| 3 | Evaluation of four final irrigation protocols for cleaning root canal walls显示文摘The aim of this study was to compare the efficiency of four final irrigation protocols in smear layer removal and bacterial inhibition in root canal systems.Thirty roots inoculated with Enterococcus faecalis were prepared with ProTaper Universal files.The teeth were disinfected by conventional needle irrigation,sonic agitation using the EndoActivator device,passive ultrasonic irrigation,or an M3 Max file.Teeth with no root canal preparation served as blank controls for the establishment of the infection baseline.Teeth with preparation but no final irrigation served as a post-instrumentation baseline.After the final irrigation,the teeth were sectioned in half.One half of each tooth was examined by scanning electron microscopy(SEM)to assess smear layer removal using a five-point scale.The other half was examined by confocal laser scanning microscopy(CLSM)using the LIVE/DEAD BackLight bacterial viability kit to evaluate the depth of bacterial survival in dentinal tubules.SEM analysis revealed no significant difference in smear layer removal throughout the whole canal among the EA,PUI,and M3 Max groups(P>0.05).CLSM revealed that PUI achieved the greatest bacterial inhibition depth in the coronal((174.27±31.63)μm),middle((160.94±37.77)μm),and apical((119.53±28.49)μm)thirds of the canal(all P<0.05 vs.other groups).According to this comprehensive SEM and CLSM evaluation,PUI appears to have the best infection control ability in root canal systems. | Qiang Li Qian Zhang Xiaoying Zou Lin Yue | 2020 | International Journal of Oral Science2020,12,3: | 6 |
| 4 | Immunogenicity and virus-like particle formation of rotavirus capsid proteins produced in transgenic plants显示文摘The human pathogen, group A rotavirus, is the most prevalent cause of acute infantile and pediatric gastroenteritis worldwide, especially in developing countries. There is an urgent demand for safer, more effective and cheaper vaccines against rotavirus infection. Plant-derived antigens may provide an exclusive way to produce economical subunit vaccines. Virus-like particles, constituting viral capsid proteins without viral nucleic acids, are considered a far safer candidate compared with live attenuated viral vaccines. In this study, the rotavirus capsid proteins VP2, VP6 and VP7 were co-expressed in transgenic tobacco plants, and their expression levels, formation of rotavirus-like particles (RV VLPs) and immunogenicity were extensively studied. Quantitative real-time RT-PCR and Western blot analysis revealed that the expression level of vp6 was the highest while vp7 was expressed at the lowest levels. The RV VLPs were purified from transgenic tobacco plants and analyzed by electron microscopy and Western blot. Results indicated that the plant-derived VP2, VP6 and VP7 proteins self-assembled into 2/6 or 2/6/7 RV VLPs with a diameter of 60-80 nm. | 6 | Small interfering RNA delivery to the neurons near the amyloid plaques for improved treatment of Alzheimer’s disease显示文摘Gene therapy represents a promising treatment for the Alzheimer’s disease(AD). However,gene delivery specific to brain lesions through systemic administration remains big challenge. In our previous work, we have developed an siRNA nanocomplex able to be specifically delivered to the amyloid plaques through surface modification with both CGN peptide for the blood–brain barrier(BBB)penetration and QSH peptide for β-amyloid binding. But, whether the as-designed nanocomplex could indeed improve the gene accumulation in the impaired neuron cells and ameliorate AD-associated symptoms remains further study. Herein, we prepared the nanocomplexes with an siRNA against β-site amyloid precursor protein-cleaving enzyme 1(BACE1), the rate-limiting enzyme of Aβ production, as the therapeutic siRNA of AD. The nanocomplexes exhibited high distribution in the Aβ deposits-enriched hippocampus, especially in the neurons near the amyloid plaques after intravenous administration. In APP/PS1 transgenic mice, the nanocomplexes down-regulated BACE1 in both mRNA and protein levels,as well as Aβ and amyloid plaques to the level of wild-type mice. Moreover, the nanocomplexes significantly increased the level of synaptophysin and rescued memory loss of the AD transgenic mice without hematological or histological toxicity. | 7 | Salivary protease spectrum biomarkers of oral cancer显示文摘Proteases are important molecules that are involved in many physiological and pathological processes of the human body,such as growth,apoptosis and metastasis cancer cells.They are potential targets in cancer diagnosis and biotherapy.In this study,we analyzed the salivary protease spectrum of patients with oral