1Mapping wetland changes in China between 1978 and 2008显示文摘Four wetland maps for all China have been produced,based on Landsat and CBERS-02B remote sensing data between 1978 and 2008 (1978,1990,2000 and 2008).These maps were mainly developed by manual interpretation and validated by substantial field investigation in 2009.Based on these maps,we analyzed the 2008 wetland distribution in China and discussed wetland changes and their drivers over the past 30 years.(i) There were about 324097 km 2 of wetlands in 2008,for which inland marshes or swamps were the most common wetland type (35%),with lakes (26%) second.Most of the wetlands were in Heilongjiang,Inner Mongolia,Qinghai and Tibet,occupying about 55% of the national wetland area.(ii) From 1978 to 2008,China's wetland area continually and significantly decreased,by about 33% based on changes in the wetland map.This was in sharp contrast to the increase in artificial wetlands,which increased by about 122%.Inland marshes accounted for the main loss of total wetlands from 1978 to 2000.From 2000 through 2008,riverine and lacustrine wetlands constituted the main wetland loss.Fortunately however,the rate of wetland loss decreased from 5523 to 831 km 2 /a.(iii) The change ratio of lost natural wetlands (including inland and coastal wetlands) to non-wetlands has decreased slightly over the past 30 years.From 1978 to 1990,nearly all natural wetlands (98%) lost were transformed into non-wetlands.However,the ratio declined to 86% from 1990 to 2000,and to 77% from 2000 to 2008.(iv) All Chinese provinces were divided into three groups according to patterns of wetland changes,which could relate to the driving forces of such changes.Tibet was completely different from other provinces,as it was one representative example in which there was a net wetland increase,because of global warming and decreased human activity since 1990.Increased economic development caused considerable wetland loss in most eastern provinces,and artificial wetlands increased.NIU ZhenGuo ZHANG HaiYing WANG XianWei YAO WenBo ZHOU DeMin ZHAO KuiYi ZHAO Hui LI NaNa HUANG HuaBing LI CongCong YANG Jun LIU CaiXia LIU Shuang WANG Lin LI Zhan YANG ZhenZhong QIAO Fei ZHENG YaoMin CHEN YanLei SHENG YongWei GAO XiaoHong ZHU WeiHong WANG WenQing WANG Hong WENG YongLing ZHUANG DaFang LIU JiYuan LUO ZhiCai CHENG Xiao GUO ZiQi GONG Peng 2012Chinese Science Bulletin2012,57,22:51

3Hi-TOM: a platform for high-throughput tracking of mutations induced by CRISPR/Cas systems显示文摘The CRISPR/Cas system has been extensively applied to make precise genetic modifications in various organisms. Despite its importance and widespread use, large-scale mutation screening remains time-consuming, labour-intensive and costly. Here, we developed Hi-TOM(available at http://gffzz103f3ce5d9b14419hw9n6qopf9bf96pq6.ffgz.tsg.suse.edu.cn/hi-tom/), an online tool to track the mutations with precise percentage for multiple samples and multiple target sites. We also described a corresponding next-generation sequencing(NGS) library construction strategy by fixing the bridge sequences and barcoding primers. Analysis of the samples from rice, hexaploid wheat and human cells reveals that the Hi-TOM tool has high reliability and sensitivity in tracking various mutations, especially complex chimeric mutations frequently induced by genome editing. Hi-TOM does not require special design of barcode primers,cumbersome parameter configuration or additional data analysis. 5The first all-season sample set for mapping global land cover with Landsat-8 data显示文摘We report the world's first all-season training and validation sample sets for global land cover classification with Landsat-8 data.Prior to this,such samples were only available at a single date primarily from the growing season.It is unknown how much limitation such a single-date sample has to mapping global land cover in other seasons of the year.To answer this question,we selected available Landsat-8 images from four seasons and collected training and validation samples from them.We compared the performances of training samples in different seasons using Random Forest algorithm.We found that the use of training samples from any individual season would result in the best overall classification accuracy when validated by samples in the same season.The global overall accuracy from combined best seasonal results was 67.2% when classifying the 11 Level-1 classes in the Finer Resolution Observation and Monitoring of Global Land Cover(FROM-GLC) classification system.The use of training samples from all seasons(named all-season training sample set hereafter) produced an overall accuracy of 67.0%.We also tested classification within 10° latitude 60° longitude zones using all-season training subsample within each zone and obtained an overall accuracy of 