| 2 | Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene and risk of ischemic stroke in Han Chinese of eastern China显示文摘Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene have been suggested to play an important role in the pathogenesis of atherosclerosis and ischemic stroke.This study was aimed to explore the association of ALOX5AP variants with ischemic stroke risk in Han Chinese of eastern China.A total of 690 ischemic stroke cases and 767 controls were recruited.The subjects were further subtyped according to the Trial of Org 10172 in Acute Stroke Treatment (TOAST) criteria.On the basis of that,two polymorphisms of the ALOX5AP gene (rs10507391 and rs12429692) were determined by TaqMan genotyping assay.In addition,plasma leukotriene B4 (LTB4) levels were analyzed in these subjects.There was no evidence of association between the two variants of ALOX5AP and the risk of ischemic stroke or its TOAST-subtypes.Haplotype analysis and stratification analysis according to sex,age,body mass index,hypertension,and diabetes also showed negative association.Analysis of LTB4 levels in a subset of cases and controls revealed that LTB4 levels were significantly higher in ischemic stroke cases than in the controls (70.06±14.75 ng/L vs 57.34±10.93 ng/L;P=0.000) and carriers of the T allele of the rs10507391 variant were associated with higher plasma LTB4 levels (P=0.000).The present study suggests there is no association of the two polymorphisms in the ALOX5AP gene with ischemic stroke risk in Han Chinese of eastern China. | Gannan Wang Yao Wang Hao Sun Weijuan Cao Jing Zhang Hang Xiao Jinsong Zhang | 2011 | The Journal of Biomedical Research2011,25,5: | 12 |

| 4 | Microelement analysis of source of raw materials of terracotta warriors and horses of Qin's Mausoleum in pits No.1 and No.2显示文摘In order to search for sources of the raw material of the Qin’s terracotta warriors and horses, we selected 20 samples of pottery warriors and pottery horses of the Qin’s Mausoleum from pit No. 1,19 samples of pottery warriors and brick from pit No. 2, 20 samples of clays near the Qin’s Mausoleum and 2 samples of Yaozhou porcelain bodies. The 32 kinds of element content of each sample were measured by neutron activation analysis (NAA). These data were analyzed by fuzzy cluster analysis, the trend cluster analysis diagram was obtained. The results showed that samples of terracotta warriors and horses from pits No.1 and No. 2 were closely related to the loam layer near Qin’s Mausoleum, but their relations with the loess layers were not so close, and they were out of all relation to Yaozhou porcelain bodies. | 5 | Astragalus injection inhibits c-Jun N terminal kinase mRNA expression following oxygen-glucose deprivation and reintroduction in rat hippocampal neurons显示文摘BACKGROUND:In studies concerning cell injury induced by cerebral ischemia-reperfusion,current experiments have primarily focused on altered protein levels.In addition,the apoptotic proteins Bax and Bcl-2 have been thoroughly studied with regard to initiating neuronal apoptosis. OBJECTIVE:To establish an in vitro model of oxygen-glucose deprivation and reintroduction in the rat hippocampus to simulate cerebral ischemia-reperfusion injury;to observe c-Jun N-terminal kinase 3(JNK3) mRNA expression in hippocampal neurons following Astragalus injection;and thus to determine changes in the signaling and downstream pathways of neuronal apoptosis at the cellular and molecular level. DESIGN,TIME AND SETTING:A randomized,controlled,cellular and molecular experiment was performed at the Department of Central Laboratory,Chengde Medical College from February to June 2008. MATERIALS:Astragalus injection,the main ingredient of astragaloside,was purchased from Chengdu Di'ao Jiuhong Pharmaceutical Manufactory,China.JNK3 mRNA probe and in situ hybridization kit were purchased from Tianjin Haoyang Biological Technology,China,and JNK3 RT-PCR primers were designed by Shanghai Bio-engineering,China. METHODS:Primary cultures of hippocampal neurons derived from Sprague Dawley rats,aged 1-2 days,were established.After 8 days,the hippocampal neurons were assigned to the following interventions:model group,Astragalus group,and vehicle control group,cells were subjected to oxygen-glucose reintroduction after oxygen-glucose deprivation for 30 minutes in sugar-free