1Structures of EV71 RNA-dependent RNA polymerase in complex with substrate and analogue provide a drug target against the hand-foot-and-mouth disease pandemic in China显示文摘Enterovirus 71(EV71),one of the major causative agents for hand-foot-and-mouth disease(HFMD),has caused more than 100 deaths among Chinese children since March 2008.The EV71 genome encodes an RNAdependent RNA polymerase(RdRp),denoted 3D^(pol),which is central for viral genome replication and is a key target for the discovery of specific antiviral therapeutics.Here we report the crystal structures of EV71 RdRp(3D^(pol))and in complex with substrate guanosine-5'-triphosphate and analog 5-bromouridine-5'-triphosphate best to 2.4Åresolution.The structure of EV71 RdRp(3D^(pol))has a wider open thumb domain compared with the most closely related crystal structure of poliovirus RdRp.And the EV71 RdRp(3D^(pol))complex with GTP or Br-UTP bounded shows two distinct movements of the polymerase by substrate or analogue binding.The model of the complex with the template:primer derived by superimposition with foot-and-mouth disease virus(FMDV)3D/RNA complex reveals the likely recognition and binding of template:primer RNA by the polymerase.These results together provide a molecular basis for EV71 RNA replication and reveal a potential target for anti-EV71 drug discovery.Yang Wu Zhiyong Lou Yi Miao Yue Yu Hui Dong Wei Peng Mark Bartlam Xuemei Li Zihe Rao 2010Protein & Cell2010,1,5:30
2Meta-analyses on prevalence of selected Parkinson’s nonmotor symptoms before and after diagnosis显示文摘Background:Nonmotor symptoms are common among patients with Parkinson’s disease(PD)and some may precede disease diagnosis.Methods:We conducted a meta-analysis on the prevalence of selected nonmotor symptoms before and after PD diagnosis,using random-effect models.We searched PubMed(1965 through October/November 2012)for the following symptoms:hyposmia,constipation,rapid eye movement sleep behavior disorder,excessive daytime sleepiness,depression,and anxiety.Eligible studies were publications in English with original data on one or more of these symptoms.Results:The search generated 2,373 non-duplicated publications and 332 met the inclusion criteria,mostly(n=320)on symptoms after PD diagnosis.For all symptoms,the prevalence was substantially higher in PD cases than in controls,each affecting over a third of the patients.Hyposmia was the most prevalent(75.5%in cases vs.19.1%in controls),followed by constipation(50%vs.17.7%),anxiety(39.9%vs.19.1%),rapid eye movement sleep behavior disorder(37.0%vs.7.0%),depression(36.6%vs.14.9%),and excessive daytime sleepiness(33.9%vs.10.5%).We observed substantial heterogeneities across studies and meta-regression analyses suggested that several factors might have contributed to this.However,the prevalence estimates were fairly robust in several sensitivity analyses.Only 20 studies had data on any symptoms prior to PD diagnosis,but still the analyses revealed higher prevalence in future PD cases than in controls.Conclusion:These symptoms are common among PD patients both before and after diagnosis.Further studies are needed to understand the natural history of nonmotor symptoms in PD and their etiological and clinical implications.Honglei Chen Edward J Zhao Wen Zhang Yi Lu Rui Liu Xuemei Huang Anna J Ciesielski-Jones Michele A Justice Deborah S Cousins Shyamal Peddada 2015Translational Neurodegeneration2015,4,1:18
3Buyang Huanwu Decoction regulates neural stem cell behavior in ischemic brain显示文摘The traditional Chinese medicine Buyang Huanwu Decoction has been shown to improve the neurological function of patients with stroke.However,the precise mechanisms underlying its effect remain poorly understood.In this study,we established a rat model of cerebral ischemia by middle cerebral artery occlusion and intragastrically administered 5 g/kg Buyang Huanwu Decoction,once per day,for 1,7,14 and 28 days after cerebral ischemia.Immunohistochemical staining revealed a number of cells positive for the neural stem cell marker nestin in the cerebral cortex,the subventricular zone and the ipsilateral hippocampal dentate gyrus in rat models of cerebra ischemia.Buyang Huanwu Decoction significantly increased the number of cells positive for5-bromodeoxyuridine(BrdU),a cell proliferation-related marker,microtubule-associated protein-2,a marker of neuronal differentiation,and growth-associated protein 43,a marker of synaptic plasticity in the ischemic rat cerebral regions.The number of positive cells peaked at 14 and 28 days after intragastric administration of Buyang Huanwu Decoction.These findings suggest that Buyang Huanwu Decoction can promote the proliferation and differentiation of neural stem cells and enhance synaptic plasticity in ischemic rat brain tissue.Baiyan Liu Guangxian Cai Jian Yi Xuemei Chen 2013Neural Regeneration Research2013,8,25:17
