| 1 | Abiogenic hydrocarbons in commercial gases from the Songliao Basin,China显示文摘This paper discusses the kinetic fractionation, composition and distribution characteristics of carbon and hydrogen isotopes for various alkane gases formed in different environments, by different mecha- nisms and from different sources in nature. It is demonstrated that the biodegradation or thermode- gradation of complex high-molecule sedimentary organic material can form microbial gas or thermogenic gas. The δ 13C1 value ranges from -110‰ to -50‰ for microbial gases but from -50‰ to -35‰ (even heavier) for thermogenic gases. Controlled by the kinetic isotope fractionation, both microbial and thermogenic gases have δ 13C and δ D values characterized by normal distribution, i.e. δ 13C1< δ 13C2< δ 13C3< δ 13C4 and δ DCH4< δ DC2H6< δ DC3H8<δ DC4H10, and by a positive correlation between the δ 13C and δ D values. Simple carbonbearing molecules (CH4, CO and CO2) can form abiogenic alkane gases via polymerization in the abiological chemical process in nature, with δ 13C1 heavier than -30‰. Moreover, controlled by the kinetic isotope fractionation, abiogenic alkane gases are characterized by a reverse distribution of δ 13C values and a normal trend of δ D values, namely δ 13C1> δ 13C2> δ 13C3> δ 13C4 and δ DCH4<δ DC2H6< δ DC3H8< δ DC4H10. The δ 13C values and δ D values are negatively correlated. Natural gases from 26 commercial gas wells distributed in the Xujiaweizi and Yingshan-Miaotaizi faulted depressions in the Songliao Basin, China, show δ13C1 values ranging from -30.5‰ to -16.7‰ with a very narrow δ D range between -203‰―-196‰. These gases are characterized by a reverse distribution of δ 13C values but a normal distribution of δ D values, and a negative correlation between their δ 13C and δ D values, indicating an abiological origin. The present study has revealed that abiogenic hydrocarbons not only exist in nature but also can make significant contribution to commercial gas reserviors. It is estimated that the reserve volume of alkane gases with abiogenic characteristics in these 26 gas wells in the Songliao Basin is over 500×108 m3. | 3 | Axonal autophagy during regeneration of the rat sciatic nerve显示文摘BACKGROUND: The removal of degenerated axonal debris during Wallerian degeneration is very important for nerve regeneration. However, the mechanism by which debris is removed is not been completely understood. Considerable controversy remains as to the clearance pathway and cells that are involved. OBJECTIVE: To investigate axonal autophagy during removal of degenerated axonal debris by transecting the sciatic nerve in a rat Wallerian degeneration model. DESIGN, TIME AND SETTING: Experimental neuropathological analysis. The experiment was conducted at the Laboratory Animal Service Center of the Southern Medical University between January and June 2005. MATERIALS: Fifty-four adult, Wistar rats of either sex, weighing 180-250 g, were obtained from the Laboratory Animal Service Center of the Southern Medical University. Animals were randomly divided into nine groups of six rats. METHODS: Wallerian degeneration was induced by transecting the rat sciatic nerve, and tissue samples from the distal stump were obtained 0.2, 0.4, 1, 2, 3, 4, 7, 10, and 15 days post-transection. Ultrathin sections were prepared for electron microscopy to study ultrastructure and enzyme cytochemistry staining. MAIN OUTCOME MEASURES: Ultrastructure (axon body, autophagic body, and cystoskeleton) of axons and myelin sheaths observed with electron microscopy; acidic phosphatase activity detected by Gomori staining using electron microscopy. RESULTS: The major changes of degenerating axons after transection were axoplasm swelling and separation of axons from their myelin sheath between five hours and two days post-transection. At four days post-transection, the axoplasm condensed and axons were completely separated from the myelin sheath, forming dissociative axon bodies. Vacuoles of different sizes formed in axons during the early phase after lesion. Larger dissociative axon bodies were formed when the axons were completely separated from the myelin sheath during a late phase. The axolemma surrounding the axon body was derived from the neuronal cell membrane; the condensed axoplasm contained many autophagic vacuoles at all levels. A large number of neurofilaments, microtubules, and microfilaments were arranged in a criss-cross pattern. The autophagic vacuoles exhibited acidic phosphatase activity. Axonal bodies were absorbed after