1Personalized neoantigen pulsed dendritic cell vaccine for advanced lung cancer显示文摘Neoantigens are considered to be ultimate target of tumor immunotherapy due to their high tumor specificity and immunogenicity.Dendritic cell(DCs)vaccines based on neoantigens have exciting effects in treatment of some malignant tumors and are a promising therapeutic modality.Lung cancer is a lethal disease with the highest morbidity and mortality rate in the world.Despite the rapid development of targeted therapy and immune checkpoint inhibitors for lung cancer in recent years,their efficacy is still unsatisfactory overall.Therefore,there is an urgent unmet clinical need for lung cancer treatment.Here,we attempted to treat lung cancer using a personalized neoantigen peptide-pulsed autologous DC vaccine and conducted a single-arm,2 medical centers,pilot study initiated by the investigator(ChiCTR-ONC 16009100,NCT02956551).The patients enrolled were patients with heavily treated metastatic lung cancer.Candidate neoantigens were derived from whole-exome sequencing and RNA sequencing of fresh biopsy tissues as well as bioinformatics analysis.A total of 12 patients were enrolled in this study.A total of 85 vaccine treatments were administered with a median value of 5 doses/person(range:3-14 doses/person).In total,12-30 peptide-based neoantigens were selected for each patient.AIl treatment-related adverse events were grade 1-2 and there were no delays in dosing due to toxic effects.The objective effectiveness rate was 25%;the disease control rate was 75%;the median progression-free survival was 5.5 months and the median overall survival was 7.9 months.This study provides new evidence for neoantigen vaccine therapy and new therapeutic opportunities for lung cancer treatment.Zhenyu Ding Qing Li Rui Zhang Li Xie Yang shu Song Gao Peipei Wang Xiaoqing Su Yun Qin Yuelan Wang Juemin Fang Zhongzheng Zhu Xuyang Xia Guochao Wei Hui Wang Hong Qian Xianling Guo Zhibo Gao Yu Wang Yuquan Wei Qing Xu Heng Xu Li Yang 2021Signal Transduction and Targeted Therapy2021,6,2:3
2Mechanical properties of material in a mine dump at the Shengli#1 Surface Coal Mine,China显示文摘In-situ experiments were conducted to investigate the mechanical properties of the soil-rock mixture in the internal dump of the Shengli #1 Surface Coal Mine, China. Based on the experimental results, this study used comparative analysis and found that the shear strength of the soil-rock mixture in the dump was greater than the residual shear strength of the original rock. The results showed that the material presented in the dump as large blocks was the main factor affecting the strength of the soil-rock mixture.Numerical simulation was carried out for the analyses of three factors: different combinations of shear failure, rolling failure along with different large-block radius ratios, and mixture densities. The results illustrated that the cohesion and angle of internal friction of the soil-rock mixture are 12 kPa and 32.26°. However, in some cases the bench angle in the dump was controlled by a coupling relationship of rocks in the material. 3Creep-Induced Phase Instability and Microstructure Evolution of a Nearly Lamellar Ti–45Al–8.5Nb–(W, B, Y) Alloy显示文摘Microstructure degradation and stress-induced transformation of a high Nb-containing TiAl alloy with nearly lamellar microstructure during creep were investigated.Tensile creep experiments were performed at 800,850 and 900℃ under 150 MPa in air.Microstructures before and after creep tests were examined using scanning and transmission electron microscopy(SEM and TEM).Dislocations within the lamellar structure andβo(ω)region and twin intersection in massiveγgrains were investigated.Dislocation sliding played a critical role in the deformation ofωo phase,which preferentially occurred on the(0002)ωo plane.Possible deformation mechanisms were revealed.A stress-inducedγ→α2 phase transformation took place during the creep test at 850 and 900℃.α2 lamella could directly decompose into theωo phase at 850℃.The instability of high-temperature microstructure can weaken the creep resistance and promote the plastic deformation of the lamellar matrix,thus could be detrimental to the creep properties.The correlations between creep properties and microstructure instability were discussed.Xuyang Wang Jieren Yang Rui Hu Zitong Gao Jinguang Li Hengzhi Fu 2020Acta Metallurgica Sinica(English Letters)2020,33,12:3

