1Newly discovered hydrothermal fields along the ultraslow-spreading Southwest Indian Ridge around 63°E显示文摘The ultraslow-spreading Southwest Indian Ridge(SWIR) to the east of the Melville fracture zone is characterized by very low melt supply and intensive tectonic activity. Due to its weak thermal budget and extremely slow spreading rate, the easternmost SWIR was considered to be devoid of hydrothermal activity until the discovery of the inactive Mt. Jourdanne hydrothermal field(27°51′S, 63°56′E) in 1998. During the COMRA DY115-20 cruise in2009, two additional hydrothermal fields(i.e., the Tiancheng(27°51′S, 63°55′E) and Tianzuo(27°57′S, 63°32′E)fields) were discovered. Further detailed investigations of these two hydrothermal sites were conducted by Chinese manned submersible Jiaolong in 2014–2015. The Tiancheng filed can be characterized as a lowtemperature(up to 13.2°C) diffuse flow hydrothermal field, and is hosted by fractured basalts with hydrothermal fauna widespread on the seafloor. The Tianzuo hydrothermal field is an inactive sulfide field, which is hosted by ultramafic rocks and controlled by detachment fault. The discovery of the three hydrothermal fields around Segment #11 which receives more melt than the regional average, provided evidence for local enhanced magmatism providing heat source to drive hydrothermal circulation. 3Accelerated generation of oligodendrocyte progenitor cells from human induced pluripotent stem cells by forced expression of Sox10 and Olig2显示文摘Oligodendrocyte progenitor cells(OPCs) hold great promise for treatment of dysmyelinating disorders, such as multiple sclerosis and cerebral palsy. Recent studies on generation of human OPCs mainly use human embryonic stem cells(hESCs) or neural stem cells(NSCs) as starter cell sources for the differentiation process. However, NSCs are restricted in availability and the present method for generation of oligodendrocytes(OLs) from ESCs often requires a lengthy period of time. Here, we demonstrated a protocol to efficiently derive OPCs from human induced pluripotent stem cells(hiPSCs) by forced expression of two transcription factors(2TFs), Sox10 and Olig2. With this method, PDGFRα+ OPCs can be obtained in 14 days and O4^+ OPCs in 56 days.Furthermore, OPCs may be able to differentiate to mature OLs that could ensheath axons when co-cultured with rat cortical neurons. 16Neighbouring carbonyl group-assisted sequential 1,2-azide and1,4-oxygen migrations of vinyl azides leading to α-azido ketones显示文摘The development of azide migration reactions is a formidable challenge due to potential competition from side processes driven by the release of molecular nitrogen. Here, we show a novel neighbouring carbonyl group-assisted sequential 1,2-azide and 1,4-oxygen migration reaction of α-vinyl azides for the efficient preparation of a range of α-azido ketones. This represents the first transformation of α-vinyl azides into α-azido ketones. Notable features of this method are its high efficiency, broad substrate scope, excellent functional group compatibility, and high yields. The computational studies gave a key insights into the regioselective 1,2-azide and 1,4-oxygen migrations assisted by the neighbouring carbonyl group. Further conversions into a variety of nitrogen-containing compounds demonstrated the synthetic utility of the α-azido ketone products. 18Cloning,Sequencing and Analysis of DDX3Y Gene in Five Male Equus Animals显示文摘[Objective] This study analyzed the differences in DDX3 Y gene sequences among male horse,male donkey,male mule,male hinny and male foal to investigate the relationship between DDX3 Y gene structure and the mechanism of infertility in male Equus ferus × asinus. [Method] Partial sequences of DDX3 Y gene in male horse,male donkey,male mule,male hinny and male foal were obtained by genome comparison,sequencing and cloning,and compared by bioinformatics methods. [Result] The CDS regions varied in 5 base pairs and two amino acids. DDX3 Y protein was predicted to be an unstable protein. In addition,the results also showed that DDX3 Y gene in male horse displayed a closer phylogenetic relationship to male mule and male foal; DDX3 Y gene in male donkey displayed a closer phylogenetic relationship to male hinny. 19Oocyte Transfer as an Analytical Tool:Vintage Experiments in Domestic Farm Animals显示文摘Four transplant studies are described that focus on fertilisation and early development or the progression of unfertilised oocytes(eggs) in the oviduct.(1) Pig eggs transplanted from ovulations induced during the luteal phase of the estrous cycle were fertilised in the oviducts of inseminated recipient animals in estrus.By contrast,pig eggs from donors in estrus became highly polyspermic when transplanted to the oviducts of animals force-mated during the luteal phase.(2) Pig embryos at the stage of hatched blastocysts(days 7 and 8) could be transplanted successfully to synchronous recipients and full embryonic development demonstrated to between days 19 and 23 of pregnancy.Thus,the exposed trophectoderm of developing embryos could withstand the physical manipulation of recovery and transplantation,and the lifespan of corpora lutea in the unmated recipients could be prolonged by transfer of day 7 and 8 blastocysts.(3) Bovine oocytes aspirated from 2 to 6 mm diameter Graafian follicles and matured in vitro were fertilized normally in the oviducts of inseminated recipient heifers,demonstrating the potential of slaughterhouse ovaries for the generation of embryos.(4) Transplanting equine eggs to a pig oviduct,in which egg descent to the uterus requires only 46 to 48 h,did not reveal a retarded progress of degenerating unfertilised horse eggs,suggesting the involvement of non-physical factors in equine embryo progression to the uterus.Prostaglandins of embryonic origin are now known to be a key.A final section examines the post-ovulatory role of ovarian follicular cells on the secretory activity of the oviductal epithelium.R. 20A Comparison of the Effects of Pentoxifylline on Quality of Fresh and Frozen-thawed Bull Spermatozoa显示文摘[Object]This study aimed to examine the effects of 3,7-dimethyl-1-(5-oxo-hexyl)-xanthine(pentoxifylline,PF)on the motility,mitochondrial activity,acrosome integrity and fertilization rate of spermatozoa in both fresh and frozen-thawed bull semen.[Method]Fresh and frozen-thawed bull spermatozoa were exposed to 5 mmol/L PF with untreated samples as controls.[Result]Fresh spermatozoa showed reduced(P<0.05)motility after 2 h incubation with PF whereas,surprisingly,frozen-thawed samples exhibited increased sperm motility(P<0.05)after 2 h incubation with PF and they also showed enhanced longevity compared to controls.Mitochondrial activity in both fresh and frozen-thawed bull spermatozoa increased(P<0.05)during 4 h incubation with PF whereas acrosome integrity remained unchanged in both types of semen.However,treatment with 5 mmol/L PF did not influence the in vitro fertilization efficiency of fresh spermatozoa but improved significantly(P<0.05)that of frozen-thawed spermatozoa.[Conclusion]These results indicate that PF can improve sperm quality of frozenthawed bull semen,and may improve pregnancy rates in bovine artificial insemination programmes employing frozen semen.Jian ZHANG Huamin GUO Jie SU Lixia ZHAO Yunxia LI Wei SUN Hongmei HAN Shuxiang HU Gaoping ZHAO Yao LI Yanfeng DAI Xihe LI 2014Agricultural Biotechnology2014,3,3:0
