| 2 | Effects of nitrogen dioxide and its acid mist on reactive oxygen species production and antioxidant enzyme activity in Arabidopsis plants显示文摘Nitrogen dioxide(NO2) is one of the most common and harmful air pollutants. To analyze the response of plants to NO2 stress, we investigated the morphological change, reactive oxygen species(ROS) production and antioxidant enzyme activity in Arabidopsis thaliana(Col-0) exposed to 1.7, 4, 8.5, and 18.8 mg/m3NO2. The results indicate that NO2 exposure affected plant growth and chlorophyll(Chl) content, and increased oxygen free radical(O2-)production rate in Arabidopsis shoots. Furthermore, NO2 elevated the levels of lipid peroxidation and protein oxidation, accompanied by the induction of antioxidant enzyme activities and change of ascorbate(As A) and glutathione(GSH) contents. Following this, we mimicked nitric acid mist under experimental conditions, and confirmed the antioxidant mechanism of the plant to the stress. Our results imply that NO2 and its acid mist caused pollution risk to plant systems. | 3 | Effectiveness of Qingre Lishi Yishen decoction on the glomerular fibrosis of immunoglobulin A nephropathy in a rat’s model显示文摘OBJECTIVE: To investigate the effect of the clinical effective prescription of Qingre Lishi Yishen decoction(QRLS) on the activation of mesangial cells in immunoglobulin A nephropathy(IgAN) rats.METHODS: IgAN rat’s model was established by combine with intragastric administration of bovine serum albumin(BSA) + intravenous injection of lipopolysaccharide(LPS) by + subcutaneous injection of carbon tetrachloride(CCL4). Then the animals were randomly divided into four groups: control group, IgAN model group, IgAN model with Valsartan(Val) treatment group and IgAN model with QRLS treatment group. To observe the indexes of 24-h urine protein, renal function, deposition of immune complexes, expression of activation factor, fibrosis marker and inflammatory cytokines in four different groups.RESULTS: The Val or QRLS treatment group:(a) it reduced the immune complexes deposition of IgA in glomerular mesangial and inhibited mesangial cell proliferation;(b) it decreased the expression of smooth muscle actin(α-SMA), fibronectin(FN) and tumor necrosis factor alpha(TNF-α).CONCLUSION: The study suggested that QRLS ameliorate renal structure and function in IgAN rat’s model. | 11 | Study on screening potential allergenic proteins from infant milk powders based on human mast cell membrane chromatography and histamine release assays显示文摘Cow's milk allergy is mainly observed in infants and young children. Most allergic reactions affect the skin, followed by the gastrointestinal and respiratory systems. Conventional diagnosis is based on positive allergy studies and evaluation of parameters including IgE and IgG1 levels, acute allergic skin response and anaphylactic shock reactions. We developed a cell membrane chromatographic(CMC)method based on human mast cells(HMC-1) for screening potential allergens in infant formula milk powders(IFMP). HMC-1 cell membranes were extracted and mixed with silica to prepare cell membrane chromatography columns(10 mm ? 2 mm i.d., 5 mm). Under the conditions of 0.2 mL/min flow rate and214 nm detection wavelength, human breast milk showed no retention. However, IFMP showed clear retention. The retained fractions were collected and analyzed through matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS). Four major milk proteins, i.e., α-casein, β-casein, α-lactalbumin, and β-lactoglobulin A, were identified. Furthermore, these proteins and β-lactoglobulin B showed clear retention on HMC-1/CMC columns. To test the degranulation effects of the five proteins, histamine and β-hexosaminidase release assays were carried out. All five proteins induced HMC-1 cells to release histamine and β-hexosaminidase. Also, we established a reversed phase liquid chromatographic(RPLC) method for the determination of the five proteins in IFMP and the results showed that 90% proteins in IFMP were α-casein and β-casein. We concluded that cow's milk proteins may be potential allergens and caseins cause more β-casein allergic risk than other