| 1 | Syndecan-2, negatively regulated by miR-20b-5p, contributes to 5-fluorouracil resistance of colorectal cancer cells via the JNK/ERK signaling pathway显示文摘5-Fluorouracil (5-FU) resistance has been long considered as an obstacle to the efficacy of chemotherapy in colorectal cancer (CRC). In this study, we demonstrated the role of miR-20b-5p-regulated syndecan-2 (SDC2) in 5-FU resistance of CRC cells. 5-FU-resistant SW480 CRC cells were established by treatment of SW480 cells with stepwise increase of 5-FU concentration. The results showed that SDC2 was expressed significantly higher in SW480/5-FU cells than in SW480/WT cells as revealed by quantitative real-time polymerase chain reaction and western blot analysis. MTT assay and BrdU assay showed that SDC2 overexpression led to increased cell survival rate, while SDC2 knockdown reversed the drug resistance of SW480/5-FU cells. Wound healing and transwell invasion assays revealed that knockdown of SDC2 inhibited the migratory and invasive ability of SW480/5-FU cells. Moreover, animal experiments indicated that si-SDC2 plays a suppressive role in tumor growth in vivo. We also confirmed that miR-20b-5p interacted with SDC2, which reversed the effect of SDC2 in SW480/5-FU cells via the c-Jun N-terminal kinase (JNK)/extracellular regulated protein kinases (ERK) signaling pathway. | 2 | Mitochondrial uncoupler triclosan induces vasorelaxation of rat arteries显示文摘Our previous studies found that mitochondrial uncouplers induced vasodilation. Triclosan, the broad spectrum antibacterial agent, is the active ingredient in soaps and toothpastes. It was reported that triclosan induced mitochondrial uncoupling, so we aim to investigate the effects of triclosan on vascular function of rat mesenteric arteries and aorta. The isometric tension of rat mesenteric artery and thoracic aorta was recorded by multi-wire myograph system. The cytosolic [Ca^(2+)]_i, mitochondrial reactive oxygen species(mitoROS), and mitochondrial membrane potential of smooth muscle cells(A10 cells) were measured using laser scanning confocal microscopy. Triclosan treatment relaxed phenylephrine(PE)-and high K^(+)(KPSS)-induced constriction, and pre-treatment with triclosan inhibited PE-and KPSS-induced constriction of rat mesenteric arteries. In rat thoracic aorta, triclosan also relaxed PE-and KPSS-induced constriction. Triclosan induces vasorelaxation without involving KATPchannel activation in smooth muscle cells of arteries.Triclosan treatment increased cytosolic [Ca^(2+)]_i, mitochondrial ROS production and depolarized mitochondrial membrane potential in A10 cells. In conclusion, triclosan induces mitochondrial uncoupling in vascular smooth muscle cells and relaxes the constricted rat mesenteric arteries and aorta of rats. | 4 | Heat treatment of hot-isostatic-pressed 60NiTi shape memory alloy:Microstructure,phase transformation and mechanical properties显示文摘60NiTi alloy is considered to be a promising material for specialized bearing and gear applications due to its high hardness,strength,and low modulus.However,fabricating 60NiTi through conventional processing methods is challenging due to the brittleness and poor workability.In this study,60NiTi with high relative density was successfully fabricated directly from pre-alloyed powder through hot isostatic pressing.The effects of solution and aging treatments on microstructure and mechanical properties were systematically studied by advanced characterization techniques.The hot-isostatic-pressed 60NiTi showed low average hardness and elastic strain due to the formation of a soft Ni_(3)Ti phase and B2 NiTi matrix.Solution treatment above 1000℃dissolved the Ni_(3)Ti phase and promoted the formation of nanoscale Ni_(4)Ti_(3)precipitates,which significantly improved the hardness,strength,and elastic strain of 60NiTi.The formation of the Ni_(4)Ti_(3) phase can be mainly attributed to the driving forces induced by the chemical supersaturation and mechanical stress concentration.Finally,the phase transformation mechanisms during heat treatment and compression test were discussed. | Yinghao Zhou Xiyu Yao Wenfei Lu Dandan Liang Xiaodi Liu Ming Yan Jun Shen | 2022 | Journal of Materials Science & Technology2022,,12: | 1 |
