| 1 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 3 | Efficacy of Xuebijing Injection(血必净注射液)on Cardiopulmonary Bypass-Associated Pulmonary Injury:A Prospective,Single-center,Double Blinded,Randomized Controlled Trial显示文摘Objective: To evaluate the efficacy of Xuebijing Injection(血必净注射液, XBJ) on the lung injury induced by cardiopulmonary bypass(CPB). Methods: Fifty patients undergoing CPB were randomized to either the saline group or XBJ group according to a random number table(25 cases in each group). The patients in the saline group received saline and patients in XBJ group received XBJ at 12 h prior to the operation, at the beginning of the operation, and at 12 h after the second injection. The PaO_2/Fi O2 at extubation 3 days post-operation, duration of ventilation in the intensive care unit(ICU), and lengths of stay in the ICU and hospital were recorded. The levels of inflammatory mediators including interleukin(IL)-1β, IL-8, IL-10, and C-reactive protein(CRP) in bronchoalveolar lavage fluid(BALF) and plasma were measured. The neutrophil count and elastase neutrophil elastase in BALF were also measured. In addition, adverse events were monitored. Results: The PaO-2/FiO_2 in the XBJ group was higher than that in the saline group from 12 to 72 h post-operation(all P<0.05). The blood levels of IL-1β, IL-8, and CRP in the XBJ group from 12 to 72 h were all significantly lower than those in the saline group(all P<0.05). In contrast, the level of the anti-inflammatory cytokine IL-10 was significantly higher in the XBJ group than in the saline group(P<0.05). In addition, 4 patients presented with atelectasis in the saline group and none in the XBJ group. Ten patients experienced mild acute respiratory distress syndrome(ARDS) during hospitalization, and 5 patients with mild ARDS were in the XBJ group(P<0.05). | 5 | CXCR7/CXCL12 axis is involved in lymph node and liver metastasis of gastric carcinoma显示文摘AIM To investigate the role of CXC chemokine receptor(CXCR)-7 and CXCL12 in lymph node and liver metastasis of gastric carcinoma.METHODS In 160 cases of gastric cancer, the expression of CXCR7 and CXCL12 in tumor and matched tumoradjacent non-cancer tissues, in the lymph nodes around the stomach and in the liver was detected using immunohistochemistry to analyze the relationship between CXCR7/CXCL12 expression and clinicopathological features and to determine whether CXCR7 and CXCL12 constitute a biological axis to promote lymph node and liver metastasis of gastric cancer. Furthermore, the CXCR7 gene was silenced and overexpressed in human gastric cancer SGC-7901 cells, and cell proliferation, migration and invasiveness were measured by the MTT, wound healing and Transwell assays, respectively. RESULTS CXCR7 expression was up-regulated in gastric cancer tissues(P = 0.011). CXCR7/CXCL12 expression was significantly related to poor tumor differentiation, high tumor stage and lymph node(r = 0.338, P = 0.000) and liver metastasis(r = 0.629, P = 0.000). The expression of CXCL12 in lymph node and liver metastasis was higher than that in primary gastric cancer tissues(χ2 = 6.669, P = 0.010; χ2 = 25379, P = 0.000), and the expression of CXCL12 in lymph node and liver metastasis of gastric cancer was consistent with the positive expression of CXCR7 in primary gastric cancer(r = 0.338, P = 0.000; r = 0.629, P = 0.000). Overexpression of the CXCR7 gene promoted cell proliferation, migration and invasion. Silencing of the CXCR7 gene suppressed SGC-7901 cell proliferation, migration and invasion. Human gastric cancer cell lines expressed CXCR7 and showed vigorous proliferation and migratory responses to CXCL12.CONCLUSION The CXCR7/CXCL12 axis is involved in lymph node and liver metastasis of gastric cancer. | 6 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 7 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 8 | Efficacy of chitosan and sodium alginate scaffolds for repair of spinal cord injury in rats显示文摘Spinal cord injury results in the loss of motor and sensory pathways and spontaneous regeneration of adult mammalian spinal cord neurons is limited. Chitosan and sodium alginate have good biocompatibility, biodegradability, and are suitable to assist the recovery of damaged tissues, such as skin, bone and nerve. Chitosan scaffolds, sodium alginate scaffolds and chitosan-sodium