1Blockade of Tim-3 Pathway Ameliorates Interferon-γ Production from Hepatic CD8^+ T Cells in a Mouse Model of Hepatitis B Virus Infection显示文摘T cell immunoglobulin-and mucin-domain-containing molecule-3(Tim-3) has been reported to participate in the pathogenesis of inflammatory diseases. However,whether Tim-3 is involved in hepatitis B virus(HBV) infection remains unknown. Here,we studied the expression and function of Tim-3 in a hydrodynamics-based mouse model of HBV infection. A significant increase of Tim-3 expression on hepatic T lymphocytes,especially on CD8+ T cells,was demonstrated in HBV model mice from day 7 to day 18. After Tim-3 knockdown by specific shRNAs,significantly increased IFN-γ production from hepatic CD8+ T cells in HBV model mice was observed. Very interestingly,we found Tim-3 expression on CD8+ T cells was higher in HBV model mice with higher serum anti-HBs production. Moreover,Tim-3 knockdown influenced anti-HBs production in vivo. 4Identification and Fine Mapping of a Gene Related to Pale Green Leaf Phenotype near the Centromere Region in Rice(Oryza sativa)显示文摘A thermo-insensitive pale green leaf mutant (pgl2) was isolated from T-DNA inserted transgenic lines of rice (Oryza sativa L. subsp. japonica cv. Nipponbare). Genetic analysis indicated that the phenotype was caused by a recessive mutation in a single nuclear-encoded gene. To map the PGL2 gene, an F2 population was constructed by crossing the mutant with Longtefu (Oryza sativa L. subsp. indica). The PGL2 locus was roughly linked to SSR marker RM331 on chromosome 8. To finely map the gene, 14 new InDel markers were developed around the marker, and PGL2 was further mapped to a 2.37 Mb centromeric region. Analysis on chlorophyll contents of leaves showed that there was no obvious difference between the mutant and the wild type in total chlorophyll (Chl) content, while the ratio of Chl a / Chl b in the mutant was only about 1, which was distinctly lower than that in the wild type, suggesting that the PGL2 gene was related to the conversion between Chl a and Chl b. 5Triptolide, A Potential Autophagy Modulator显示文摘As a major active component extracted from traditional Chinese herb Tripterygium wilfordii Hook F, triptolide exhibits multiple pharmacological effects. Autophagy is an evolutionary conserved intracellular catabolic process involved in cytoplasmic materials degradation. Autophagic dysfunction contributes to the pathologies of many human diseases, which makes it a promising therapeutic target. Recent studies have shown that triptolide exerts neuroprotection, anti-tumor activities, organ toxicity, and podocyte protection by modulating autophagy. 7Aggregate-associated changes in nutrient properties,microbial community and functions in a greenhouse vegetable field based on an eight-year fertilization experiment of China显示文摘Soil aggregation,microbial community,and functions(i.e.,extracellular enzyme activities;EEAs)are critical factors affecting soil C dynamics and nutrient cycling.We assessed soil aggregate distribution,stability,nutrients,and microbial characteristics within>2,0.25-2,0.053-0.25,and<0.053 mm aggregates,based on an eight-year field experiment in a greenhouse vegetable field in China.The field experiment includes four treatments:100%N fertilizer(CF),50%substitution of N frtilizer with manure(M),straw(S),and manure plus straw(MS).The amounts of nutrient(N,P20,and K20)input were equal in each treatment.Results showed higher values of mean weight diameter in organic amended soils(M,MS,and S,2.43-2.97)vs.CF-amended soils(1.99).Relative to CF treatment,organic amendments had positive effects on nutrient(i.e.,available N,P,and soil organic C(SOC))conditions,microbial(e.g,bacterial and fungal)growth,and EEAs in the>0.053 mm aggregates,but not in the<0.053 mm aggregates.The 0.25-0.053 mm aggregates exhibited better nutrient conditions and hydrolytic activity,while the<0.053 mm aggregates had poor nutrient conditions and higher oxidative activity among aggregates,per SOC,available N,available P,and a series of enzyme activities.These results indicated that the 0.25-0.053 mm(<0.053 mm)aggregates provide suitable microhabitats for hydrolytic(oxidative)activity.Interestingly,we