4CMHX008,a PPAR γ partial agonist,enhances insulin sensitivity with minor influences on bone loss显示文摘Traditional thiazolidinediones(TZDs),such as rosiglitazone,are peroxisome proliferator-activated receptor g(PPARg)potent agonists that can be used to treat type 2 diabetes but carry unwanted effects,including increased risk for fracture.The present work aimed to compare the insulin-sensitizing efficacies and bone-loss side effects of CMHX008,a novel TZDs-like PPARg partial agonist,with those of rosiglitazone.A TR-FRET PPARg competitive binding assay was used to compare the binding affinity between CMHX008 and rosiglitazone.Mice were administered vehicle,CMHX008 or rosiglitazone for 16 weeks.Mesenchymal stem cells(MSCs)were used to examine differences in differentiation into osteoblasts after compounds treatment.TR-FRET showed lower affinity to PPARg by CMHX008 compared with rosiglitazone.Mice treated with CMHX008 showed insulin sensitization similar to that of mice treated with rosiglitazone,which was related to the significant inhibition of PPARg Ser273 phosphorylation and improved insulin sensitivity by facilitating the phosphorylation of insulin receptor and Akt in adipose tissues.Micro-CT and histomorphometric analyses demonstrated that the degree of trabecular bone loss after treatment with CMHX008 was weaker than that observed with rosiglitazone,as evidenced by consistent changes in BV/TV,Tb.N,Tb.Th,Tb.Sp,and the mineral apposition rate.MSCs treated with CMHX008 showed higher ALP activity and mRNA levels of bone formation markers than did cells treated with rosiglitazone in the osteoblast differentiation test.Thus,CMHX008 showed insulin-sensitizing effects similar to those of rosiglitazone with a lower risk of bone loss,suggesting that PPARg sparing eliminates the skeletal side effects of TZDs while maintaining their insulin-sensitizing properties.Yi Hou Xuemei Cao Xiangnan Hu Xinyu Li Xiaoqin Shi Hongying Wang Chuan Peng Jiayu Li Jibin Li Qifu Li Chaodong Wu Xiaoqiu Xiao 2018Genes & Diseases2018,5,3:6
5Investigation of microstructure changes in Al_(2)O_(3)-YSZ coatings and YSZ coatings and their effect on thermal cycle life显示文摘Yttria-stabilized zirconia(YSZ)coatings and Al_(2)O_(3)–YSZ coatings were prepared by atmospheric plasma spraying(APS).Their microstructural changes during thermal cycling were investigated via scanning electron microscopy(SEM)equipped with electron backscatter diffraction(EBSD)and X-ray diffraction(XRD).It was found that the microstructure and microstructure changes of the two coatings were different,including crystallinity,grain orientation,phase,and phase transition.These differences are closely related to the thermal cycle life of the coatings.There is a relationship between crystallinity and crack size.Changes in grain orientation are related to microscopic strain and cracks.Phase transition is the direct cause of coating failure.In this study,the relationship between the changes in the coating microstructure and the thermal cycle life is discussed in detail.The failure mechanism of the coating was comprehensively analyzed from a microscopic perspective.Meiqi DAI Xuemei SONG Chucheng LIN Ziwei LIU Wei ZHENG Yi ZENG 2022Journal of Advanced Ceramics2022,11,2:4
6Impact of Asarum Essential Oils on Physiological and Biochemical Indicators of Two Agricultural Pests显示文摘[Objective]The paper was to study the effects of asarum essential oils on physiological and biochemical indices of two agricultural pests. [Method]Asarum essential oils were extracted by different methods,and their effects on physiological and biochemical indices related to the structure of body wall( chitin and phenoloxidase),the physiological and biochemical indices acting on the nervous system( Acetylcholinesterase),and the physiological and biochemical indices acting on the digestive system( protease,amylase and lipase) of Ostrinia furnacalis and Mythimna seperata were studied. [Result]Asarum essential oils had no effect no chitin content of body wall under the experimental concentration,and had no inhibitory effect but activated effect on polyphenol oxidase activity. Asarum essential oils could act on the nervous system of insect,and had strong inhibitory effect against acetylcholinesterase activity of O. furnacalis larvae,with inhibition rates of 24. 86%- 38. 79%. Asarum essential oils showed a significant activated effect on pepsin activity in midgut,but the activated amplitude became smaller with the prolongation of treatment time. Asarum essential oils showed a significant inhibitory effect on activities of amylase and lipase,indicating that it could play an insecticidal effect by inhibiting activity of amylase and lipase,and it could act on digestive system of insect. 8Are current Chinese national ambient air quality standards on 24-hour averages for particulate matter sufficient to protect public health?