1Elf-1和Ki-67在非小细胞肺癌中的表达及其相关性研究(英文)显示文摘Objective: Elf-1 is a member of the proto-oncogenes Ets-related transcription factor family and over-expressed in many human tumors, Ki-67 is an important nuclear antigen associated with cell proliferation. This study investigated the expression of Elf-1 and Ki-67 in non-small-cell lung cancer(NSCLC) and studied their correlation with the clinicopathological features. Methods: Tissue microarray from 64 cases lung cancer tissue and 10 cases normal lung tissue was constructed, immunohistochemical method was used to evaluate the protein expressions of Elf-1 and Ki-67, correlations of the expression of Elf-1 and Ki-67 to clinicopathological features of NSCLC were analyzed. Results: Expression of Elf-1 and Ki-67 in NSCLC tissues were significantly higher than in normal lung tissues(P < 0.05), the positive rate of Elf-1 and Ki-67 was 73.44% and 64.06% in NSCLC group, Overexpression of Elf-1 in NSCLC was significantly related to histopathological grading, different clinical staging and the intensity of ELF-1 expression was significantly higher in the group with lymph node metastasis than that without(P < 0.05). Overexpression of Ki-67 was also closely related to tumor differentiation, clinical stages and lymph node metastasis(P < 0.05). In addition positive correlation was found between the expressive intensity of Elf-1 and Ki-67(τ = 0.295, P = 0.018). 2Targeting EZH1/2 induces cell cycle arrest and inhibits cell proliferation through reactivation of p57CDKN1C and TP53INP1 in mantle cell lymphoma显示文摘Objective: To explore the effect of dysregulation of epigenetic regulator EZH1 and EZH2 on the proliferation in MCL and the underlying mechanisms.Methods: In this study, we elucidated the role of EZH1 and EZH2 overexpression by immunohistochemistry and correlated them to clinical outcome in 41 MCL patients.Quantitative real-time PCR and Western blot were applied to confirm the level of EZH1 and EZH2 in well-characterized MCL cell lines which were compared to those of na?ve B cells.Then we manipulated the expression of EZH1 and EZH2 in MCL cells using CRISPR/Cas9 system to directly investigate their functional roles in MCL.We also evaluated the effect of two small molecule selective inhibitors, EPZ005687 and UNC1999, on MCL cell proliferation, cell cycle distribution and apoptosis in vitro.Finally, we performed RNA-sequencing(RNA-Seq) and Chromatin immunoprecipitation(ChIP) assay to further gain insight into the underlying molecular mechanisms.Results: We found that EZH2 protein is overexpressed in approximately half of this cohort of MCL cases.More importantly, the overexpression of EZH2 is associated with poor OS in the patients.Nevertheless, simple EZH2 depletion in vitro has little impact on the viability of MCL cells, predominantly because of the consequent up-regulation of EZH1.Consistently, UNC1999, a dual EZH1/2 inhibitor, unlike the EZH2 selective inhibitor EPZ005687, exerts a potent inhibitory effect on MCL cells.Furthermore, we discover CDKN1C and TP53 INP1 as the two important cell cycle regulators, the expression of which are repressed by EZH1/2 mediated epigenetic regulation and are restored by EZH1/2 dual inhibition.Conclusions: Our study suggests that EZH2 participates in the pathogenesis of MCL which may serve as a potential biomarker for prognosis prediction.The dual inhibition of EZH1/2 is a promising therapeutic strategy for MCL.Wei Li Chengfeng Bi Yating Han Tian Tian Xianhuo Wang Huijing Bao Xiaoying Xu Xuhan Zhang Lu Liu Weiwei Zhang Hai Gao Huaqing Wang Huilai Zhang Bin Meng Xi Wang Kai Fu 2019Cancer Biology & Medicine2019,16,3:2

4Ultracompact bandwidth-tunable filter based on subwavelength grating-assisted contra-directional couplers显示文摘An ultracompact,bandwidth-tunable filter has been demonstrated using a silicon-on-insulator(SOI)wafer.The device is based on cascaded grating-assisted contra-directional couplers(GACDCs).It also involves the use of a subwavelength grating(SWG)structure.By heating one of the heaters on GACDCs,a bandwidth tunability of~6 nm is achieved.Owing to the benefit of having a large coupling coefficient between SWG and strip waveguides,the length of the coupling region is only 100 pm.Moreover,the combination of the curved SWG and the tapered strip waveguides effectively suppresses the sidelobes.The filter possesses features of simultaneous wavelength tuning with no free spectral range(FSR)limitation.A maximum bandwidth of 10 nm was experimentally measured with a high out-of-band contrast of 25 dB.Similarly,the minimum bandwidth recorded is 4 nm with an out-of-band contrast of 15 dB.Kangnian WANG Yuan WANG Xuhan GUO Yong ZHANG An HE Yikai SU 2021Frontiers of Optoelectronics2021,14,3:1
