| 1 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. From the propagation model of malicious code, it is easy to find that quickly making peers get their patched and upgraded anti-virus system is the key way of immunization and damage control. To distribute patches and immune modules efficiently, a new exponential tree plus (ET+) and vaccine distribution algorithm based on ET+ are also proposed. | 5 | Seeing permeability from images: fast prediction with convolutional neural networks显示文摘Fast prediction of permeability directly from images enabled by image recognition neural networks is a novel pore-scale modeling method that has a great potential. This article presents a framework that includes (1) generation of porous media samples,(2) computation of permeability via fluid dynamics simulations,(3) training of convolutional neural networks (CNN) with simulated data, and (4) validations against simulations. Comparison of machine learning results and the ground truths suggests excellent predictive performance across a wide range of porosities and pore geometries, especially for those with dilated pores. Owning to such heterogeneity, the permeability cannot be estimated using the conventional Kozeny–Carman approach. Computational time was reduced by several orders of magnitude compared to fluid dynamic simulations. We found that, by including physical parameters that are known to affect permeability into the neural network, the physics-informed CNN generated better results than regular CNN. However, in high density flexible crossbar RRAM array, crosstalk issue that currents from the neighboring unselected cell lead to failure of write and read operations, still keeps a main bottleneck. Therefore, flexible selector compatible with the flexibility of the RRAM array should be focused on to configure one selector-one resistor(1 S1 R) system,which is immune to crosstalk issue. In this paper, flexible cation-based threshold switching(TS) selectors(Pt/Ag/Hf O_2/Pt/Ti/parylene) are fabricated and the compressive performance is studied systematically.The device shows excellent bidirectional volatile TS characteristics, including high selectivity ratio(10~9), low operating voltages(|VTH|<1 V), ultra-low leakage current(~10^(-13) A) and good flexibility. | 12 | Exploring the Degradation Potential of Halomonas Bacteria from Oil-contaminated Marine Environment显示文摘The degradation potential of a diesel-degrading bacteria(HDMP1) isolated from the oil-contaminated marine environment was explored systematically by analyzing the environmental conditions and synergistic action with other dieseldegrading bacteria. The result indicated that HDMP1 was confirmed as a strain of Halomonas by the method of strain identification. And, HDMP1 showed good diesel degradation performance with a diesel degradation rate of up to 79.59%after 7 days. By analyzing the effect of environmental conditions on HDMP1, the best carbon and nitrogen sources were found to be lactose and peptone, respectively, at a pH value of 7.5 and a salinity of 4 g/(100 mL). Additionally, the synergistic effect of HDMP1 combined with other diesel-degrading bacteria was analyzed by orthogonal experimental design. | 13 | Geochemistry and Geochronology of Diorite in Pengshan Area of Jiangxi Province:Implications for Magmatic Source and Tectonic Evolution of Jiangnan Orogenic Belt显示文摘Magmatic activities associated with tectonic events play a significant role in understanding the evolution of an orogenic belt.The Jiangnan orogenic belt has been regarded as the collisional suture zone between the Yangtze Block and the Cathaysia Block.Although the magmatic activities during the period of intra-plate extension after the collision have been well studied in recent years,some remaining issues,including source nature and geodynamic mechanism,need to be further addressed.In this paper,based on a detailed field geological,petrological,geochemical and geochronological study,we focus our work on diorites in the Pengshan area located at the northwestern margin of the Jiangnan orogenic belt.The mineral assemblages are mainly composed of plagioclase(55 vol.%-65 vol.%)and hornblende(35 vol.%-45 vol.