| 1 | Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. | 3 | Charge structures and cloud-to-ground lightning discharges characteristics in two supercell thunderstorms显示文摘The charge structures and temporal and spatial characteristics of cloud-to-ground (CG) light- ning discharges in two supercell thunderstorms have been analyzed based on the data of three-dimen- sional VHF radiation sources with high time and space resolution produced by lightning discharges. The results indicate that the charge structures in main part (convective region) of the thunderstorms were inverted tripole while a number of positive CG lightning discharges were occurring in the two thun- derstorms. The positive CG lightning discharges oc- curred in main part of the thunderstorms and origi- nated from the positive charge region located at the middle part of the thunderstorms. While a number of negative CG lightning discharges were occurring, the negative CG lightning discharges occurred in the anvil of the thunderstorm. The charge structure is inverted dipole in the region due to the slant of charge structure in main region toward anvil region. The negative charge region located at the upper part of anvil produced a lot of negative CG lightning dis- charges. No or less CG lighting was produced di- rectly by the charge region located at the lower part of the thunderstorm. | 4 | DNA image cytometry test for primary screening of esophageal cancer: a population-based multi-center study in high-risk areas in China显示文摘Objective: To evaluate the feasibility of DNA image cytometry(DNA-ICM) as a primary screening method for esophageal squamous cell cancer(ESCC).Methods: A total of 5,382 local residents aged 40–69 years from three high-risk areas in China(Linzhou in Henan province, Feicheng in Shandong province and Cixian in Hebei province) from 2008 to 2011 were recruited in this population-based screening study. And 2,526 subjects declined to receive endoscopic biopsy examination with Lugol's iodine staining, while 9 and 815 subjects were excluded from liquid-based cytology and DNA-ICM test respectively due to slide quality. Finally, 2,856, 5,373 and 4,567 subjects were enrolled in the analysis for endoscopic biopsy examination, liquid-based cytology and DNA-ICM test, respectively. Sensitivity(SE), specificity(SP), negative predictive values(NPV) and positive predictive values(PPV) as well as their 95% confidence intervals(95% CI) for DNA-ICM, liquid-based cytology and the combination of the two methods were calculated. Receiver operating characteristic(ROC) curves were applied to determine the cutoff point of DNA-ICM for esophageal cancer.Results: DNA-ICM results were significantly correlative with esophageal cancer and precancer lesions(χ~2=18.016, P<0.001). The cutoff points were 5,802, 5,803 and 8,002 based on dissimilar pathological types of low grade intraepithelial neoplasia(LGIN), high grade intraepithelial neoplasia(HGIN), and ESCC, respectively, and 5,803 was chosen in this study considering the SE and SP. | 9 | Serum Uric Acid is Associated with the Predicted Risk of Prevalent Cardiovascular Disease in a Community-dwelling Population without Diabetes显示文摘Objective To examine the association between serum uric acid levels and cardiovascular disease risk among individuals without diabetes. Methods We investigated the association between serum uric acid levels and the risk of prevalent cardiometabolic diseases, 10-year Framingham risk for coronary heart disease, and 10-year risk for atherosclerotic cardiovascular diseases(ASCVD) among 8,252 participants aged ≥ 40 years without diabetes from Jiading district, Shanghai, China. Results Body mass index, waist circumference, blood glucose, glycated hemoglobin, blood pressure, and serum lipids increased progressively across the sex-specific quartiles of uric acid(all P trend < 0.05). Compared with individuals in the lowest quartile, those in the higher quartiles had a significantly higher prevalence of obesity, hypertension, and dyslipidemia(all P trend < 0.05). A fully adjusted logistic regression analysis revealed that individuals in the highest quartile had an increased risk of predicted cardiovascular disease compared with those in the lowest quartile of uric acid. The multivariate adjusted odds ratios(ORs) [95% confidence intervals(CIs)] for the highest quartiles for high Framingham risk were 3.00(2.00-4.50) in men and 2.95(1.08-8.43) in women. The multivariate adjusted ORs(95% CIs) for the highest quartile for high ASCVD risk were 1.93(1.17-3.17) in men and 4.53(2.57-7.98) in women. | 10 | Elevated IL-37 levels in the plasma of patients with severe coronary arterycalcification显示文摘Background Coronary artery calcification (CAC) is a predictor of cardiovascular events and plaque burden and is closely associatedwith chronic inflammation. Intedeukin (IL)-37 is a newly discovered member of the IL-1 family and is considered an anti-inflammatorycytokine. Our recent study on mice indicated that IL-37 could attenuate atherosclerosis and vascular calcification, which suggests that IL-37could be associated with the development of atherosclerosis and related diseases. The aim of this study was to investigate if IL-37 plays arole in the progression of CAC in patients. Methods Two hundred participants with suspected cardiovascular disease were recruited. Thelevels of plasma IL-37, osteoprotegerin (OPG), hypersensitive C-reactive protein (hsCRP) together with other biochemical parameters weremeasured, and a coronary calcium assessment was carried out by multi-detector row CT. A score of 〈 10 AU (Agatston units) denotes anabsence of CAC, a score of 11-100 AU denotes mild CAC, 101-400 denotes moderate CAC, and 〉 400 AU denotes severe CAC. ResultsOur initial data showed that there were no apparent differences in plasma IL-37 levels among patients with or without mild or moderate CAC.However, IL-37 levels were significantly increased in patients with severe CAC (P 〈 0.001). Similar results were observed for plasma OPGand hsCRP levels. When IL-37 levels in patients with severe calcification were compared with that in all of the other non-severe CAC groups,it became apparent that there was a significant positive correlation between IL-37 level and severe CAC (r = 0.360, P 〈 0.001; OR = 1.033)using Spearrnan's correlation and binary logistic regression analysis. | 19 | Association between large artery stenosis,cerebral small vessel disease and risk of ischemic stroke显示文摘We aimed to assess the associations of large artery stenosis(LAS)and cerebral small vessel disease(CSVD)with the risk of ischemic stroke and to investigate their respective and combined contributions.In the prospective population-based Shunyi Study,1,082 stroke-free participants aged 55.9±9.1 years were included.Participants were followed for incident stroke throughout the study period(2013-2019).Total small vessel disease score was used to measure CSVD burden.Cervico-cerebral large artery stenosis was evaluated via brain magnetic resonance angiography and carotid ultrasound.We estimated the risk of ischemic stroke in relation to LAS and CSVD with Cox regression models.During a mean follow-up of 4.2 years,34 participants(3.1%)experienced at least one ischemic stroke.Severe LAS(≥50% stenosis versus no stenosis:HR=3.27(95%CI:1.31-8.18))and high CSVD burden(total small vessel disease score 2-4 versus 0 point:HR=12.73(4.83-33.53))were associated with increased stroke risk independently.In multivariate models,CSVD burden(7.72%)explained a larger portion of the variation in stroke risk than severity of LAS(3.49%).Our findings identified that both LAS and CSVD were associated with future ischemic stroke in asymptomatic subjects,while those with high CSVD burden deserve more attention in primary prevention of stroke. | Fei Han Ding-Ding Zhang Fei-Fei Zhai Jun Xue Jiang-Tao Zhang Shuang Yan Li-Xin Zhou Jun Ni Ming Yao Meng Yang Ming-Li Li Zheng-Yu Jin Qing Dai Shu-Yang Zhang Li-Ying Cui Yi-Cheng Zhu | 2021 | Science China(Life Sciences)2021,64,9: | 9 |
