2Genome Editing Enables Next-Generation Hybrid Seed Production Technology显示文摘The next-generation hybrid seed technology enables the successful production of sortable hybrid seeds from genic male sterile(GMS)lines and maintainers;however,it requires multiple laborious and complicated steps.Here,we designed a simple next-generation hybrid seed production strategy that takes advantage of the CRISPR/Cas9 technology to create a Manipulated GMS Maintainer(MGM)system via a single transformation.Under this schema,the maize male fertility gene ZmMS26 was nullified by removal of its fifth exon using the CRISPR/Cas9 system on a vector,and a second vector carrying a functional ZmMS26 cDNA was co-transformed to restore fertility.The second vector also contains a male gametophyte inactivation gene(ZmAA1)encoding maizeα-amylase driven by the pollen-specific promoter PG47 and an endosperm fluorescent marker(DsRED)driven by the barley endosperm aleurone-specific promoter Ltp2.The derived single-copy hemizygous MGM lines bore a mutated MS26 gene,leading to complete male sterility but normal vegetative growth and grain yield.The MGM system could prevent genetic transmission of the MGM elements via male gametophytes,providing an efficient method for sorting maintainer seeds labeled by DsRED.This strategy can be extended to any GMS gene and to hybrid crops other than maize.Xiantao Qi Congsheng Zhang Jinjie Zhu Changlin Liu Changling Huang Xinhai Li Chuanxiao Xie 2020Molecular Plant2020,13,9:18
3Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 4Inhibitory Activity of Honeysuckle Extracts against Influenza A Virus In Vitro and In Vivo显示文摘Honeysuckle has been used in the treatment of influenza virus infection for thousands of years in China.However,its main active components and the functional mechanisms remain to be elucidated.Here,four honeysuckle extracts,including acids extract,flavonoids extract,total extract and acids-flavonoids mixture,were prepared to clarify the main active antiviral components.The cytopathic effect reduction assay showed that all the four extracts inhibited the replication of influenza viruses H1N1,H3N2 and the oseltamivir-resistant mutant strain H1N1-H275Y.The acids-flavonoids mixture had the strongest inhibitory effects in vitro with EC_(50) values of 3.8,4.1,and >20 lg/m L against H1N1,H3N2 and H1N1-H275Y,respectively,showing competitive antiviral activity with oseltamivir and ribavirin.Honeysuckle acids extract also showed the most significant antiviral activity in vivo.Oral administration of the acids extract at a dosage of 600 mg/kg/d effectively alleviated viral pneumonia,maintained body weight and improved the survival rate to 30% of the mice infected with a lethal dose of H1N1.The results of time-of-drug addition experiment and neuraminidase (NA) inhibition assay showed that honeysuckle extracts had a broad-spectrum inhibitory effect against influenza virus NAs.The flavonoid extract showed the strongest inhibitory effect on the NA of influenza virus H7N9 with an IC_(50) of 24.7 lg/m L.These results suggested that these extracts might exert their antiviral activity by suppressing the release of influenza viruses.Briefly,our findings demonstrate that acids and flavonoids extracts of honeysuckle are the major antiviral active components,and the acids extract has the potential to be developed into an antiviral agent against influenza virus,especially for oseltamivirresistant viruses.Mengwei Li Yuxu Wang Jing Jin Jie Dou Qinglong Guo Xue Ke Changlin Zhou Min Guo 2021Virologica Sinica2021,36,3:11
5Effect of extrusion speed on microstructure and mechanical properties of the Mg-Ca binary alloy显示文摘This work reported the effect of extrusion speeds on the microstructures and mechanical properties of Mg-Ca binary alloy.The results showed that yield strength of the as-extruded Mg-1.2wt.%Ca alloys decrease from∼360MPa to∼258MPa as the ram speed increases from 0.4mm/s to 2.4 mm/s,and the elongation increases from∼3.9%to∼12.2%.The microstructure changes from bimodal grain feature to the complete dynamical recrystallization(DRX)with increase of the extrusion speed.The ultrafine DRXed grains in size of∼0.85μm,the numerous nano-Mg_(2)Ca particles dispersing along the grain boundaries and interiors,as well as the high density of residual dislocations,should account for the high strength.It is believed that the high degree of dynamic recrystallization and the resulting texture randomization play the critical roles in the ductility enhancement of the high-speed extruded Mg alloys.Jingren Li Aiyue Zhang Hucheng Pan Yuping Ren Zhuoran Zeng Qiuyan Huang Changlin Yang Lifeng Ma Gaowu Qin 2021Journal of Magnesium and Alloys2021,9,4:9
