhibition of FNDC5 transcription.Either inhibition of miR-135a-5p or upregulation of FNDC5 may be a therapeutically strategy in attenuating vascular remodeling and hypertension.Chao Ye Ying Tong Nan Wu Guo-wei Wan Fen Zheng Jing-yu Chen Jian-zhen Lei Hong Zhou Ai-dong Chen Jue-jin Wang Qi Chen Yue-hua Li Yu-ming Kang Guo-qing Zhu 2021Acta Pharmacologica Sinica2021,42,11:4
9Immunological Evaluation of a Novel Mycobacterium tuberculosis Antigen Rv0674显示文摘Objective This study aimed to characterize the diagnostic and vaccine potential of a novel Mycobacterium tuberculosisantigen Rv0674. Methods To evaluate thediagnostic potential and antigenicity of Rv0674, IgG was evaluated using ELISA and interferon (IFN)-γ was done by using ELISpot assay among TB patients and healthy donors. For immunogenicity evaluation, BALB/c mice were immunized with Rv0674. Cytokine production was determined by cytokine release assay using an ELISA kit, and the antibodies were tested using ELISA. Results The results of serum Elisa tests showed that Rv0674 specific immunoglobulin G (IgG) response was higher in TB patients than negative controls. And Rv0674 had good performance in serological test with sensitivity and specificity of 77.1% and 81.1%, respectively. While it shows poor sensitivity and specificity of 26.23% and 79.69% for IFN-γ tests. In BALB/c mice, Rv0674 adjuvant by DDA/PolyI:C could also induce a high level of IFN-γ, interleukin-2 and interleukin-6 as well as a high IgG titer in both high-and low-dose groups indicating that Rv0674 is essential in humoral and cellular immunity. Moreover, the cytokine profile and IgG isotypecharacterized Rv0674 as a Th1/Th2-mixed-type protective immunity with the predominance of Th1 cytokines. 10Comparison of survival between adolescent and young adult vs older patients with hepatocellular carcinoma显示文摘BACKGROUND Due to the special clinical features and biologic characteristics of adolescent and young adult(AYA)cancers,AYA cancers are different from cancers in children and elderly individuals.However,there are few reports on AYA hepatocellular carcinoma(HCC).AIM To investigate the overall survival(OS)of AYA(15-39 years)and elderly(40-74 years)patients with HCC.METHODS The data of all the HCC cases were extracted from the Surveillance,Epidemiology,and End Results database from 2004 to 2015 and were then divided into two groups based on age:AYA group(15-39 years)and older group(40-74 years).Kaplan-Meier curves and log-rank tests were used to compare the OS of the two groups.Propensity score matching(PSM)was employed to analyze the OS difference between the two groups.The Cox proportional hazards regression model was used to perform multivariate analysis to explore the risk factors for OS of HCC patients.RESULTS Compared to elderly cancer patients,AYA patients with HCC had a worse Surveillance,Epidemiology,and End Results stage,including the distant stage(22.1%vs 15.4%,P<0.001),and a more advanced American Joint Committee on Cancer(AJCC)stage,including AJCC III and IV(49.2%vs 38.3%,P<0.001),and were more likely to receive surgery(64.5%vs 47.5%,P<0.001).Before PSM,the AYA group had a longer survival in months(median:20.00,interquartile range[IQR]:5.00-62.50)than the older group(median:15.00,IQR:4.00-40.00)(P<0.001).After PSM,the AYA group still had a longer survival in months(median:21.00,IQR:5.00-64.50)than the older group(median:18.00,IQR:6.00-53.00)(P<0.001).The Cox proportional hazards regression model showed that advanced age(hazard ratio[HR]=1.405,95%CI:1.218-1.621,P<0.001)was a risk factor for OS of HCC patients.In the subgroup analysis,the Cox proportional hazards regression model showed that in AJCC I/II HCC patients,advanced age(HR=1.749,95%CI:1.352-2.263,P<0.001)was a risk factor for OS,while it was not a risk factor in AJCC III/IV HCC patients(HR=1.186,95%CI:0.997-1.410,P=0.054)before PSM.After PSM,advanced age(HR=1.891,95%CI:1.356-2.637,P<0.001)was still a risk factor for OS in AJCC I/II HCC patients,but was not a risk factor for OS in AJCC III/IV HCC patients(HR=1.192,95%CI:0.934-1.521,P=0.157)after PSM.CONCLUSION AYA patients with HCC have different clinical characteristics from older adults.In different AJCC stages,the two groups of patients have different OS:In AJCC I/II HCC patients,advanced age is a risk factor for OS,but it is not a risk factor for OS in the AJCC III/IV HCC patient group.Jie Ren Ying-Mu Tong Rui-Xia Cui Zi Wang Qing-Lin Li Wei Liu Kai Qu Jing-Yao Zhang Chang Liu Yong Wan 2020World Journal of Gastrointestinal Oncology2020,12,12:3
