2Recent advances in cell sheet technology for bone and cartilage regeneration: from preparation to application显示文摘Bone defects caused by trauma,tumour resection,infection and congenital deformities,together with articular cartilage defects and cartilage–subchondral bone complex defects caused by trauma and degenerative diseases,remain great challenges for clinicians.Novel strategies utilising cell sheet technology to enhance bone and cartilage regeneration are being developed.The cell sheet technology has shown great clinical potential in regenerative medicine due to its effective preservation of cell–cell connections and extracellular matrix and its scaffold-free nature.This review will first introduce several widely used cell sheet preparation systems,including traditional approaches and recent improvements,as well as their advantages and shortcomings.Recent advances in utilising cell sheet technology to regenerate bone or cartilage defects and bone–cartilage complex defects will be reviewed.The key challenges and future research directions for the application of cell sheet technology in bone and cartilage regeneration will also be discussed.Yuezhi Lu Wenjie Zhang Jie Wang Guangzheng Yang Shi Yin Tingting Tang Chunhua Yu Xinquan Jiang 2019International Journal of Oral Science2019,11,2:14
3Marginal sealing around integral bilayer scaffolds for repairing osteochondral defects based on photocurable silk hydrogels显示文摘Osteochondral repair remains a major challenge in current clinical practice despite significant advances in tissue engineering.In particular,the lateral integration of neocartilage into surrounding native cartilage is a difficult and inadequately addressed problem that determines the success of tissue repair.Here,a novel design of an integral bilayer scaffold combined with a photocurable silk sealant for osteochondral repair is reported.First,we fabricated a bilayer silk scaffold with a cartilage layer resembling native cartilage in surface morphology and mechanical strength and a BMP-2-loaded porous subchondral bone layer that facilitated the osteogenic differentiation of BMSCs.Second,a TGF-β3-loaded methacrylated silk fibroin sealant(Sil-MA)exhibiting biocompatibility and good adhesive properties was developed and confirmed to promote chondrocyte migration and differentiation.Importantly,this TGF-β3-loaded Sil-MA hydrogel provided a bridge between the cartilage layer of the scaffold and the surrounding cartilage and then guided new cartilage to grow towards and replace the degraded cartilage layer from the surrounding native cartilage in the early stage of knee repair.Thus,osteochondral regeneration and superior lateral integration were achieved in vivo by using this composite.These results demonstrate that the new approach of marginal sealing around the cartilage layer of bilayer scaffolds with Sil-MA hydrogel has tremendous potential for clinical use in osteochondral regeneration.Xiaolin Wu Mingliang Zhou Fei Jiang Shi Yin Sihan Lin Guangzheng Yang Yuezhi Lu Wenjie Zhang Xinquan Jiang 2021Bioactive Materials2021,6,11:7
4Long-term anodal block stimulation at sacral anterior roots promoted recovery of neurogenic bladder function in a rabbit model of complete spinal cord injury显示文摘A complete spinal cord injury model was established in experimental rabbits using the spinal cord clip compression method.Urodynamic examination was performed 2 weeks later to determine neurogenic bladder status.The rabbits were treated with anodal block stimulation at sacral anterior roots for 4 weeks.Electrical stimulation of sacral anterior roots improved urodynamic parameters of neurogenic bladder in rabbit models of complete spinal cord injury,effectively promoted urinary function,and relieved urinary retention.Immunohistochemistry results showed that a balance was achieved among expression of muscarinic receptor subunits M2,M3,ATP-gated ion channel P2X3 receptors,and β2-adrenergic receptor,and nerve growth factor expression decreased.These results suggested that long-term sacral anterior root stimulation of anodal block could be used to treat neurogenic bladder in a rabbit model of complete spinal cord injury.Xiaoran Wang Qi Gao Xiaoyu Yang Weihua Wang Xinquan Gu Guifeng Liu Peng Yan Ge Gao Xin Yu Yongjie Wang Jihu Lian Chaoling Shi Yao Wang Li Fan 2012Neural Regeneration Research2012,7,5:6
