| 1 | Restoration of electrical microenvironment enhances bone regeneration under diabetic conditions by modulating macrophage polarization显示文摘Macrophage-mediated inflammation compromises bone repair in diabetic patients.Electrical signaling cues are known to regulate macrophage functions.However,the biological effects of electrical microenvironment from charged biomaterials on the immune response for regulating osteogenesis under diabetic conditions remain to be elucidated.Herein the endogeneous electrical microenvironment of native bone tissue was recapitulated by fabricating a ferroelectric BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membrane.In vitro,the polarized BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membranes inhibited high glucose-induced M1-type inflammation,by effecting changes in cell morphology,M1 marker expression and pro-inflammatory cytokine secretion in macrophages.This led to enhanced osteogenic differentiation of human bone marrow mesenchymal stem cells(BM-MSCs).In vivo,the biomimetic electrical microenvironment recapitulated by the polarized nanocomposite membranes switched macrophage phenotype from the pro-inflammatory(M1)into the pro-healing(M2)phenotype,which in turn enhanced bone regeneration in rats with type 2 diabetes mellitus.Mechanistic studies revealed that the biomimetic electrical microenvironment attenuated pro-inflammatory M1 macrophage polarization under hyperglycemic conditions by suppressing expression of AKT2 and IRF5 within the PI3K-AKT signaling pathway,thereby inducing favorable osteo-immunomodulatory effects.Our study thus provides fundamental insights into the biological effects of restoring the electrical microenvironment conducive for osteogenesis under DM conditions,and offers an effective strategy to design functionalized biomaterials for bone regeneration therapy in diabetic patients. | Xiaohan Dai Boon Chin Heng Yunyang Bai Fuping You Xiaowen Sun Yiping Li Zhangui Tang Mingming Xu Xuehui Zhang Xuliang Deng | 2021 | Bioactive Materials2021,6,7: | 9 |

| 5 | lncR-GAS5 upregulates the splicing factor SRSF10 to impair endothelial autophagy,leading to atherogenesis显示文摘Long noncoding RNAs(lncRNAs)play a critical role in the regulation of atherosclerosis.Here,we investigated the role of the lncRNA growth arrest-specific 5(lncR-GAS5)in atherogenesis.We found that the enforced expression of lncR-GAS5 contributed to the development of atherosclerosis,which presented as increased plaque size and reduced collagen content.Moreover,impaired autophagy was observed,as shown by a decreased LC3II/LC3I protein ratio and an elevated P62 level in lncR-GAS5-overexpressing human aortic endothelial cells.By contrast,lncR-GAS5 knockdown promoted autophagy.Moreover,serine/arginine-rich splicing factor 10(SRSF10)knockdown increased the LC3II/LC3I ratio and decreased the P62 level,thus enhancing the formation of autophagic vacuoles,autolysosomes,and autophagosomes.Mechanistically,lncR-GAS5 regulated the downstream splicing factor SRSF10 to impair autophagy in the endothelium,which was reversed by the knockdown of SRSF10.Further results revealed that overexpression of the lncR-GAS5-targeted gene miR-193-5p promoted autophagy and autophagic vacuole accumulation by repressing its direct target gene,SRSF10.Notably,miR-193-5p overexpression decreased plaque size and increased collagen content.Altogether,these findings demonstrate that lncR-GAS5 partially contributes to atherogenesis and plaque instability by impairing endothelial autophagy.In conclusion,lncR-GAS5 overexpression arrested endothelial autophagy through the miR-193-5p/SRSF10 signaling pathway.Thus,miR-193-5p/SRSF10 may serve as a novel treatment target for atherosclerosis. | Yuhua Fan Yue Zhang Hongrui Zhao Wenfeng Liu Wanqing Xu Lintong Jiang Ranchen Xu Yue Zheng Xueqing Tang Xiaohan Li Limin Zhao Xin Liu Yang Hong Yuan Lin Hui Chen Yong Zhang | 2023 | Frontiers of Medicine2023,17,2: | 1 |

