4Development of a Drilling and Coring Test-bed for Lunar Subsurface Exploration and Preliminary Experiments显示文摘Drill sampling has been widely employed as an effective way to acquire deep samples in extraterrestrial exploration. A novel sampling method, namely, flexible-tube coring, was adopted for the Chang'e mission to acquire drilling cores without damaging stratification information. Since the extraterrestrial environment is uncertain and different from the terrestrial environment, automated drill sampling missions are at risk of failure. The principles of drilling and coring for the lunar subsurface should be fully tested and verified on earth before launch. This paper proposes a test-bed for conducting the aforementioned experiments on earth. The test-bed comprises a rotary-percussive drilling mechanism, penetrating mechanism, drilling medium container, and signal acquisition and control system. For granular soil, coring experiments indicate that the sampling method has a high coring rate greater than 80%. For hard rock, drilling experiments indicate that the percussive frequency greatly affects the drilling efficiency. A multi-layered simulant composed of granular soil and hard rock is built to test the adaptability of drilling and coring. To tackle complex drilling media, an intelligent drilling strategy based on online recognition is proposed to improve the adaptability of the sampling drill. 5Building a sequence map of the pig pan-genome from multiple de novo assemblies and Hi-C data显示文摘Pigs were domesticated independently in the Near East and China,indicating that a single reference genome from one individual is unable to represent the full spectrum of divergent sequences in pigs worldwide.Therefore,12 de novo pig assemblies from Eurasia were compared in this study to identify the missing sequences from the reference genome.As a result,72.5 Mb of nonredundant sequences(~3% of the genome)were found to be absent from the reference genome(Sscrofa11.1)and were defined as pan-sequences.Of the pan-sequences,9.0 Mb were dominant in Chinese pigs,in contrast with their low frequency in European pigs.One sequence dominant in Chinese pigs contained the complete genic region of the tazarotene-induced gene 3(TIG3)gene which is involved in fatty acid metabolism.Using flanking sequences and Hi-C based methods,27.7% of the sequences could be anchored to the reference genome.The supplementation of these sequences could contribute to the accurate interpretation of the 3D chromatin structure.A web-based pan-genome database was further provided to serve as a primary resource for exploration of genetic diversity and promote pig breeding and biomedical research.Xiaomeng Tian Ran Li Weiwei Fu Yan Li Xihong Wang Ming Li Duo Du Qianzi Tang Yudong Cai Yiming Long Yue Zhao Mingzhou Li Yu Jiang 2020Science China(Life Sciences)2020,63,5:7
6Multi-state autonomous drilling for lunar exploration显示文摘Due to the lack of information of subsurface lunar regolith stratification which varies along depth, the drilling device may encounter lunar soil and lunar rock randomly in the drilling process. To meet the load safety requirements of unmanned sampling mission under limited orbital resources, the control strategy of autonomous drilling should adapt to the indeterminable lunar environments. Based on the analysis of two types of typical drilling media(i.e., lunar soil and lunar rock), this paper proposes a multi-state control strategy for autonomous lunar drilling. To represent the working circumstances in the lunar subsurface and reduce the complexity of the control algorithm, lunar drilling process was categorized into three drilling states: the interface detection, initiation of drilling parameters for recognition and drilling medium recognition. Support vector machine(SVM) and continuous wavelet transform were employed for the online recognition of drilling media and interface, respectively. Finite state machine was utilized to control the transition among different drilling states. To verify the effectiveness of the multi-state control strategy, drilling experiments were implemented with multi-layered drilling media constructed by lunar soil simulant and lunar rock simulant. 7Effects of surfactants on the combined toxicity of TiO_2 nanoparticles and cadmium to Escherichia coli显示文摘The combined ecological toxicity of TiO_2 nanoparticles(nano-TiO_2) and heavy metals has been paid more attention. As the common pollutants in water environment, surfactants could affect the properties of nanoparticles and heavy metals, and thus further influence the combined toxicity of nano-TiO_2 and heavy metals. In this study, the effects of sodium dodecyl benzene sulfonate(SDBS) and Tween 80 on the single and combined toxicities of Cd^(2+) and nano-TiO_2 to Escherichia coli(E. coli) were examined, and the underlying influence mechanism was further discussed. The results showed both SDBS and Tween 80 enhanced the toxicity of Cd^(2+) to E. coli in varying degrees. The reaction of SDBS and Cd^(2+) could increase the outer membrane permeability and the bioavailability of Cd, while Tween 80 itself could enhance the outer membrane permeability. The combined toxicity of nano-TiO_2 and Cd^(2+) to E. coli in absence of surfactant was antagonistic because of the adsorption of Cd^(2+) to nanoTiO_2 particles. However, in the presence of SDBS, both SDBS and nano-TiO_2 influenced the toxicity of Cd^(2+) , and also SDBS could adsorb to nano-TiO_2 by binding to Cd^(2+) . The combined toxicity was reduced at Cd^(2+) lower than 4 mg/L and enhanced at Cd^(2+) higher than 4 mg/L under multiple interactions. Tween 80 enhanced the combined toxicity of nano-TiO_2 and Cd^(2+) by increasing the outer membrane permeability. 