found that hydrolytic and oxidative activities were mainly impacted by fertilization(58.5%,P<0.01)and aggregate fractions(50.5%,P<0.01),respectively.The hydrolytic and oxidative activities were significantly(P<0.01)associated with nutrients(SOC and available N)and pH,electrical conductivity,respectively.Furthermore,SOC,available N,and available P closely(P<0.05)afected microbial communities within>0.25,0.25-0.053,and<0.053 mm aggregates,respectively.These findings provide several insights into microbial characteristics within aggregates under dfferent frilization modes in the greenhouse vegetable production system in China.LUAN Hao-an GAO Wei TANG Ji-wei LI Ruo-nan LI Ming-yue ZHANG Huai-zhi CHEN Xin-ping Dainius MASILIUNAS HUANG Shao-wen 2020Journal of Integrative Agriculture2020,19,10:7

Some theorems and lemmas are shown in order to give procedures for generating maximally consistent subsets. Then a complete procedure and an incomplete procedure for generating the maximal consistent subsets are presented, and the correctness of the procedures is also shown. Furthermore, a way to implement knowledge base revision is presented, and a prototype system is introduced. 9Biofilm-inspired adhesive and antibacterial hydrogel with tough tissue integration performance for sealing hemostasis and wound healing显示文摘Uncontrolled bleeding and infection can cause significant increases in mortalities.Hydrogel sealants have attracted extensive attention for their ability to control bleeding.However,because interfacial water is a formidable barrier to strong surface bonding,a challenge remains in finding a product that offers robust tissue adhesion combined with anti-infection properties.Inspired by the strong adhesive mechanism of biofilm and mussels,we report a novel dual bionic adhesive hydrogel(DBAH)based on chitosan grafted with methacrylate(CS-MA),dopamine(DA),and N-hydroxymethyl acrylamide(NMA)via a facile radical polymerization process.CS-MA and DA were simultaneously included in the adhesive polymer for imitating the two key adhesive components:polysaccharide intercellular adhesin(PIA)of staphylococci biofilm and 3,4-dihydroxy-L-phenylalanine(Dopa)of mussel foot protein,respectively.DBAH presented strong adhesion at 34 kPa even upon three cycles of full immersion in water and was able to withstand up to 168 mm Hg blood pressure,which is significantly higher than the 60–160 mm Hg measured in most clinical settings.Most importantly,these hydrogels presented outstanding hemostatic capability under wet and dynamic in vivo movements while displaying excellent antibacterial properties and biocompatibility.Therefore,DBAH represents a promising class of biomaterials for high-efficiency hemostasis and wound healing.Wei Han Bo Zhou Kai Yang Xin Xiong Shifang Luan Yu Wang Zheng Xu Peng Lei Zhengshan Luo Jian Gao Yijing Zhan Guopu Chen Lei Liang Rui Wang Sha Li Hong Xu 2020Bioactive Materials2020,5,4:6
10Effects of a decade of organic fertilizer substitution on vegetable yield and soil phosphorus pools, phosphatase activities, and the microbial community in a greenhouse vegetable production system显示文摘Partial substitution of chemical fertilizers by organic amendments is adopted widely for promoting the availability of soil phosphorus(P)in agricultural production.However,few studies have comprehensively evaluated the effects of longterm organic substitution on soil P availability and microbial activity in greenhouse vegetable fields.A 10-year(2009–2019)field experiment was carried out to investigate the impacts of organic fertilizer substitution on soil P pools,phosphatase activities and the microbial community,and identify factors that regulate these soil P transformation characteristics.Four treatments included 100%chemical N fertilizer(4 CN),50%substitution of chemical N by manure(2 CN+2 MN),straw(2 CN+2 SN),and combined manure with straw(2 CN+1 MN+1 SN).Compared with the 4 CN treatment,organic substitution treatments increased celery and tomato yields by 6.9-13.8%and 8.6-18.1%,respectively,with the highest yields being in the 2 CN+1 MN+1 SN treatment.After 10 years of fertilization,organic substitution treatments reduced total P and inorganic