degradation from day 7 onwards, and macrophages were observed rarely in the formative cavity. | 6 | Functional analysis of the emerging roles for the KISS1/KISS1R signaling pathway in cancer metastasis显示文摘Cancer metastasis, a process that primary tumor cells disseminate to secondary organs, is the most lethal and least effectively treated characteristic of human cancers. Kisspeptins are proteins encoded by the KISS1 gene that was originally described as a melanoma metastasis suppressor gene. Then, Kisspeptins were discovered as the natural ligands of the G-protein-coupled receptor 54(GPR54) that is also called KISS1R. The KISS1/KISS1R signaling is essential to control Gn RH secretion during puberty and to establish mammalian reproductive function through the hypothalamic-pituitary-gonadal(HPG) axis. Although KISS1primarily plays a suppressive role in the metastasis progression in several cancer types, emerging evidence indicates that the physiological effect of KISS1/KISS1R in cancer metastasis is tissue context-dependent and still controversial. | 7 | The enhanced genomic 6mA metabolism contributes to the proliferation and migration of TSCC cells显示文摘In contrast to the well-established genomic 5-methylcytosine(5mC),the existence of N^(6)-methyladenine(6 mA)in eukaryotic genomes was discovered only recently.Initial studies found that it was actively regulated in cancer cells,suggesting its involvement in the process of carcinogenesis.However,the contribution of 6 mA in tongue squamous cell carcinoma(TSCC)still remains uncharacterized.In this study,a pan-cancer type analysis was first performed,which revealed enhanced 6mA metabolism in diverse cancer types.The study was then focused on the regulation of 6 mA metabolism,as well as its effects on TSCC cells.To these aspects,genome 6mA level was found greatly increased in TSCC tissues and cultured cells.By knocking down 6mA methylases N6AMT1 and METTL4,the level of genomic 6mA was decreased in TSCC cells.This led to suppressed colony formation and cell migration.By contrast,knockdown of 6mA demethylase ALKBH1 resulted in an increased 6 mA level,enhanced colony formation,and cell migration.Further study suggested that regulation of the NF-κB pathway might contribute to the enhanced migration of TSCC cells.Therefore,in the case of TSCC,we have shown that genomic 6mA modification is involved in the proliferation and migration of cancer cells. | Lei Xi Ying Yang Ying Xu Fangming Zhang Jinghui Li Xiyang Liu Zhenxi Zhang Quan Du | 2022 | International Journal of Oral Science2022,14,1: | 0 |

| 9 | Scattering Inversion Study for Suspended Label-Free Lymphocytes with Complex Fine Structures显示文摘Objective and Impact Statement.Distinguishing malignant lymphocytes from normal ones is vital in pathological examination.We proposed an inverse light scattering(ILS)method for label-free suspended lymphocytes with complex fine structures to identify their volumes for pathological state.Introduction.Light scattering as cell’s“fingerprint”provides valuable morphology information closely related to its biophysical states.However,the detail relationships between the morphology with complex fine structures and its scattering characters are not fully understood.Methods.To quantitatively inverse the volumes of membrane and nucleus as the main scatterers,clinical lymphocyte morphologies were modeled combining the Gaussian random sphere geometry algorithm by 750 reconstructed results after confocal scanning,which allowed the accurate simulation to solve ILS problem.For complex fine structures,the specificity for ILS study was firstly discussed(to our knowledge)considering the differences of not only surface roughness,posture,but also the ratio of nucleus to the cytoplasm and refractive index.Results.The volumes of membrane and nucleus were proved theoretically to have good linear relationship with the effective area and entropy of forward scattering images.Their specificity deviations were less than 3.5%.Then,our experimental results for microsphere and clinical leukocytes showed the Pearson product-moment correlation coefficients(PPMCC)of this linear relationship were up to 0.9830~0.9926.Conclusion.Our scattering inversion method could be effectively applied to identify suspended label-free lymphocytes without destructive sample pretreatments and complex experimental systems. | Lu Zhang Huijun Wang Jianyi Liu Shuang Chen He Yang Zewen Yang Zhenxi Zhang Hong Zhao Li Yuan Lifang Tian Bo Zhong Xiaolong Liu | 2022 | Biomedical Engineering Frontiers2022,3,1: | 0 |