| 5 | Structural characterization of SARS-CoV-2 dimeric ORF9b reveals potential fold-switching trigger mechanism显示文摘The constant emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants indicates the evolution and adaptation of the virus.Enhanced innate immune evasion through increased expression of viral antagonist proteins,including ORF9b,contributes to the improved transmission of the Alpha variant;hence,more attention should be paid to these viral proteins.ORF9b is an accessory protein that suppresses innate immunity via a monomer conformation by binding to Tom70.Here,we solved the dimeric structure of SARS-CoV-2 ORF9b with a long hydrophobic tunnel containing a lipid molecule that is crucial for the dimeric conformation and determined the specific lipid ligands as monoglycerides by conducting a liquid chromatography with tandem mass spectrometry analysis,suggesting an important role in the viral life cycle.Notably,a long intertwined loop accessible for host factor binding was observed in the structure.Eight phosphorylated residues in ORF9b were identified,and residues S50 and S53 were found to contribute to the stabilization of dimeric ORF9b.Additionally,we proposed a model of multifunctional ORF9b with a distinct conformation,suggesting that ORF9b is a fold-switching protein,while both lipids and phosphorylation contribute to the switching.Specifically,the ORF9b monomer interacts with Tom70 to suppress the innate immune response,whereas the ORF9b dimer binds to the membrane involving mature virion assembly.Our results provide a better understanding of the multiple functions of ORF9b. | Xiyue Jin Xue Sun Yan Chai Yu Bai Ying Li Tianjiao Hao Jianxun Qi Hao Song Catherine CL Wong George F.Gao | 2023 | Science China(Life Sciences)2023,66,1: | 0 |
| 6 | A fluid flow model in the lacunar-canalicular system under the pressure gradient and electrical field driven loads显示文摘The lacunar-canalicular system(LCS)is acknowledged to directly participate in bone tissue remodeling.The fluid flow in the LCS is synergic driven by the pressure gradient and electric field loads due to the electro-mechanical properties of bone.In this paper,an idealized annulus Maxwell fluid flow model in bone canaliculus is established,and the analytical solutions of the fluid velocity,the fluid shear stress,and the fluid flow rate are obtained.The results of the fluid flow under pressure gradient driven(PGD),electric field driven(EFD),and pressure-electricity synergic driven(P-ESD)patterns are compared and discussed.The effects of the diameter of canaliculi and osteocyte processes are evaluated.The results show that the P-ESD pattern can combine the regulatory advantages of single PGD and EFD patterns,and the osteocyte process surface can feel a relatively uniform shear stress distribution.As the bone canalicular inner radius increases,the produced shear stress under the PGD or P-ESD pattern increases slightly but changes little under the EFD pattern.The increase in the viscosity makes the flow slow down but does not affect the fluid shear stress(FSS)on the canalicular inner wall and osteocyte process surface.The increase in the high-valent ions does not affect the flow velocity and the flow rate,but the FSS on the canalicular inner wall and osteocyte process surface increases linearly.In this study,the results show that the shear stress sensed by the osteocyte process under the P-ESD pattern can be regulated by changing the pressure gradient and the intensity of electric field,as well as the parameters of the annulus fluid and the canaliculus size,which is helpful for the osteocyte mechanical responses.The established model provides a basis for the study of the mechanisms of electro-mechanical signals stimulating bone tissue(cells)growth. | Xiaogang WU Xiyu WANG Chaoxin LI Zhaowei WANG Yuqin SUN Yang YAN Yixian QIN Pengcui LI Yanqin WANG Xiaochun WEI Weiyi CHEN | 2022 | Applied Mathematics and Mechanics(English Edition)2022,43,6: | 0 |