alginate scaffolds were separately transplanted into rats with spinal cord hemisection. Basso-Beattie-Bresnahan locomotor rating scale scores and electrophysiological results showed that chitosan scaffolds promoted recovery of locomotor capacity and nerve transduction of the experimental rats. Sixty days after surgery, chitosan scaffolds retained the original shape of the spinal cord. Compared with sodium alginate scaffolds-and chitosan-sodium alginate scaffolds-transplanted rats, more neurofilament-H-immunoreactive cells(regenerating nerve fibers) and less glial fibrillary acidic protein-immunoreactive cells(astrocytic scar tissue) were observed at the injury site of experimental rats in chitosan scaffold-transplanted rats. Due to the fast degradation rate of sodium alginate, sodium alginate scaffolds and composite material scaffolds did not have a supporting and bridging effect on the damaged tissue. | 9 | A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. | 11 | Spatial Distribution and Risk Assessment of Heavy Metals in Paddy Soils of Yongshuyu Irrigation Area from Songhua River Basin,Northeast China显示文摘There is an increasing concern for potentially hazardous metals pollution, which can threaten crops production and human health. In this study, the spatial distribution and environmental risks of eight heavy metals in surface soil samples collected from the paddy fields in Yongshuyu irrigation area, Northeast China were investigated. The mean concentrations of Pb, Cr, Cu, Ni, Zn, Cd, Hg and As were 34.6 ± 4.67, 82.8 ± 9.51, 17.3 ± 4.09, 21.2 ± 12.0, 88.6 ± 17.9, 0.18 ± 0.15, 0.22 ± 0.07 and 8.77 ± 2.47 mg/kg, respectively, which were slightly higher than their corresponding background values of Jilin Province, indicating enrichment of these metals in the paddy soils, especially for Ni, Cd and Hg. The spatial distribution of heavy metals was closely correlated with local anthropogenic activities, such as agricultural production, mining and transportation. The hot-spot areas of As and Cd were mainly concentrated in the up-midstream where were associated with agricultural activities. Cr and Cu showed similar spatial distributions with hot-spot areas distributed the whole irrigation area uniformly. Ni was mainly distributed in the downstream where Ni quarries concentrated, while the spatial distribution patterns of Hg was mainly located in the upstream and downstream where the soil was significantly influenced by irrigation and coal mining emission. The spatial distributions of Pb and Zn were mainly concentrated along the highway side. The pollution levels of Yongshuyu irrigation area were estimated through index of geo-accumulation(Igeo), Nemerow integrated pollution index(NIPI) and potential ecological risk index(PERI). The results showed that Cd and Hg were the main pollutants in the study area. Health risk assessment results indicated that children were in higher non-carcinogenic and carcinogenic risks than adults with the carcinogenic metal of As. Ingestion was the main exposure pathway to non-carcinogenic and carcinogenic risk for both adults and children. Principal component analysis(PCA) indicated that Cr and Cu were mainly from parent materials, while Cd and As were mainly affected by agricultural activities. Pb and Zn were controlled by traffic activities, and the accumulations of Ni and Hg were associated with mining activities. | 12 | Repeatability and Reproducibility of Quantitative Assessment of the Retinal Microvasculature Using Optical Coherence Tomography Angiography Based on Optical Microangiography显示文摘Objective The aim of this study was to determine the repeatability and reproducibility of optical coherence tomography angiography(OCTA) based on optical microangiography(OMAG) measurements of macular vessels in normal eyes. Methods In this prospective cohort study, 40 eyes of 40 healthy volunteers underwent repeated OCTA(Cirrus HD-OCT 5000 angiography system, Carl Zeiss Meditec, Inc.) scans on two separate visit days. On each visit day, the eyes were scanned three times. The following parameters were used to quantitatively describe the OCTA images of the superficial vascular network: vessel area density(VAD), vessel skeleton density(VSD), vessel diameter index(VDI), vessel perimeter index(VPI), vessel complexity index(VCI), flux, and foveal avascular zone(FAZ). Coefficient