8High-operating-temperature mid-infrared photodetectors via quantum dot gradient homojunction显示文摘Due to thermal carriers generated by a narrow mid-infrared energy gap,cooling is always necessary to achieve ideal photodetection.In quantum dot(QD),the electron thermal generation should be reduced with quantum confinement in all three dimensions.As a result,there would be a great potential to realize high-operating-temperature(HOT)QD mid-IR photodetectors,though not yet achieved.Taking the advantages of colloidal nanocrystals’solution processability and precise doping control by surface dipoles,this work demonstrates a HOT mid-infrared photodetector with a QD gradient homojunction.The detector achieves background-limited performance with D^(*)=2.7×1011 Jones on 4.2μm at 80 K,above 10^(11) Jones until 200 K,above 10^(10 )Jones until 280 K,and 7.6×10^(9) Jones on 3.5μm at 300 K.The external quantum efficiency also achieves more than 77%with responsivity 2.7 A/W at zero bias.The applications such as spectrometers,chemical sensors,and thermal cameras,are also approved,which motivate interest in low-cost,solution-processed and high-performance mid-infrared photodetection beyond epitaxial growth bulk photodetectors.Xiaomeng Xue Menglu Chen Yuning Luo Tianling Qin Xin Tang Qun Hao 2023Light(Science & Applications)2023,12,3:2

10Activating transcription factor 4 protects mice against sepsis-induced intestinal injury by regulating gut-resident macrophages differentiation显示文摘Background: Gut-resident macrophages (gMacs) supplemented by monocytes-to-gMacs differentiation play a critical role in maintaining intestinal homeostasis. Activating transcription factor 4 (ATF4) is involved in immune cell differentiation. We therefore set out to investigate the role of ATF4-regulated monocytes-to-gMacs differentiation in sepsis-induced intestinal injury.Methods: Sepsis was induced in C57BL/6 wild type (WT) mice andAtf4-knockdown (Atf4+/-) mice by cecal ligation and puncture or administration of lipopolysaccharide (LPS). Colon, peripheral blood mononuclear cells, sera, lung, liver, and mesenteric lymph nodes were collected for flow cytometry, hematoxylin and eosin staining, immunohistochemistry, quantitative reverse transcription polymerase chain reaction, and enzyme-linked immunosorbent assay, respectively.Results: CD64, CD11b, Ly6C, major histocompatibility complex-II (MHC-II), CX3CR1, Ly6G, and SSC were identified as optimal primary markers for detecting the process of monocytes-to-gMacs differentiation in the colon of WT mice. Monocytes-to-gMacs differentiation was impaired in the colon during sepsis and was associated with decreased expression of ATF4 in P1 (Ly6Chi monocytes), the precursor cells of gMacs.Atf4 knockdown exacerbated the impairment of monocytes-to-gMacs differentiation in response to LPS, resulting in a significant reduction of gMacs in the colon. Furthermore, compared with WT mice,Atf4+/- mice exhibited higher pathology scores, increased expression of inflammatory factor genes (TNF-α, IL-1β), suppressed expression of CD31 and vascular endothelial-cadherin in the colon, and increased translocation of intestinal bacteria to lymph nodes and lungs following exposure to LPS. 11Tectonic Evolution and Key Geological Issues of the ProtoSouth China Sea显示文摘There are numerous controversies surrounding the tectonic properties and evolution of the Proto-South China Sea(PSCS).By combining data from previously published works with our geological and paleontological observations of the South China Sea(SCS),we propose that the PSCS should be analyzed within two separate contexts:its paleogeographic location and the history of its oceanic crust.With respect to its paleogeographic location,the tectonic properties of the PSCS vary widely from the Triassic to the mid-Late Cretaceous.In the Triassic,the Paleo-Tethys and the Paleo-Pacific Oceans were the major causes of tectonic changes in the SCS,while the PCSC may have been a remnant sea residing upon Tethys or Paleo-Pacific oceanic crust.In the Jurassic,the Meso-Tethys and the Paleo-Pacific oceans joined,creating a PSCS back-arc basin consisting of Meso-Tethys and/or Paleo-Pacific oceanic crust.From the Early Cretaceous to the midLate Cretaceous,the Paleo-Pacific Ocean was the main tectonic body affecting the SCS;the PSCS may have been a marginal sea or a back-arc basin with Paleo-Pacific oceanic crust.With respect to its oceanic crust,due to the subduction and retreat of the Paleo-Pacific plate in Southeast Asia at the end of the Late Cretaceous,the SCS probably produced new oceanic crust,which allowed the PSCS to formally emerge.At this time,the PSCS was most likely a combination of a new marginal sea and a remnant sea;its oceanic crust,which eventually subducted and became extinct,consisted of both new oceanic crust and remnant oceanic crust from the Paleo-Pacific Ocean.In the present day,the remnant PSCS oceanic crust is located in the southwestern Nansha Trough.TIAN Zhiwen TANG Wu WANG Pujun ZHAO Zhigang SUN Xiaomeng TANG Huafeng 2021Acta Geologica Sinica(English Edition)2021,95,1:1

