1Diversity and redundancy of the ripening regulatory networks revealed by the fruitENCODE and the new CRISPR/Cas9 CNR and NOR mutants显示文摘Tomato is considered as the genetic model for climacteric fruits,in which three major players control the fruit ripening process:ethylene,ripening transcription factors,and DNA methylation.The fruitENCODE project has now shown that there are multiple transcriptional circuits regulating fruit ripening in different species,and H3K27me3,instead of DNA methylation,plays a conserved role in restricting these ripening pathways.In addition,the function of the core tomato ripening transcription factors is now being questioned.We have employed CRISPR/Cas9 genome editing to mutate the SBP-CNR and NAC-NOR transcription factors,both of which are considered as master regulators in the current tomato ripening model.These plants only displayed delayed or partial non-ripening phenotypes,distinct from the original mutant plants,which categorically failed to ripen,suggesting that they might be gain-of-function mutants.Besides increased DNA methylation genome-wide,the original mutants also have hyper-H3K27me3 in ripening gene loci such as ACS2,RIN,and TDR4.It is most likely that multiple genetic and epigenetic factors have contributed to their strong non-ripening phenotypes.Hence,we propose that the field should move beyond these linear and twodimensional models and embrace the fact that important biological processes such as ripening are often regulated by highly redundant network with inputs from multiple levels.Ying Gao Ning Zhu Xiaofang Zhu Meng Wu Cai-Zhong Jiang Donald Grierson Yunbo Luo Wei Shen Silin Zhong Da-Qi Fu Guiqin Qu 2019Horticulture Research2019,6,1:7
2Isolation and oral immunogenicity assessment of porcine epidemic diarrhea virus NH-TA2020 strain:One of the predominant strains circulating in China from 2017 to 2021显示文摘Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is one of the most devastating diseases in the global pig industry due to its high mortality rate in piglets.Maternal vaccines can effectively enhance the gut-mammary gland-secretory IgA axis to boost lactogenic immunity and passive protection of nursing piglets against PEDV challenge.From 2017 to 2021,we collected 882 diarrhea samples from 303 farms in China to investigate the epidemiology of PEDV.The result showed that about 52.15%(158/303)of the farms were positive for PEDV with an overall detection rate of 63.95%(564/882)of the samples.The S1 fragments of S gene from 104 strains were sequenced for the phylogenetic analysis.A total of 71 PEDV strains(68.27%)sequenced in this study were clustered into the predominant G2c subgroup,while the newly-defined G2d strains(9.62%)were identified in three provinces of China.The NH-TA2020 strain of G2c subgroup was isolated and cultured,and its infection to piglets caused watery diarrhea within 24 h,indicating its strong pathogenicity.Oral administration of NH-TA2020 strain to pregnant gilts stimulated high levels of IgA antibody in colostrum.The piglets fed by the gilts above were challenged with NH-TA2020 strain or CH-HeB-RY-2020 strain from G2d subgroup,and the clinical symptoms and virus shedding were significantly reduced compared to the mock group.Our findings suggest that G2c subgroup is the predominant branch circulating in China from 2017 to 2021.Oral administration of NH-TA2020 enhances maternal IgA and lactogenic immune responses,which confer protection against the homologous and emerging G2d PEDV strains challenges in neonates.Xiaowen Li Yang Li Jiapei Huang Yali Yao Wenying Zhao Yunjing Zhang Jie Qing Jing Ren Zhong Yan Zewei Wang Xiaofang Hu Duli Kang Hongqiang Liu Zhichun Yan 2022Virologica Sinica2022,37,5:6
3Generality and characteristics of genetic and epigenetic changes in newly synthesized allotetraploid wheat lines显示文摘Previous studies have shown rapid and extensive genomic instability associated with early stages of allopolyploidization in wheat.However,these studies are based on either a few pre-selected genomic loci or genome-wide analysis of a single plant individual for a given cross combination,thus making the extent and generality of the changes uncertain.To further study the generality and characteristics of allopolyploidization-induced genomic instability in wheat,we investigated genetic and epigenetic changes from a genome-wide perspective(by using the AFLP and MSAP markers) in four sets of newly synthesized allotetraploid wheat lines with various genome constitutions,each containing three randomly chosen individual plants at the same generation.We document that although general chromosomal stability was characteristic of all four sets of allotetraploid wheat lines,genetic and epigenetic changes at the molecular level occurred in all these plants,with both kinds of changes classifiable into two distinct categories,i.e.,stochastic and directed.The abundant type of genetic change is loss of parental bands while the prevalent cytosine methylation pattern alteration is hypermethylation at the CHG sites.Our results have extended previous studies regarding allopolyploidization-induced genomic dynamics in wheat by demonstrating the generality of both genetic and epigenetic changes associated with multiple nascent allotetraploid wheat lines,and providing novel insights into the characteristics of the two kinds of induced genomic instabilities.Bao Qi 1,Xiaofang Zhong 1,Bo Zhu,Na Zhao,Liying Xu,Huakun Zhang,Xiaoming Yu,Bao Liu Key Laboratory of Molecular Epigenetics of MOE and Institute of Genetics & Cytology,Northeast Normal University,Changchun 130024,China 2010Journal of Genetics and Genomics2010,37,11:5