70.2%.This indicates that properly grouped subsamples in space can help improve classification accuracies.All the results in this study seem to suggest that it is possible to use an all-season training sample set to reach global optimality with universal applicability in classifying images acquired at any time of a year for global land cover mapping.Congcong Li Peng Gong Jie Wang Zhiliang Zhu Gregory S. 8Characteristics of the water-soluble components of aerosol particles in Hefei,China显示文摘Size-classified daily aerosol mass concentrations and concentrations of water-soluble inorganic ions were measured in Hefei,China,in four representative months between September 2012 and August 2013.An annual average mass concentration of 169.09μg/m^3 for total suspended particulate(TSP)was measured using an Andersen Mark-II cascade impactor.The seasonal average mass concentration was highest in winter(234.73μg/m^3)and lowest in summer(91.71μg/m^3).Water-soluble ions accounted for 59.49%,32.90%,48.62%and 37.08%of the aerosol mass concentration in winter,spring,summer,and fall,respectively,which indicated that ionic species were the primary constituents of the atmospheric aerosols.The four most abundant ions were NO_3^-,SO_4^(2-),Ca^(2+) and NH_4^+.With the exception of Ca^(2+),the mass concentrations of water-soluble ions were in an intermediate range compared with the levels for other Chinese cities.Sulfate,nitrate,and ammonium were the dominant fine-particle species,which were bimodally distributed in spring,summer and fall;however,the size distribution became unimodal in winter,with a peak at 1.1–2.1μm.The Ca^(2+) peak occurred at approximately 4.7–5.8μm in all seasons.The cation to anion ratio was close to 1.4,which suggested that the aerosol particles were alkalescent in Hefei.The average NO_3^-/SO_4^(2-)mass ratio was 1.10 in Hefei,which indicated that mobile source emissions were predominant.Significant positive correlation coefficients between the concentrations of NH_4^+ and SO_4^(2-),NH_4^+ and NO_3^-,SO_4^(2-)and NO_3^-,and Mg^(2+) and Ca^(2+) were also indicated,suggesting that aerosol particles may be present as(NH_4)_2SO_4,NH_4HSO_4,and NH_4NO_3.Xue-liang Deng Chun-e Shi Bi-wen Wu Yuan-jian Yang Qi Jin Hong-lei Wang Song Zhu Caixia Yu 2016Journal of Environmental Sciences2016,28,4:18
9Collagen type Ⅱ suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction显示文摘Hypertrophic differentiation is not only the terminal process of endochondral ossification in the growth plate but is also an important pathological change in osteoarthritic cartilage.Collagen type II(COL2A1)was previously considered to be only a structural component of the cartilage matrix,but recently,it has been revealed to be an extracellular signaling molecule that can significantly suppress chondrocyte hypertrophy.However,the mechanisms by which COL2A1 regulates hypertrophic differentiation remain unclear.In our study,a Col2a1 p.Gly1170Ser mutant mouse model was constructed,and Col2a1 loss was demonstrated in homozygotes.Loss of Col2a1 was found to accelerate chondrocyte hypertrophy through the bone morphogenetic protein(BMP)-SMAD1 pathway.Upon interacting with COL2A1,integrinβ1(ITGB1),the major receptor for COL2A1,competed with BMP receptors for binding to SMAD1 and then inhibited SMAD1 activation and nuclear import.COL2A1 could also activate ITGB1-induced ERK1/2 phosphorylation and,through ERK1/2-SMAD1 interaction,it further repressed SMAD1 activation,thus inhibiting BMP-SMAD1-mediated chondrocyte hypertrophy.Moreover,COL2A1 expression was downregulated,while chondrocyte hypertrophic markers and BMP-SMAD1 signaling activity were upregulated in degenerative human articular cartilage.Our study reveals novel mechanisms for the inhibition of chondrocyte hypertrophy by COL2A1 and suggests that the degradation and decrease in COL2A1 might initiate and promote osteoarthritis progression.Chengjie Lian Xudong Wang Xianjian Qiu Zizhao Wu Bo Gao Lei Liu Guoyan Liang Hang Zhou Xiaoming Yang Yan Peng Anjing Liang Caixia Xu Dongsheng Huang Peiqiang Su 2019Bone Research2019,7,1:17
10Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. 11Fine mapping and candidate gene analysis of purple pericarp gene Pb in rice (Oryza sativa L.)