| 1 | HIV-1 Vif suppresses antiviral immunity by targeting STING显示文摘HIV-1 infection-induced cGAS–STING–TBK1–IRF3 signaling activates innate immunity to produce type I interferon(IFN).The HIV-1 nonstructural protein viral infectivity factor(Vif)is essential in HIV-1 replication,as it degrades the host restriction factor APOBEC3G.However,whether and how it regulates the host immune response remains to be determined.In this study,we found that Vif inhibited the production of type I IFN to promote immune evasion.HIV-1 infection induced the activation of the host tyrosine kinase FRK,which subsequently phosphorylated the immunoreceptor tyrosine-based inhibitory motif(ITIM)of Vif and enhanced the interaction between Vif and the cellular tyrosine phosphatase SHP-1 to inhibit type I IFN.Mechanistically,the association of Vif with SHP-1 facilitated SHP-1 recruitment to STING and inhibited the K63-linked ubiquitination of STING at Lys337 by dephosphorylating STING at Tyr162.However,the FRK inhibitor D-65495 counteracted the phosphorylation of Vif to block the immune evasion of HIV-1 and antagonize infection.These findings reveal a previously unknown mechanism through which HIV-1 evades antiviral immunity via the ITIM-containing protein to inhibit the posttranslational modification of STING.These results provide a molecular basis for the development of new therapeutic strategies to treat HIV-1 infection. | Yu Wang Gui Qian Lingyan Zhu Zhuo Zhao Yinan Liu Wendong Han Xiaokai Zhang Yihua Zhang Tingrong Xiong Hao Zeng Xianghui Yu Xiaofang Yu Xiaoyan Zhang Jianqing Xu Quanming Zou Dapeng Yan | 2022 | Cellular & Molecular Immunology2022,19,1: | 2 |
| 2 | KLF2 inhibits TGF-β-mediated cancer cell motility in hepatocellular carcinoma显示文摘Feedback regulation plays a pivotal role in determining the intensity and duration of TGF-p signaling and subsequently affecting the pathophysiological roles of TGF-p,including those in liver malignancy.KLF2,a member of the Kruppel-like factor(KLF)fam ily transcription factors,has been implicated in impeding hepatocellular carcinoma(HCC)development.However,the underlying molecular mechanisms are not fully understood.In the present study,we found that TGF-p stimulates the expression of KLF2 gene in several HCC cell lines.KLF2 protein is able to inhibit TGF-p/Smad signaling in HCC cells as assessed by luciferase reporter assay.Further studies indicated that KLF2 inhibits the transcriptional activity of Smad2/3 and Smad4 and ameliorates TGF-p-induced target gene expression,therefore creating a novel negative feedback loop in TGFp signaling.Functionally,stably expression of KLF2 in HCCLM3 cells attenuated TGF-卜induced cancer cell motility in wound-healing and transwell assays by interfering with TGF-p-mediated upregulation of MMP2.Together,our results revealed that KLF2 protein has a tumor-suppressive function in HCC through a negative feedback loop over TGF-p signaling. | Yining Li Shuo Tu Yi Zeng Cheng Zhang Tian Deng Weicheng Luo Lingyan Lian Ling Chen Xiangyang Xiong Xiaohua Yan | 2020 | Acta Biochimica et Biophysica Sinica2020,52,5: | 2 |
| 3 | A Target Grabbing Strategy for Telerobot Based on Improved Stiffness Display Device显示文摘Most target grabbing problems have been dealt with by computer vision system, however, computer vision method is not always enough when it comes to the precision contact grabbing problems during the teleoperation process, and need to be combined with the stiffness display to provide more effective information to the operator on the remote side. Therefore, in this paper a more portable stiffness display device with a small volume and extended function is developed based on our previous work. A new static load calibration of the improved stiffness display device is performed to detect its accuracy, and the relationship between the stiffness and the position is given. An effective target grabbing strategy is presented to help operator on the remote side to judge and control and the target is classified by multi-class SVM(supporter vector machine). The teleoperation system is established to test and verify the feasibility. | 5 | Non-target Screening and Quantitation of Organic Chlorides in Oilfield Chemicals with Comprehensive Two Dimensional Gas Chromatography Coupled with Time of Flight Mass Spectrometry显示文摘Advanced analytical methodology based on comprehensive two-dimensional gas chromatography-time of flight mass spectrometry(GC×GC-TOFMS) is proposed for investigation of organic chlorides in different oilfield chemicals.The target screening was initially carried out on 8 suspected organic chlorides by evaluating the capability of the enhanced separation and reliable identification at a trace concentration. GC×GC-TOFMS allowed for the fast and automated analysis of organic chlorides at a level of 200 μg/L. This method was subsequently applied for non-target screening of organic chlorides in different oilfield chemicals at various locations across China. 22 organic chlorides were identified and verified by comparison with pure standards in the mixed sample. Finally, this method was used to determine the content of the organic chlorides in individual samples.