P accumulation,increased the concentrations of available P,organic P,and microbial biomass P,and promoted phosphatase activities(alkaline and acid phosphomonoesterase,phosphodiesterase,and phytase)and microbial growth in comparison with the 4 CN treatment.Further,organic substitution treatments significantly increased soil C/P,and the partial least squares path model(PLS-PM)revealed that the soil C/P ratio directly and significantly affected phosphatase activities and the microbial biomass and positively influenced soil P pools and vegetable yield.Partial least squares(PLS)regression demonstrated that arbuscular mycorrhizal fungi positively affected phosphatase activities.Our results suggest that organic fertilizer substitution can promote soil P transformation and availability.Combining manure with straw was more effective than applying these materials separately for developing sustainable P management practices.ZHANG Yin-jie GAO Wei LUAN Hao-an TANG Ji-wei LI Ruo-nan LI Ming-yue ZHANG Huai-zhi HUANG Shao-wen 2022Journal of Integrative Agriculture2022,21,7:5

12Investigation on Precooling Effects of 4 K Stirling-Type Pulse Tube Cryocoolers显示文摘Stirling-type pulse tube cryocoolers(SPTCs)working at liquid-helium temperatures are appealing in space applications because of their promising advantages such as high reliability,compactness,etc.Worldwide efforts have been put in to develop SPTCs operating at liquid-helium temperatures especially with helium-4 as the working fluid.Staged structure is essential to reach such low temperatures.Generally,both the regenerator of the last section and the pulse tube together with the phase shifter are precooled by its upper stage or by external cold source to a low temperature of around 20 K.However,the precooling effects on the regenerator and the pulse tube are synthetic in previous studies,and their independent effects have not been studied clearly.In this manuscript,the precooling effects on the regenerator and on the pulse tube together with the phase shifter are tested independently on a unique-designed precooled SPTC.The tested precooling temperature is between 13.3 K and 22 K,and the no-load refrigeration temperature gets down to 3.6 K.Further analyses and numerical calculations have been carried out.It is found that the influence on the regenerator is remarkable,which is different from previous conclusions.It is also found that the precooling effects on the pulse tube are relatively weak because of the large pressure-induced enthalpy flow of a real gas working at the temperatures near to the critical point.Furthermore,the phase shifting capacity is analyzed with two cases and with both helium-4 and helium-3 as working fluids,and it keeps quite constant after optimizing the frequency and the precooling temperature for each case.The investigation on these independent effects will provide valid reference on the precooling mechanism study of SPTCs working down to liquid-helium temperatures.CAO Qiang LI Zimu LUAN Mingkai SUN Zheng TANG Xiao LI Peng JIANG Zhenhua WEI Li 2019Journal of Thermal Science2019,28,4:4
13Ablation characteristics of mosaic structure ZrC-SiC coatings on low-density, porous C/C composites显示文摘Mosaic structure ZrC-SiC coatings were fabricated on low-density, porous C/C composites via thermal evaporation and an in-situ method. ZrC was packed in a typical lamellar mode, and the mosaic structure was formed by the deposition of Zr and Si atoms on the shallow surface of the porous C/C composites.Ablation analysis showed that the defects in the coatings originate from the boundary between the ZrC and holes created by the consumption of SiC at 2500℃. After ablation for 200 s at 3000℃, a dense ZrO2 layer formed on the coating surface, and the defects were sealed owing to the continuous supply of ablative components. 