显示文摘Purple rice is a type of rice with anthocyanins deposited in its grain pericarp. The rice Pb gene control-ling purple pericarp character is known to be on chromosome 4,and the purple color is dominant over white color. In this study,we fine mapped the Pb gene using two F2 segregating populations,i.e. Pei'ai 64S(white) × Yunanheixiannuo(purple) and Pei'ai 64S × Chuanheinuo(purple) . In the first-pass map-ping,the Pb gene was located in the region downstream the SSR marker RM3820. In the fine mapping,the candidate region was saturated with InDel and CAPS markers developed specifically for this study. Eventually,the Pb gene was mapped within the 25-kb region delimited by the upstream marker RID3 and the downstream marker RID4. The delimited region contained two annotated genes,Ra and bhlh16(TIGR Rice Genome,R.5) . The former is a homologue of the Myc transcription factor Lc controlling anthocyanin biosynthesis in maize,and the latter is a homologue of the TT8 gene,which is also an Myc transcription factor gene controlling the pericarp pigmentation in Arabidopsis thaliana. Sequence analysis showed that the exon 7 of the Ra gene of Yunanheixiannuo and Chuanheinuo had a 2-bp(GT) deletion compared with those of the white rice varieties Pei'ai 64S,9311 and Nipponbare. A CAPS marker,CAPSRa,was developed according to the GT deletion for analysis of the two F2 segregating populations and 106 rice lines. The results showed that all F2 plants with white pericarp,and all non-purple rice lines(63 white and 22 red) contained no GT deletion,but all 20 purple rice lines con-tained the GT deletion. 13Highly efficient heritable genome editing in wheat using an RNA virus and bypassing tissue culture.显示文摘Genome editing provides novel strategies for improving plant traits,but relies on current genetic transformation and plant regeneration procedures,which can be inefficient.We have engineered a barley stripe mosaic virus(BSMV)-based sgRNA delivery vector(BSMV-sg)that is effective in performing heritable genome editing in Cas9-transgenic wheat plants.Mutated progenies were present in the next generation at frequencies ranging from 12.9%to 100%in three different wheat varieties,and 53.8%to 100%of mutants were virus-free.We also achieved multiplex mutagenesis in progeny using a pool of BSMV-sg vectors harboring different sgRNAs.Furthermore,we devised a virus-induced transgene-free editing procedure(VITF-Edit)to generate Cas9-free wheat mutants by crossing BSMV-infected Cas9-transgenic wheat pollen with wild-type wheat.Our study provides a robust,convenient and tissue culture-free approach for genome editing in wheat through virus infection.Tingdong Li Jiacheng Hu Yu Sun Boshu Li Dingliang Zhang Wenli Li Jinxing Liu Dawei Li Caixia Gao Yongliang Zhang Yanpeng Wang 2021Molecular Plant2021,14,11:10
14A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. 15Sequence stratigraphy, paleogeography, and coal accumulation regularity of major coal-accumulating periods in China显示文摘There are 9 major coal-accumulating periods during geological history in China,including the Early Carboniferous,Late Carboniferous-Early Permian,Middle Permian,Late Permian,Late Triassic,Early-Middle Jurassic,Early Cretaceous,Paleogene and Neogene.The coal formed in these periods were developed in different coal-accumulating areas(CAA)including the North China,South China,Northwest China,Northeast China,the Qinghai–Tibet area,and China offshore area.In this paper,we investigated depositional environments,sequence stratigraphy,lithofacies paleogeography and coal accumulation pattern of five major coal-accumulating periods including the Late Carboniferous to Middle Permian of the North China CAA,the Late Permian of the South China CAA,the Late Triassic of the South China CAA,the Early-Middle Jurassic of the North and Northwest China CAA,and the Early Cretaceous in the Northeast China CAA.According to distribution of the coal-bearing strata and the regional tectonic outlines,we have identified distribution range of the coal-forming basins,sedimentary facies types and coal-accumulating models.The sequence stratigraphic frameworks of the major coal-accumulating periods were established based on recognition of a variety of sequence boundaries.The distribution of thick coals and migration patterns of the coal-accumulating centers in the sequence stratigraphic framework were analyzed.The lithofacies paleogeography maps based on third-order sequences were reconstructed and the distribution of coal accumulation centers and coal-rich belts were predicted.Longyi Shao Xuetian Wang Dongdong Wang Mingpei Li Shuai Wang Yingjiao Li Kai Shao Chao Zhang Caixia Gao Daxiao Dong Aiguo Cheng Jing Lu Congwei Ji Di Gao 2020International Journal of Coal Science & Technology2020,7,2:9

