Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. These findings not only provide empirical evidence of OsNAC041 function, but also provide new insight into its potential application in rice resistance breeding. | WANG Bo ZHONG Zhaohui ZHANG Huanhuan WANG Xia LIU Binglin YANG Lijia HAN Xiangyan YU Deshui ZHENG Xuelian WANG Chunguo SONG Wenqin CHEN Chengbin ZHANG Yong | 2019 | Rice science2019,26,2: | 8
AN IMPROVED SPARSITY ADAPTIVE MATCHING PURSUIT ALGORITHM FOR COMPRESSIVE SENSING BASED ON REGULARIZED BACKTRACKING显示文摘Sparsity Adaptive Matching Pursuit (SAMP) algorithm is a widely used reconstruction algorithm for compressive sensing in the case that the sparsity is unknown. In order to match the sparsity more accurately, we presented an improved SAMP algorithm based on Regularized Backtracking (SAMP-RB). By adapting a regularized backtracking step to SAMP algorithm in each iteration stage, the proposed algorithm can flexibly remove the inappropriate atoms. The experimental results show that SAMP-RB reconstruction algorithm greatly improves SAMP algorithm both in reconstruction quality and computational time. It has better reconstruction efficiency than most of the available matching pursuit algorithms. | Zhao Ruizhen Ren Xiaoxin Han Xuelian Hu Shaohai | 2012 | Journal of Electronics(China)2012,29,6: | 3
Upwelling of Mantle-derived Material in Southeast China:Evidence from Noble Gas Isotopes显示文摘Shallow groundwater collected in Chaozhou,Huizhou, and Guangzhou allowed testing of concentrations and the isotope ratios of noble gases.Based on the calculated noble gas temperature(NGT)and the ratio of noble gas isotopes, the recharge temperature, recharge source, and residence time of groundwater can be calculated. In addition, the contribution of noble gas components from different sources to the sample components can be assessed. In the Huizhou area, according to the 1/Xe vs.Ne/Xe and NGT data, the shallow sandstone-confined water samples in the Shiba area and the unconfined water samples of the Huangshadong are in different temperature ranges, indicating that they have different recharge sources, both in time or space. The He components in the samples are calculated to obtain the content of radiogenic ^(4)He in the crust and to simulate the groundwater ages. The noble gas isotope ratios show the addition of mantle components into the basalt aquifers and sandstone aquifers in Chaozhou and Huizhou.Except for atmospheric and crustal sources, there is a certain proportion of mantle-derived components in the shallow underground cold water in Huizhou and Chaozhou. The noble gases in the Chaozhou groundwater have an obvious mantle signature, allowing speculation that there is a deep fluid carrying mantle characteristics. This upwelling of mantle-derived material might be caused by the India-Eurasia collision or that between the Philippine Sea Plate and the Eurasian Plate. | WANG Shuai KUANG Jian HUANG Xuelian ZHANG Hongyan ZHANG Min QI Shihua HAN Yongjie XIAO Zhicai WANG Siqi TANG Ling | 2022 | Acta Geologica Sinica(English Edition)2022,96,1: | 2

SeDeM Expert System: A review and new perspectives显示文摘The SeDeM Expert System was first known as a galenic pre-formulation system, which was based on the experimental research and quantitative determination of powdered substances. And the mathematical formula provided by the SeDeM Expert System has plays an important role in the study of powder properties. The system can be used not only to evaluate the powder direct compression (DC) of excipients and active pharmaceutical ingredients (API’s), but also to predict the possible formulations, so it can reduce unnecessary research and trials, and shorten the time of development. In this paper, the research development and application of SeDeM Expert System in DC was summarized, and the results showed that with a few exceptions, the system was skilled in predicting acceptable tablet formulations. Finally, the new application prospect of the system is presented, including the application of the Internet traffic and content management (iTCM) database and the new co-processed excipients. | Mingxian Gu Tianbing Guan Shulin Wan Han Zhang Xuelian Li Songtao Kong Jianbing Ren Huimin Sun Chuanyun Dai | 2019 | Journal of Pharmaceutical and Biopharmaceutical Research2019,1,1: | 1