Earle's solution and a hypoxia device,which contained high-purity nitrogen.The normal control group was subjected to primary culture techniques and was not treated using above-mentioned interventions. In addition,the Astragalus and vehicle control groups were treated with Astragalus injection(0.5 g/L raw drug) or sterile,deionized water at 2 hours prior to oxygen-glucose deprivation,respectively. MAIN OUTCOME MEASURES:JNK3 mRNA expression was measured by in situ hybridization and RT-PCR at 0,0.5,2,6,24,72,and 120 hours after oxygen-glucose reintroduction. RESULTS:Hippocampal neuronal morphology was normal in the normal control group. Hippocampal neurons exhibited apparent apoptosis-like pathological changes in the model,as well as the vehicle control,groups.The apoptosis-like pathological changes in the hippocampal neurons were less in the Astragalus group.Results from in situ hybridization and RT-PCR showed that JNK3 mRNA expression significantly increased in hippocampal neurons from model group,as well as the vehicle control group,compared with the normal control group(P<0.05).In addition,JNK3 mRNA expression significantly decreased in hippocampal neurons of the Astragalus group,compared with the model group and vehicle control group(P<0.05). | 6 | Nondestructive 3D Image Analysis Pipeline to Extract Rice Grain Traits Using X-Ray Computed Tomography显示文摘The traits of rice panicles play important roles in yield assessment,variety classification,rice breeding,and cultivation management.Most traditional grain phenotyping methods require threshing and thus are time-consuming and labor-intensive;moreover,these methods cannot obtain 3D grain traits.In this work,based on X-ray computed tomography,we proposed an image analysis method to extract twenty-two 3D grain traits.After 104 samples were tested,the R^(2) values between the extracted and manual measurements of the grain number and grain length were 0.980 and 0.960,respectively.We also found a high correlation between the total grain volume and weight.In addition,the extracted 3D grain traits were used to classify the rice varieties,and the support vector machine classifier had a higher recognition accuracy than the stepwise discriminant analysis and random forest classifiers.In conclusion,we developed a 3D image analysis pipeline to extract rice grain traits using X-ray computed tomography that can provide more 3D grain information and could benefit future research on rice functional genomics and rice breeding. | Weijuan Hu Can Zhang Yuqiang Jiang Chenglong Huang Qian Liu Lizhong Xiong Wanneng Yang Fan Chen | 2020 | Plant Phenomics2020,2,1: | 6 |

| 11 | Matrix stiffness exacerbates the proinflammatory responses of vascular smooth muscle cell through the DDR1-DNMT1 mechanotransduction axis显示文摘Vascular smooth muscle cell (vSMC) is highly plastic as its phenotype can change in response to mechanical cues inherent to the extracellular matrix (ECM). VSMC may be activated from its quiescent contractile phenotype to a proinflammatory phenotype, whereby the cell secretes chemotactic and inflammatory cytokines, e.g. MCP1 and IL6, to functionally regulate monocyte and macrophage infiltration during the development of various vascular diseases including arteriosclerosis. Here, by culturing vSMCs on polyacrylamide (PA) substrates with variable elastic moduli, we discovered a role of discoidin domain receptor 1 (DDR1), a receptor tyrosine kinase that binds collagens, in mediating the mechanical regulation of vSMC gene expression, phenotype, and proinflammatory responses. We found that ECM stiffness induced DDR1 phosphorylation, oligomerization, and endocytosis to repress the expression of DNA methyltransferase 1 (DNMT1), very likely in a collagen-independent manner. The DDR1-to-DNMT1 signaling was sequentially mediated by the extracellular signal-regulated kinases (ERKs) and p53 pathways. ECM stiffness primed vSMC to a proinflammatory phenotype and this regulation was diminished by DDR1 inhibition. In agreement with the in vitro findings, increased DDR1 phosphorylation was observed in human arterial stiffening. DDR1 inhibition in mouse attenuated the acute injury or adenine diet-induced vascular stiffening and inflammation. Furthermore, mouse vasculature with SMC-specific deletion of Dnmt1 exhibited proinflammatory and stiffening phenotypes.