proteins. | 12 | Genesis of the Jiajika superlarge lithium deposit,Sichuan,China:constraints from He–Ar–H–O isotopes显示文摘The Jiajika granitic-and pegmatite-type lithium deposit,which is in the Songpan-Garze Orogenic Belt in western Sichuan Province,China,is the largest in Asia.Previous studies have examined the geochemistry and mineralogy of pegmatites and their parental source rocks to determine the genesis of the deposit.However,the evolution of magmatic-hydrothermal fluids has received limited attention.We analyzed He–Ar–H–O isotopes to decipher the ore-fluid nature and identify the contribution of fluids to mineralization in the late stage of crystallization differentiation.In the Jiajika ore field,two-mica granites,pegmatites(including common pegmatites and spodumene pegmatites),metasandstones,and schists are the dominant rock types exposed.Common pegmatites derived from early differentiation of the two-mica granitic magmas before they evolved into spodumene pegmatites during the late stage of the magmatic evolution.Common pegmatites have~3He/~4He ratios that vary from 0.18 to 4.68 Ra(mean1.62 Ra),and their~(40)Ar/~(36)Ar ratios range from 426.70 to 1408.06(mean 761.81);spodumene pegmatites have~3He/~4He ratios that vary from 0.18 to 2.66 Ra(mean 0.87Ra)and their~(40)Ar/~(36)Ar ratios range from 402.13 to 1907.34(mean 801.65).These data indicate that the hydrothermal fluids were shown a mixture of crust-and mantle-derived materials,and the proportion of crustderived materials in spodumene pegmatites increases significantly in the late stage of the magmatic evolution.Theδ~(18)OH_(2)O–VSMOWvalues of common pegmatites range from 6.2‰to 10.9‰,with a mean value of 8.6‰,andδDV–SMOWvalues vary from-110‰to-72‰,with a mean o f-85‰.Theδ~(18)OH_(2)O–VSMOWvalues of spodumene pegmatites range from 5.3‰to 13.2‰,with a mean of 9.1‰,andδDV–SMOWvalues vary from-115‰to-77‰,with a mean of-91‰.These data suggest that the ore-forming fluids came from primary magmatic water gradually mixing with more meteoric water in the late stage of the magmatic evolution.Based on the He–Ar–H–O and other existing data,we propose that the oreforming metals are mainly derived from the upper continental crust with a minor contribution from the mantle,and the fluid exsolution and addition of meteoric water during the formation of pegmatite contributed to the formation of the Jiajika superlarge lithium deposit. | Tao Liu Hai Wang Shihong Tian Denghong Wang Xianfang Li Xiaofang Fu Xuefeng Hao Yujie Zhang Kejun Hou | 2023 | Acta Geochimica2023,42,3: | 0 |
| 13 | An antigen self-assembled and dendritic cell-targeted nanovaccine for enhanced immunity against cancer显示文摘The rise of nanotechnology has opened new horizons for cancer immunotherapy.However,most nano vaccines fabricated with nanomaterials suffer from carrier-related concerns,including low drug loading capacity,unpredictable metabolism,and potential systemic toxicity,which bring obstacles for their clinical translation.Herein,we developed an antigen self-assembled nanovaccine,which was resulted from a simple acryloyl modification of the antigen to induce self-assembly.Furthermore,a dendritic cell targeting head mannose monomer and a mevalonate pathway inhibitor zoledronic acid(Zol)were integrated or absorbed onto the nanoparticles(denoted as MEAO-Z)to intensify the immune response.The synthesized nano vaccine with a diameter of around 70 nm showed successful lymph node transportation,high dendritic cell internalization,promoted costimulatory molecule expression,and preferable antigen cross-presentation.In virtue of the above superiorities,MEAO-Z induced remarkably higher titers of serum antibody,stronger cytotoxic T lymphocyte immune responses and IFN-γsecretion than free antigen and adjuvants.In vivo,MEAO-Z significantly suppressed EG7-OVA tumor groth and prolonged the survival time of tumor-bearing mice.These results indicated the translation promise of our self-assembled nano vaccine for immune potentiation and cancer immunotherapy. | Yunting Zhang Min Jiangy Guangsheng Du Xiaofang Zhong Chunting He Ming Qin Yingying Hou Rong Liu Xun Sun | 2023 | Acta Pharmaceutica Sinica B2023,13,8: | 0 |