5Investigation of PM_(2.5) pollution during COVID-19 pandemic in Guangzhou,China显示文摘The COVID-19 pandemic has raised awareness about various environmental issues,includ-ing PM_(2.5) pollution.Here,PM_(2.5) pollution during the COVID-19 lockdown was traced and an-alyzed to clarify the sources and factors influencing PM_(2.5) in Guangzhou,with an emphasis on heavy pollution.The lockdown led to large reductions in industrial and traffic emissions,which significantly reduced PM_(2.5) concentrations in Guangzhou.Interestingly,the trend of PM_(2.5) concentrations was not consistent with traffic and industrial emissions,as minimum concentrations were observed in the fourth period(3/01-3/31,22.45 μg/m^(3))of the lockdown.However,the concentrations of other gaseous pollutants,e.g.,SO_(2),NO_(2) and CO,were corre-lated with industrial and traffic emissions,and the lowest values were noticed in the sec-ond period(1/24-2/0_(3))of the lockdown.Meteorological correlation analysis revealed that the decreased PM_(2.5) concentrations during COVID-19 can be mainly attributed to decreased in-dustrial and traffic emissions rather than meteorological conditions.When meteorological factors were included in the PM_(2.5) composition and backward trajectory analyses,we found that long-distance transportation and secondary pollution offset the reduction of primary emissions in the second and third stages of the pandemic.Notably,industrial PM_(2.5) emis-sions from western,southern and southeastern Guangzhou play an important role in the formation of heavy pollution events.Our results not only verify the importance of control-ling traffic and industrial emissions,but also provide targets for further improvements in PM_(2.5) pollution.Luyao Wen Chun Yang Xiaoliang Liao Yanhao Zhang Xuyang Chai Wenjun Gao Shulin Guo Yinglei Bi Suk-Ying Tsang Zhi-Feng Chen Zenghua Qi Zongwei Cai 2022Journal of Environmental Sciences2022,34,5:2
6Peering into the Milky Way by FAST:Ⅳ.Identification of two new Galactic supernova remnants G203.1+6.6and G206.7+5.9显示文摘A 5°×7°sky area containing two large radio structures of G203.1+6.6 and G206.7+5.9 with a size of about 2.°5 and 3.°5 respectively is scanned by using the L-band 19-beam receiver of the Five-hundred-meter Aperture Spherical radio Telescope(FAST).The FAST L-band receiver covers a frequency range of 1.0-1.5 GHz.Commissioning of the receiving system,including the measurements of the half-power beam width,gain,and main-beam efficiency is made by observing the calibrators.The multi-channel spectroscopy backend mounted to the receiver allows an in-band spectral-index determination.The brightness-temperature spectral indices of both objects are measured to beβ~-2.6 to-2.7.Polarized emission is detected from the archival Effelsbergλ11 cm data for all the shell structures of G203.1+6.6 and G206.7+5.9.These results clearly indicate a non-thermal synchrotron emitting nature,confirming that G203.1+6.6 and G206.7+5.9 are large shell-type supernova remnants(SNRs).Based on morphological correlation between the radio continuum emission of G206.7+5.9 and the H i structures,the kinematic distance to this new SNR is estimated to be about 440 pc,placing it in the Local Arm.XuYang Gao Wolfgang Reich XiaoHui Sun He Zhao Tao Hong ZhongSheng Yuan Patricia Reich JinLin Han 2022Science China(Physics,Mechanics & Astronomy)2022,65,12:1