squamous cell carcinoma (OSCC),oral benign masses and chronic periodontitis,as well as that of health,using human protease array kits,enzyme-linked immunosorbent assay,western blot and immunofluorescence.The salivary protease spectrum was found to be associated with oral diseases.For example,the saliva of patients with OSCC contained increased numbers of proteases than those of other oral diseases and health.The levels of matrix metalloproteinase (MMP)-1,MMP-2,MMP-10,MMP-12,A disintegrin and metalloprotease (ADAM)9,A disintegrin and metalloprotease with thrombospondin type 13 motifs (ADAMST13),cathepsin V and kallikrein 5 in the saliva of patients with OSCC were significantly increased compared with those of other groups.Taking MMP-1,cathepsin V,kallikrein 5 and ADAM9 as biomarkers of OSCC,cutoff values were199,11.34,9.29 and 202.55 pg·mL?1,respectively.From the area under the curve,sensitivity and specificity,the combination of cathepsin V/kallikrein5/ADAM9 was an optimal biomarker for diagnosing OSCC.Thus,analysis of the salivary protease spectrum may be an innovative and cost-efficient approach to evaluating the health status of the oral cavity.Specifically,increases in cathepsin V,kallikrein 5 and ADAM9 may be useful biomarkers in the screening and diagnosis of OSCC. | Yun Feng Qian Li Jiao Chen Ping Yi Xin Xu Yaping Fan Bomiao Cui Yu Yu Xiaoying Li Yue Du Qianming Chen Lingling Zhang Jingjing Jiang Xuedong Zhou Ping Zhang | 2019 | International Journal of Oral Science2019,11,2: | 2 |

| 12 | Geopolitics of the energy transition显示文摘Geopolitics of energy transition has increasingly become the frontier and hot research area of world energy geography and global political science.Different historical periods are characterised by obvious differences in energy connotations,attributes,and geopolitical characteristics.In the new energy era,energy geopolitics becomes more diversified,complex,and comprehensive.In this paper,we compare the geopolitical characteristics of energy in the fossil fuel and renewable energy periods,and provide an overview of current study trends in new energy geopolitics.Recent research shows that the global energy transition will intensify the reconstruction of geopolitical patterns,change the relationship between geopolitical security and conflict dominated by traditional energy security,alter the role of different countries in global energy geopolitical games,reshape national energy relationships formed in the traditional oil and gas era.In addition,geopolitics will be affected by new energy technologies,availability of key rare materials,and energy cybersecurity measures.Despite considerable attention to this research topic,the likely geopolitical impact of energy transition remains uncertain,and there is still room for the development and improvement of the theoretical framework,technical methods,and research perspective.Looking forward to the future,the research into geopolitics of energy transition urgently needs to strengthen its theoretical basis and rely on the scientific and quantitative methods.The practical conclusions of the research into geopolitics of energy transition should strengthen major national energy security decisions,explore the geographical effect of energy transition,and determine the impact of energy transition on energy security.Research into geopolitics of energy transition should be carried out taking into account international academic frontiers such as climate change,“carbon peak”and“carbon neutral”goals,and global energy governance,to enrich the research perspective of world energy geography. | YANG Yu XIA Siyou QIAN Xiaoying | 2023 | Journal of Geographical Sciences2023,33,4: | 1 |

| 15 | The molecular tumor burden index as a response evaluation criterion in breast cancer显示文摘Circulating tumor DNA(ctDNA)is a potential biomarker of prognosis and therapeutic response.We conducted this study to explore the role of the molecular tumor burden index(mTBI)in ctDNA as a therapeutic response and prognostic biomarker in a larger cohort prospective phase III randomized multicenter study.We collected 291 plasma samples from 125 metastatic breast cancer patients from the CAMELLIA study(NCT01917279).Target-capture deep sequencing of 1021 genes was performed to detect somatic variants in ctDNA from the plasma samples.The pretreatment mTBI value was correlated with tumor burden(P=0.025).Patients with high-level pretreatment mTBI had shorter overall survival than patients with low-level pretreatment mTBI,and the median overall survival was 40.9 months and 68.4 months,respectively(P=0.011).Patients with mTBI decrease to less than 0.02%at the first tumor evaluation had longer progression-free survival and overall survival(P<0.001 and