14Laminar Distribution of Neurochemically-Identified Interneurons and Cellular Co-expression of Molecular Markers in Epileptic Human Cortex显示文摘Inhibitory GABAergic interneurons are fundamental elements of cortical circuits and play critical roles in shaping network activity. Dysfunction of interneurons can lead to various brain disorders, including epilepsy,schizophrenia, and anxiety. Based on the electrophysiological properties, cell morphology, and molecular identity,interneurons could be classified into various subgroups. In this study, we investigated the density and laminar distribution of different interneuron types and the coexpression of molecular markers in epileptic human cortex.We found that parvalbumin(PV) and somatostatin(SST)neurons were distributed in all cortical layers except layer I, while tyrosine hydroxylase(TH) and neuropeptide Y(NPY) were abundant in the deep layers and white matter.Cholecystokinin(CCK) neurons showed a high density in layers IV and VI. Neurons with these markers constituted*7.2%(PV), 2.6%(SST), 0.5%(TH), 0.5%(NPY), and4.4%(CCK) of the gray-matter neuron population. Doubleand triple-labeling revealed that NPY neurons were also SST-immunoreactive(97.7%), and TH neurons were more likely to express SST(34.2%) than PV(14.6%). A subpopulation of CCK neurons(28.0%) also expressed PV, but none contained SST. 15Microstructure and corrosion behavior of Zr–1.0Cr–0.4Fe–xMo alloys显示文摘Possibility of using Mo as an alloying element in Zr-based alloys was attractive in terms of microstructure refinement and mechanical properties strengthening. In this research, Zr–1.0Cr–0.4Fe–xMo(0, 0.2, 0.4, and 0.6) alloys with different Mo contents were prepared by vacuum arcmelting method, the microstructure and the corrosion resistance of these alloys were investigated. Addition of Mo has a refinement effect on the microstructure; with the increase of Mo content, the a-laths in the as-cast samples and the grain size in the annealed samples decrease. Zr–1.0Cr–0.4Fe–xMo alloys have large numbers of fine second-phase particles(SPPs) in the matrix, the area fraction of the SPPs is more than 10 %. With the increase of Mo content, the population density of the SPPs increases significantly, while the average diameter of the SPPs decreases. Mo addition also affects the texture; the intensity of basal pole texture aligning normal direction decreases with the increase of Mo content in the alloys.Compared with Zr-4 and Zr–1Nb, Zr–1.0Cr–0.4Fe–xMo alloys have excellent corrosion resistance in 500 °C/10.3 MPa steam. 17Stimuli-responsive nanocarriers for bacterial biofilm treatment显示文摘Bacterial biofilm infections have been threatening the human’s life and health globally for a long time because they typically cause chronic and persistent infections.Traditional antibiotic therapies can hardly eradicate biofilms in many cases,as biofilms always form a robust fortress for pathogens inside,inhibiting the penetration of drugs.To address the issues,many novel drug carriers emerged as promising strategies for biofilm treatment.Among them,stimuli-responsive nanocarriers have attracted much attentions for their intriguing physicochemical properties,such as tunable size,shape and surface chemistry,especially smart drug release characteristic.Based on the microenvironmental difference between biofilm infection sites and normal tissue,many stimuli,such as bacterial products accumulating in biofilms(enzymes,glutathione,etc.),lower pH and higher H_(2)O_(2)levels,have been employed and proved in favor of“on-demand”drug release for biofilm elimination.Additionally,external stimuli including light,heat,microwave and magnetic fields are also able to control the drug releasing behavior artificially.In this review,we summarized recent advances in stimuli-responsive nanocarriers for combating biofilm infections,and mainly,focusing on the different stimuli that trigger the drug release.Meng Ding Wei Zhao Ling-Jie Song Shi-Fang Luan 2022Rare Metals2022,41,2:3
18Optomechanically-induced-transparency cooling of massive mechanical resonators to the quantum ground state显示文摘Ground state cooling of massive mechanical objects remains a difficult task restricted by the unresolved mechanical sidebands. We propose an optomechanically-induced-transparency cooling scheme to achieve ground state cooling of mechanical motion without the resolved sideband condition in a pure optomechanical system with two mechanical modes coupled to the same optical cavity mode. We show that ground state cooling is achievable for sideband resolution ωm/κ as low as~0.003.