5cDNA cloning and expression of erythropoietin in the plateau zokor(Myospalax baileyi) from the Qinghai-Tibet Plateau显示文摘All organisms respond to variation in their environments and manage environmental stress through metabolic adjustments.The plateau zokor(Myospalax baileyi) is an endemic and keystone subterranean rodent species that inhabits the Qinghai-Tibet Plateau between 2800 and 4200 m above sea level.It is a hypoxic-tolerant mammal with a high ratio of oxygen utilization that enables it to cope with its harsh surroundings.To explore the molecular mechanism of altitude acclimatization of the plateau zokor,we cloned the zokor erythropoietin(Epo) gene and used real-time PCR to compare Epo mRNA levels in zokors inhabiting 16 different altitudes.The full-length zokor Epo open reading frame was 579 bp that encoded a precursor peptide of 192 amino acids with a signal peptide of 26 residues.The Epo gene of the plateau zokor was 81%-95% homologous to that of human,mouse,rat,root vole and the Golan Heights blind mole rat,with the highest homology(95%) to species of the genus Spalax.Epo mRNA was detected mainly in the zokor kidney and spleen among 8 selected tissues.The level of Epo mRNA increased in the liver and kidney with increases in altitude.The increase in the kidney was 5 times that in the liver.Remarkably,expression of Epo mRNA in the kidney of zokors living at the highest altitude(4268 m) was 12-fold higher than that of zokors living at the lowest(2492 m) altitude.These findings provide essential information for understanding the possible role of Epo in adaptation to hypoxia in the plateau zokor.WANG ZhenLong CHEN Yan YANG Jie CHEN WeiJun ZHANG YanMing ZHAO XinQuan 2012Chinese Science Bulletin2012,57,9:4
6Redefinition and Formation Age of the Tanjianshan Group in Xitieshan Region, Qinghai显示文摘The Tanjianshan Group, which was previously divided into a, b, c and d formations, has been controversial for a long time. It mainly distributes in the northern margin of Qaidam Basin and is an important early Paleozoic greenschist facies metamorphic volcanic sedimentary rock formation. Detailed field investigation and zircon LA-ICPMS U-Pb dating of the key strata suggest that the original lower part of a Formation(a-1) versus the original middle upper of d Formation(d-3 and d-4), the original upper part of a Formation(a-2) and b Formation versus the original lower part of d Formation(d-1 and d-2) of Tanjianshan Group are contemporaneous heterotopic facies volcanicclasolite deposit, respectively. The former formations formed during the middle-late Ordovician(463–458 Ma), while the latter ones formed in the late Ordovician(about 445 Ma). The original c formation of Tanjianshan Group, which formed after 430 Ma, is similar to the Maoniushan Formation of Kunlun Mountains and north Qaidam Basin. According to the rules of stratigraphic division and naming, new stratum formations of Tanjianshan Group are re-built and divided into Duancenggou(O1-2td), Zhongjiangou(O2-3tz) and Xitieshan(O3tx) formations. The original c Formation is separated from Tanjianshan Group and is renamed as the Wuminggou Formation(S3-D1w), which shows a discordant contact with underlying Tanjianshan Group and overlying Amunike Formation(D3a). 7Dinosaur eggs and dinosaur egg-bearing deposits(Upper Cretaceous) of Henan Province,China:Occurrences,palaeoenvironments,taphonomy and preservation显示文摘The Upper Cretaceous dinosaur egg-bearing deposits in Henan Province,central China are divided into three formations in ascending order:Gaogou,Majiacun and Sigou.The Gaogou Formation belongs to alluvial fan deposits containing the fossil dinosaur egg assemblage of Faveoloolithus,Dendroolithus,Dictyoolithus,Paraspheroolithus and Longiteresoolithus.The Majiacun Formation is interpreted as braided stream to meandering stream deposits with assemblage of Ovaloolithus,Paraspheroolithus,Placoolithus,Dendroolithus,Prismatoolithus, rare Youngoolithus and Nanhiungoolithus.The Sigou Formation is shallow lacustrine/palustrine to low-sinuosity river sedimentary rocks with assemblage of Macroolithus,Elongatoolithus,Ovaloolithus and Paraspheroolithus. To date,37 oospecies,13 oogenera and 8 oofamilies of dinosaur eggs have been distinguished.Autochthonous dinosaur eggs are preserved in the floodplain deposits,whereas allochthonous and parautochthonous dinosaur eggs are preserved in the alluvial fans.This suggests that river floodplains are the best environments for the preservation of numerous autochthonous dinosaur eggs.The preservation of most dinosaur eggs in brown to red calcic palaeosoils,muddy siltstones or mudstones suggests that the paleoclimate of the nesting area was semiarid.The presence of gray and green mudstone and coal layers,however,indicates that there existed an ephemeral subhumid climate during the middle and Late Cretaceous.It is suggested that such a climate was favorable for the development of meandering streams in a vegetated environment.Xinquan Liang Shunv Wen Dongsheng Yang Shiquan Zhou Shichong Wu 2009Progress in Natural Science:Materials International2009,19,11:3