of variation(CV) and intraclass correlation coefficient(ICC) were calculated for evaluating intravisit and intervisit repeatability, as well as interobserver reproducibility. Results The measurements showed high repeatability [CVs ≤ 4.2%(intravisit) and ≤ 4.6%(intervisit)] and interobserver reproducibility(ICCs ≥ 0.923) for all parameters. | 14 | Donepezil,a drug for Alzheimer’s disease,promotes oligodendrocyte generation and remyelination显示文摘Myelin sheaths play important roles in neuronal functions.In the central nervous system(CNS),the myelin is formed by oligodendrocytes(OLs),which are differentiated from oligodendrocyte precursor cells(OPCs).In CNS demyelinating disorders such as multiple sclerosis(MS),the myelin sheaths are damaged and the remyelination process is hindered.Small molecule drugs that promote OPC to OL differentiation and remyelination may provide a new way to treat these demyelinating diseases.Here we report that donepezil,an acetylcholinesterase inhibitor(AChEI)developed for the treatment of Alzheimer's disease(AD);significantly promotes OPC to OL differentiation.Interestingly,other AChEls,including huperzine A,rivastigmine,and tacrine,have no such effect indicating that donepezil's effect in promoting OPC differentiation is not dependent on the inhibition of AChE.Donepezil also facilitates the formation of myelin sheaths in OPC-DRG neuron co-culture.More interestingly,donepezil also promotes the repair of the myelin sheaths in vivo and provides significant therapeutic effect in a cuprizone-mediated demyelination animal model.Donepezil is a drug that has been used to treat AD safely for many years;our findings suggest that it might be repurposed to treat CNS demyelinating diseases such as MS by promoting OPC to OL differentiation and remyelination. | Xue Cui Yu-e Guo Jia-hui Fang Chang-jie Shi Na Suo Ru Zhang Xin Xie | 2019 | Acta Pharmacologica Sinica2019,40,11: | 7 |
| 15 | The association between the genetic polymorphisms of LMP2/LMP7 and the outcomes of HCV infection among drug users显示文摘Objective:To investigate a possible association of LMP2/LMP7 genes with chronic hepatitis C virus(HCV) infection,and to assess whether LMP2/LMP7 genes could influence the outcomes of HCV infection among drug users.Methods:Genomic DNAs of 362 anti-HCV sero-positive drug users and 225 control drug users were extracted from the peripheral blood leukocytes.The sero-positive patients were divided into those who had persistent infection and those who had spontaneously cleared the infection.Polymorphisms of LMP genes were determined by PCR combined with restriction fragment length polymorphism(RFLP).Results:The distribution of LMP2 genotypes among the control,persistent infection and spontaneous clearance groups were not different.However,the LMP7 codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in control group(OR=1.75,95%CI=1.06~2.90;OR=3.16,95%CI=1.23-8.12;OR=1.94,95%CI=1.21-3.12,respectively).Similarly,the frequencies of the codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in the spontaneous clearance group(OR=1.64,95%CI=1.04-2.57;OR=2.40,95%CI=1.09-5.28;OR=1.76,95%CI=1.152.69,respectively).Stratified analysis indicated that combined genotype Gln/Lys+Lys/Lys of the LMP7 gene was related to an increasing susceptibility to HCV infection(OR=1.91,95%CI=1.02-3.55;OR=2.19,95%CI=1.243.89;OR=1.91,95%CI=1.05-3.48,OR=2.86,95%CI=1.41-5.78,respectively)and the risk of persistent HCV infection(OR=1.94,95%CI=1.12-3.34;OR=2.02,95%CI=1.21-3.38;OR=1.78,95%CI=1.12-2.85,OR=2.23,95%CI=1.09-4.58,respectively)among>30-year-old,males,the injection drug user(IDU)subjects and/or the shorter duration drug users(≤5 y).Conclusion:These results suggest that polymorphism of the LMP7 gene may have an influence on the outcomes of HCV infection,and is one of the factors accounting for the genetic susceptibility to HCV infection among drug users. | Qian Cui Yongxiang Zhang Jing Su Chao Shi Na Lei Keqin Ding Jun Li Rongbin Yu L u Wang Ning Wang | 2010 | The Journal of Biomedical Research2010,24,5: | 6 |