| 10 | In-situ alloyed CoCrFeMnNi high entropy alloy:Microstructural development in laser powder bed fusion显示文摘In-situ alloying has the potential to combine the compositional flexibility of high entropy alloys(HEAs)and the advanced forming capability of laser powder bed fusion(LPBF).This study fundamentally investigated the elemental homogenisation and grain development in the in-situ alloying process of CoCrFeMnNi HEA,by analysing the basic units,i.e.,tracks and layers,and introducing Mn as an alloying element to the base Co Cr Fe Ni HEA.Different modelling methods were employed to predict meltpool dimensions,and the results indicated the dependence of the modelling on practical meltpool modes.Delimitation of elemental distribution was found in keyhole meltpools since an intensive flow was generated due to recoil pressure.The homogeneity of in-situ alloyed Mn in single tracks was insufficient whether operated in conduction mode or keyhole mode,which required remelting from adjacent tracks and following layers to promote homogenisation significantly.The preferred orientation in single tracks along scanning directions changed from<001>to<101>as the scanning speed increased,although the cross-sections were similar in size with identical linear energy density.Such preference can be inherited during the printing process and lead to different textures in three-layer samples.It was also observed that applying hatch spacing smaller than a half meltpool width could coarsen the grains in a layer.The results from this study provide structure-parameter correlations for future microstructural tailoring and manipulation. | Peng Chen Xiyu Yao Moataz M.Attallah Ming Yan | 2022 | Journal of Materials Science & Technology2022,,28: | 0 |
| 11 | Laser Wire Additive Manufacturing of Ti-6Al-4V Alloy and Its Machine Learning Study for Parameters Optimization(Invited)显示文摘Ti-6Al-4V is a benchmark Ti alloy. Laser wire additive manufacturing(LWAM) offers advanced manufacturing capability to the alloy for applications possibly including exploration of outer space. As a typical multiple-variable process, LWAM is complex, which, however, can be analyzed, predicated or even optimized by artificial intelligence(AI) methods such as machine learning(ML). In this study, printing parameters of the Ti-6Al-4V is firstly optimized using single-track-single-layer experiments, and then single-track-multiple-layer samples are printed, whose properties in terms of hardness and compressive strength are analyzed subsequently by both experiments and ML. The two ML approaches, artificial neural network(ANN) and support vector machine(SVM), are employed to predict the experimental results, whose coefficients of determination R2 show good values. Further optimized properties are realized by adopting genetic algorithm(GA) and simulated annealing(SA) approaches, which contribute to high mechanical properties achieved, for instance, an engineering compressive strength of about 1694 MPa. | 13 | Comparative studies on carbon origin of dawsonite between Hailaer Basin in China and Bowen-Gunnedah-Sydney basin system in Australia显示文摘Dawsonite, NaAlCO3(OH)2, is widespread as a cement, replacement and cavity filling in Hailaer Basin in China and Bowen-Gunnedah-Sydney (BGS) basin system in Australia. The origin of dawsonite is emphatically contrasted and analyzed through stable isotopic composition. Dawsonite δ13C values ranging from -4.0×10-3 to 4.1×10-3 are remarkably consistent through the BGS basin system. The calculated δ13C values of CO2 gas in isotopic equilibrium with dawsonite range from -11.3×10-3 to -4.6×10-3. These values indicate carbon of dawsonite came from inorganic CO2 gas accompanied by magmatic activity. In Hailaer Basin, the Dawsonite δ13C values ranging from -4.64×10-3 to 2.12×10-3 are also consistent. The calculated δ13C values of CO2 gas in isotopic equilibrium with dawsonite range from -11.82×10-3 to -5.11×10-3. According to the coincidence of dawsonite-bearing well and CO2 gas well with mantle source,lying along deep fracture within or adjacent to Yanshanian granite,it is concluded that CO2 gas forming dawsonite is derived from mantle related to magmatic process during the Yanshanian.
