2Updates in version 2019 of CSCO guidelines for colorectal cancer from version 2018显示文摘According to the latest data from cancer registration center, there are estimated to be 3,929,000 individuals newly diagnosed with cancer and 2,338,000 deaths from this disease in China in 2015 (1). Among them, colorectal cancer (CRC) with 388,000 newly diagnosed cases and 187,000 deaths, stands the third place in incidence and the fifth place in mortality. The Chinese Society of Clinical Oncology (CSCO) has organized an expert committee to write and publish the Guideline for Colorectal Cancer in 2017. And its English version has already been published in March 2019 (2). 3m6A RNA methylation-mediated HNF3γreduction renders hepatocellular carcinoma dedifferentiation and sorafenib resistance显示文摘Hepatocyte nuclear factor 3γ(HNF3γ)is a hepatocyte nuclear factor,but its role and clinical significance in hepatocellular carcinoma(HCC)remain unclear.Herein,we report that HNF3γexpression is downregulated in patient HCC and inversely correlated with HCC malignancy and patient survival.Moreover,our data suggested that the HNF3γreduction in HCC could be mediated by METTL14-dependent m6A methylation of HNF3γmRNA.HNF3γexpression was increased during hepatic differentiation and decreased in dedifferentiated HCC cells.Interestingly,HNF3γdelivery promoted differentiation of not only HCC cells but also liver CSCs,which led to suppression of HCC growth.Mechanistic analysis suggested an HNF3γ-centered regulatory network that includes essential liver differentiation-associated transcription factors and functional molecules,which could synergistically facilitate HCC cell differentiation.More importantly,enforced HNF3γexpression sensitized HCC cells to sorafenib-induced growth inhibition and cell apoptosis through transactivation of OATP1B1 and OATP1B3 expression,which are major membrane transporters for sorafenib uptake.Clinical investigation showed that patient-derived HCC xenografts with high HNF3γexpression exhibited a sorafenib response and patients with high HCC HNF3γlevels benefited from sorafenib therapy.Together,these results suggest that HNF3γplays an essential role in HCC differentiation and may serve as a therapeutic target and predictor of sorafenib benefit in patients.Tengfei Zhou Shichao Li Daimin Xiang Junyu Liu Wen Sun Xiuliang Cui Beifang Ning Xiao Li Zhuo Cheng Weiqi Jiang Cheng Zhang Xijun Liang Liang Li Xin Cheng Liu Hui Hongyang Wang Jin Ding 2020Signal Transduction and Targeted Therapy2020,5,1:4
4Chimeric Newcastle Disease Virus-like Particles Containing DC-Binding Peptide-Fused Haemagglutinin Protect Chickens from Virulent Newcastle Disease Virus and H9N2 Avian Influenza Virus Challenge显示文摘Newcastle disease virus(NDV)and H9N2 subtype Avian influenza virus(AIV)are two notorious avian respiratory pathogens that cause great losses in the poultry industry.Current inactivated commercial vaccines against NDV and AIV have the disadvantages of inadequate mucosal responses,while an attenuated live vaccine bears the risk of mutation.Dendritic cell(DC)targeting strategies are attractive for their potent mucosal and adaptive immune-stimulating ability against respiratory pathogens.In this study,DC-binding peptide(DCpep)-decorated chimeric virus-like particles(cVLPs),containing NDV haemagglutinin–neuraminidase(HN)and AIV haemagglutinin(HA),were developed as a DC-targeting mucosal vaccine candidate.DCpep-decorated cVLPs activated DCs in vitro,and induced potent immune stimulation in chickens,with enhanced secretory immunoglobulin A(sIgA)secretion and splenic T cell differentiation.40μg cVLPs can provide full protection against the challenge with homologous,heterologous NDV strains,and AIV H9N2.In addition,DCpep-decorated cVLPs could induce a better immune response when administered intranasally than intramuscularly,as indicated by robust s IgA secretion and a reduced virus shedding period.Taken together,this chimericVLPs are a promising vaccine candidate to control NDV and AIV H9N2 and a useful platform bearing multivalent antigens.Xiaohong Xu Jing Qian Lingsong Qin Jindou Li Cong Xue Jiaxin Ding Weiqi Wang Wei Ding Renfu Yin Ningyi Jin Zhuang Ding 2020Virologica Sinica2020,35,4:4