8Orchestration of energy metabolism and osteogenesis by Mg^(2+)facilitates low-dose BMP-2-driven regeneration显示文摘The clinical application of bone morphogenetic protein-2(BMP-2)is limited by several factors,including ineffectiveness at low doses and severe adverse effects at high doses.To address these efficacy and safety limitations,we explored whether orchestration of energy metabolism and osteogenesis by magnesium ion(Mg^(2+))could reduce the dose and thereby improve the safety of BMP-2.Our results demonstrated that rapid metabolic activation triggered by BMP-2 was indispensable for subsequent osteogenesis.Moreover,inadequate metabolic stimulation was shown to be responsible for the ineffectiveness of low-dose BMP-2.Next,we identified that Mg^(2+),as an'energy propellant',substantially increased cellular bioenergetic levels to support the osteogenesis via the Akt-glycolysis-Mrs2-mitochondrial axis,and consequently enhanced the osteoinductivity of BMP-2.Based on the mechanistic discovery,microgel composite hydrogels were fabricated as low-dose BMP-2/Mg^(2+)codelivery system through microfluidic and 3D printing technologies.An in vivo study further confirmed that rapid and robust bone regeneration was induced by the codelivery system.Collectively,these results suggest that this bioenergetic-driven,cost-effective,low-dose BMP-2-based strategy has substantial potential for bone repair.Sihan Lin Shi Yin Junfeng Shi Guangzheng Yang Xutao Wen Wenjie Zhang Mingliang Zhou Xinquan Jiang 2022Bioactive Materials2022,7,12:2
9A shedding soluble form of interleukin-17 receptor D exacerbates collagen-induced arthritis through facilitating TNF-α-dependent receptor clustering显示文摘Rheumatoid arthritis(RA)is exacerbated by TNF-alpha signaling.However,it remains unclear whether TNF-α-activated TNFR1 and TNFR2 are regulated by extracellular factors.Here,we showed that soluble glycosylated interleukin-17 receptor D(sIL-17RD),which was produced by proteolytic cleavage,enhanced TNF-α-induced RA.We revealed that IL-17RD shedding was induced by the proteolytic enzyme TACE and enhanced by TNF-αexpression in macrophages.Intriguingly,sIL-17RD was elevated in the sera of arthritic mice and rats.Recombinant sIL-17RD significantly enhanced the TNF-α-induced proinflammatory response by promoting TNF-α-TNFR-sIL-17RD complex formation and receptor clustering,leading to the accelerated development of collagen-induced arthritis.Our observations revealed that ectodomain shedding of IL-17RD occurred in RA to boost the TNF-α-induced inflammatory response.Targeting sIL-17RD may provide a new strategy for the therapy of RA.Sihan Liu Yanxia Fu Kunrong Mei Yinan Jiang Xiaojun Sun Yinyin Wang Fangli Ren Congshan Jiang Liesu Meng Shemin Lu Zhihai Qin Chen Dong Xinquan Wang Zhijie Chang Shigao Yang 2021Cellular & Molecular Immunology2021,18,8:1

12Bidirectional differentiation of BMSCs induced by a biomimetic procallus based on a gelatin-reduced graphene oxide reinforced hydrogel for rapid bone regeneration显示文摘Developmental engineering strategy needs the biomimetic composites that can integrate the progenitor cells,biomaterial matrices and bioactive signals to mimic the natural bone healing process for faster healing and reconstruction of segmental bone defects.We prepared the gelatin-reduced graphene oxide(GOG)and constructed the composites that mimicked the procallus by combining the GOG with the photo-crosslinked gelatin hydrogel.The biological effects of the GOG-reinforced composites could induce the bi-differentiation of bone marrow stromal cells(BMSCs)for rapid bone repair.The proper ratio of GOG in the composites regulated the composites’mechanical properties to a suitable range for the adhesion and proliferation of BMSCs.Besides,the GOG-mediated bidirectional differentiation of BMSCs,including osteogenesis and angiogenesis,could be activated through Erk1/2 and AKT pathway.The methyl vanillate(MV)delivered by GOG also contributed to the bioactive signals of the biomimetic procallus through priming the osteogenesis of BMSCs.During the repair of the calvarial defect in vivo,the initial hypoxic condition due to GOG in the composites gradually transformed into a well-vasculature robust situation with the bi-differentiation of BMSCs,which mimicked the process of bone healing resulting in the rapid bone regeneration.As an inorganic constituent,GOG reinforced the organic photo-crosslinked gelatin hydrogel to form a double-phase biomimetic procallus,which provided the porous extracellular matrix microenvironment and bioactive signals for the bi-directional differentiation of BMSCs.These show a promised application of the bio-reduced graphene oxide in biomedicine with a developmental engineering strategy.Delong Jiao Ao Zheng Yang Liu Xiangkai Zhang Xiao Wang Jiannan Wu Wenjun She Kaige Lv Lingyan Cao Xinquan Jiang 2021Bioactive Materials2021,6,7:1
