| 1 | A combination of follicle stimulating hormone, estradiol and age is associated with the pregnancy outcome for women undergoing assisted reproduction: a retrospective cohort analysis显示文摘To investigate the effect of basal hormone levels including follicle stimulating hormone(FSH) and estradiol(E2) and age on the pregnancy outcomes in women undergoing in vitro fertilization and embryo transfer(IVF-ET), we designed a retrospective cohort analysis and collected information of patients undergoing IVF between January 2014 and April 2017. We found that young women had much better pregnancy outcomes than older women regardless of the hormone levels. The pregnancy outcomes declined in young women with high basal E2, even though they had normal FSH. Older women with higher FSH had a worse outcome of pregnancy, especially with increased E2 level. The results suggest that a combination of FSH, E2 and age could effectively predict the pregnancy outcome for women undergoing IVF-ET, and we should encourage infertile women with diminished ovarian reserve, especially young infertile women, to try the assisted reproduction as early as possible. | 2 | Defining a critical period in calvarial development for Hedgehog pathway antagonist-induced frontal bone dysplasia in mice显示文摘The Hedgehog(Hh) signalling pathway is essential for cellular proliferation and differentiation during embryonic development.Gain and loss of function of Hh signalling are known to result in an array of craniofacial malformations. To determine the critical period for Hh pathway antagonist-induced frontal bone hypoplasia, we examined patterns of dysmorphology caused by Hh signalling inhibition. Pregnant mice received a single oral administration of Hh signalling inhibitor GDC-0449 at 100 or 150 mg·kg^(-1) body weight at preselected time points between embryonic days(E)8.5 and 12.5. The optimal teratogenic concentration of GDC-0449 was determined to be 150 mg·kg^(-1). Exposure between E9.5 and E10.5 induced frontal bone dysplasia, micrognathia and limb defects, with administration at E10.5 producing the most pronounced effects. This model showed decreased ossification of the frontal bone with downregulation of Hh signalling. The osteoid thickness of the frontal bone was significantly reduced. | 3 | Characterization of a novel regulatory pathway for mannitol metabolism and its coordination with biofilm formation in Mycobacterium smegmatis显示文摘Biofilm formation has been implicated to be tightly regulated in bacteria. Mycobacterial species possess a unique cell-wall structure; however, the underlying regulation mechanism for their biofilm formation remains largely unclear. In this study, we characterized a hypothetical mannitol metabolism and transportation gene cluster(Ms5571-Ms5576), designated as mmt operon, whose expression significantly contributes to the biofilm formation in Mycobacterium smegmatis. We showed that in the operon the Ms5575 gene encodes a GntR-like transcriptional repressor and the Ms5576 gene encodes a mannitol2-dehydrogenase which can produce D-mannitol from D-mannose. Strikingly, the D-mannitol molecule can derepress the negative regulation of Ms5575 on the mmt operon to stimulate the operon's expression. Consistently, addition of D-mannitol into the medium can obviously induce mycobacterial biofilm formation. Furthermore, we found that Ms0179 positively regulates the mmt operon through its downstream regulator Ms0180. Ms0180 directly binds the mmt operon to positively regulate its expression. Both Ms0179 and Ms0180 significantly affect the mycobacterial biofilm formation. Taken together, we explored a regulatory pathway for the mannitol metabolism and its coordination with the biofilm formation in M. smegmatis. | 5 | Emergence of hypervirulent Pseudomonas aeruginosa pathotypically armed with co-expressed T3SS effectors ExoS and ExoU显示文摘Pseudomonas aeruginosa is a significant pathogen mainly causing healthcare-associated infections(HAIs).Newly emerging high-risk clones of P.aeruginosa with elevated virulence profiles furtherly cause severe community-acquired infections(CAIs).Usually,it is not common for P.aeruginosa to co-carry exoU and exoS genes,encoding two type III secretion system(T3SS)effectors.The pathogenicity mechanism of exoS+/exoU+strains of P.aeruginosa remains unclear.Here,we provide detailed evidence for a subset of hypervirulent P.aeruginosa strains,which abundantly co-express and secrete the T3SS effectors ExoS and ExoU.The exoS+/exoU+P.aeruginosa strains were available to cause both HAIs and CAIs.The CAI-associated strains could elicit severe inflammation and hemorrhage,leading to higher death rates in a murine acute pneumonia model,and had great virulence potential in establishing chronic infections,demonstrating hypervirulence when compared to PAO1(exoS+/exoU-)and PA14(exoS-/exoU+).Both ExoS and ExoU were co-expressed and co-secreted in abundance in exoS+/exoU+strains.Their abundant protein secretion could boost exoS+/exoU+strains’potentials for cytotoxicity in vitro and pathogenicity in vivo.Genomic evidence indicates that exoU acquisition is likely mediated by horizontal gene transfer(HGT)of the pathogenicity island PAPI-2,while deletion of exoU was sufficient to mitigate virulence in the exoS+/exoU+strains.Furthermore,bioinformatics analysis showed that such exoS+/exoU+P.aeruginosa strains turned out to be widely distributed across the globe.Overall,the research provide detailed evidence for the high virulence and epidemicity of exoS+/exoU+strains of P.aeruginosa,highlighting an urgent need for surveillance against these high-risk hypervirulent strains. | Yuqin Song Yongqi Mu Nai-Kei Wong Zhuo Yue Juan Li Min Yuan Xiong Zhu Jinshu Hu Gang Zhang Dawei Wei Chao Wang Weihui Wu Fang Bai Jie Feng | 2023 | hLife2023,1,1: | 0 |

| 10 | A preliminary study on volatile organic compounds and odor in university dormitories:Situation,contribution,and correlation显示文摘Many dormitories have a high population density,and occupants exposed to volatile organic compounds(VOCs)could suffer severe physical and mental issues,so it is important to ensure good indoor air quality(IAQ).Currently,indoor VOC analysis using only chemical concentration cannot accurately reflect the effect of odor on human comfort.Therefore,the sensory evaluation of indoor odor acts as a supplementary method for an IAQ assessment.Here,we measured indoor VOC concentrations in 10 dormitories under occupied and unoccupied conditions.The contribution of building materials and human-related emissions was analyzed.An odor activity value(OAV)was calculated using the VOC concentration and odorant threshold.In addition,recruited sensory odor panel members assessed the odor intensity of each dormitory and the correlation between odor intensity and chemical measurements was analyzed.The indoor total VOC(TVOC)concentration under occupied conditions ranged from 242.2 to 1063.1μg/m^(3),with a mean value of 454.1μg/m^(3).The TVOC concentration under unoccupied conditions varied from 97.2 to 1055.2μg/m^(3),with a mean value of 342.2μg/m^(3).The TVOC concentration of 90% of the dormitories under the two conditions met the national IAQ standards.An average of 53.6% indoor TVOC emissions are related to building materials,the outdoor-related TVOC concentration accounts for 15.7%,and the average human-related TVOC emissions are 1519.4μg/(h·person),which accounts for 30.7%.Compared to TVOC concentration,odor intensity-OAV(OAV_(sum),OAV_(max))could better characterize subjective and objective correlations.However,the goodness-of-fit is not satisfactory.Weber–Fencher's law has limitations in characterizing the correlation between odor intensity and OAV when applied to the indoor air mixture,based on this small-scale study. | Yibing Hu Ling Xu Weihui Liang | 2023 | Building Simulation2023,16,3: | 0 |