6Genetic characterization and linkage disequilibrium mapping of resistance to gray leaf spot in maize(Zea mays L.)显示文摘Gray leaf spot(GLS),caused by Cercospora zeae-maydis,is an important foliar disease of maize(Zea mays L.)worldwide,resistance to which is controlled by multiple quantitative trait loci(QTL).To gain insights into the genetic architecture underlying the resistance to this disease,an association mapping population consisting of 161 inbred lines was evaluated for resistance to GLS in a plant pathology nursery at Shenyang in 2010 and 2011.Subsequently,a genome-wide association study,using 41,101 single-nucleotide polymorphisms(SNPs),identified 51 SNPs significantly(P<0.001)associated with GLS resistance,which could be converted into 31 QTL.In addition,three candidate genes related to plant defense were identified,including nucleotidebinding-site/leucine-rich repeat,receptor-like kinase genes similar to those involved in basal defense.Two genic SNPs,PZE-103142893 and PZE-109119001,associated with GLS resistance in chromosome bins 3.07 and 9.07,can be used for marker-assisted selection(MAS)of GLS resistance.These results provide an important resource for developing molecular markers closely linked with the target trait,enhancing breeding efficiency.Liyu Shi Xiangling Lv Jianfeng Weng Hanyong Zhu Changlin Liu Zhuanfang Hao Yu Zhou Degui Zhang Mingshun Li Xiaoke Ci Xinhai Li Shihuang Zhang 2014The Crop Journal2014,2,Z1:9

8Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 9Preparation,characterization and photocatalytic performance of Mo-doped ZnO photocatalysts显示文摘A series of Mo-doped ZnO photocatalysts with different Mo-dopant concentrations have been prepared by a grinding-calcination method.The structure of these photocatalysts was characterized by a variety of methods,including N 2 physical adsorption,X-ray diffraction(XRD),scanning electron microscopy(SEM),Fourier transform infrared(FT-IR) spectroscopy,photoluminescence(PL) emission spectroscopy,and UV-vis diffuse reflectance spectroscopy(DRS).It was found that Mo 6+ could enter into the crystal lattice of ZnO due to the radius of Mo 6+(0.065 nm) being smaller than that of Zn 2+(0.083 nm).XRD results indicated that Mo 6+ suppressed the growth of ZnO crystals.The FT-IR spectroscopy results showed that the ZnO with 2 wt.% Mo-doping has a higher level of surface hydroxyl groups than pure ZnO.PL spectroscopy indicated that ZnO with 2 wt.% Mo-doping also exhibited the largest reduction in the intensity of the emission peak at 390 nm caused by the recombination of photogenerated hole-electron pairs.The activities of the Mo-doped ZnO photocatalysts were investigated in the photocatalytic degradation of acid orange II under UV light(λ = 365 nm) irradiation.It was found that ZnO with 2 wt.% Mo-doping showed much higher photocatalytic activity and stability than pure ZnO.The high photocatalytic performance of the Mo-doped ZnO can be attributed to a great improvement in the surface properties of ZnO,higher crystallinity and lower recombination rate of photogenerated hole-electron(e-/h+) pairs.Moreover,the undoped Mo species may exist in the form of MoO3 and form MoO3 /ZnO heterojunctions which further favors the separation of e-/h+ pairs.YU ChangLin YANG Kai SHU Qing YU Jimmy C CAO FangFang LI Xin ZHOU XiaoChun 2012Science China Chemistry2012,55,9:8