13A rapidly magnetically assembled stem cell microtissue with“hamburger”architecture and enhanced vascularization capacity显示文摘With the development of magnetic manipulation technology based on magnetic nanoparticles(MNPs),scaffold-free microtissues can be constructed utilizing the magnetic attraction of MNP-labeled cells.The rapid in vitro construction and in vivo vascularization of microtissues with complex hierarchical architectures are of great importance to the viability and function of stem cell microtissues.Endothelial cells are indispensable for the formation of blood vessels and can be used in the prevascularization of engineered tissue constructs.Herein,safe and rapid magnetic labeling of cells was achieved by incubation with MNPs for 1 h,and ultrathick scaffold-free microtissues with different sophisticated architectures were rapidly assembled,layer by layer,in 5 min intervals.The in vivo transplantation results showed that in a stem cell microtissue with trisection architecture,the two separated human umbilical vein endothelial cell(HUVEC)layers would spontaneously extend to the stem cell layers and connect with each other to form a spatial network of functional blood vessels,which anastomosed with the host vasculature.The“hamburger”architecture of stem cell microtissues with separated HUVEC layers could promote vascularization and stem cell survival.This study will contribute to the construction and application of structural and functional tissues or organs in the future.Yuezhi Lu Chun-Hua Yu Guangzheng Yang Ningjia Sun Fei Jiang Mingliang Zhou Xiaolin Wu Jiaxin Luo Cui Huang Wenjie Zhang Xinquan Jiang 2021Bioactive Materials2021,6,11:1
14Correlation between X-ray cross-complementing group 1 polymorphisms and the onset risk of glioma A meta-analysis显示文摘OBJECTIVE:To evaluate the association of X-ray cross-complementing group 1(XRCC1)Arg399Gln,Arg194Trp and Arg280His polymorphisms with the risk of glioma.DATA SOURCES:A systematic literature search of papers published from January 2000 to August2012 in PubMed,Embase,China National Knowledge Infrastructure database,and Wanfang database was performed.The key words used were'glioma','polymorphism',and'XRCC1 or X-ray repair cross-complementing group 1'.References cited in the retrieved articles were screened manually to identify additional eligible studies.STUDY SELECTION:Studies were identified according to the following inclusion criteria:case-control design was based on unrelated individuals;and genotype frequency was available to estimate an odds ratio(OR)and 95%confidence interval(CI).Meta-analysis was performed for the selected studies after strict screening.Dominant and recessive genetic models were used and the relationship between homozygous mutant genotype frequencies and mutant gene frequency and glioma incidence was investigated.We chose the fixed or random effect model according to the heterogeneity to calculate OR and 95%CI,and sensitivity analyses were conducted.Publication bias was examined using the inverted funnel plot and the Egger’s test using Stata 12.0 software.MAIN OUTCOME MEASURES:Association of XRCC1 Arg399Gln,Arg194Trp,and Arg280His polymorphisms with the risk of glioma,and subgroup analyses were performed according to different ethnicities of the subjects.RESULTS:Twelve articles were included in the meta-analysis.Eleven of the articles were concerned with the Arg399Gln polymorphism and glioma onset risk.Significantly increased glioma risks were found only in the dominant model(Gln/Gln+Gln/Arg versus Arg/Arg:OR=1.26,95%CI=1.03-1.54,P=0.02).In the subgroup analysis by ethnicity,significantly increased risk was found in Asian subjects in the recessive(OR=1.46,95%CI=1.04-2.45,P=0.03)and dominant models(OR=1.40,95%CI=1.10-1.78,P=0.007),and homozygote contrast(OR=1.69,95%CI=1.17-2.45,P=0.005),but not in Caucasian subjects.For association of the Arg194Trp(eight studies)and Arg280His(four studies)polymorphisms with glioma risk,the meta-analysis did not reveal a significant effect in the allele contrast,the recessive genetic model,the dominant genetic model,or homozygote contrast.CONCLUSION:The XRCC1 Arg399Gln polymorphism may be a biomarker of glioma susceptibility,especially in Asian populations.The Arg194Trp and Arg280His polymorphisms were not associated with overall glioma risk.Xinquan Gu Hongyan Sun Liping Chang Ran Sun Hongfeng Yang Xuewen Zhang Xianling Cong 2013Neural Regeneration Research2013,8,26:1
