1Clinical characteristics of 24 asymptomatic infections with COVID-19 screened among close contacts in Nanjing,China显示文摘Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections.This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China,from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records,laboratory assessments,and chest CT scans were reviewed.As a result,none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever,cough,fatigue,etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs.The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization.These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died.The median communicable period,defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests,was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members,which even caused severe COVID-19 pneumonia.Overall,the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization.However,the communicable period could be up to three weeks and the communicated patients could develop severe illness.These results highlighted the importance of close contact tracing and longitudinally surveillance via virus nucleic acid tests.Further isolation recommendation and continuous nucleic acid tests may also be recommended to the patients discharged.Zhiliang Hu Ci Song Chuanjun Xu Guangfu Jin Yaling Chen Xin Xu Hongxia Ma Wei Chen Yuan Lin Yishan Zheng Jianming Wang Zhibin Hu Yongxiang Yi Hongbing Shen 2020Science China(Life Sciences)2020,63,5:77
2PM_(2.5)-bound polycyclic aromatic hydrocarbons(PAHs) in Beijing:Seasonal variations,sources,and risk assessment显示文摘Polycyclic aromatic hydrocarbons(PAHs) have been of health concern due to its carcinogenesis and mutagenesis. In this study, we aimed to assess the variations, sources, and lifetime excessive cancer risk(ECR) attributable to PAHs bound to ambient particulate matters with aerodynamic diameter less than 2.5 μm(PM_(2.5)) in metropolitan Beijing, China. We collected24-hour integrated PM_(2.5) samples on daily basis between November 2014 and June 2015 across both central heating(cold months) and non-heating(warm months) seasons, and further analyzed the PAH components in these daily PM_(2.5) samples. Our results showed that total concentrations of PM_(2.5)-bound PAHs varied between(88.6 ± 75.4) ng/m^3 in the cold months and(11.0 ± 5.9) ng/m^3 in the warm months. Benzo[a]pyrene(Ba P), the carcinogenic marker of PAHs,averaged at 5.7 and 0.4 ng/m^3 in the cold and warm months, respectively. Source apportionment analyses illustrated that gasoline, biomass burning, diesel, coal combustion and cooking were the major contributors, accounting for 12.9%, 17.8%, 24.7%, 24.3% and 6.4% of PM_(2.5)-bound PAHs, respectively. The BaP equivalent lifetime ECR from inhalation of PM_(2.5)-bound PAHs was 16.2 cases per million habitants. Our results suggested that ambient particulate reduction from energy reconstruction and adaption of clean fuels would result in reductions PM_(2.5)-bound PAHs and its associated cancer risks. 3Total pollution effect of urban surface runoff显示文摘For pollution research with regard to urban surface runoff,most sampling strategies to date have focused on differences in land usage. With single land-use sampling,total surface runoffpollution effect cannot be evaluated unless every land usage spot is monitored. 4Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. 7Prognostic assessment of apoptotic gene polymorphisms in non-small cell lung cancer in Chinese显示文摘Apoptosis plays a key role in inhibiting tumor growth, progression and resistance to anti-tumor therapy. We hypothesized that genetic variants in apoptotic genes may affect the prognosis of lung cancer. To test this hypothesis, we selected 38 potentially functional single nucleotide polymorphisms (SNPs) from 12 genes (BAX, BCL2, BID, CASP3, CASP6, CASP7, CASP8, CASP9, CASP10, FAS, FASLG and MCL1) involved in apoptosis to assess their prognostic significance in lung cancer in a Chinese case cohort with 568 non-small cell lung cancer (NSCLC) patients. Thirty-five SNPs passing quality control underwent association analyses, 11 of which were shown to be significantly associated with NSCLC survival (P<0.05). After Cox stepwise regression analyses, 3 SNPs were independently associated with the outcome of NSCLC (BID rs8190315: P=0.003; CASP9 rs4645981: P=0.007 and FAS rs1800682: P=0.016). A favorable survival of NSCLC was significantly associated with the genotypes of BID