A positive feedback circuit for ROP-mediated polar growth显示文摘Tip growth is a special type of polarized growth in which a single and unique polarization site is established and maintained.Rho of Plants(ROP)proteins, which represent the only class of Rho GTPases in plants, regulate tip growth. The dynamic and asymmetric distribution of ROPs is critical for the establishment and maintenance of tip growth, and requires at least one positive feedback loop, which is still elusive. Here,we report a positive feedback circuit essential for tip growth of root hairs,in which ROPs,ROP activators and effectors, and AGC1.5 subfamily kinases are interconnected by sequential oligomerization and phosphorylation.AGC1.5 subfamily kinases interact with and phosphorylate two guanine nucleotide exchange factors(GEFs)of ROPs,RopGEF4 and RopGEF10.They also interact with two ROP effectors,ICR2/RIP3 and MIDD1/RIP4,which bridge active ROPs with AGC1.5.Functional loss of the AGC1.5 subfamily kinases or ICR2 and MIDD1 compromised root hair growth due to reduced ROP signaling. We found that asymmetric targeting of RopGEF4 and RopGEF10 is controlled by AGC1.5-dependent phosphorylation.Interestingly,we discovered that the ROP effectors recruit AGC1.5 to active ROP domains at the plasma membrane during root hair growth and are critical for AGC1.5-dependent phosphorylation of RopGEFs.Given the large number of AGC kinases in plants, this positive feedback circuit may be a universal theme for plant cell polar growth. | En Li Yu-Ling Zhang Xuelian Shi Han Li Xuefeng Yuan Sha Li Yan Zhang | 2021 | Molecular Plant2021,14,3: | 1
Research status and prospect of wind-vehicle-bridge coupling vibration system显示文摘To promote and develop the theoretical basis and application of the wind-vehicle-bridge coupling vibration system, the corresponding research status and prospects are reviewed and discussed from five aspects,i.e.,the analytical framework, the aerodynamic interference, the evaluation criteria, the design loads of long-span bridge and the double-deck railcum-road bridge. The refining process of analysis system is reviewed from the aspects of simulation wind load, vehicle load and bridge structure, and the corresponding coupling relationship. For aerodynamic interference, the development process is summarized from the simulative precision of the elements(wind, vehicle and bridge),the load cases and the object of interference. For evaluation criteria, the corresponding development course is summarized from the certain evaluation method to uncertain one.For long-span bridge design load, the wind and vehicle loads are reviewed and summarized from current multinational codes and theoretical research. For double-deck rail-cum-road bridge, the mechanism of multi-element coupling relationship and corresponding aerodynamic interference are both reviewed.By comprehensive review and summary, the analytical framework is in the process from simplification to refinement. The simulation and consideration of the objects of structural interference gradually become complex. The corresponding simulation theory, wind tunnel scale, test equipment and technology are the key factors to limit its development. For systematic evaluation of vehicle and bridge, the structural and systemic security are the basis of the evaluation, and the auxiliary components and functional evaluation need to be paid more attention. The evaluation criterion will be developed from certain method to reliability assessment. For design load of long-span bridge, the vehicle load is gradually transferred from the simple application of the design load of small-medium span bridge into a complex model considering the load characteristics. For double-deck rail-cum-road bridge, the basic theory and experimental study on coupling mechanism and aerodynamic interference need to be developed. | Wanshui Han Xiaodong Liu Xuelian Guo Shizhi Chen Gan Yang Huanju Liu | 2022 | Journal of Traffic and Transportation Engineering(English Edition)2022,9,3: | 1
Hypercompact CRISPR–Cas12j2 (CasF) enables genome editing, gene activation, and epigenome editing in plants显示文摘CRISPR-Cas9,-Cas12a,-Cas12b,and-Cas13 have been harnessed for genome engineering in human and plant cells(Liu et al.,2022).However,the large size of these Cas proteins(e.g.190 kDa for SpCas9)makes them difficult to deliver into cells via a viral vector. The development of smaller Cas proteins will lead to reduced viral vector sizes that can be more widely adopted in versatile genome engineering systems.Recently,a CRISPR-Cas12j2(CasF)system was discovered in huge phages and developed into a hypercompact genome editor due to the small size of Cas12j2(80 kDa)(Pausch et al.,2020).Unfortunately,the gene editing efficiency of Cas12j2 in Arabidopsis protoplasts using ribonucleoprotein delivery was less than one percent(Pausch et al.,2020).Further optimization of this system is clearly required if CRISPR-Cas12j2-mediated editing in plant genomes is to be adopted by the plant sciences community. | Shishi Liu Simon Sretenovic Tingting Fan Yanhao Cheng Gen Li Aileen Qi Xu Tang Yang Xu Weijun Guo Zhaohui Zhong Yao He Yanling Liang Qinqin Han Xuelian Zheng Xiaofeng Gu Yiping Qi Yong Zhang | 2022 | Plant Communications2022,3,6: | 0