10Creation of targeted inversion mutations in plants using an RNA-guided endonuclease显示文摘Inversions are DNA rearrangements that are essential for plant gene evolution and adaptation to environmental changes. We demonstrate the creation of targeted inversions and previously reported targeted deletion mutations via delivery of a pair of RNA-guided endonucleases(RGENs) of CRISPR/Cas9. The efficiencies of the targeted inversions were2.6% and 2.2% in the Arabidopsis FLOWERING TIME(At FT) and TERMINAL FLOWER 1(At TFL1)loci, respectively. 11Coagulation factors VII, IX and X are effective antibacterial proteins against drug-resistant Gram-negative bacteria显示文摘Infections caused by drug-resistant“superbugs”pose an urgent public health threat due to the lack of effective drugs;however, certain mammalian proteins with intrinsic antibacterial activity might be underappreciated. Here, we reveal an antibacterial property against Gram-negative bacteria for factors VII, IX and X, three proteins with well-established roles in initiation of the coagulation cascade. These factors exert antibacterial function via their light chains (LCs). Unlike many antibacterial agents that target cell metabolism or the cytoplasmic membrane, the LCs act by hydrolyzing the major components of bacterial outer membrane, lipopolysaccharides, which are crucial for the survival of Gram-negative bacteria. The LC of factor VII exhibits in vitro efficacy towards all Gram-negative bacteria tested, including extensively drug-resistant (XDR) pathogens, at nanomolar concentrations. It is also highly effective in combating XDR Pseudomonas aeruginosa and Acinetobacter baumannii infections in vivo. Through decoding a unique mechanism whereby factors VII, IX and X behave as antimicrobial proteins, this study advances our understanding of the coagulation system in host defense, and suggests that these factors may participate in the pathogenesis of coagulation disorder-related diseases such as sepsis via their dual functions in blood coagulation and resistance to infection. 12Enhancement of innate immune system in monocot rice by transferring the dicotyledonous elongation factor Tu receptor EFR显示文摘The elongation factor Tu(EF-Tu) receptor(EFR) in cruciferous plants specifically recognizes the N-terminal acetylated elf18 region of bacterial EF-Tu and thereby activates plant immunity.It has been demonstrated that Arabidopsis EFR confers broad-spectrum bacterial resistance in the EFR transgenic solanaceous plants.Here,the transgenic rice plants(Oryza sativa L.ssp.japonica cv.Zhonghua 17) and cell cultures with constitutive expression of AtEFR were developed to investigate whether AtEFR senses EF-Tu and thus enhances bacterial resistance in the monocot plants.We demonstrated that the Xanthomonas oryzae-derived elf18 peptide induced oxidative burst and mitogen-activated protein kinase activation in the AtEFR transgenic rice cells and plants,respectively.Pathogenesis-related genes,such as OsPBZ1,were upregulated dramatically in transgenic rice plant and cell lines in response to elf18 stimulation.Importantly,pretreatment with elf18 triggered strong resistance to X.oryzae pv.oryzae in the transgenic plants,which was largely dependent on the AtEFR expression level.These plants also exhibited enhanced resistance to rice bacterial brown stripe,but not to rice fungal blast.Collectively,the results indicate that the rice plants with heterologous expression of AtEFR recognize bacterial EF-Tu and exhibit enhanced broad-spectrum bacterial disease resistance and that pattern recognition receptor-mediated immunity may be manipulated across the two plant classes,dicots and monocots.Fen Lu Huiqin Wang Shanzhi Wang Wendi Jiang Changlin Shan Bin Li Jun Yang Shiyong Zhang Wenxian Sun 2015Journal of Integrative Plant Biology2015,57,7:6