rs8190315 AG/GG (adjusted HR=0.65, 95% CI: 0.49-0.88), CASP9 rs4645981 AA (HR=0.22, 95% CI: 0.07-0.69) and FAS rs1800682 GG (adjusted HR=0.67, 95% CI: 0.46-0.97). Time-dependent receptor operation curve (ROC) analysis revealed that the area under curve (AUC) at year 5 was significantly increased from 0.762 to 0.819 after adding the risk score of these 3 SNPs to the clinical risk score. The remaining 32 SNPs were not significantly associated with NSCLC prognosis after adjustment for these 3 SNPs. 8Function of Ca^(2+)-/calmodulin-dependent protein kinase Ⅳ in Ca^(2+)-stimulated neuronal signaling and behavior显示文摘The activity of Ca2+/calmodulin-dependent protein kinase IV(Ca MKIV) is sensitive to activity-dependent changes in the level of intracellular Ca2+.Following neuronal stimulation,the activation of Ca MKIV may trigger synaptic modifications and transcriptional responses,both of which are involved in regulating cognitive and emotional behavior.Here,we used Ca MKIV knockout(KO) neurons and mice to examine the function of Ca MKIV in Ca2+-stimulated intracellular signaling and animal behavior,respectively.Following NMDA receptor activation or membrane depolarization,the up-regulation of CREB(c AMP responsive element binding protein) and its target gene Bdnf(brain-derived neurotrophic factor) was intact in cortical neurons obtained from Ca MKIV KO mice.Ca MKIV KO mice displayed severe impairment in contextual fear memory but normal locomotor activity and anxiety level in the contextual training chamber.Although Ca MKIV KO mice showed normal memory in the standard passive avoidance task,they were defective in learning the temporal dissociative passive avoidance task.As indicated by the light/dark test and marble-burying test data,Ca MKIV KO mice showed less anxiety and normal perseveration.In the voluntary wheel-running test,Ca MKIV KO mice showed normal running time and distance but higher maximal running speed.Our results demonstrate the function of Ca MKIV in regulating different forms of fear memory,anxiety,and certain aspect of motor function.SONG ZiXiang CHEN Qi DING Qi ZHENG Fei LI ChengWei XU LePing WANG HongBing 2015Science China(Life Sciences)2015,58,1:6
9A 5'-flanking region polymorphism in toll-like receptor 4 is associated with gastric cancer in a Chinese population显示文摘Objective:Inflammation induced by H.pylori colonization in the stomach is related to the development of gastric cancer and the genetic variations of the genes involved in the immune responses modify the host response to the infection. The aim of this study was to evaluate whether polymorphisms in the toll-like receptor 4 (TLR4) gene, a key regulator of both innate and adaptive immunity, were related to the susceptibility togastric cancer in a Chinese population. Methods: Two variations in the 5'-flanking region of TLR4 (rs1927914 T>C and rs10759932 T>C) were genotyped by using the PCR-restriction fragment length polymorphism (RFLP) assay in a case-control study of 1,053 incident gastric cancer cases and 1,100 cancer-free controls in a Chinese population. Results: Individuals carrying the C allele of rs10759932 had a significantly reduced risk of gastric cancer (adjusted OR=0.81; 95%CI=0.67-0.96), compared with the wild-type homozygote (TT), and the protective effect was not significantly different among subgroups stratified by age, sex, smoking, drinking and H.pylori infection status (P for heterogeneity >0.05). No significant association was observed between rs1927914 and gastric cancer risk in this study population. Conclusion: The T to C allele substitution of rs10759932 may play a protective role in gastric carcinogenesis in a Chinese population. 10Moho offset beneath the central Bangong-Nujiang suture of Tibetan Plateau显示文摘By analyzing teleseismic waveforms recorded by 53 stations of Hi-Climb profile passing through the central Bangong-Nujiang suture(BNS) ,a total of 4764 high-quality receiver functions are obtained.The average crustal thickness and Poisson's ratio beneath each station are estimated using the travel time of Ps and PpPs of the Moho.The discontinuities such as the Moho,the 410and 660-km interfaces are also studied using the common converted points(CCP) time to depth migration of receiver functions. The main results are as follows:(1) The Moho of Lhasa terrane and that of Qiangtang terrane nearby BNS are overridden and offset by~10 km.The structural geometry shows a northward uplifting and the southward deepening for the Moho of Lhasa terrane and Qiangtang terrane,respectively,which is related to the reactivated structure beneath BNS since Cenozoic era.