| 3 | Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats. | Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang | 2022 | Science China(Life Sciences)2022,65,10: | 5 |

| 5 | Differential transcriptome analysis between Populus and its synthesized allotriploids driven by second-division restitution显示文摘In this report, we compared transcriptomic differences between a synthetic Populus section Tacamahaca triploid driven by second-division restitution and its parents using a high-throughput RNA-seq method. A total of 4,080 genes were differentially expressed between the high-growth vigor allotriploids(SDR-H) and their parents, and 719 genes were non-additively expressed in SDR-H. Differences in gene expression between the allotriploid and male parent were more signi ficant than those between the allotriploid and female parent, which may be caused by maternal effects. We observed3,559 differentially expressed genes(DEGs) between the SDR-H and male parent. Notably, the genes were mainly involved in metabolic process, cell proliferation, DNA methylation, cell division, and meristem and developmental growth. Among the1,056 DEGs between SDR-H and female parent, many genes were associated with metabolic process and carbon utilization.In addition, 1,789 DEGs between high- and low-growth vigor allotriploid were mainly associated with metabolic process,auxin poplar transport, and regulation of meristem growth.Our results indicated that the higher poplar ploidy level can generate extensive transcriptomic diversity compared with its parents. | 7 | Do trace metal(loid)s in road soils pose health risks to tourists?A case of a highly-visited national park in China显示文摘Nowadays,more people tend to spend their recreational time in large national parks,and trace metal(loid)s in soils have attracted long-term attention due to their possible harm to human health.To investigate the pollution levels,potential sources and health risks of trace metal(loid)s in road soils,a total of eight trace metal(loid)s(including As,Cd,Cr,Cu,Ni,Pb,Zn and Hg)from 47 soil samples along roads were studied in the Huangshan National Park in Southeast China.The results showed that the concentrations of As,Cd,Pb,Zn and Hg appeared different degrees of pollution compared with their corresponding background values.According to the pollution indices,Hg and Cd were recognized as significant pollutants presenting moderate to high ecological risk.Combining principal component analysis and positive matrix factorization model,the results showed that traffic,industrial,agricultural and natural sources were the potential origins of trace metal(loid)s in this area,with contribution rates of 39.93%,25.92%,10.53%and 23.62%,respectively.Non-carcinogenic risks were all negligible,while the carcinogenic risk of As was higher than the limit(1×10^(-6)).Moreover,children were more susceptible to trace metal(loid)s by ingestion which appeared to be a more important exposure pathway than dermal contact and inhalation.The contribution rates of different sources to non-carcinogenic risks and carcinogenic risks were similar among children and adults,while traffic and industrial sources have a significant impact on health risks.This study will give more insights to control the environmental risks of trace metal(loid)s in national parks. | Jingling Huang Yuying Wu Yanyao Li Jiaxun Sun Yujing Xie Zhengqiu Fan | 2022 | Journal of Environmental Sciences2022,34,1: | 2 |

| 10 | Human cytomegalovirus miR-US5-1 inhibits viral replication by targeting Geminin mRNA显示文摘Viruses commonly create favorable cellular conditions for their survival through multiple mechanisms. Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. | 12 | Efficient catalytic hydrogen generation by intermetallic platinum-lead nanostructures with highly tunable porous feature显示文摘The water-gas shift(WGS) reaction is an essential industrial reaction for upgrading hydrogen(H2) by removing carbon monoxide(CO), while highly efficient platinum(Pt)-based catalysts for WGS with simultaneously high activity and stability are still yet to be developed due to the poisoning issue during the reaction. Herein, we report on the porous PtPb peanut nanocrystals(porous PtPb PNCs) and porous PtPb octahedron nanocrystals(porous PtPb ONCs) with controllable ratios of Pt/Pb as extremely active and stable catalysts towards WGS reaction. It exhibits the composition-dependent activity with porous PtPb PNCs-40/ZnO being the most active for WGS to H_2, 16.9 times higher than that of the commercial Pt/C. The porous PtPb PNCs-40/ZnO also display outstanding durability with barely activity decay and negligible structure and composition changes after ten successive reaction cycles. X-ray photoelectron spectroscopy(XPS) results reveal that the suitable binding energy of Pt 4f_(7/2) and the high ratio of Pt(0)to Pt(II) in porous PtPb PNCs/ZnO and porous PtPb ONCs/ZnO are crucial for the enhanced WGS activity.The CO stripping results indicate the optimized CO adsorption strength on the Pt surface ensure the excellent WGS activity and the outstanding durability.