11Preliminary Study on Drug Susceptibility Profile and Resistance Mechanisms to Macrolides of Clinical Isolates of Non-tuberculous Mycobacteria from China显示文摘Objective Macrolide susceptibility and drug resistance mechanisms of clinical non-tuberculous mycobacteria(NTM) isolates were preliminarily investigated for more accurate diagnosis and treatment of the infection in China. Methods Four macrolides, including clarithromycin(CLAR), azithromycin(AZM), roxithromycin(ROX), and erythromycin(ERY), were used to test the drug susceptibility of 310 clinical NTM isolates from six provinces of China with the broth microdilution method. Two resistance mechanisms, 23 S r RNA and erm, were analyzed with nucleotide sequence analysis. Results Varied effectiveness of macrolides and species-specific resistance patterns were observed. Most Mycobacterium abscessus subsp. massiliense were susceptible and all M. fortuitum were highly resistant to macrolides. All the drugs, except for erythromycin, exhibited excellent activities against slow-growing mycobacteria, and drug resistance rates were below 22.2%. Only four highly resistant strains harbored 2,058/2,059 substitutions on rrl and none of other mutations were related to macrolide resistance. G2191 A and T2221 C on rrl were specific for the M. abscessus complex(MABC). Seven sites, G2140 A, G2210 C, C2217 G, T2238 C, T2322 C, T2404 C, and A2406 G, were specifically carried by M. avium and M. intracellulare. Three sites, A2192 G, T2358 G, and A2636 G, were observed only in M. fortuitum and one site G2152 A was specific for M. gordonae. The genes erm(39) and erm(41) were detected in M. fortuitum and M. abscessus and inducible resistance was observed in relevant sequevar. Conclusion The susceptibility profile of macrolides against NTM was demonstrated. 12Entire Solutions for Nonlocal Dispersal Equations with Bistable Nonlinearity: Asymmetric Case显示文摘This paper mainly focuses on the entire solutions of a nonlocal dispersal equation with asymmetric kernel and bistable nonlinearity. Compared with symmetric case, the asymmetry of the dispersal kernel function makes more diverse types of entire solutions since it can affect the sign of the wave speeds and the symmetry of the corresponding nonincreasing and nondecreasing traveling waves.We divide the bistable case into two monostable cases by restricting the range of the variable, and obtain some merging-front entire solutions which behave as the coupling of monostable and bistable waves. Before this, we characterize the classification of the wave speeds so that the entire solutions can be constructed more clearly. 17Fault Tolerant Design of Large-Scale Digital Beam Forming in SRAM-FPGAs for Software Defined Satellite Platforms显示文摘Large scale digital beamforming(LS-DBFs)are widely used in satellite communications for spectrum reuse and transmission enhancement.SRAM-FPGAs are a popular option for software defined satellite platforms due to their rich computation resources and high flexibility.However,they are sensitive to soft errors,which limit their application in space.This paper discusses the application of coding based fault tolerance schemes for the protection of LS-DBFs on software defined payloads.Since multiple FPGAs are usually needed to support the whole LS-DBFs system,different decomposition schemes are compared in terms of resource efficiency and reliability when the coding based scheme is applied to protect the DBFs on a FPGA.Theoretical analysis and hardware experiments shows that resource efficiency and reliability are a pair of contradictory requirements for decomposition schemes.The protection with vertical decomposition could improve the reliability by 96%with 1.5x redundancy.And the protection with horizontal decomposition could improve the reliability by 85%with 1.2 x redundancy.Zhen Gao Jinhua Zhu Tong Yan Linghua Guo Xiangping Chen Yinqiao Li Xiaolei Wan 2020China Communications2020,17,7:2

20Key regulators of intestinal stem cells:diet,microbiota,and microbial metabolites显示文摘Interactions between diet and the intestinal microbiome play an important role in human health and disease development.It is well known that such interactions,whether direct or indirect,trigger a series of metabolic reactions in the body.Evidence suggests that intestinal stem cells(ISCs),which are phenotypic precursors of various intestinal epithelial cells,play a significant role in the regulation of intestinal barrier function and homeostasis.The advent and evolution of intestinal organoid culture techniques have presented a key opportunity to study the association between the intestinal microenvironment and ISCs.As a result,the effects exerted by dietary factors,intestinal microbiomes,and their metabolites on the metabolic regulation of ISCs and the potential mechanisms underlying such effects are being gradually revealed.This review summarises the effects of different dietary patterns on the behaviour and functioning of ISCs and focuses on the crosstalk between intestinal microbiota,related metabolites,and ISCs,with the aim of fully understanding the relationship between these three factors and providing further insights into the complex mechanisms associated with ISCs in the human body.Gaining an understanding of these mechanisms may lead to the development of novel dietary interventions or drugs conducive to intestinal health.Chensi Yao Xiaowen Gou Chuanxi Tian Lijuan Zhou Rui Hao Li Wan Zeyu Wang Min Li Xiaolin Tong 2023Journal of Genetics and Genomics2023,50,10:1
