| 1 | Exercise-induced irisin in bone and systemic irisin administration reveal new regulatory mechanisms of bone metabolism显示文摘Irisin is a polypeptide hormone derived from the proteolytic cleavage of fibronectin-type III domaincontaining 5(FNDC5) protein. Once released to circulation upon exercise or cold exposure, irisin stimulates browning of white adipose tissue(WAT) and uncoupling protein 1(UCP1) expression, leading to an increase in total body energy expenditure by augmented UCP1-mediated thermogenesis. It is currently unknown whether irisin is secreted by bone upon exercise or whether it regulates bone metabolism in vivo. In this study, we found that 2 weeks of voluntary wheel-running exercise induced high levels of FNDC5 messenger RNA as well as FNDC5/irisin protein expression in murine bone tissues. Increased immunoreactivity due to exercise-induced FNDC5/irisin expression was detected in different regions of exercised femoral bones,including growth plate, trabecular bone, cortical bone, articular cartilage, and bone–tendon interface. Exercise also increased expression of osteogenic markers in bone and that of UCP1 in WAT, and led to bodyweight loss. Irisin intraperitoneal(IP) administration resulted in increased trabecular and cortical bone thickness and osteoblasts numbers, and concurrently induced UCP1 expression in subcutaneous WAT. Lentiviral FNDC5 IP administration increased cortical bone thickness. In vitro studies in bone cells revealed irisin increases osteoblastogenesis and mineralization, and inhibits receptor activator of nuclear factor-k B ligand(RANKL)-induced osteoclastogenesis. | 3 | Xuanbai Chengqi decoction(宣白承气汤) plus Western Medicine in treatment of severe pneumonia with symptom pattern of phlegm-heat obstructing lung:a Meta-analysis显示文摘OBJECTIVE: To evaluate the effectiveness and safety of Xuanbai Chengqi decoction(宣白承气汤, XBCQD) plus Western Medicine(WM) in treatment of severe pneumonia with symptom pattern of phlegm-heat obstructing lung.METHODS: Randomized controlled trials(RCTs) investigating the effect of XBCQD on severe pneumonia with symptom pattern of phlegm-heat obstructing lung, were included in this study. Seven electronic databases were searched up to March 2019.Meta-analysis was conducted by Review Manager5.3 software. Risk ratio(RR) and mean difference(MD) with 95% confidence interval(CI) were used as effect estimation.RESULTS: Eleven RCTs were included, involving992 participants. Meta-analysis showed that XBCQD combined with WM achieved better effectiveness than WM alone in terms of total effective rate[RR = 1.23, 95%CI(1.16, 1.30)], clinical pulmonary infection score [CPIS, MD =-2.02, 95%CI(-2.42,-1.63)], acute physiology and chronic health evaluation Ⅱ [APACHE Ⅱ, MD =-6.81, 95% CI(-8.26,5.37)], mechanical ventilation time [MD =-101.41,95%CI(-140.47,-62.34)], and lactic acid content in arterial blood [MD =-2.41, 95%CI(-2.64,-2.18)].CONCLUSION: XBCQD combined with WM had better benefit than WM alone to the patients of severe pneumonia with the symptom pattern of phlegm-heat obstructing lung. | 5 | Nash Model Parameter Uncertainty Analysis by AM-MCMC Based on BFS and Probabilistic Flood Forecasting显示文摘A hydrologic model consists of several parameters which are usually calibrated based on observed hy-drologic processes. Due to the uncertainty of the hydrologic processes, model parameters are also uncertain, which further leads to the uncertainty of forecast results of a hydrologic model. Working with the Bayesian Forecasting System (BFS), Markov Chain Monte Carlo simulation based Adaptive Metropolis method (AM-MCMC) was used to study parameter uncertainty of Nash model, while the probabilistic flood forecasting was made with the simu-lated samples of parameters of Nash model. The results of a case study shows that the AM-MCMC based on BFS proposed in this paper is suitable to obtain the posterior distribution of the parameters of Nash model according to the known information of the parameters.