5In situ investigation of SiC powder's microwave sintering by SR-CT technique显示文摘Microwave sintering is being developed as a novel technique for the preparation of dense structural ceramics,but the mature theory has not been established due to the technical difficulties.The synchrotron radiation X-ray computed tomography(SR-CT) technique was introduced for the first time into the study of microwave sintering to in-situ observe the microstructure evolution of silicon carbide(SiC) material in this paper.By applying the SR-CT technique,the reconstructed 2D and 3D images of the specimen were obtained and the double logarithm curve of mean neck size and time(Ln(x)-Ln(t)) were obtained from these reconstructed images.Various sintering phenomena including sintering neck growth during microwave treatment were observed from the reconstructed images.Furthermore,the differences in microstructure evolution and sintering kinetics between microwave and conventional sintering were analyzed based on the reconstructed images and the Ln(x)-Ln(t) curve.1) The sharp surface of grains near the contact region distinctly grew blunt and the sintering neck growth between these grains were obviously observed at the early stage.Besides,the larger particles grew faster than smaller ones.The main reason for these phenomena may be the micro-focusing effect of electric fields.2) During each of the three sintering stages,the sintering kinetics curve of double logarithm relationship between mean neck size and time shows a good linear relationship,but at the middle stage the slope of the curve increases dramatically,which is quite larger than conventional sintering.The preliminary interpretation for these extraordinary phenomena has been discussed in details.LI YongCun XU Feng HU XiaoFang QU HongYan MIAO Hong ZHANG Zhong XIAO TiQiao 2011Science China(Technological Sciences)2011,54,6:4

17Gut microbiome,cognitive function and brain structure:a multi-omics integration analysis显示文摘Background:Microbiome-gut-brain axis may be involved in the progression of age-related cognitive impairment and relevant brain structure changes,but evidence from large human cohorts is lacking.This study was aimed to investigate the associations of gut microbiome with cognitive impairment and brain structure based on multi-omics from three independent populations.Methods:We included 1430 participants from the Guangzhou Nutrition and Health Study(GNHS)with both gut microbiome and cognitive assessment data available as a discovery cohort,of whom 272 individuals provided fecal samples twice before cognitive assessment.We selected 208 individuals with baseline microbiome data for brain magnetic resonance imaging during the follow-up visit.Fecal 16S rRNA and shotgun metagenomic sequencing,tar-geted serum metabolomics,and cytokine measurements were performed in the GNHS.The validation analyses were conducted in an Alzheimer’s disease case-control study(replication study 1,n=90)and another community-based cohort(replication study 2,n=1300)with cross-sectional dataset.Results:We found protective associations of specific gut microbial genera(Odoribacter,Butyricimonas,and Bac-teroides)with cognitive impairment in both the discovery cohort and the replication study 1.Result of Bacteroides was further validated in the replication study 2.Odoribacter was positively associated with hippocampal volume(β,0.16;95%CI 0.06-0.26,P=0.002),which might be mediated by acetic acids.Increased intra-individual alterations in gut microbial composition were found in participants with cognitive impairment.We also identified several serum metabolites and inflammation-associated metagenomic species and pathways linked to impaired cognition.Conclusions:Our findings reveal that specific gut microbial features are closely associated with cognitive impair-ment and decreased hippocampal volume,which may play an important role in dementia development.Xinxiu Liang Yuanqing Fu Wen-ting Cao Zhihong Wang Ke Zhang Zengliang Jiang Xiaofang Jia Chun-ying Liu Hong-rou Lin Haili Zhong Zelei Miao Wanglong Gou Menglei Shuai Yujing Huang Shengdi Chen Bing Zhang Yu-ming Chen Ju-Sheng Zheng 2022Translational Neurodegeneration2022,11,1:1