17CRISPR editing-mediated antiviral immunity: a versatile source of resistance to combat plant virus infections显示文摘Virus infection poses a constant threat on global crop productivity.It is estimated that virus diseases account for at least 10% of crop losses worldwide.Development of resistant crops using naturally evolved resistance genes has been a main measure for controlling plant virus epidemics.However,isolation of plant resistance genes is costly and time-consuming.Furthermore,most plant resistance genes are pathogen race-specific,with very few of them being broadly specific (Langner et al.,2018).Consequently,artificial immunity with engineered resistance to virus infections has received substantial research,which includes pathogenderived resistance,introducing single or stacked transgene into crops,and RNA interference (Shepherd et al.,2009).However,these strategies have so far achieved limited success due to narrow availability and/or fitness costs on host plants.Therefore,it is urgent to develop novel and more efficient strategies to combat plant virus infections.Recent breakthrough of CRISPR/Cas-based DNA and RNA editing tools provides a promising direction for engineering artificial immunity to plant viruses.There are now increasing reports demonstrating that CRISPR/Cas systems can be harnessed to develop antiviral immunity in plants with high efficiency and broad specificity (Table S1 in Supporting Information).Xiang Ji Daowen Wang Caixia Gao 2019Science China(Life Sciences)2019,62,9:6

19Myofiber necroptosis promotes muscle stem cell proliferation via releasing Tenascin-C during regeneration显示文摘Necroptosis,a form of programmed cell death,is characterized by the loss of membrane integrity and release of intracellular contents,the execution of which depends on the membrane-disrupting activity of the Mixed Lineage Kinase Domain-Like protein(MLKL)upon its phosphorylation.Here we found myofibers committed MLKL-dependent necroptosis after muscle injury.Either pharmacological inhibition of the necroptosis upstream kinase Receptor Interacting Protein Kinases 1(RIPK1)or genetic ablation of MLKL expression in myofibers led to significant muscle regeneration defects.By releasing factors into the muscle stem cell(MuSC)microenvironment,necroptotic myofibers facilitated muscle regeneration.Tenascin-C(TNC),released by necroptotic myofibers,was found to be critical for MuSC proliferation.The temporary expression of TNC in myofibers is tightly controlled by necroptosis;the extracellular release of TNC depends on necroptotic membrane rupture.TNC directly activated EGF receptor(EGFR)signaling pathway in MuSCs through its N-terminus assembly domain together with the EGF-like domain.These findings indicate that necroptosis plays a key role in promoting MuSC proliferation to facilitate muscle regeneration.Shen'ao Zhou Wei Zhang Gaihong Cai Yingzhe Ding Caixia Wei Sheng Li Yu Yang Jie Qin Dan Liu Hao Zhang Xiexiang Shao Jianhua Wang Hongye Wang Wenjun Yang Huating Wang She Chen Ping Hu Liming Sun 2020Cell Research2020,30,12:4
20Ablative fractional CO_(2)laser surgery improving sleep quality,pain and pruritus in adult hypertrophic scar patients:a prospective cohort study显示文摘Background:Poor sleep quality is associated with a decrease in quality of life in patients with major burn scars,combined with pruritus and pain.Few interventions have been reported to improve the sleep quality of patients with scars.In the current prospective cohort study,we investigated the efficacy of CO_(2)-ablative fractional laser(AFL)surgery vs conventional surgery in post-burn patients with hypertrophic scars with sleep quality as the primary study outcome.Methods:In total 68 consecutive patients undergoing scar surgical treatment were recruited,including a CO_(2)-AFL surgery cohort(n=35)and a conventional surgery cohort(n=33).A subgroup from the AFL cohort was selected.Sleep quality,pain and pruritus were evaluated.Multiple linear regression analyses were performed to reveal the effect of CO_(2)-AFL surgery.Results:The CO_(2)-AFL surgery cohort had significantly lower Pittsburgh sleep quality index(PSQI)global scores than the conventional surgery cohort after the last surgical treatment.In the subgroup of patients receiving hardware sleep monitoring,CO_(2)-AFL markedly increased deep sleep time,deep sleep efficiency and reduced initial sleep latency.Compared to the conventional surgery cohort,the CO_(2)-AFL cohort presented significantly lower pain and pruritus scores.Correlation analysis showed pain and pruritus were significantly associated with PSQI scores,and there were also significant correlations between pain and pruritus scores.Multiple linear regression analysis showed that surgery method was negatively linearly correlated with visual analog scale(VAS)pain score,brief pain inventory(BPI)total,VAS pruritus score,5-D itch scale total,four-item itch questionnaire(FIIQ)total and PSQI total.Conclusions:CO_(2)-AFL surgery significantly improved sleep quality and reduced pain and pruritus of hypertrophic scar patients.The alleviation of sleep disorder was associated with improvement of deep sleep quality including deep sleep time and deep sleep deficiency.Trial registration:The Chinese Clinical Trial Registry(ChiCTR200035268)approved retrospectively registration on 5 Aug 2020.Kaiyang Lv Huazhen Liu Haiting Xu Caixia Wang Shihui Zhu Xiaozhen Lou Pengfei Luo Shichu Xiao Zhaofan Xia 2021Burns & Trauma2021,9,1:4