| 7 | Expanding the application of a UV-visible reporter for transient gene expression and stable transformation in plants显示文摘Green fl uorescent protein(GFP)has been widely used for monitoring gene expression and protein localization in diverse organisms.However,highly sensitive imaging equipment,like fl uorescence microscope,is usually required for the visualization of GFP,limitings its application to fi xed locations in samples.A reporter that can be visualized in realtime regardless the shape,size and location of the target samples will increase the fl exibility and ef fi ciency of research work.Here,we report the application of a GFP-like protein,called eYGFPuv,in both transient expression and stable transformation,in two herbaceous plant species(Arabidopsis and tobacco)and two woody plant species(poplar and citrus).We observed bright fl uorescence under UV light in all of the four plant species without any effects on plant growth or development.eYGFPuv was shown to be effective for imaging transient expression in leaf and root tissues.With a focus on in vitro transformation,we demonstrated that the transgenic events expressing 1x eYGFPuv could be easily identi fi ed visually during the callus stage and the shoot stage,enabling early and ef fi cient selection of transformants.Furthermore,whole-plant level visualization of eYGFPuv revealed its ubiquitous stability in transgenic plants.In addition,our transformation experiments showed that eYGFPuv can also be used to select transgenic plants without antibiotics.This work demonstrates the feasibility of utilizing 1x eYGFPuv in studies of gene expression and plant transformation in diverse plants. | Guoliang Yuan Haiwei Lu Dan Tang Md Mahmudul Hassan Yi Li Jin-Gui Chen Gerald A.Tuskan Xiaohan Yang | 2021 | Horticulture Research2021,8,1: | 1 |
| 8 | Associations of insulin resistance and beta-cell function with abnormal lipid profile in newly diagnosed diabetes显示文摘Background:Abnormal lipids are strong predictors of cardiovascular disease in type 1 diabetes mellitus(T1DM)and type 2 diabetes mellitus(T2DM).However,the potential associations of insulin resistance(IR)and beta-cell function(BCF)with abnormal lipids in newly diagnosed T1DM or T2DM patients are not fully understood.Methods:A cross-sectional survey of 15,928 participants was conducted.Homeostasis model assessment and postprandial C-peptide levels were used to estimate IR and BCF.A restricted cubic spline(RCS)nested in binary logistic regression was used to examine the associations of IR and BCF with abnormal lipids.Results:High triglyceride(TG),low high-density lipoprotein cholesterol,and high low-density lipoprotein cholesterol(LDL-C)accounted for 49.7%,47.8%,and 59.2%of the participants,respectively.In multivariable analysis,high IR was associated with an increased risk of high TGs(P for trend<0.001)in T1DM and is associated with an elevated risk of high TG and low HDL-C(all P for trend<0.01)in T2DM.Low BCF was not associated with risks of dyslipidemia in patients with T1DM or T2DM after adjustment for potential confounders.Conclusion:High IR had different associations with the risk of dyslipidemia in newly diagnosed T1DM and T2DM patients,suggesting that early treatment that improves IR may benefit abnormal lipid metabolism. | Xiaohan Tang Xiang Yan Houde Zhou Gan Huang Xiaohong Niu Hongwei Jiang Heng Su Xilin Yang Xia Li Zhiguang Zhou | 2022 | Chinese Medical Journal2022,135,21: | 1 |
| 9 | UltraStar:A Lightweight Simulator of Ultra-Dense LEO Satellite Constellation Networking for 6G显示文摘The mega-constellation network has gained significant attention recently due to its great potential in providing ubiquitous and high-capacity connectivity in sixth-generation(6G)wireless communication systems.However,the high dynamics of network topology and large scale of mega-constellation pose new challenges to the constellation simulation and performance evaluation.In this paper,we introduce UltraStar,a lightweight network simulator,which aims to facilitate the complicated simulation for the emerging mega-constellation of unprecedented scale.Particularly,a systematic and extensible architecture is proposed,where the joint requirement for network simulation,quantitative evaluation,data statistics and visualization is fully considered.For characterizing the network,we make lightweight abstractions of physical entities and models,which contain basic representatives of networking nodes,structures and protocol stacks.Then,to consider the high dynamics of Walker constellations,we give a two-stage topology maintenance method for constellation initialization and orbit prediction.Further,based on the discrete event simulation(DES)theory,a new set of discrete events is specifically designed for basic network processes,so as to maintain network state changes over time.Finally,taking the first-generation Starlink of 11927 low earth orbit(LEO)satellites as an example,we use UltraStar to fully evaluate its network performance for different deployment stages,such as characteristics of constellation topology,performance of end-to-end service and effects of network-wide traffic interaction.The simulation results not only demonstrate its superior performance,but also verify the effectiveness of UltraStar. | Xiaoyu Liu Ting Ma Zhixuan Tang Xiaohan Qin Haibo Zhou Xuemin(Sherman)Shen | 2023 | IEEE/CAA Journal of Automatica Sinica2023,10,3: | 1 |

| 15 | Combination therapy with saxagliptin and vitamin D for the preservation ofβ-cell function in adult-onset type 1 diabetes:a multi-center,randomized,controlled trial显示文摘Disease modifying therapies aiming to preserveβ-cell function in patients with adult-onset autoimmune type 1 diabetes are lacking.Here,we conducted a multi-centre,randomized,controlled trial to assess theβ-cell preservation effects of saxagliptin alone and saxagliptin combined with vitamin D as adjunctive therapies in adult-onset autoimmune type 1 diabetes.In this 3-arm trial,301 participants were randomly assigned to a 24-month course of the conventional therapy(metformin with or without insulin)or adjunctive saxagliptin or adjunctive saxagliptin plus vitamin D to the conventional therapy.The primary endpoint was the change from baseline to 24 months in the fasting C-peptide.The secondary endpoints included the area under the concentration-time curve(AUC)for C-peptide level in a 2-h mixed-meal tolerance test,glycemic control,total daily insulin use and safety,respectively.The primary endpoint was not achieved in saxagliptin plus vitamin D group(P=0.18)and saxagliptin group(P=0.26).However,compared with the conventional therapy,2-h C-peptide AUC from 24 months to baseline decreased less with saxagliptin plus vitamin D(-276 pmol/L vs.-419 pmol/L;P=0.01),and not to the same degree with saxagliptin alone(-314 pmol/L;P=0.14).Notably,for participants with higher glutamic acid decarboxylase antibody(GADA)levels,the decline ofβ-cell function was much lower in saxagliptin plus vitamin D group than in the conventional therapy group(P=0.001).Insulin dose was significantly reduced in both active treatment groups than in the conventional therapy group despite all groups having similar glycemic control.In conclusion,the combination of saxagliptin and vitamin D preserves pancreaticβ-cell function in adult-onset autoimmune type 1 diabetes,an effect especially efficacious in individuals with higher GADA levels.Our results provide evidence for a novel adjunct to insulin and metformin as potential initial treatment for adult-onset type 1 diabetes.(ClinicalTrials.gov identifier:NCT02407899). | Xiang Yan Xia Li Bingwen Liu Jiaqi Huang Yufei Xiang Yuhang Hu Xiaohan Tang Ziwei Zhang Gan Huang Zhiguo Xie Houde Zhou Zhenqi Liu Xiangbing Wang Richard David Leslie Zhiguang Zhou | 2023 | Signal Transduction and Targeted Therapy2023,8,5: | 0 |