| 16 | Neuroprotective mechanisms of DNA methyltransferase in a mouse hippocampal neuronal cell line after hypoxic preconditioning显示文摘Hypoxic preconditioning has been shown to improve hypoxic tolerance in mice,accompanied by the downregulation of DNA methyltransferases(DNMTs)in the brain.However,the roles played by DNMTs in the multiple neuroprotective mechanisms associated with hypoxic preconditioning remain poorly understood.This study aimed to establish an in vitro model of hypoxic preconditioning,using a cultured mouse hippocampal neuronal cell line(HT22 cells),to examine the effects of DNMTs on the endogenous neuroprotective mechanisms that occur during hypoxic preconditioning.HT22 cells were divided into a control group,which received no exposure to hypoxia,a hypoxia group,which was exposed to hypoxia once,and a hypoxic preconditioning group,which was exposed to four cycles of hypoxia.To test the ability of hypoxic preadaptation to induce hypoxic tolerance,cell viability was measured using the 3-(4,5-dimethylthiazol-2-yl)-5(3-carboxymethonyphenol)-2-(4-sulfophenyl)-2H-tetrazolium assay.Cell viability improved in the hypoxic preconditioning group compared with that in the hypoxia group.The effects of hypoxic preconditioning on the cell cycle and apoptosis in HT22 cells were examined by western blot assay and flow cytometry.Compared with the hypoxia group,the expression levels of caspase-3 and spectrin,which are markers of early apoptosis and S-phase arrest,respectively,noticeably reduced in the hypoxic preconditioning group.Finally,enzyme-linked immunosorbent assay,real-time polymerase chain reaction,and western blot assay were used to investigate the changes in DNMT expression and activity during hypoxic preconditioning.The results showed that compared with the control group,hypoxic preconditioning downregulated the expression levels of DNMT3A and DNMT3B mRNA and protein in HT22 cells and decreased the activities of total DNMTs and DNMT3B.In conclusion,hypoxic preconditioning may exert anti-hypoxic neuroprotective effects,maintaining HT22 cell viability and inhibiting cell apoptosis.These neuroprotective mechanisms may be associated with the inhibition of DNMT3A and DNMT3B. | Na Liu Xiao-Lu Zhang Shu-Yuan Jiang Jing-Hua Shi Jun-He Cui Xiao-Lei Liu Li-Hong Han Ke-Rui Gong Shao-Chun Yan Wei Xie Chun-Yang Zhang Guo Shao | 2020 | Neural Regeneration Research2020,15,12: | 6 |

Real-time qPCR and immunohistochemistry were applied to detect dectin-1 expression in corneal epithelium of both groups. HCE cells were cultured with aspergillus fumigatus(AF) antigens in vitro. The expression of dectin-1 mRNA was measured by real-time qPCR at the stimulation of 0,4,8 and 24h separately. Dectin-1 protein was detected by immunocytochemistry at 0 and 24h separately. ·RESULTS: Dectin-1 expressed in corneal epithelium of normal persons and FK patients. Vitro cellular experiment showed that the expression of dectin-1 mRNA in HCE cells began to increase after stimulation of AF antigens at 4h,and dectin-1 protein expression increased after stimulation at 24h. · CONCLUSION: Dectin-1 expressed in corneal epithelium of normal persons. | 18 | Curcuminoids Target Decreasing Serum Adipocyte-fatty Acid Binding Protein Levels in Their Glucose-lowering Effect in Patients with Type 2 Diabetes显示文摘Whether supplementation of curcuminoids decreases serum adipocyte-fatty acid binding protein(A-FABP) level and whether this decrease benefits glucose control is unclear.One-hundred participants(n=50 administered curcuminoids,n=50 administered placebo) from our previous report on the effect of curcuminoids on type 2 diabetes in a 3-month intervention were assessed for levels of serum A-FABP,oxidative stress,and inflammatory biomarkers.Curcuminoids supplementation led to significant decreases in serum A-FABP,C-reactive protein(CRP),tumor necrosis factor-α,and interleukin-6 levels.Curcuminoids supplementation also significantly increased serum superoxide dismutase(SOD) activity.The change in serum A-FABP levels showed positive correlations with changes in levels of glucose,free fatty acids(FFAs),and CRP in subjects supplemented with curcuminoids.Further stepwise regression analysis showed that A-FABP was an independent predictor for levels of FFAs,SOD,and CRP.These results suggest that curcuminoids may exert anti-diabetic effects,at least in part,by reductions in serum A-FABP level.A-FABP reduction is associated with improved metabolic parameters in human type 2 diabetes. | NA Li Xin YAN Bo Lin JIANG Shuo CUI Hong Li LI Ying SUN Chang Hao | 2014 | Biomedical and Environmental Sciences2014,27,11: | 6 |
