1Genetics and Molecular Marker Identification of a Resistance to Glomerella Leaf Spot in Apple显示文摘Apple Glomerella leaf spot(GLS) is a destructive fungal disease that damages apple leaves during the summer in China.Breeding new diseaseresistant varieties is considered to be the best way of controlling GLS.A genetic study of resistance to Glomerella leaf spot(GLS) in apple was conducted by using four F_1 hybrid groups(‘Fuji' בGolden Delicious',‘Golden Delicious' בFuji',‘Gala' בFuji',and ‘Fuji' בQF-2') generated from two highly resistant varieties or selections,‘Fuji' and ‘QF-2',and two highly susceptible varieties,‘Golden Delicious' and ‘Gala'.The results showed that the separation ratios of resistant plants to the susceptible ones in the four F_1 hybrid groups were statistically consistent with the theoretical ratios of 1:1,1:1,0:1,and 1:0.Comprehensive analysis enabled us to generate the following conclusions:GLS resistance in apple may be controlled by a single recessive gene.The genotype of the resistant plants was rr,whereas the genotypes of the susceptible ones were RR and Rr.By using ‘Golden Delicious' בFuji' F_1 hybrid groups and the bulked segregation analysis(BSA) method,the marker S0506206-243 bp associated with disease resistance character to GLS was identified through screening 500 SSR primers encompassing the entire apple genome with even coverage,and the genetic distance between the marker and the GLS resistance gene was 9.8 cM.LIU Yuanxia LI Baohua WANG Caihong LIU Chunxiao KONG Xianghua ZHU Jun DAI Hongyi 2016Horticultural Plant Journal2016,2,3:6
2Impact of Asarum Essential Oils on Physiological and Biochemical Indicators of Two Agricultural Pests显示文摘[Objective]The paper was to study the effects of asarum essential oils on physiological and biochemical indices of two agricultural pests. [Method]Asarum essential oils were extracted by different methods,and their effects on physiological and biochemical indices related to the structure of body wall( chitin and phenoloxidase),the physiological and biochemical indices acting on the nervous system( Acetylcholinesterase),and the physiological and biochemical indices acting on the digestive system( protease,amylase and lipase) of Ostrinia furnacalis and Mythimna seperata were studied. [Result]Asarum essential oils had no effect no chitin content of body wall under the experimental concentration,and had no inhibitory effect but activated effect on polyphenol oxidase activity. Asarum essential oils could act on the nervous system of insect,and had strong inhibitory effect against acetylcholinesterase activity of O. furnacalis larvae,with inhibition rates of 24. 86%- 38. 79%. Asarum essential oils showed a significant activated effect on pepsin activity in midgut,but the activated amplitude became smaller with the prolongation of treatment time. Asarum essential oils showed a significant inhibitory effect on activities of amylase and lipase,indicating that it could play an insecticidal effect by inhibiting activity of amylase and lipase,and it could act on digestive system of insect. 3Evolution Behaviors of Oxides in Severely Plastic Deformed Region of AISI 52100 Steel during Dry Sliding Wear显示文摘Under dry sliding wear, the evolution of oxides in severely plastic deformed(SPD) regions of metals has a great impact on the wear behaviors. To study the evolution behaviors of oxides in the SPD region, an SPD region was prefabricated on the surface of AISI 52100 steel by supersonic ?ne particle bombarding(SFPB) treatment. Dry sliding wear tests were carried out on both of the SFPB-treated and original samples.Wear volume loss of the SPBF-treated samples were compared with those of the original samples at different loads. Microstructure, element composition and oxides distribution in the SPD region were characterized by scanning electron microscopy(SEM), transmission electron microscopy(TEM) and an electron probe microanalysis(EPMA). The results show that the evolution behaviors of the oxides in the SPD region change signi?cantly with the load. Under low loads, oxides are usually formed on the contact surface. It inhibits adhesive wear on the steel. However, under high loads, oxides are apt to distribute along the cracks in the subsurface layer. 