1Qingnaoyizhi decoction suppresses the formation of glial fibrillary acidic protein-positive cells in cultured neural stem cells by inhibiting the Janus kinase 2/signal transducer and activator of transcription 3 signaling pathway显示文摘OBJECTIVE: Inactivation of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) signaling axis plays a crucial role in determining the fate of neural stem cells(NSCs).Qingnaoyizhi decoction(QNYZD) has been used for the treatment of vascular dementia and has shown to improve synaptic remodeling. The aim of this study was to evaluate the effect of cerebrospinal fluid(CSF) containing QNYZD(CSF-QNYZD) on the differentiation of cultured NSCs and the involvement of the JAK2/STAT3 pathway.METHODS: The protein expression levels of glial fibrillary acidic protein(GFAP), tubulin, drosophila mothers against decapentaplegic protein(SMAD-1), STAT3, and phosphorylated-STAT3 were detected by western immunoblot analysis in the groups: control, CSF, JAK/STAT inhibitor(AG490),CSF-QNYZD, and CSF-XDZ(CSF-Xidezhen). The differentiation of NSCs was determined by immunofluorescence staining. The proliferation of NSCs was measured using the Cell Counting Kit-8 proliferation assay.RESULTS: Compared with the control group,CSF-QNYZD and AG490 significantly increased the number and expression of tubulin-positive cells, reduced the number and expression of GFAP-positive cells, and down-regulated the expression of p-STAT3. However, CSF-QNYZD also decreased the expression of SMAD-1 and STAT3.CONCLUSION: Enhanced neuronal differentiation may be associated with the down-regulation of glial differentiation instead of promoting proliferationin treated NSCs. Furthermore, QNYZD may play a direct role in suppressing the formation of GFAP-positive cells and enhancing neuronal differentiation by inhibiting JAK2/STAT3 activation. 2Axonal autophagy during regeneration of the rat sciatic nerve显示文摘BACKGROUND: The removal of degenerated axonal debris during Wallerian degeneration is very important for nerve regeneration. However, the mechanism by which debris is removed is not been completely understood. Considerable controversy remains as to the clearance pathway and cells that are involved. OBJECTIVE: To investigate axonal autophagy during removal of degenerated axonal debris by transecting the sciatic nerve in a rat Wallerian degeneration model. DESIGN, TIME AND SETTING: Experimental neuropathological analysis. The experiment was conducted at the Laboratory Animal Service Center of the Southern Medical University between January and June 2005. MATERIALS: Fifty-four adult, Wistar rats of either sex, weighing 180-250 g, were obtained from the Laboratory Animal Service Center of the Southern Medical University. Animals were randomly divided into nine groups of six rats. METHODS: Wallerian degeneration was induced by transecting the rat sciatic nerve, and tissue samples from the distal stump were obtained 0.2, 0.4, 1, 2, 3, 4, 7, 10, and 15 days post-transection. Ultrathin sections were prepared for electron microscopy to study ultrastructure and enzyme cytochemistry staining. MAIN OUTCOME MEASURES: Ultrastructure (axon body, autophagic body, and cystoskeleton) of axons and myelin sheaths observed with electron microscopy; acidic phosphatase activity detected by Gomori staining using electron microscopy. RESULTS: The major changes of degenerating axons after transection were axoplasm swelling and separation of axons from their myelin sheath between five hours and two days post-transection. At four days post-transection, the axoplasm condensed and axons were completely separated from the myelin sheath, forming dissociative axon bodies. Vacuoles of different sizes formed in axons during the early phase after lesion. Larger dissociative axon bodies were formed when the axons were completely separated from the myelin sheath during a late phase. The axolemma surrounding the axon body was derived from the neuronal cell membrane; the condensed axoplasm contained many autophagic vacuoles at all levels. A large number of neurofilaments, microtubules, and microfilaments were arranged in a criss-cross pattern. The autophagic vacuoles exhibited acidic phosphatase activity. Axonal bodies were absorbed after degradation from day 7 onwards, and macrophages were observed rarely in the formative cavity. 14Different Burial-Cooling History of Triassic Strata between the Western Weibei Uplift and the Northwestern Weihe Basin in Northwest China显示文摘Analysis of tectonothermal history of the Yanchang Formation in the western Weibei Uplift and in the northwestern Weihe Basin can reconstruct the cooling history of the southwest most remained Upper Triassic source rock of the North China Plate. Apatite fission-track(AFT) and(U-ThSm)/He(AHe) analysis were used to recover the cooling and uplift history of the Upper Triassic here. Ten sandstones from the Middle–Upper Triassic strata yield AFT ages between 179.8 ± 7.4 and 127.6 ± 8.1 Ma. AHe ages of two sandstones have the value of 37.7 ± 2.3–131.1 ± 8.1 and 45.7 ± 2.8–83.5 ± 5.2 Ma. Time-temperature modeling results showed that tectonothermal history of the Yanchang Formation was initially different in time-space relationships but then became almost identical through time followed by different cooling rate. Modeling results of the Triassic strata in the Qianyang area and the Yaojiagou area revealed three different uplift-cooling stages commencing in the Late Jurassic at ~165 Ma and in Early Cretaceous at ~110 Ma, respectively, both followed by first similar cooling histories to the Early Miocene at ~20 – 23 Ma and then different since the Late Miocene. Uplift-cooling rate since the Late Miocene at ~8 Ma were different between the Western Weibei Uplift and the Northwestern Weihe Basin. The timing, cooling-uplift rates of the Yaojiagou area, which was mainly controlled by movements related to the Liupanshan Mountains, the Qinling Orogens and the Weibei Uplift, had the earliest onset of uplift-cooling for the Upper Triassic series compared to other regions within the Weibei Uplift.