Expert consensus on prevention and cardiopulmonary resuscitation for cardiac arrest in COVID-19显示文摘Background:Cardiopulmonary resuscitation(CPR)strategies in COVID-19 patients differ from those in patients suffering from cardiogenic cardiac arrest.During CPR,both healthcare and non-healthcare workers who provide resuscitation are at risk of infection. The Working Group for Expert Consensus on Prevention and Cardiopulmonary Resuscitation for Cardiac Arrest in COVID-19 has developed this Chinese Expert Consensus to guide clinical practice of CPR in COVID-19 patients.Main recommendations:1)A medical team should be assigned to evaluate severe and critical COVID-19 for early monitoring of cardiac-arrest warning signs.2)Psychological counseling and treatment are highly recommended, since sympathetic and vagal abnormalities induced by psychological stress from the COVID-19 pandemic can induce cardiac arrest.3)Healthcare workers should wear personal protective equipment(PPE).4)Mouth-to-mouth ventilation should be avoided on patients suspected of having or diagnosed with COVID-19.5)Hands-only chest compression and mechanical chest compression are recommended.6)Tracheal-intubation procedures should be optimized and tracheal-intubation strategies should be implemented early.7)CPR should be provided for 20-30 min.8)Various factors should be taken into consideration such as the interests of patients and family members, ethics, transmission risks, and laws and regulations governing infectious disease control.Changes in management:The following changes or modifications to CPR strategy in COVID-19 patients are proposed:1)Healthcare workers should wear PPE.2)Hands-only chest compression and mechanical chest compression can be implemented to reduce or avoid the spread of viruses by aerosols.3)Both the benefits to patients and the risk of infection should be considered.4)Hhealthcare workers should be fully aware of and trained in CPR strategies and procedures specifically for patients with COVID-19. | Wei Song Jie Wei Xiangdong Jian Deren Wang Yanhong Ouyang Yuanshui Liu Xianjin Du Ying Chen Yingqi Zhang Heping Xu Shuming Xianyu Qiong Ning Xiang Li Xiaotong Han Feng Zhan Tao Yu Wenteng Chen Jun Zhang Wenwei Cai Sheng’ang Zhou Shengyang Yi Yu Cao Xiaobei Chen Shunjiang Xu Zong’an Liang Duohu Wu Fen Ai Zhong Wang Qingyi Meng Yuhong Mi Sisen Zhang Rongjia Yang Shouchun Yan Wenbin Han Yong Lin Chuanyun Qian Wenwu Zhang Yan Xiong Jun Lv Baochi Liu Yan Cao Xiaojun He Xuelian Sun Yufang Cao Tian’en Zhou | 2021 | Asian Pacific Journal of Tropical Medicine2021,14,6: | 0
Molecular genetic analysis of Dongzhou-period ancient human of Helingeer in Inner Mongolia,China显示文摘The mtDNA hypervariable region I(HVR-I)of 10 ancient individuals from Dongzhou-period ancient human populations in Helingeer county of Inner Mongolia were amplified and sequenced to investigate the genetic structure. The relationships between the ancient population and related extant populations,as well as its possible origin at the molecular level, were also studied. Moreover, phylogenetic analysis and multi-dimensional scaling analysis were also performed based on the mtDNA data of the ancient population in Helingeer and the related Eurasian population. The results showed that the ancient population in Helingeer were closer to the northern Asian populations than to the other compared populations in matrilineal lineage.Combining the research results of archaeology and anthropology as well as molecular biology,we inferred that they were nomads who migrated from Mongolia plateau and cis-Baikal region to Helingeer in Inner Mongolia,China. | Yuqin FU Chengzhi XIE Han ZHAO Xuelian XU Yinqiu CUI Quanchao ZHANG Hui ZHOU Hong ZHU | 2008 | Frontiers in Biology2008,3,1: | 0
Disease X Testing:The results of an international external quality assessment exercise显示文摘The United Nations Secretary-General Mechanism(UNSGM)for investigation of the alleged use of chemical and biological weapons is the only established international mechanism of this type under the UN.The UNGSM may launch an international investigation, relying on a roster of expert consultants, qualified experts, and analytical laboratories nominated by the member states.Under the framework of the UNSGM,we organized an external quality assurance exercise for nominated laboratories, named the Disease X Test,to improve the ability to discover and identify new pathogens that may cause possible epidemics and to determine their animal origin.The“what-if”scenario was to identify the