5DVL mutations identified from human neural tube defects and Dandy-Walker malformation obstruct the Wnt signaling pathway显示文摘Wnt signaling pathways,including the canonical Wnt/β-catenin pathway,planar cell polarity pathway,and Wnt/Ca2+signaling pathway,play important roles in neural development during embryonic stages.The DVL genes encode the hub proteins for Wnt signaling pathways.The mutations in DVL2 and DVL3 were identified from patients with neural tube defects(NTDs),but their functions in the pathogenesis of human neural diseases remain elusive.Here,we sequenced the coding regions of three DVL genes in 176 stillborn or miscarried fetuses with NTDs or Dandy-Walker malformation(DWM)and 480 adult controls from a Han Chinese population.Four rare mutations were identified:DVL1 p.R558 H,DVL1 p.R606 C,DVL2 p.R633 W,and DVL3 p.R222 Q.To assess the effect of these mutations on NTDs and DWM,various functional analyses such as luciferase reporter assay,stress fiber formation,and in vivo teratogenic assay were performed.The results showed that the DVL2 p.R633 W mutation destabilized DVL2 protein and upregulated activities for all three Wnt signalings(Wnt/β-catenin signaling,Wnt/planar cell polarity signaling,and Wnt/Ca2+signaling)in mammalian cells.In contrast,DVL1 mutants(DVL1 p.R558 H and DVL1 p.R606 C)decreased canonical Wnt/β-catenin signaling but increased the activity of Wnt/Ca2+signaling,and DVL3 p.R222 Q only decreased the activity of Wnt/Ca2+signaling.We also found that only the DVL2 p.R633 W mutant displayed more severe teratogenicity in zebrafish embryos than wild-type DVL2.Our study demonstrates that these four rare DVL mutations,especially DVL2 p.R633 W,may contribute to human neural diseases such as NTDs and DWM by obstructing Wnt signaling pathways.Lingling Liu Weiqi Liu Yan Shi Ling Li Yunqian Gao Yunping Lei Richard Finnell Ting Zhang Feng Zhang Li Jin Huili Li Wufan Tao Hongyan Wang 2020Journal of Genetics and Genomics2020,47,6:2

14AgPdCo hollow nanospheres electrocatalyst with high activity and stability toward the formate electrooxidation显示文摘The widespread commercial application of direct formate fuel cell(DFFC)is limited by the lack of efficient electrocatalysts for the formate oxidation reaction(FOR).AgPdCo hollow nanospheres(H-NSs)with jagged surfaces are successfully synthesized via a facile method involving the wet-chemical synthesis of AgPdCo nanospheres(NSs)and galvanic replacement reaction between Pd salt and AgPdCo NSs.Surpassing Ag_(30)Pd_(69)Co_(1) NSs and most of previously reported electrocatalysts,Ag_(9)Pd_(90)Co_(1) H-NSs exhibit extremely high FOR activity with a peak current density of 3.08 A·mg_(Pd)^(−1).Apart from the competitive activity,Ag_(9)Pd_(90)Co_(1) H-NSs show greatly improved chronoamperometric and cycling stability,whereby the current density retains about 0.24 A·mg_(Pd)^(−1) after 3,600 s electrocatalysis and the mass activity maintains 54.06%of the initial value after 500 cycles.The unique hollow nanosphere and synergistic effect are responsible for the enhanced activity and stability.This study will provide new clues for the development of outstanding electrocatalysts.Qiao Wang Fuyi Chen Quan Tang Longfei Guo Tao Jin Bowei Pan Junpeng Wang Zhen Li Bo Kou Weiqi Bian 2021Nano Research2021,14,7:1
