| 3 | TLR9 agonist enhances radiofrequency ablation-induced CTL responses, leading to the potent inhibition of primary tumor growth and lung metastasis显示文摘Radiofrequency ablation(RFA)is the most common approach to thermal ablation for cancer therapy.Unfortunately,its efficacy is limited by incomplete ablation,and further optimization of RFA is required.Here,we demonstrate that incubation at 65°C triggers more EG7 tumor cell death by necrosis than treatment at 45°C,and the 65°C-treated cells are more effective at inducing antigen-specific CD8^(+)cytotoxic T lymphocyte(CTL)responses after injection in mice than the 45°C-treated ones.Dendritic cells(DCs)that phagocytose 65°C-treated EG7 cells become mature with upregulated MHCII and CD80 expression and are capable of efficiently inducing effector CTLs in mouse tumor models.RFA(65°C)therapy of EG7 tumors induces large areas of tumor necrosis and stimulates CTL responses.This leads to complete regression of small(~100 mm^(3))tumors but fails to suppress the growth of larger(~350 mm^(3))tumors.The administration of the Toll-like receptor-9(TLR9)agonist unmethylated cytosine-phosphorothioate-guanine oligonucleotide(CpG)to DCs phagocytosing 65°C-treated EG7 cells enhances the expression of MHCII and CD40 on DCs as well as DC-induced stimulation of CTL responses.Importantly,the intratumoral administration of CpG following RFA also increases the frequencies of tumor-associated immunogenic CD11b−CD11c^(+)CD103^(+)DC2 and CD11b+F4/80+MHCII+M1 macrophages and increases CD4^(+)and CD8^(+)T-cell tumor infiltration,leading to enhanced CD4^(+)T cell-dependent CTL responses and potent inhibition of primary RFA-treated or distant untreated tumor growth as well as tumor lung metastasis in mice bearing larger tumors.Overall,our data indicate that CpG administration,which enhances RFA-induced CTL responses and ultimately potentiates the inhibition of primary tumor growth and lung metastasis,is a promising strategy for improving RFA treatment,which may assist in optimizing this important cancer therapy. | Aizhang Xu Lifeng Zhang Jingying Yuan Fatma Babikr Andrew Freywald Rajni Chibbar Michael Moser Wenjun Zhang Bing Zhang Zhaoying Fu Jim Xiang | 2019 | Cellular & Molecular Immunology2019,16,10: | 3 |

| 5 | Analysis of GNSS radio occultation data from satellite ZH-01显示文摘The electromagnetic satellite Zhangheng 01(ZH-01) was successfully launched on February 2, 2018. The GNSS Radio Occultation(GRO) receiver on board the satellite is able to observe the occultation events of GPS and BeiDou navigation satellites. We analyzed the data acquired during the in-orbit testing period. We concludes that the GRO ionosphere inversion results are reasonable,the trend is correct, the satellite can observe about 600 ionosphere occultation events each day. The global coverage of more than 30000 consecutive GRO events in more than two months were analyzed and compared with COSMIC observations: both the GRO and COSMIC occultation can realize global coverage: the Nm F2 and Hm F2 global distributions are similar and change obviously with latitude. | 6 | Experimental Investigation on the Pressure Propagation Mechanism of Tight Reservoirs显示文摘Low permeability tight sandstone reservoirs have a high filtrational resistance and a very low fluid flow rate.As a result,the propagation speed of the formation pressure is low and fluid flow behaves as a non-Darcy flow,which typically displays a highly non-linear behavior.In this paper,the characteristics and mechanism of pressure propagation in this kind of reservoir are revealed through a laboratory pressure propagation experiment and through data from an actual tight reservoir development.The main performance mechanism is as follows:A new pressure cage concept is proposed based on the pressure variation characteristics of the laboratory experiments.There are two methods of energy propagation in the actual water injection process:one is that energy is transmitted to the deep reservoir by the fluid flowing through the reservoir,and the other is that energy is transmitted by the elasticity of the reservoir.For one injection well model and one production well model,the pressure distribution curve between the injection and production wells,as calculated by the theoretical method,has three section types,and they show an oblique“S”shape with a straight middle section.However,the actual pressure distribution curve is nonlinear,with an obvious pressure advance at the front.After the injection pressure increases to a certain level,the curve shape is an oblique and reversed“S”shape.Based on the research,this paper explains the deep-seated reasons for the difference in pressure distribution and proposes that it is an effective way to develop low permeability tight reservoirs using the water injection supplement energy method. | Jing Sun Dehua Liu Xiang Zhu Wenjun Huang Liang Cheng | 2020 | Fluid Dynamics & Materials Processing2020,16,3: | 2 |