%).One diorite sample yields zircon^(206)Pb/^(238)U mean age of 768±8 Ma(MSWD=0.29).The diorites have enriched large ion lithophile elements(Ba,K and Rb)and incompatible elements(Th and U),and are depleted in high field-strength elements including Ta,Ti and Nb.Diorites in this study have relatively high MgO content(6.56 wt.%-7.58 wt.%,7.07 wt.%on average)and Mg number values(65-67,65.8 on average).The diorites are metaluminous,high K calc-alkaline series rocks with high contents of K_2O(1.59 wt.%-1.97 wt.%)and total alkali(Na_2O+K_2O=5.56 wt.%-6.05 wt.%).The Nd/Th ratio(4.34-5.27)is higher than that of crust-derived rocks and lower than mantle-derived rocks.The Rb/Sr ratio(0.19-0.22)is slightly lower than crust,but significantly higher than upper mantle.Based on the above geochemical and geochronological analyses,we suggest that the diorites in the Pengshan area were mainly derived from crustal materials with a small amount of mantleoriginated materials involved,and possibly produced from an extensional tectonic setting after the collision between the Yangtze Block and Cathaysia Block. | Hongfeng Shi Junpeng Wang Yuan Yao Jing Zhang Song Jin Yingxin Zhu Kang Jiang Xiaolong Tian Deng Xiao Wenbin Ning | 2020 | Journal of Earth Science2020,31,1: | 2 |

| 17 | A comparison on the phytoremediation ability of triazophos by different macrophytes显示文摘The strategy of choosing suitable plants should receive great performance in phytoremediation of surface water polluted by triazophos(O,O-diethyl-O-(1-phenyl-1,2,4-triazol-3-base) sulfur phosphate, TAP), which is an organophosphorus pesticide widespread applied for agriculture in China and moderately toxic to higher animal and fish. The tolerance, uptake, transformation and removal of TAP by twelve species of macrophytes were examined in a hydroponic system and a comprehensive score(CS) of five parameters(relative growth rate(RGR), biomass, root/shoot ratio, removal capacity(RC), and bio-concentration factor(BCF)) by factor analysis was employed to screen the potential macrophyte species for TAP phytoremediation. The results showed that Thalia dealbata, Cyperus alternifolius, Canna indica and Acorus calamus had higher RGR values, indicating these four species having stronger growth capacity under TAP stress. The higher RC loading in Iris pseudacorus and Cyperus rotundus were 42.11 and 24.63 μg/(g fw·day), respectively. The highest values of BCF occurred in A. calamus(1.17), and TF occurred in Eichhornia crassipes(2.14). Biomass and root/shoot ratio of plant showed significant positive correlation with first-order kinetic constant of TAP removal in the hydroponic system, indicating that plant biomass and root system play important roles in remediation of TAP. | 18 | Natural astaxanthin enhanced antioxidant capacity and improved semen quality through the MAPK/Nrf2 pathway in aging layer breeder roosters显示文摘Background:Natural astaxanthin(ASTA)has strong antioxidant properties and has been widely used as a health product to improve human health.However,the effects of ASTA on the reproductive performance of aging roosters have been poorly studied.We aimed to investigate the effects of dietary ASTA on semen quality and antioxidant capacity in aging roosters and to explore the potential mechanism of semen quality change via antioxidation defense system.Methods:In the present study,9653-week-old Jinghong No.1 layer breeder roosters were fed a corn-soybean meal basal diet containing 0,25,50,or 100 mg/kg ASTA for 6 weeks.Results:Semen quality in the ASTA groups remarkably improved than that in the control group,and antioxidant activities,the abilities to scavenge hydroxyl radicals and superoxide anions,increased gradually with ASTA addition(P<0.05).In addition,the mRNA levels of antioxidant enzymes as well as the mRNA and protein levels of the mitogen-activated protein kinase(MAPK)and nuclear factor-erythroid 2-related factor 2(Nrf2)were markedly increased in the 50-100 mg/kg ASTA group(P<0.05).Conclusions:Collectively,these results demonstrate that dietary ASTA may improve semen quality by increasing antioxidant enzyme activities and the ability to scavenge hydroxyl radicals,which may be related to upregulation of the MAPK/Nrf2 pathway. | Shan Gao Nuo Heng Fang Liu Yong Guo Yu Chen Liang Wang Hemin Ni Xihui Sheng Xiangguo Wang Kai Xing Longfei Xiao Xiaolong Qi | 2022 | Journal of Animal Science and Biotechnology2022,13,2: | 2 |