7An Effective Method of Prompting Juvenile Rainbow Trout (Oncorhynchus mykiss) to Cope with Heat Stress显示文摘In the present study,juvenile rainbow trout(Oncorhynchus mykiss)were acclimated at 5 temperatures.Fish in the control group(C0)were reared at a constant temperature(16℃);trout in four other treatments(A0,A3,A6,and A9)were acclimated to a high temperature(22℃)for 0,3,6,and 9 d,respectively,and then returned to normal temperature(16℃)for 7d.The temperature was then raised to 20℃and the fish were cultured for 40 d.The results showed that the growth rates of A3,A6,and A9 were higher than that of A0 but lower than that of C0.The growth rate of A9 was the highest among the 4 acclimation groups.The activities of serum transaminase and liver antioxidant enzyme significantly increased during acclimation.At the beginning of growth,the activities of enzymes were lower in A3,A6,and A9 with the lowest in A9.In addition,the transcript abundance of heat shock protein(HSP)60 gene in A9 was not significantly different from that of C0 during growth.HSP70 in A9 significantly increased at the beginning and returned to that of C0 at the end.Our findings indicated that pre-acclimation improved the high temperature tolerance with the best effectiveness observed at 22℃for 9 d.A possible mechanism underlining such phenomenon is the improvement of antioxidant defense system.JIANG Xuyang DONG Shuanglin ZHOU Yangen GAO Qinfeng PAN Zhe 2020Journal of Ocean University of China2020,19,1:1
8Peering into the Milky Way by FAST:Ⅱ.Ionized gas in the inner Galactic disk revealed by the piggyback line observations of the FAST GPPS survey显示文摘As one of the major components of the interstellar medium,the ionized gas in our Milky Way,especially the low-density diffuse component,has not been extensively observed in the radio band.The Galactic Plane Pulsar Snapshot(GPPS)survey covers the sky area within the Galactic latitude of±10°around the Galactic plane visible by the Five-hundred-meter Aperture Spherical radio Telescope(FAST),and the spectral line data are simultaneously recorded during the pulsar survey observations.With an integration time of 5 min for each beam,the GPPS survey project provides the most sensitive piggyback spectra for tens of radio recombination lines(RRLs)in the band of 1000-1500 MHz for Hnα,Henα,Cnα,as well as Hnβand Hnγ.We processed the spectral data of RRLs,and obtained a sensitive averaged HnαRRL map of a sky area of 88 square degrees in the inner Galaxy of 33°≤l≤55°and∣b∣≤2°0.The final spectral data of the HnαRRLs have a spatial resolution of∼3′,a spectral resolution of 2.2 km s^(−1),and a typical spectral RMS noise of 0.25 mJy beam−1 or 6.3 mK in main-beam brightness temperature.The FAST GPPS HnαRRL observations are sensitive down to an emission measure of about 200 cm^(−6) pc if a 3σdetection limit is required.The new HnαRRL map shows complex structural features dominated by a number of HII regions and large extended diffuse ionized gas regions.We detect about 94%of the known HII regions and confirm 43 WISE HII regions in the observed sky area.Several large HII regions or star-forming complexes in the distant outer Galaxy are resolved in the map of HnαRRLs.Extended RRL features of the diffuse ionized gas are detected.In addition,the GPPS piggyback spectral-line data also provide sensitive detection for other kinds of RRLs,such as the Henα,Cnα,Hnβ,and HnγRRLs.The RRL data products of the GPPS survey will be published and updated at http://gffzz656c80eee53645cchx5b9bxx0unxn6qf5.ffgz.tsg.suse.edu.cn/MilkyWayFAST/.LiGang Hou JinLin Han Tao Hong XuYang Gao Chen Wang 2022Science China(Physics,Mechanics & Astronomy)2022,65,12:0
