| 1 | Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics. | Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng | 2018 | Genes & Diseases2018,5,2: | 8 |
| 2 | Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions. | Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan | 2018 | Genes & Diseases2018,5,2: | 5 |

| 4 | Flexible lead-free BFO-based dielectric capacitor with large energy density,superior thermal stability,and reliable bending endurance显示文摘As an essential energy-stored device,the inorganic dielectric film capacitor plays an irreplaceable role in high-energy pulse power technology area.In this work,propelled by the challenge of overcoming the bottlenecks of inflexibility and inferior energy storage density of the pure BiFeO3 films,the mica with high bendability and thermal stability is adopted as substrate,and the relaxor ferroelectric(Sr_(0.7)Bi_(0.2))TiO_(3) is introduced to form solid solution to introduce relaxor behavior.The subsequently fabricated 0.3Bi(Fe_(0.95)Mn_(0.05))O_(3)-0.7(Sr_(0.7)Bi_(0.2))TiO_(3)(BFMO-SBT)thin film capacitor exhibits a high recoverable energy storage density(W_(rec)=61 J cm^(-3))and a high efficiency(η=75%)combined with a fast discharging rate(23.5 μs)due to the large polarization difference(ΔP=59.4 μC cm^(-2)),high breakdown strength(E_(b)=3000 kV cm^(-1)),and the strong relaxor dispersion(γ=1.78).Of particular importance is the capacitor presents excellent stability of energy storage performance,including a wide working temperature window of -50-200℃,fatigue endurance of 108 cycles,and frequency range of 500 Hz-20 kHz.Furthermore,there are no obviously deteriorations on energy storage capability under various bending states and after 104 times of mechanical bending cycles.All these results indicate that BFMO-SBT on mica film capacitor has potential application in the future flexible electronics. | Changhong Yang Jin Qian Panpan Lv Haitao Wu Xiujuan Lin Kun Wang Jun Ouyang Shifeng Huang Xin Cheng Zhenxiang Cheng | 2020 | Journal of Materiomics2020,6,1: | 4 |
| 5 | Efficacy of Lidan Tang on high-fat-diet induced gallstone in mice and possible mechanism显示文摘OBJECTIVE:To investigate efficacy of Lidan Tang(LDT)on gallstone induced by high fat diet in mice,and to study its underlying mechanism.METHODS:Mice were fed with high fat diet every day and treated with LDT(9.01 times of human clinic dosage).Mice were randomly divided into 6 groups as control group,gallstone model group(high-fat diet),positive control ursodeoxycholic acid(UDCA)group(80 mg·kg^-1·d^-1,i.g.),LDT low dose group(6 kg/d,i.g.),LDT middle dose group(12 kg/d,i.g.),and LDT high dose group(24 kg/d,i.g.).The whole experiment was lasted for 4 weeks.The levels of ALT,AST,LDH,CHO,HDL-C and LDL-C in serum were measured,the pathological sections were observed by hematoxylin-eosin staining,the activities of antioxidant enzymes were measured by kits,and the proteins related to oxidative stress and lipid transport were detected by Western blot analysis.RESULTS:LDT could significantly reduce the contents of ALT and AST in serum and improve the pathological tissue of liver.LDT could significantly reduce the content of MDA and LPO,and increase the level of GSH and GSH-PX in liver tissue.The data of Western blot showed that LDT had antioxidant effect promoting Keap1/Nrf2 pathway and regulated the process of lipid transport,which was statistically significant.In addition,LDT treatment inhibited the expression of ATP-binding cassette transports ABCG5/8 in liver,and reduced cholesterol transport from the hepatocytes to the gallbladder.CONCLUSION:LDT has protective effect on gallstones induced by high fat diet in mice,which might be based on the protective effect on liver,including enhancing the antioxidant capacity of liver and reducing the production of lipid peroxides. | Gao Yan Liu Caipin Li Juntong Zhai Yingying Lin Meiyu Wu Qinglin Chu Shifeng Zhang Zhao Li Jianping Zhou Xin Li Yueting Chen Naihong | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 4 |

| 8 | Crystal structure and enhanced microwave dielectric properties of the Ce_(2)[Zr_(1−x)(Al_(1/2)Ta_(1/2))_(x)]_(3)(MoO_(4))_(9) ceramics at microwave frequency显示文摘Dense microwave dielectric ceramics of Ce_(2)[Zr_(1−x)(Al_(1/2)Ta_(1/2))_(x)]_(3)(MoO_(4))_(9)(CZMAT) (x = 0.02–0.10) were prepared by the conventional solid-state route. The effects of (Al1/2Ta1/2)^(4+) on their microstructures, sintering behaviors, and microwave dielectric properties were systematically investigated. On the basis of the X-ray diffraction (XRD) results, all the samples were matched well with Pr_(2)Zr_(3)(MoO_(4))_(9) structures, which belonged to the space group R3¯c. The lattice parameters were obtained using the Rietveld refinement method. The correlations between the chemical bond parameters and