15Arterial relaxation is coupled to inhibition of mitochondrial fission in arterial smooth muscle cells:comparison of vasorelaxant effects of verapamil and phentolamine显示文摘Mitochondria are morphologically dynamic organelles which undergo fission and fusion processes. Our previous study found that arterial constriction was always accompanied by increased mitochondrial fission in smooth muscle cells, whereas inhibition of mitochondrial fission in smooth muscle cells was associated with arterial relaxation. Here, we used the typical vasorelaxants, verapamil and phentolamine, to further confirm the coupling between arterial constriction and mitochondrial fission in rat aorta. Results showed that phentolamine but not verapamil induced vasorelaxation in phenylephrine(PE)-induced rat thoracic aorta constriction. Verapamil, but not phentolamine, induced vasorelaxation in high K^+(KPSS)-induced rat thoracic aorta constriction. Pre-treatment with phentolamine prevented PEbut not KPSS-induced aorta constriction and pre-treatment with verapamil prevented both PE-and KPSSinduced aorta constriction. Transmission electron microscopy(TEM) results showed that verapamil but not phentolamine inhibited KPSS-induced excessive mitochondrial fission in aortic smooth muscle cells,and verapamil prevented both PE-and KPSS-induced excessive mitochondrial fission in aortic smoothmuscle cells. Verapamil inhibited KPSS-induced excessive mitochondrial fission in cultured vascular smooth muscle cells(A10). 16Somatosensory Neuron Typing with High-Coverage Single-Cell RNA Sequencing and Functional Analysis显示文摘Different physical and chemical stimuli are detected by the peripheral sensory receptors of dorsal root ganglion(DRG) neurons, and the generated inputs are transmitted via afferent fibers into the central nervous system. The gene expression profiles of DRG neurons contribute to the generation, transmission, and regulation of various somatosensory signals. Recently, the single-cell transcriptomes, cell types, and functional annotations of somatosensory neurons have been studied. In this review,we introduce our classification of DRG neurons based on single-cell RNA-sequencing and functional analyses, and discuss the technical approaches. 17Mechanism and effect of Shijueming (Concha Haliotidis) on serum calcium in spontaneously hypertensive rats显示文摘OBJECTIVE: To observe the impact of Shijueming (Concha Haliotidis) on spontaneously hypertensive rats via blood pressure, serum calcium, vascular smooth muscle membrane L-type calcium channel α1 C subunit (CaL-α1C), plasma membrane calcium-ATPase (PMCA) mRNA expression, and the L-type calcium channel in vascular smooth muscle cells. METHODS:Twelve-week-old male rats with spontaneous hypertension were divided into three groups: a Shijueming (Concha Haliotidis) group (group 1), a nifedipine group (group 2), and a distilled water group (group 3). All were given a four-week treatment. Blood pressure and dissociative serum calcium were examined before treatment. Blood pressure was taken every week during treatment. Atomic absorption spectrometry was used to examine dissociative serum calcium. Re-verse transcription-polymerase chain reaction was used to examine the expression of CaL-α1C and PM-CA1 mRNA. The patch clamp technique was used to examine the electrophysiological characteristics of the vascular smooth muscle cell calcium channels. RESULTS: After treatment, blood pressure of the Shijueming (Concha Haliotidis) group lowered but not significantly (P>0.05). Blood pressure of the nifedipine group lowered significantly (P<0.05). Blood pressure of the distilled water group remained high. The concentration of serum calcium in the Shijueming (Concha Haliotidis) and the distilled water groups lowered (P<0.05). Expression of CaL-α1C mRNA in the nifedipine group decreased compared with the distilled water group (P<0.01). There was the decreasing trend in the Shijueming (Concha Haliotidis) group, but it was not statistically significant. Shijueming (Concha Haliotidis) also had effects on the expression of PMCA1 mRNA but with- out statistical significance. However, there was a significant decreasing effect on vascular smooth muscle cell I Ca-L flow. CONCLUSION: This study indicated that Shijueming (Concha Haliotidis) could increase serum calcium and decrease blood pressure. 