9Peering into the Milky Way by FAST:Ⅰ.Exquisite HI structures in the inner Galactic disk from the piggyback line observations of the FAST GPPS survey显示文摘Neutral hydrogen(Hi)is the fundamental component of the interstellar medium.The Galactic Plane Pulsar Snapshot(GPPS)survey is designed for hunting pulsars by using the Five-hundred-meter Aperture Spherical radio Telescope(FAST)from the visible Galactic plane within|b|≤10°.The survey observations are conducted with the L-band 19-beam receivers in the frequency range of 1.0-1.5 GHz,and each pointing has an integration time of 5 min.The piggyback spectral data simultaneously recorded during the FAST GPPS survey are great resources for studies on the Galactic Hi distribution and ionized gas.We process the piggyback Hi data of the FAST GPPS survey in the region of 33°≤l≤55°and|b|≤2°.The rms of the data cube is found to be approximately 40 m K at a velocity resolution of 0.1 km s^(-1),placing it the most sensitive observations of the Galactic Hi by far.The high velocity resolution and high sensitivity of the FAST GPPS Hi data enable us to detect weak exquisite Hi structures in the interstellar medium.Hi absorption line with great details can be obtained against bright continuum sources.The FAST GPPS survey piggyback Hi data cube will be released and updated on the web:http://gffzz656c80eee53645cchx5b9bxx0unxn6qf5.ffgz.tsg.suse.edu.cn/Milky Way FAST/.Tao Hong JinLin Han LiGang Hou XuYang Gao Chen Wang Tao Wang 2022Science China(Physics,Mechanics & Astronomy)2022,65,12:0

11Digestion performance improvement of tributyrin through nano-emulsion preparation technology显示文摘The current research aimed to optimize the preparation technology of tributyrin nano-emulsion(TBNE)to improve its digestion properties, which were investigated through digestion models in vitro and in vivo. The content of each component of TBNE was optimized by response surface methodology(RSM) to improve the z-average particle size and stability of TBNE. The optimized TBNE was evaluated for its digestion properties by in vitro and in vivo models. The preparation conditions of TBNE optimized by RSM were as follows: 61.3% of sorbitol, 32.7% of tributyrin, and 6.0% of modified phospholipid(MP). The predicted z-average particle size of TBNE was(246.02 ± 18.10) nm. The results of the verification test showed that the z-average particle size, zeta potential, conductivity, emulsification activity index, and emulsification stability index of TBNE were(250.02 ± 7.18) nm,(-40.23 ± 0.76) m V,(31.80 ± 2.09) μS/cm,(848.00 ± 84.53) min and(1.14 ± 0.02) m^(2)/g, respectively. The in vitro digestion experiment results showed that the TBNE remained stable in the stomach and was released in the intestine, while the size of TBNE in the gastrointestinal tact was significantly smaller than that of tributyrin(P < 0.05), which made it easier to be digested and absorbed. Compared with tributyrin treatment, TBNE significantly promoted the average body weight at the 7th day, average daily feed intake,average daily gain, feed/gain, ileum weight, and organ index of ileum of Hy-Line BROWN chicks(P < 0.05), and the butyric acid content in the ileal chyme from TBNE and tributyrin treatment were172.18 mg/m L and 100.85 mg/m L(P < 0.01). 12Intermetallic ferric nickel silicide alloy derived from magadiite by magnesiothermic reaction as bifunctional electrocatalyst for overall water splitting显示文摘As the cleanest energy source,hydrogen energy is regarded as the most promising fuel.Water electrolysis,as the primary means of hydrogen production,has constantly been the focus of attention in the energy conversion field.Developing eco-friendly,cheap,safe and efficient catalysts for electrochemical water splitting(EWS)is the key challenge.Herein,the intermetallic silicide alloy is first synthesized via a facile magnesiothermic reduction and employed as bifunctional electrocatalysts for EWS.Ferric-nickel silicide(denoted as FeNiSi)alloy is designed and shows a good electrocatalytic performance for EWS.The lattice distortions of FeNiSi enhance the electrocatalytic activity.Besides,the porous structure affords more active sites and improves the reaction kinetics.As a consequence,FeNiSi delivers an excellent performance with overpotential