etiological agent responsible for an outbreak that has tested negative for many known pathogens, including viruses and bacteria.Three microbes were added to the samples,Dabie bandavirus,Mammarenavirus, and Gemella spp.,of which the last two have not been taxonomically named or published. The animal samples were from Rattus norvegicus,Marmota himalayana,New Zealand white rabbit, and the tick Haemaphysalis longicornis.Of the 11 international laboratories that participated in this activity, six accurately identified pathogen X as a new Mammarenavirus, and five correctly identified the animal origin as R.norvegicus.These results showed that many laboratories under the UNSGM have the capacity and ability to identify a new virus during a possible international investigation of a suspected biological event. The technical details are discussed in this report. | Tian Qin Han Zheng Xuelian Luo Wen Zhang Jing Yang Yamin Sun Na Han Yuanhai You Liang Lu Xin Lu Di Xiao Shenshen Jiang Xuexin Hou Jinxing Lu Biao Kan Jianzhong Zhang Jianguo | 2022 | Journal of Biosafety and Biosecurity2022,4,2: | 0
Characteristics of Hydrogen and Oxygen Isotopes and Noble Gas Isotopes in the Groundwater of Weishan, Wudalianchi, Northeast China显示文摘According to the hydrochemical characteristics, hydrogen and oxygen isotope characteristics and the ratio of noble gas isotopes of the sandstone aquifer and basalt aquifer, this study calculated the recharge temperature and residence time of groundwater in the Weishan area of Wudalianchi, also calculating the contribution of noble gas components from different sources to the samples. Based on the characteristics of hydrogen and oxygen isotopes and noble gases Xe and Ne, the recharge altitude and recharge temperature of the two aquifers were estimated, and the recharge temperature fitting with the NGT model as verified, the results showing that the main recharge altitude of groundwater in the region was 500–600 m, the recharge temperature being 2–7°C. He_(eq) and He_(ea) of the samples have been simulated using the OD model, the content of radioactive ~4He in the crust being obtained, the groundwater ages under the two conditions(closed condition and open condition) both being simulated. The results show that groundwater from the sandstone layer water is older than groundwater from the basalt layer. Hydrochemical characteristics and noble gas isotope ratios indicate that in the basalt aquifer and sandstone aquifer in the Weishan area, in addition to atmospheric and crustal helium, there is also an input of mantle-derived helium. The fault constitutes the uplift channel for groundwater containings mantle components, which results in the mantle source composition in water samples near the fault being much higher than those form non-fault areas. | WANG Shuai HUANG Xuelian QI Shihua HAN Yongjie KUANG Jian WANG Siqi WANG He XIAO Zhicai | 2022 | Acta Geologica Sinica(English Edition)2022,96,5: | 0
The prospects for bioprinting tumor models:recent advances in their applications显示文摘Three-dimensional(3D)tumor models prepared from patient-derived cells have been reported to imitate some of the biological development processes of in situ tumors in vitro. These 3D tumor models share several important characteristics with their in vivo tumor counterparts.Accordingly, their applications in tumor modeling, drug screening, and precision-targeted treatment are promising. However, the establishment of tumormodels is subject to several challenges, including advancements in scale size, repeatability, structural precision in time and space, vascularization, and the tumor microenvironment.Recently, bioprinting technologies enabling the editorial arrangement of cells, factors, and materials have improved the simulation of tumormodels in vitro.Among the 3D bioprinted tumor models, the organoidmodel has been widely appreciated for its advantages of maintaining high heterogeneity and capacity for simulating the developmental process of tumor tissues. In this review,we outline approaches and potential prospects for tumor model bioprinting and discuss the existing bioprinting technologies and bioinks in tumor model construction. The multidisciplinary combination of tumor pathology, molecular biology, material science, and additive manufacturing will help overcome the barriers to tumor model construction by allowing consideration of the structural and functional characteristics of in vitro models and promoting the development of heterogeneous tumor precision therapies. | Changru Zhang Xiaoqin Qiu Yu Dai Weiqing Kong Yihao Liu Haoyi Niu Chengwei Wang Xuelian Mi Hui Wang Ya Ren Han Yang Kerong Dai Jinwu Wang | 2023 | Bio-Design and Manufacturing2023,6,6: | 0