18Neuro-protective effects of CNTF on hippocampal neurons via an unknown signal transduction pathway显示文摘In our previous study, we proposed that there may be an unknown pathway in the upper stream of the known signal transduction pathway of Ciliary neurotrophic factor (CNTF) that mediates the neuro-protective function of CNTF. In the present experiment, we observed that the neuro-protective function of the non-classic signal transduction path- way in a L-NMDA (a glutamic acid ion type receptor atagonist) induced hippocampal neuron injury model, using primary culture rat hippocampal neurons, con- tinuous photography and gp130 immunohistochemi- cal assay. The results showed that L-NMDA induced injurious reaction of hippocampal neurons, and CNTF was able to inhibit the toxic action of L-NMDA on hippocampal neurons. Additionally, when JAK/STATs in the known classic signal transduction pathway of CNTF were blocked by PTPi-2, the protective effect of CNTF against L-NMDA injury still existed. L-NMDA caused a rapid increase in the concentration of hip- pocampal intracellular free [Ca2+]i. CNTF was able to attenuate L-NMDA-induced elevation of [Ca2+]i, and blocking JAK/STATs in the known classic signal trans- duction pathway of CNTF did not affect L-NMDA- induced elevation of [Ca2+]i, indicating that, apart from the known classic signal transduction pathway, there may be some other transduction pathways for CNTF to exert the protective effect on hippocampal neurons, and this pathway is related to [Ca2+].LI Ping WANG Zongwen YAN Jin Li Zhaoshen JIANG Chunlei NI Xin YANG Yongji LIU Fang LU Changlin 2006Chinese Science Bulletin2006,51,1:4
19Presynaptic inhibition of nociceptive neurotransmission by somatosensory neuron-secreted suppressors显示文摘Noxious stimuli cause pain by activating cutaneous nociceptors.The Aδ-and C-fibers of dorsal root ganglion(DRG) neurons convey the nociceptive signals to the laminae Ⅰ—Ⅱ of spinal cord.In the dorsal horn of spinal cord,the excitatory afferent synaptic transmission is regulated by the inhibitory neurotransmitter γ-aminobutyric acid and modulators such as opioid peptides released from the spinal interneurons,and by serotonin,norepinepherine and dopamine from the descending inhibitory system.In contrast to the accumulated evidence for these central inhibitors and their neural circuits in the dorsal spinal cord,the knowledge about the endogenous suppressive mechanisms in nociceptive DRG neurons remains very limited.In this review,we summarize our recent findings of the presynaptic suppressive mechanisms in nociceptive neurons,the BNP/NPR-A/PKG/BK_(Ca) channel pathway,the FSTL1/α1Na^+-K^+ ATPase pathway and the activin C/ERK pathway.These endogenous suppressive systems in the mechanoheat nociceptors may also contribute differentially to the mechanisms of nerve injury-induced neuropathic pain or inflammation-induced pain.Kaicheng Li Bing Cai Changlin Li Xu Zhang 2017Science China(Life Sciences)2017,60,9:3
20Experimental and mechanistic understanding of photo-oxidation of methanol catalyzed by CuO/TiO2-spindle nanocomposite:Oxygen vacancy engineering显示文摘We report experimental and mechanistic understanding of methanol oxidation to produce methyl formate using CuO/Ti02-spindle composite as a promising photocatalyst under mild conditions with over 97%conversion and 83%selectivity.The catalysts are obtained via precise depositing of CuO nanoclusters(size:~3.5 nm)at the{101}facet of the TiO2 to optimally tune exciton recombination through oxygen vacancies generation,evidenced by photoluminescence and Raman spectroscopy measurements.The turnover frequency(TOF)and the apparent quantum efficiency(AQE)of the 7%CuO/TiO2-spindle composites reach up to 23.8 molmethanol·gcat^-1·h^-1 and 55.2%at 25℃,respectively,which are substantially higher than these previously reported photocatalysts.Further,the in-situ attenuated total reflection infrared spectroscopy analysis reveals that the methanol oxidation most likely takes place through the conversion of adsorbed methoxy(CH30^*)to formaldehyde(CHO^*)intermediate,a subject of major debate for a long time.The adsorbed formaldehyde(CHO^*)thus produced reacts with another CH30^*species in its close proximity to form the final product of methyl formate.Results of this study provide insights into the reaction mechanism,and offer guidelines to systematically develop and apply photocatalysts for methanol conversion and related reactions via surface engineering.Quanquan Shi Zhaoxian Qin Changlin Yu Ammara Waheed Hui Xu Yong Gao Hadi Abroshan Gao Li 2020Nano Research2020,13,4:3