of 308 mV for oxygen evolution reaction(OER)and 386 mV for hydrogen evolution reaction(HER)at 10 mA·cm−2 in 1 M KOH.The stability structure of intermetallic silicide achieves an outstanding durability with an unchanged potential of 1.66 V for overall water splitting at 10 mA·cm−2 for 15 h.This work not only provides a facile method for the synthesis of intermetallic silicide with considerable porous structures,but also develops the potential of intermetallic silicide alloy as bifunctional electrocatalysts for EWS,which opens up a new avenue for the design and application of intermetallic silicide alloy.Xuyang Jing Yang Mu Zhanming Gao Xueying Dong Changgong Meng Chi Huang Yifu Zhang 2024Nano Research Energy2024,3,2:0
13Novel thieno[2,3-b]quinoline-procaine hybrid molecules:A new class of allosteric SHP-1 activators evolved from PTP1B inhibitors显示文摘Small molecule activators could equally provide powerful tools as inhibitors do for interrogating cellular signal transduction.However,targeted protein activation is chemically challenging.Developing activators against Src homology region 2 domain-containing phosphatase-1(SHP-1)to block STAT3 pathway represents a promising strategy for DLBCL therapy.Here we reported a new class of thieno[2,3-b]quinolineprocaine hybrid molecules as SHP-1 allosteric activators.The representative hybrid compound 3b displayed SHP-1 activating effect with EC50 of 5.48±0.28μmol/L.Further investigations confirmed that 3b allosterically interacted with SHP-1,switched it from close to open conformation,blocked SHP-1/p-STAT3 pathway,induced apoptosis and inhibited ABC-DLBCL cell proliferation in vitro,and delayed tumor growth in the xenograft model of SU-DHL-2.Overall,this work offered a novel paradigm to develop SHP-1 allosteric activators through chemical space evolution of PTPs inhibitors,and firstly validated the therapeutic strategy that directly activating SHP-1 alone could be a potential therapy against ABC-DLBCL via blocking STAT3 pathway.Lei Xu Xuyang Mu Minmin Liu Zhijia Wang Chao Shen Qianwen Mu Bo Feng Yechun Xu Tingjun Hou Lixin Gao Haini Jiang Jia Li Yubo Zhou Wenlong Wang 2023Chinese Chemical Letters2023,34,8:0
14A steady composite molecular anode Ru1/MWCNTsCOOH/GC for robust catalytic water oxidation显示文摘Development of efficient molecular devices for overall water splitting has always been a hotspot of research to realize clean and sustainable energy conversion. In the case, long-term durability of molecular devices is a crucial factor to determine their practical application. Here, an efficient composite molecular anode was assembled by immobilization of charge-neutral mononuclear Ru1 complex on COOHfunctionalized multiwalled carbon nanotubes(MWCNTs COOH) sticking on a glassy carbon(GC) electrode(Ru1/MWCNTsCOOH/GC). The prepared hybrid anode showed a low onset overpotential of only 380 mV for electrocatalytic water oxidation. Moreover, the anode displayed a steady catalytic current density of1.25 mA/cm^2 for more than 5 h at the overpotential of 580 m V, a high TON of 186000(5 h), a TOF of10.3 s-1 and a faradic efficiency of 96%, indicating the significant efficiency and extremely durability.The excellent durability of the molecular anode should be attributed to the key charge-neutral catalytic intermediates(Ru1 ad) of the catalytic step and the two long flexible carbon chains of the catalyst.Yan Gao Yu Wei Zhongkai Lu Xuyang Chen Dongping Wang 2019Journal of Energy Chemistry2019,28,8:0