8Identification of neutral genome integration sites with high expression and high integration efficiency in Fusarium venenatum TB01显示文摘CRISPR/Cas9-mediated homology-directed recombination is an efficient method to express target genes.Based on the above method,providing ideal neutral integration sites can ensure the reliable,stable,and high expression of target genes.In this study,we obtained a fluorescent transformant with neutral integration and high expression of the GFP expression cassette from the constructed GFP expression library and named strain FS.The integration site mapped at 4886 bp upstream of the gene FVRRES_00686 was identified in strain FS based on a Y-shaped adaptor-dependent extension,and the sequence containing 600 bp upstream and downstream of this site was selected as the candidate region for designing sgRNAs(Sites)for CRISPR/Cas9-mediated homology-directed recombination.PCR analysis showed that the integration efficiency of CRISPR/Cas9-mediated integration of target genes in designed sites reached 100%.Further expression stability and applicability analysis revealed that the integration of the target gene into the above designed sites can be stably inherited and expressed and has no negative effect on the growth of F.venenatum TB01.These results indicate the above designed neutral sites have the potential to accelerate the development of F.venenatum TB01 through overexpression of target genes in metabolic engineering.Sheng Tong Kexin An Wuxi Chen Mengdan Chai Yuanxia Sun Qinhong Wang Demao Li 2023Synthetic and Systems Biotechnology2023,8,1:0
9Effect of Cluster Nursing in Preventing Urinary Tract Infection in Postoperative Patients with Indwelling Urinary Catheter显示文摘Objective:This paper expounds and analyzes the effect of cluster nursing in preventing urinary tract infection in postoperative patients with indwelling urinary catheter.Methods:A total of 400 postoperative patients with indwelling urinary catheter,treated in Yancheng No.1 People’s Hospital in recent two years,were recruited as research subjects.The patients were equally divided into group A and group B,with 200 patients in each group,and all received routine anti-infectives.The patients in group A(200 cases)received routine nursing.On the basis of group A,cluster nursing was implemented for patients in group B(200 cases).Results:The indwelling time of urinary catheter,the disappearance time of bladder irritation symptoms,and the probability of urinary tract infection were compared between both groups,which showed that the nursing effect of group B was better than that of group A(P<0.05).Conclusion:The implementation of cluster nursing in postoperative patients with indwelling urinary catheter can improve the deficiency of routine nursing,consolidate the treatment effect,and reduce the complication rate of urinary tract infection.Longhui Zhang Yuanxia Wang Jianying Huang 2022Journal of Clinical and Nursing Research2022,6,2:0
10Effects of Astragalus membranaceus on Energy Metabolism and Expression of CNTF Protein in Skeletal Muscle of Exercise-induced Fatigue Rats显示文摘[Objectives]This study was conducted to investigate the effects of Astragalus membranaceus in different groups on energy metabolism and CNTF protein expression in skeletal muscle of exercise-induced fatigue rats.[Methods]Thirty-five clean male SD rats were randomly divided into a normal group,and low-,meddle-and high-dose groups of A.membranaceus aqueous solution,with 7 rats in each group.The low-dose,medium-dose and high-dose groups were given by gavage at 0.65,1.3 and 2.6 g/kg,respectively,while the normal group and the model group were given normal food and water.The weight of rats was observed.The contents of serum urea,lactate,muscle glycogen,liver glycogen and CNTF expression were detected.[Results]After modeling,compared with the normal group,the serum lactate and urea contents of rats in the model group significantly increased(P<0.01),while the muscle glycogen content(P<0.01)and liver glycogen content(P<0.05)of the skeletal muscle significantly decreased.Compared with the model group,the low-,meddle-and high-dose groups of A.membranaceus significantly reduced the levels of lactate and urea in serum(P<0.01),while the levels of muscle glycogen and liver glycogen in the skeletal muscle significantly increased(P<0.01,P<0.05).[Conclusions]This study provides a good research foundation for the treatment of exercise-induced fatigue using traditional Chinese herb A.membranaceus in modern clinical practice.Yueqi ZHOU Xue ZHANG Zelin ZHAO Yuanxia SHEN Li YANG Song WANG Junying TIAN Sibu MA Shiyan HUANG 2023Agricultural Biotechnology2023,12,6:0