| 7 | A policy gradient algorithm integrating long and short-term rewards for soft continuum arm control显示文摘The soft continuum arm has extensive application in industrial production and human life due to its superior safety and flexibility. Reinforcement learning is a powerful technique for solving soft arm continuous control problems, which can learn an effective control policy with an unknown system model. However, it is often affected by the high sample complexity and requires huge amounts of data to train, which limits its effectiveness in soft arm control. An improved policy gradient method, policy gradient integrating long and short-term rewards denoted as PGLS, is proposed in this paper to overcome this issue. The shortterm rewards provide more dynamic-aware exploration directions for policy learning and improve the exploration efficiency of the algorithm. PGLS can be integrated into current policy gradient algorithms, such as deep deterministic policy gradient(DDPG). The overall control framework is realized and demonstrated in a dynamics simulation environment. Simulation results show that this approach can effectively control the soft arm to reach and track the targets. Compared with DDPG and other model-free reinforcement learning algorithms, the proposed PGLS algorithm has a great improvement in convergence speed and performance. | 8 | Weighted joint calibration for interferometric SAR显示文摘Calibration is a processing procedure for across-track interferometric synthetic aperture radar(InSAR) to achieve an accurate three-dimensional location. A calibration technique,called weighted joint calibration, for the generation of wide-area geocoded digital elevation models(DEMs) is proposed. It calibrates multiple InSAR scenes simultaneously, and allows reducing the number of required ground control points(GCPs) by using tie points(TPs). This approach may ensure the continuity of threedimensional location among adjacent scenes, which is necessary for mosaic and fusion of data coming from different scenes. In addition, it introduces weights to calibration to discriminate GCPs and TPs with different coherences and locations. | 18 | Origin of dolomites in the Permian dolomitic reservoirs of Fengcheng Formation in Mahu Sag,Junggar Basin, NW China显示文摘Origin of authigenic dolomites in the dolomitic reservoir of the Permian Fengcheng Formation in the Mahu Sag of Junggar Basin is unclear.Occurrence and genetic evolution of the authigenic dolomites in dolomitic rock reservoir of the Fengcheng Formation in the Mahu Sag were analyzed by polarized and fluorescence thin sections,scanning electron microscope(SEM),electron microprobe(EMP),C,O and Sr isotopes analysis,and other techniques.(1)Dolomites were mainly precipitated in three stages:penecontemporaneous-shallow burial stage(early stage of the Middle Permian),middle burial stage(middle stage of the Middle Permian),and middle-deep burial stage,with the former two stages in dominance.(2)Dolomitization fluid was high-salinity brine originating from alkaline lake.In the penecontemporaneous-shallow burial stage,Mg^(2+)was mainly supplied by alkaline-lake fluid and devitrification of volcanic glass.In the middle burial stage,Mg^(2+)mainly came from the transformation of clay minerals,devitrification of volcanic glass and dissolution of aluminosilicates such as feldspar.(3)Regular changes of Mg,Mn,Fe,Sr,Si and other elements during the growth of dolomite were mainly related to the alkaline-lake fluid,and to different influences of devitrification and diagenetic alteration of volcanic materials during the burial.(4)In the penecontemporaneous stage,induced by alkaline-lake microorganisms,the micritic-microcrystalline dolomites were formed by primary precipitation,replacement of aragonite and high-Mg calcite,and other processes;in the shallow burial stage,the silt-sized dolomites were formed by continuous growth of micritic-microcrystalline dolomite and replacement of calcites,tuffs and other substances;in the middle burial stage,the dolomites,mainly silt-and fine-sized,were formed by replacement of volcanic materials.The research results are referential for investigating the formation mechanism and distribution patterns of tight dolomitic reservoirs in the Mahu Sag and other similar oil and gas bearing areas. | TANG Yong LYU Zhengxiang HE Wenjun QING Yuanhua LI Xiang SONG Xiuzhang YANG Sen CAO Qinming QIAN Yongxin ZHAO Xinmei | 2023 | Petroleum Exploration and Development2023,50,1: | 0 |