14Performance Comparison of Computational Methods for the Prediction of the Function and Pathogenicity of Non-coding Variants显示文摘Non-coding variants in the human genome significantly influence human traits and complex diseases via their regulation and modification effects.Hence,an increasing number of computational methods are developed to predict the effects of variants in human non-coding sequences.However,it is difficult for inexperienced users to select appropriate computational methods from dozens of available methods.To solve this issue,we assessed 12 performance metrics of 24 methods on four independent non-coding variant benchmark datasets:(1)rare germline variants from clinical relevant sequence variants(ClinVar),(2)rare somatic variants from Catalogue Of Somatic Mutations In Cancer(COSMIC),(3)common regulatory variants from curated expression quantitative trait locus(eQTL)data,and(4)disease-associated common variants from curated genomewide association studies(GWAS).All 24 tested methods performed differently under various conditions,indicating varying strengths and weaknesses under different scenarios.Importantly,the performance of existing methods was acceptable for rare germline variants from ClinVar with the area under the receiver operating characteristic curve(AUROC)of 0.4481–0.8033 and poor for rare somatic variants from COSMIC(AUROC=0.4984–0.7131),common regulatory variants from curated eQTL data(AUROC=0.4837–0.6472),and disease-associated common variants from curated GWAS(AUROC=0.4766–0.5188).We also compared the prediction performance of 24 methods for non-coding de novo mutations in autism spectrum disorder,and found that the combined annotation-dependent depletion(CADD)and context-dependent tolerance score(CDTS)methods showed better performance.Summarily,we assessed the performance of 24 computational methods under diverse scenarios,providing preliminary advice for proper tool selection and guiding the development of new techniques in interpreting non-coding variants.Zheng Wang Guihu Zhao Bin Li Zhenghuan Fang Qian Chen Xiaomeng Wang Tengfei Luo Yijing Wang Qiao Zhou Kuokuo Li Lu Xia Yi Zhang Xun Zhou Hongxu Pan Yuwen Zhao Yige Wang Lin Wang Jifeng Guo Beisha Tang Kun Xia Jinchen Li 2023Genomics, Proteomics & Bioinformatics2023,21,3:0

16Plant-derived nanovesicles: Further exploration of biomedical function and application potential显示文摘Extracellular vesicles(EVs)are phospholipid bilayer vesicles actively secreted by cells,that contain a variety of functional nucleic acids,proteins,and lipids,and are important mediums of intercellular communication.Based on their natural properties,EVs can not only retain the pharmacological effects of their source cells but also serve as natural delivery carriers.Among them,plant-derived nanovesicles(PNVs)are characterized as natural disease therapeutics with many advantages such as simplicity,safety,eco-friendliness,low cost,and low toxicity due to their abundant resources,large yield,and low risk of immunogenicity in vivo.This review systematically introduces the biogenesis,isolation methods,physical characterization,and components of PNVs,and describes their administration and cellular uptake as therapeutic agents.We highlight the therapeutic potential of PNVs as therapeutic agents and drug delivery carriers,including anti-inflammatory,anticancer,wound healing,regeneration,and antiaging properties as well as their potential use in the treatment of liver disease and COVID-19.Finally,the toxicity and immunogenicity,the current clinical application,and the possible challenges in the future development of PNVs were analyzed.We expect the functions of PNVs to be further explored to promote clinical translation,thereby facilitating the development of a new framework for the treatment of human diseases.Aixue Li Dan Li Yongwei Gu Rongmei Liu Xiaomeng Tang Yunan Zhao Fu Qi Jifu Wei Jiyong Liu 2023Acta Pharmaceutica Sinica B2023,13,8:0

18Propylamine hydrobromide passivated tin-based perovskites to efficient solar cells显示文摘The development of tin-based devices with low toxicity is critical for the commercial viability of perovskite solar cells.However because tin halide is a stronger Lewis acid,its crystallization rate is extremely fast,resulting in the formation of numerous defects that affect the device performance of tin-based perovskite solar cells.Herein,propylamine hydrobromide(PABr)was added to the perovskite precursor solution as an additive to passivate defects and fabricate more uniform and dense perovskite films.Because propylamine cations are too large to enter the perovskite lattices,they only exist at the grain boundary to passivate surface defects and promote crystal growth in a preferred orientation.The PABr additive raises the average short-circuit current density from 19.45 to 25.47 mA·cm^(-2)by reducing carrier recombination induced by defects.Furthermore,the device’s long-term illumination stability is improved after optimization,and the hysteresis effect is negligible.The addition of PABr results in a power conversion efficiency of 9.35%.Xiaomeng Li Pengcheng Jia Fanwen Meng Xingyu Zhang Yang Tang Bo Song Chang Gao Liang Qin Feng Teng Yanbing Hou 2023International Journal of Minerals,Metallurgy and Materials2023,30,10:0