显示文摘With rapid economic development and urbanization in recent decades, China has experienced the worsening of ambient air quality. For better air quality management to protect human health, Chinese government revised national ambient air quality standards(NAAQS) for particulate matter(PM) in 2012(GB3095-2012). To assess the effectiveness of current NAAQS for PM on public health in Chinese population, we conducted a metaanalysis on published studies examining the mortality risk of short-term exposure to PM with aerodynamic diameters less than 10 and 2.5 μm(PM_(10) and PM_(2.5)) in China. The reported24-hour concentrations of PM_(10) and PM_(2.5) in studies ranged from 43.5 to 150.1 μg/m^3 and 37.5 to 176.7 μg/m^3. In the pooled excess, mortality risk estimates of short-term exposure to PM.In specific, per 10 μg/m^3 increase in PM_(10), we observed increases of 0.40%(95%CI: 0.33%,0.47%), 0.57%(95%CI: 0.44%, 0.70%) and 0.49%(95%CI: 0.40%, 0.58%) in total, respiratory and cardiovascular mortality, per 10 μg/m^3 increase in PM_(2.5), we observed increases of 0.51%(95%CI: 0.38%, 0.63%), 0.62%(95%CI: 0.52%, 0.73%) and 0.75%(95%CI: 0.54%, 0.95%) in total,respiratory and cardiovascular mortality. Finally, we derived 125 μg/m^3 for PM_(10) and 62.5 μg/m^3 for PM_(2.5) as 24-hour recommendation values based on the pooled estimates. Our results indicated that current Chinese NAAQS for PM could be sufficient in mitigating the excess mortality risk from short-term exposure to ambient PM. 9Dry-regulated hydrogels with anisotropic mechanical performance and ionic conductivity显示文摘Nature consists of various soft tissues with well-ordered hierarchical anisotropic structures, which play essential roles in biological systems to exhibit particular functions. Mimicking bio-tissues, synthetic hydrogels with anisotropic structures have received considerable attention in recent years. However, existing approaches to fabricate anisotropic hydrogels often require complicated procedures, which are timeconsuming and labor-demanding. Inspired by the dry-induced crystallization phenomenon, we report a simple yet effective prestretching-drying-swelling method to afford anisotropic crystalline polyvinyl alcohol hydrogels. Owing to the distinct anisotropic microstructure, the hydrogels demonstrate excellent mechanical properties with noticeable directional distinction. It is revealed that both the enhancing of pre-orientation strain and the extending of heating time make the hydrogels with better mechanical properties and more remarkable anisotropicity. Owing to the anisotropically aligned structure, the hydrogels exhibit remarkably differential ionic conductivity: the difference between the parallel and vertical conductivity of the same sample can reach as high as 6.6 times, making the materials possible candidates as nano-conductive materials. 12Salt and pepper noise removal in surveillance video based on low-rank matrix recovery显示文摘This paper proposes a new algorithm based on low-rank matrix recovery to remove salt &pepper noise from surveillance video. Unlike single image denoising techniques, noise removal from video sequences aims to utilize both temporal and spatial information. By grouping neighboring frames based on similarities of the whole images in the temporal domain, we formulate the problem of removing salt &pepper noise from a video tracking sequence as a lowrank matrix recovery problem. The resulting nuclear norm and L1-norm related minimization problems can be efficiently solved by many recently developed methods. To determine the low-rank matrix, we use an averaging method based on other similar images. Our method can not only remove noise but also preserve edges and details. 13OTUB1 promotes osteoblastic bone formation through stabilizing FGFR2显示文摘Bone homeostasis is maintained by the balance between osteoblastic bone formation and osteoclastic bone resorption.Dysregulation of this process leads to multiple diseases,including osteoporosis.However,the underlying molecular mechanisms are not fully understood.Here,we show that the global and conditional osteoblast knockout of a deubiquitinase Otub1 result in low bone mass and poor bone strength due to defects in osteogenic differentiation and mineralization.Mechanistically,the stability of FGFR2,a crucial regulator of osteogenesis,is maintained by OTUB1.OTUB1 attenuates the E3 ligase SMURF1-mediated FGFR2 ubiquitination by inhibiting SMURF1’s E2 binding.In the absence of OTUB1,FGFR2 is ubiquitinated excessively by SMURF1,followed by lysosomal degradation.Consistently,adeno-associated virus serotype 9(AAV9)-delivered FGFR2 in knee joints rescued the bone mass loss in osteoblast-specific Otub1-deleted mice.Moreover,Otub1 mRNA level was significantly downregulated in bones from osteoporotic mice,and restoring OTUB1 levels through an AAV9-delivered system in ovariectomy-induced osteoporotic mice attenuated osteopenia.Taken together,our results suggest that OTUB1 positively regulates osteogenic differentiation and mineralization in bone homeostasis by controlling FGFR2 stability,which provides an optical therapeutic strategy to alleviate osteoporosis.Qiong Zhu Yesheng Fu Chun-Ping Cui Yi Ding Zhikang Deng Chao Ning Fan Hu Chen Qiu Biyue Yu Xuemei Zhou Guan Yang Jiang Peng Weiguo Zou Cui Hua Liu Lingqiang Zhang 2023Signal Transduction and Targeted Therapy2023,8,5:1