(2) The variation range of Poisson's ratios along the profile is between 0.237 and 0.280,indicating that the crust is mainly composed of felsic and intermediate rocks.The anti-correlation between the crustal thickness and Poisson's ratio suggests that thicker crust beneath the southern Qiangtang terrane may be related to the successive thrust of felsic and intermediate rock of Lhasa terrane.(3) The thickness of the mantle transition zone along the profile remains about 255 km,implying that the tectonic activities caused by the India-Asia collision are confined to the depths above 410 km.XU Qiang ZHAO JunMeng CUI ZhongXiong PEI ShunPing LIU HongBing 2010Chinese Science Bulletin2010,55,7:4

12Prognostic value of systemic immune- inflammation index in acute/subacute patients with cerebral venous sinus thrombosis显示文摘Objective To evaluate the prognosis values of systemic immune-inflammation index(SII)in non-chronic cerebral venous sinus thrombosis(CVST).Methods patients with CVST,admitted to the First Affiliated Hospital of Zhengzhou University,were retrospectively identified from January 2013 to December 2018.We selected patients in acute/subacute phase from database.Functional outcomes of patients were evaluated with the modified Rankin Scale(mRS)-mRS 3-6 as poor outcomes and mRS 6 as death.The overall survival time was defined as the date of onset to the date of death or last follow-up date.Survival analysis was described by the Kaplan-Meier curve and Cox regression analysis.Multivariate logistic regression analysis assessed the relationship between SII and poor functional outcome.The area under the Receiver Operating Curve curve(AUC)was estimated to evaluate the ability of SII in prediction.Results A total of 270 patients were included and their duration of follow-up was 22 months(6-66 months),of whom 31 patients had poor outcomes and 24 patients dead.Cox regression analysis showed that SII(HR=1.304,95%CI:1.101 to 1.703,p=0.001)was a predictor of death in non-chronic CVST.Patients with higher SII presented lower survival rates(p=0.003).The AUC of SII was 0.792(95%CI:0.695 to 0.888,p=0.040)with a sensitivity of 69.6%and specificity of 80.1%.Subgroups analysis demonstrated that SII was an important predictor of poor outcomes in male(OR=1.303,95%CI:1.102 to 1.501,p=0.011)and pregnancy/puerperium female(OR=1.407,95%CI:1.204 to 1.703,p=0.034).Conclusions SII was a potential predictor in the poor prognosis of patients with acute/subacute CVST,especially in male and pregnancy/puerperium female.Shen Li Kai Liu Yuan Gao Lu Zhao Rui Zhang Hui Fang Yongli Tao Hongbing Liu Jiawei Zhao Zongping Xia Yuming Xu Bo Song 2020Stroke & Vascular Neurology2020,5,4:3
13Enzymatic hydrolysis of silkworm pupa and its allergenicity evaluation by animal model with different immunization routes显示文摘Silkworm pupa is a nourishing food with high nutritional value,but its consumption has been greatly limited given its allergenicity.Enzyme hydrolytic technique is recognized as an effective method to reduce the allergenicity of protein.In this study,we aimed to investigate the effect of enzymolysis on the allergenicity of silkworm pupa.Crude silkworm pupa protein was extracted through alkali extraction and acid precipitation,which included 5 proteins with the molecular weights ranging from 34 kDa to 76 kDa,and silkworm pupa were then hydrolyzed by alkaline protease.The allergenicity of silkworm pupa protein and its enzymatic hydrolysates was evaluated by establishing BALB/c mice model,and the mice were immunized via intragastric gavage and intraperitoneal injection,respectively.The results indicated that the intraperitoneal inj ection immunization route induced more by detecting with antibodies,histamine and Th2-related cytokines.Moreover,mice treated with silkworm pupa protein peptide displayed no obvious allergic symptoms,indicating that enzyme hydrolytic technique could significantly reduce the allergenicity of silkworm pupa.Yan Dai Meijia Huang Yujuan Xu Lixia Mu Jingyan Gao Hongbing Chen Zhihua Wu Anshu Yang Yong Wu Xin Li 2023Food Science and Human Wellness2023,12,3:3