15DropLab:an automated magnetic digital microfluidic platform for sample-to-answer point-of-care testing—development and application to quantitative immunodiagnostics显示文摘In point-of-care testing(POCT),tests are performed near patients and results are given rapidly for timely clinical decisions.Immunodiagnostic assays are one of the most important analyses for detecting and quantifying protein-based biomarkers.However,existing POCT immunodiagnostics mainly rely on the lateral flow assay(LFA),which has limited sensitivity or quantification capability.Although other immunodiagnostic assays,such as enzyme-linked immunosorbent assays(ELISAs),offer more sensitive and quantitative results,they require complex liquid manipulations that are difficult to implement in POCT settings by conventional means.Here,we show the development of DropLab,an automated sample-in-answer-out POCT immunodiagnostic platform based on magnetic digital microfluidic(MDM)technology.DropLab performs microbead-based ELISA in droplets to offer more sensitive and quantitative testing results.The intricate liquid manipulations required for ELISA are accomplished by controlling droplets with magnetic microbeads using MDM technology,which enables us to achieve full automation and easy operations with DropLab.Four ELISAs(the sample in triplicates and a negative control)can be run in parallel on the thermoformed disposable chip,which greatly improves the throughput and accuracy compared to those of other POCT immunodiagnostic devices.DropLab was validated by measuring two protein targets and one antibody target.The testing results showed that the limit of detection(LOD)of DropLab matched that of the conventional ELISA in a microwell plate.DropLab brings MDM one step closer to being a viable medical technology that is ready for real-world POCT applications.Xuyang Hu Xiangyu Gao Songlin Chen Jinhong Guo Yi Zhang 2023Microsystems & Nanoengineering2023,9,1:0
16Natural anti-oxLDL IgM monoclonal antibody in the pathogenesis of atherosclerosis显示文摘Objective To explore the role and the possible molecular mechanisms of natural anti-oxLDL IgM monoclonal antibody played and involved in pathogenesis of atherosclerosis. Methods Natural anti-oxLDL IgM monoclonal antibody 3A6 was generated by using standard hybridoma production techniques. Influence of 3A6 on formation of foam cells was observed by Oil Red O staining and affinity of Na125I-conjugated oxLDL on the naive and LPS-activated macrophages. After LPS stimulation on macrophages, anti-TLR4 neutralizing mAb, p38MAPK specific inhibitor SB203580, NF-kB specific inhibitor PDTC or RNAi targeting Fcα/μ receptor (Fcamr) were applied, respectively. Results Natural anti-oxLDL IgM monoclonal antibody 3 A6 were found specifically inhibit the binding of CuoxLDL to naive macrophages but not the binding of CuoxLDL to LPS-activated macrophages. It also promoted the formation of CuoxLDL-mediated foam macrophages. 3A6 F(ab')2 or pre-incubation with un-related IgM inhibited the binding of 3A6/CuoxLDL complex to LPS-activated macrophages. LPS up-regulated the expression of Fcamr in macrophages in a dose- and time-dependent manner, which was attenuated by treatment with anti-TLR4. LPS induced the phosphorylation of p38MAPK and translocation of NF-kB p65, contributing to the up-regulated expression of Fcα/μ receptor in macrophages. Conclusions Natural anti-oxLDL IgM monoclonal antibody 3A6 specifically inhibited the binding of CuoxLDL to naive macrophages in vitro. However, LPS, through the Toll-like receptor (TLR)4 receptor, activated the p38MAPK and NF-kB pathways and up-regulated the expression of Fcα/μ receptor in macrophages, which promoted the binding of 3A6/CuoxLDL complex to macrophages through binding with Fc fragments and the formation of foam macrophages. 18Protonation effect on catalytic water oxidation activity of a mononuclear Ru catalyst containing a free pyridine unit显示文摘Two efficient single-site Ru water oxidation catalysts [Ru(bda)(pic)(Ln)](bda = 2,2'-bipyridine-6,6'-dicarboxylic acid, pic = picoline, L1 = 4,5-bipyridine-2,7-di-tert-butyl-9,9-dimethylxanthene, L2 = 4-pyridine-5-phenyl-2,7-di-tert-butyl-9,9-dimethylxanthene) were only synthesized containing different xanthene ligands at the axial site. These complexes have been thoroughly characterized by spectroscopic(UV-vis, NMR) and electrochemical(CV and DPV) techniques.
