1Programmed Death Ligand-1 on Microglia Regulates Th1 Differentiation via Nitric Oxide in Experimental Autoimmune Encephalomyelitis显示文摘Microglia are considered to be potential antigen-presenting cells and have the ability to present antigen under pathological conditions. Nevertheless, whether and how microglia are involved in immune regulation are largely unknown. Here, we investigated the suppressive activity of microglia during experimental autoimmune encephalomyelitis(EAE) induced by myelin oligodendrocyte glycoprotein, with the goal of understanding their role in regulating the T cell reaction. Using flow cytometric analysis, we found that microglia were characterized by increased cell number and up-regulated programmed death ligand-1(PD-L1) at the peak phase of EAE. Meanwhile,both the CD^(4+)T cells and microglia that infiltrated the central nervous system expressed higher levels of PD1, the receptor for PD-L1, accompanied by a decline of Th1 cells.In an ex vivo co-culture system, microglia from EAE mice inhibited the proliferation of antigen-specific CD^(4+)T cells and the differentiation of Th1 cells, and this was significantly inhibited by PD-L1 blockade. Further,microglia suppressed Th1 cells via nitric oxide(NO), the production of which was dependent on PD-L1. 4The analysis of genetic diversity and differentiation of six Chinese cattle populations using microsatellite markers显示文摘A total of 321 individuals from six cattle populations of four species in a bovine subfamily in China were studied using 12 pairs of mi- crosatellite markers. The genetic diversities within and between populations were calculated. The phylogenetic trees were constructed by (δμ) 2 and DA distances, and the divergence times between populations were estimated by (δμ) 2. Altogether, 144 microsatellite alleles were detected including 24 private alleles and nine shared alleles. Chinese Holstein had the largest number of private alleles (10), whereas, Bohai black and Buffalo had the smallest number of private alleles (2). Chinese Holstein showed the highest genetic variability. Its ob- served number of alleles (Na), mean effective number of alleles (MNA), and mean heterozygosity (He) were 7.7500, 4.9722, and 0.7719, respectively, whereas, the Buffalo and Yak showed low genetic variability. In the phylogenetic trees, Luxi and Holstein grouped first, followed by Bohai and Minnan. Yak branched next and buffalo emerged as the most divergent population from other cattle populations. Luxi and Bohai were estimated to have diverged 0.039-0.105 million years ago (MYA), however, buffalo and Holstein diverged 0.501-1.337 MYA. 15Fabrication of Bi-Bi3O4Cl plasmon photocatalysts for removal of aqueous emerging contaminants under visible light显示文摘Photocatalytic oxidation of emerging contaminants(ECs) in water has recently gained extensive attentions. In this study, bismuth oxychloride-based plasmon photocatalysts(BiBiOCl) exhibiting high performance were successfully developed by reducing Bion the surface of BiOCl. Consequently, the photocatalysts were used to remove ECs from water.The effects of developmental process and Bi metal plasmon resonance on the photoelectric performances of Bi-BiOCl were investigated through a series of characterizations. The UV-vis diffuse reflection and photoluminescence spectra revealed that the light absorption range of the photocatalyst gradually increased and the electron recombination rate gradually decreased with the introduction of Bi metals. The optimal removal rates of ciprofloxacin and tetrabromobisphenol A by Bi-BiOCl were 93.8% and 96.4%;the respective reaction rate constants were 5.48 and 4.93 times higher than that of BiOCl. The mechanism study indicated that main reactants in the photocatalytic system were ·O-2radicals and photogenerated holes, and the existence of oxygen vacancies and Bi metals promoted electron transfer in photocatalyst. 19In vivo skin penetration and metabolic path of quantum dots显示文摘The skin is the largest organ of the body and is a potential route of exposure to sunscreens and cosmetics containing nanoparticles;however,the permeability of the skin to these nanoparticles is currently unknown.In this paper,we studied the transdermal delivery capacity through mouse skin of water-soluble CdSeS quantum dots(QDs) and the deposition of these QDs in the body.QD solution was coated onto the dorsal hairless skin of male ICR mice.Fluorescence microscopy and transmission electron microscopy(TEM) were used to observe the distribution of QDs in the skin and organs,and inductively coupled plasma-mass spectrometry(ICP-MS) was used to measure the 111Cd content to indicate the concentration of QDs in plasma and organs.Experimental results indicate that QDs can penetrate into the dermal layer and are limited to the uppermost stratum corneum layers and the hair follicles.Through blood circulation,QDs deposit mostly in liver and kidney and are difficult to clear.111Cd concentration was greater than 14 ng g-1 in kidney after 120 h after 0.32 nmol QDs was applied to a mouse.These results suggest that QDs have in vivo transdermal delivery capacity through mouse skin and are harmful to the liver and kidney.TANG Lei ZHANG ChunLing SONG GuangMing JIN Xun XU ZhongWei 2013Science China(Life Sciences)2013,56,2:1