14Heat flow pattern,base of methane hydrates stability zones and BSRs in Shenhu Area,northern South China Sea显示文摘Using the collected 433 heat flow values,we estimated the bases of methane hydrate stability zone (BHSZ),in northern South China Sea (NSCS).Through comparing BHSZs with the depths of bottom simulating reflectors (BSRs),in Shenhu Area (SA),we found that there are big differences between them.In the north of SA,where the water depth is shallow,many slumps developed and the sedimentation rate is high,it appears great negative difference (as large as-192%).However,to the southeast of SA,where the water depth is deeper,sedimentation rate is relatively low and uplift basement topography exists,it changes to positive difference (as large as +45%).The differences change so great,which haven't been observed in other places of the world.After considering the errors from the process of heat flow measurement,the BSR depth,the relationship of thermal conductivity with the sediments depth,and the fluid flow activities,we conclude that the difference should be not caused by these errors.Such big disagreement may be due to the misunderstanding of BSR.The deviant 'BSRs' could represent the paleo-BSRs or just gas-bearing sediment layers,such as unconformities or the specific strata where have different permeability,which are not hydraterelated BSRs.ZHANG Yi HE Lijuan WANG Jiyang XU Xing SHA Zhibing GONG Yuehua WANG Hongbing LIANG Jinqiang 2011Acta Oceanologica Sinica2011,30,1:2
15Regional Metabolic Patterns of Abnormal Postoperative Behavioral Performance in Aged Mice Assessed by XH-NMR Dynamic Mapping Method显示文摘Abnormal postoperative neurobehavioral performance(APNP)is a common phenomenon in the early postoperative period.The disturbed homeostatic status of metabolites in the brain after anesthesia and surgery might make a significant contribution to APNP.The dynamic changes of metabolites in different brain regions after anesthesia and surgery,as well as their potential association with APNP are still not well understood.Here,we used a battery of behavioral tests to assess the effects of laparotomy under isoflurane anesthesia in aged mice,and investigated the metabolites in 12 different sub-regions of the brain at different time points using proton nuclear magnetic resonance('H-NMR)spectroscopy.The abnormal neurobehavioral performance occurred at 6 h and/or 9 h,and recovered at 24 h after anesthesia/surgery.Compared with the control group,the altered metabolite of the model group at 6 h was aspartate(Asp),and the difference was mainly displayed in the cortex;while significant changes at 9 h occurred predominantly in the cortex and hippocampus,and the corresponding metabolites were Asp and glutamate(Glu).All changes returned to baseline at 24 h.The altered metabolic changes could have occurred as a result of the acute APNP,and the metabolites Asp and Glu in the cortex and hippocampus could provide preliminary evidence for understanding the APNP process.Taotao Liu Zhengqian Li Jindan He Ning Yang Dengyang Han Yue Li Xuebi Tian Huili Liu Anne Manyande Hongbing Xiang Fuqiang Xu Jie Wang Xiangyang Guo 2020Neuroscience Bulletin2020,36,1:2
16Recombinant soluble TRAIL induces apoptosis of cancer cells显示文摘TRAIL is a tumor necrosis factor family member that selectively induces apoptosis of cancer cells but not of normal cells. To develop TRAIL into a potential cancer drug, three different sizes of soluble TRAIL fragments, including sTRAIL(74 - 281), sTRAIL(95 - 281) and sTRAIL(101-281), were expressed in E. coli and purified to homogeneity. Apoptosis assays indicated that sTRAIL(95-281) and sTRAIL(101-281), but not sTRAIL(74-281), can potently induce apoptosis of various cancer cell lines in 6 h, suggesting that the N-terminal fragment of aa101 has inhibitory effect on TRAIL-induced apoptosis. 17Computer Simulation of Thermal-Mechanical Processing显示文摘Supported by the high-speed SGI engineering workstation and the general MARCTM software, a system is developed to simulate the hot deformation process and the cooling process after deformation in the forging of Cr12 steel on the basis of elastic-plastic FEM techniques with coupled thermal-mechanical-microstructural alteration. In this system, a mathematical model, obtained by plentiful physical simulation experiments on Gleeble 1500 thermomechanical simulator, is introduced to describe the changes in microstructure and properties of steels. This system is proved to be reliable to simulate the thermal field and stress field as well as the microstructure of Cr12 steel during the forging process. 19Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