| 19 | Distinct roles but cooperative effect of TLR3/9 agonists and PD-1 blockade in converting the immunotolerant microenvironment of irreversible electroporation-ablated tumors显示文摘Irreversible electroporation(IRE)is a new cancer ablation technology,but methods to improve IRE-induced therapeutic immunity are only beginning to be investigated.We developed a mouse model bearing large primary(300 mm^(3))and medium distant(100 mm^(3))EG7 lymphomas engineered to express ovalbumin(OVA)as a nominal tumor antigen.We established experimental protocols including IRE alone and IRE combined with Toll-like receptor(TLR)3/9 agonists(poly I:C/CpG)(IRE+pIC/CpG),PD-1 blockade(IRE+PD-1 blockade),or both(IRE+Combo)to investigate therapeutic effects on primary and distant EG7 tumors and conversion-promoting effects on the immunotolerant tumor microenvironment(TME).We demonstrated that IRE alone simulated very weak OVA-specific CD8^(+)T cell responses and did not inhibit primary tumor growth.IRE+pIC/CpG synergistically stimulated more efficient OVA-specific CD8^(+)T cell responses and primary tumor growth inhibition than IRE+PD-1 blockade.IRE+pIC/CpG played a major role in the modulation of immune cell profiles but a minor role in the downregulation of PD-L1 expression in the TME and vice versa for IRE+PD-1 blockade.IRE+Combo cooperatively induced potent OVA-specific CD8^(+)T cell immunity and rescued exhausted intratumoral CD8^(+)T cells,leading to eradication of not only primary tumors but also untreated concomitant distant tumors and lung metastases.IRE+Combo efficiently modulated immune cell profiles,as evidenced by reductions in immunotolerant type-2(M2)macrophages,myeloid-derived suppressor-cells,plasmacytoid dendritic cells,and regulatory T cells and by increases in immunogenic M1 macrophages,CD169^(+)macrophages,type-1 conventional dendritic cells,and CD8^(+)T cells,leading to conversion of immunotolerance in not only primary TMEs but also untreated distant TMEs.IRE+Combo also showed effective therapeutic effects in two breast cancer models.Therefore,our results suggest that IRE+Combo is a promising strategy to improve IRE ablation therapy in cancer. | Fatma Babikr Jiangbo Wan Aizhang Xu Zhaojia Wu Shahid Ahmed Andrew Freywald Rajni Chibbar Yue Wu Michael Moser Gary Groot Wenjun Zhang Bing Zhang Jim Xiang | 2021 | Cellular & Molecular Immunology2021,18,12: | 0 |
| 20 | Suppression of cell pyroptosis by omeprazole through PDE4-mediated autophagy in gastric epithelial cells显示文摘Helicobacter pylori is a risk factor for the development of peptic ulcers with autophagy dysfunction.Omeprazole was widely known as the first-line regimen for H.pylori-associated gastritis.Objectives:The objective of this work was to assess the role of omeprazole on cell pyroptosis and autophagy.Methods:The clinical samples were collected.Quantitative polymerase chain reaction,western blotting,enzyme linked immunosorbent assay,and immunofluorescence(IF)analysis were conducted to reveal the mechanism of omeprazole on cell pyroptosis and autophagy.Results:The results revealed that omeprazole could decrease cell pyroptosis,which was attributed to the downregulation of cleaved caspase-1 expression,resulting in the inhibition of gasdermin E and interleukin-18/1βmaturation and secretion as well as the resolution of inflammation.Mechanistically,omeprazole treatment led to drastic downregulation of mammalian target of rapamycin(mTOR)activity was observed in BGC823 cells,leading to enhanced autophagy characterized by increased LC3II expression,which further reduced cell pyroptosis.This omeprazole-mediated phenomenon was enhanced after phosphodiesterase-4(PDE4)inhibitor dipyridamole(DIP)treatment.In addition,activation of mTOR by MHY1485 could rescue the suppression of cell pyroptosis induced by omeprazole.Most importantly,IF analysis suggested that phosphorylation of mTOR and PDE4 activity and caspase-1 were enhanced in H.pylori-infected gastric mucosa.Conclusion:These findings indicate that omeprazole suppresses cell pyroptosis through PDE4-mediated autophagy in gastric epithelial cells,and DIP enhanced the omeprazole-mediated inhibition of cell pyroptosis,implying that DIP is an alternative combined therapy strategy in improving the treatment of patients with H.pylori infection. | LIPING YE HUIYAN SUN XINHUA LIANG WENXU PAN LI XIANG WENJUN DU LANLAN GENG WANFU XU SITANG GONG | 2023 | BIOCELL2023,47,12: | 0 |