microwave dielectric properties were calculated and analyzed by using the Phillips—Van Vechten—Levine (P—V—L) theory. | 9 | Identification of berbamine dihydrochloride from barberry as an anti-adipogenic agent by high-content imaging assay显示文摘Objective:Lipid droplet(LD)deposition in adipose tissue is a critical factor leading to metabolic dysfunction.Various herbal medicines in traditional Chinese medicine(TCM)are used to treat hyperlipidemia,type 2 diabetes,obesity,and other diseases.The objective of this study was to identify potential anti-adipogenic agents from TCM herbal compounds.Methods:One hundred and twenty compounds were evaluated in terms of their effect on adipocyte differentiation through image-based high content screening.Anti-adipogenic effects of identified hits were further confirmed at various concentrations.In addition,drug-induced liver injury assay was performed with HepG2 cells to test the hepatotoxicity of hit compounds.Results:Berbamine(BBM),a chemical isolated from barberry,and a derivative of BBM,berbamine dihydrochloride(BBMD),reduced LDs formation by more than 50%.Dose-dependent effects were observed and the IC50 values of the two hits,BBM and BBMD,were determined as 1.88 mM and 0.95 mM,respectively.Moreover,BBM induced mild HepG2 cell injury,while its dihydrochloridedBBMD did not exhibit hepatotoxicity within 40 mM.Conclusion:This study demonstrates that BBMD may be a potential therapeutic candidate for disorders associated with elevated LDs accumulation. | Shifeng Wang Qiao Zhang Yuxin Zhang Yanling Zhang Qinghua Wu Shiyou Li Yanjiang Qiao | 2016 | Journal of Traditional Chinese Medical Sciences2016,3,2: | 2 |
| 10 | Utilization of Bayer Red Mud Derived from Bauxite for Belite-Ferroaluminate Cement Production显示文摘Bayer red mud(BRM)is a kind of industrial solid waste characterized by huge volume and high alkalinity.Its disposal generates serious environmental pollution and occupies a large number of farmland.The utilization and recycling of BRM is currently a crucial issue and needs to be addressed as soon as possible.The chemical composition of BRM is similar to cement clinker.In this study,the feasibility of preparing Belite-ferroaluminate clinker(BFAC)with different BRM was explored.The physical properties,mechanics performance,radioactivity levels and trace harmful metals leaching were measured.XRD,BEI and EDS were used to characterize the mineral formation,and SEM is used to reveal the solidified mechanism of trace harmful metal.The results show that the preparation of BFAC using a certain amount of BRM was feasible.The formed phases in clinkers mainly included C_(4)A_(3)Š,C_(2)S and C_(4)AF.The flexural strength and compressive strength of BFAC at 3 days increased whereas 28 and 90 days decreased with the increase of BRM due to the formation of higher C_(4)AF and lower C_(2)S.The formation of large amounts of Al_(2)O_(3)·3H_(2)O gel and Fe_(2)O_(3)·3H_(2)O gel in hydration products enhanced the adsorption capability to heavy metals and other ions.The trace harmful metal concentration in the leaching solution was much less than the upper limits.The radioactivity level of leaching solution was close to natural radioactive background.BRM is safe as raw material of BFAC. | Yanrong Zhao Ping Chen Shifeng Wang Yaxiong Ji Yuanhao Wang Bolin Wu Rongjin Liu | 2020 | Journal of Renewable Materials2020,8,11: | 2 |
| 11 | Differential response of multiple zebrafish hepatic F-box protein genes to 17α-ethinylestradiol treatment显示文摘Estrogens are accumulating in environment and their effects on a variety of reproductive processes and tumorigenesis were reported by previous study, but the mechanism of estrogen promoting neoplasia was still not clear. F-box protein (FBP) is the component of E3 ubiquitin ligase which takes part in a variety of key biological processes. In this study, using mature male zebrafish, which are more sensitive to estrogen treatment, we examined influence of 17α-ethinylestradiol (EE2) exposure on the expression of a series of hepatic FBP genes, which take part in a variety of biological processes, including tumorigenesis. The influence of EE2 on the expression of hepatic mRNA concentrations of FBP genes were quantified based on the expression of the optimal internal control gene in male zebrafish after 7-day exposure to EE2, from a low-dose concentration (1 ng/L) to environmentally relevant concentrations (10, 100 ng/L). Our results showed that EE2 exposure reduced the expression of fbxl14a, fbxl14b, fbxo25 and β-TRCP2b, but enchanced the expression of skp2. While the alterations in fbxl2, fbxw7, fbxo9, β-TRCP2a, fbxl18 and fbxo45 mRNA levels were not observed after EE2 exposure. Thus, our results showed that the expression of hepatic FBP genes exhibited differentially in male zebrafish exposed EE2.