15Surface form inspection with contact coordinate measurement:a review显示文摘Parts with high-quality freeform surfaces have been widely used in industries,which require strict quality control during the manufacturing process.Among all the industrial inspection methods,contact measurement with coordinate measuring machines or computer numerical control machine tool is a fundamental technique due to its high accuracy,robustness,and universality.In this paper,the existing research in the contact measurement field is systematically reviewed.First,different configurations of the measuring machines are introduced in detail,which may have influence on the corresponding sampling and inspection path generation criteria.Then,the entire inspection pipeline is divided into two stages,namely the pre-inspection and post-inspection stages.The typical methods of each sub-stage are systematically overviewed and classified,including sampling,accessibility analysis,inspection path generation,probe tip radius compensation,surface reconstruction,and uncertainty analysis.Apart from those classical research,the applications of the emerging deep learning technique in some specific tasks of measurement are introduced.Furthermore,some potential and promising trends are provided for future investigation.Yijun Shen Jieji Ren Nuodi Huang Yang Zhang Xinquan Zhang Limin Zhu 2023International Journal of Extreme Manufacturing2023,5,2:1
16IABC: An iteratively automatic machine learning boosted hand-eye calibration method for laser displacement sensor显示文摘An on-machine measuring(OMM)system with a laser displacement sensor(LDS)is designed for measuring free-form surfaces of hypersonic aircraft’s radomes.To improve the measurement accuracy of the OMM system,a novel Iteratively Automatic machine learning Boosted hand-eye Calibration(IABC)method is proposed.Both the hand-eye relationship and LDS measurement errors can be calibrated in one calibration process without any hardware changes via IABC.Firstly,a new objective function is derived,containing analytical parameters of the handeye relationship and LDS errors.Then,a hybrid calibration model composed of two kernels is proposed to solve the objective function.One kernel is the analytical kernel designed for solving analytical parameters.Another kernel is the automatic machine learning(AutoML)kernel designed to model LDS errors.The two kernels are connected with stepwise iterations to find the best calibration results.Compared with traditional methods,hand-eye experiments show that IABC reduces the calibration RMSE by about 50%.Verification experiments show that IABC reduces the measurement deviations by about 25%-50%and RMSEs within 40%.Even when the training data are obviously less than the test data,IABC performs well.Experiments demonstrate that IABC is more accurate than traditional hand-eye methods.Helin LI Bin LIN Tianyi SUI Liang XU Chen ZHANG Shuai YAN Yang WANG Xinquan HAO Deyu LOU 2023Chinese Journal of Aeronautics2023,36,8:0

19Biotinylated dextran amine is an ideal anterograde tracer for the corticospinal tract in a goat model of ischemic corticospinal tract injury显示文摘Existing visualized tracer studies of the corticospinal tract have been focused on rodents, which have markedly different spinal cord structures compared with humans. In this study, the segmental artery feeding the spinal cord was embolized with digital subtraction angiography to establish a goat model of ischemic spinal cord injury. Biotinylated dextran amine was injected into the motor function areas of the cortex in goats with ischemic spinal cord injury. The corticospinal tract originates from the cerebral cortex motor function area, and travels towards the lateral funiculus at the contralateral spinal dorsal horn after decussation at the pyramid. The number of corticospinal tract positive fibers was found to be gradually reduced. These findings indicate that digital subtraction angiography can be applied to a goat model of ischemic spinal cord injury. Biotinylated dextran amine visualizes the course of the goat corticospinal tract in the spinal cord, which is similar to the human spinal cord.