P=0.007,respectively).The mTBI has good sensitivity to identify complete response/partial response and progressive disease based on computed tomography scans(88.5%and 87.5%,respectively).The patients classified as molecular responders had longer progression-free survival and overall survival than the nonmolecular responders in the overall cohort(P<0.001 and P=0.036,respectively),as well as in the cohort in which computed tomography scans were defined as representing stable disease(P=0.027 and P=0.015,respectively).The mTBI in ctDNA detected in liquid biopsies is a potential biomarker of therapeutic response and prognosis in patients with metastatic breast cancer. | Zongbi Yi Fei Ma Guohua Rong Binliang Liu Yanfang Guan Jin Li Xiaoying Sun Wenna Wang Xiuwen Guan Hongnan Mo Jiani Wang Haili Qian Binghe Xu | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 1 |

| 19 | Exogenous Levodopa Increases the Neuro-retinal Dopamine of Guinea Pig Myopic Eyes in vitro显示文摘Purpose:In our previous work,it has been shown that intraperitoneal injection of L-DOPA can inhibit the development of occlusion myopia in guinea pigs,and increase levels of retinal dopamine.The aim of this study was to investigate whether exogenous L-DOPA can be converted into dopamine in cultured retina of guinea pig eyes subjected to visual deprivation,and to evaluate whether müller cells are involved in the processing of retinal dopamine induced by L-DOPA.Methods:Fifty-eight guinea pigs were randomly divided into 2 groups at the age of 4 weeks:normal control and visual deprivation.Form deprivation was induced with translucent eye shields over the right eye,and lasted for ten days.Corneal curvature,refraction and axial length were measured in all animals.In vitro,neuro-retina and müller cell were cultured,and L-DOPA was added to the culture medium at three concentrations:1 μM,10 μMand 100 μM.Subsequently,dopamine content was evaluated by high-performance liquid chromatography,and apoptotic cells were identified by TUNEL staining.Results:Ten days of occlusion caused the affected eyes to elongate and become myopic in guinea pigs.Compared with the deprivation group,10 μML-DOPA treament significantly raised dopamine content in cultured retina and müller cells (P<0.05).However,1 μMand 100 μML-DOPA treatment caused no increase in dopamine levels(P>0.05).Apoptotic nuclei were detected in the ganglion cell layer (92.5%±8.3%) and inner nuclear layer (46.8%±9.1%)of cultured retina treated with 100 μML-DOPA.Moreover,100 μML-DOPA also caused apoptosis of retinal müller cells,at a mean rate of 59.4 ±11.3%.Conclusion:.Our results suggest that exogenous L-DOPA can cause an increase in retinal dopamine in form-deprived guinea pig eyes in vitro,and that müller cells are involved in the increase in retinal dopamine. | Junfeng Mao Shuangzhen Liu Wenjuan Qin Qian Xiang Xiaoying Wu | 2011 | Eye Science2011,26,4: | 1 |

We performed this study to search for molecular markers of endocrine therapy efficacy and to explore the clinical value of ctDNA to guide precise endocrine therapy for HR-positive/human epidermal growth factor receptor-2(HER-2)-negative metastatic breast cancer patients.Methods: In this open-label, multicohort, prospective study, patients were assigned to four parallel cohorts and matched according to mutations identified in ctDNA: 1) activation of the phosphatidylinositol-3-kinase(PI3K)/AKT/mammalian target of rapamycin(mTOR) signaling pathway preferred mTOR inhibitor combined with endocrine therapy;2) estrogen receptor 1(ESR1) mutation preferred fulvestrant;3) HER-2 mutations preferred pyrotinib;and 4) no actionable mutations received treatment according to the clinical situation. In all cohorts, patients were divided into compliance group and violation group. The primary outcome measure was progression-free survival(PFS), and the secondary outcome measure was overall survival(OS).Results: In all cohorts, the combined median PFS was 4.9 months, and median PFS for the compliance and violation groups was 6.0 and 3.0 months, respectively [P=0.022, hazard ratio(HR)=0.57]. Multivariate Cox regression model showed the risk of disease progression was lower in compliance group than in violation group(P=0.023, HR=0.55). Among the patients with HER-2 mutations, the median PFS was 11.1 months in the compliance group and 2.2 months in the violation group(P=0.011, HR=0.20). There was no significant difference in the median PFS between patients who did and did not comply with the treatment protocol in patients with activation of the PI3K/AKT/mTOR or ESR1 mutation.Conclusions: The results suggest that ctDNA may help to guide the optimal endocrine therapy strategy for metastatic breast cancer patients and to achieve a better PFS.