5GSH-responsive camptothecin prodrug-based hybrid micellar nanoparticles enable antitumor chemo-immunotherapy by PD-L1 knockdown显示文摘The combinational chemo-immunotherapy as a novel treatment strategy has been widely studied and applied in clinic to enhance antitumor therapeutic efficacy and relieve side effects.RNA interference(RNAi)targeting PD-L1 via inhibiting novo production of PD-L1 will overcome the innate and adaptive PD-L1 expression during chemotherapy,thus enable sustained and efficient immune checkpoint blockade(ICB)to active antitumor immune response.Herein,we designed a glutathione(GSH)-responsive camptothecin(CPT)prodrug-based hybrid micellar nanoparticles(siPD-L1@HM-CPT)to achieve synergistic antitumor chemoimmunotherapy by PD-L1 knockdown.siPD-L1@HM-CPT derived from the one-step loading PD-L1 siRNA(siPD-L1)into the CPT prodrug-based hybrid micelles(HM-CPT)which were co-assembled from biodegradable polyphosphoesters-based prodrug CPT-ss-PAEEP15 and stabilizer DSPE-PEG,showed high loading efficiency,GSH-responsive drug release,and excellent stability and biosafety.siPD-L1@HM-CPT achieved simultaneously the co-delivery of CPT and siPD-L1 in vitro and in vivo,high accumulation at the tumor sites,and rapid intracellular release to promote antitumor efficacy via sensitizing CPT chemotherapy,inducing strong immunogenic cell death(ICD)and sustained ICB to improve intratumoral CD8+T cells infiltration.In addition,the antitumor immunity response limited by the differentiated immunogenicity,intrinsic PD-L1 expression,and intracellular GSH level was facilitated by efficient ICD and ICB from silencing PD-L1 and synergistic CPT chemosensitization in our experimental B16-F10 and 4T1 tumor models.Our study might offer a perspective on designing novel co-delivery nanoparticles by convenient and controllable preparation for antitumor chemo-immunotherapy.Xi Tan Hong Zhou Chenhui Wang Xuhan Liu Xiangliang Yang Wei Liu 2023Nano Research2023,16,1:0
6Comparative Study on Corrosion Behavior and Mechanism of As-Cast Mg-Zn-Y and Mg-Zn-Gd Alloys显示文摘Rare earth(RE)elements have large solid solubility in magnesium and are widely used to regulate the microstructure and property of advanced magnesium alloys.However,different kinds of RE elements have different effects on microstructure and property of the alloy.In this study,a Mg-Zn-Y alloy and a Mg-Zn-Gd alloy with alloying elements of the same atomic percentage were designed to clarify the effect of yttrium(Y)and gadolinium(Gd)on the corrosion behavior of as-cast MgZn_(2)_Y(2.66) and MgZn_(2)Gd_(2.66)alloys.The results show that the MgZn_(2)_Y(2.66) alloy is mainly composed ofα-Mg phase and long period stacking ordered(LPSO)phase,while MgZn_(2)Gd_(2.66)alloy is mainly composed ofα-Mg phase and(Mg,Gd)_(3)Zn phase(W phase).Generally speaking,the corrosion phenomena of the two alloys in 3.5 wt%NaCl solution are similar.In the early stages of exposure,the alloys underwent uniform corrosion at a relatively low corrosion rate.With prolonged exposure,localized corrosion became dominated and the corrosion rate was greatly increased.However,the corrosion rate of the MgZn_(2)_Y(2.66) alloy,in terms of the corrosion current density,is about one order of magnitude lower than that of the MgZn_(2)Gd_(2.66)alloy.The high corrosion resistance of the MgZn_(2)_Y(2.66) alloy is mainly attributed to the presence of LPSO phase in form of continuous networks and the relatively high corrosion resistance of the corrosion product layer on the alloy.Guoqiang Xi Xuhan Zhao Yanlong Ma Yu Mou Ju Xiong Kai Ma Jingfeng Wang 2023Acta Metallurgica Sinica(English Letters)2023,36,2:0

8Noise canceled graphene-microcavity fiber laser sensor for ultrasensitive gas detection显示文摘Optical microcavities offer a promising platform for highly efficient light–matter interactions.Recently,the combination of microresonators and 2D materials in the nanoscale has further enriched the optoelectronics of microcavity geometries,spurring broad advances including lasers,nonlinear converters,modulators,and sensors.Here,we report the concept of compact dual-laser cogeneration in a graphene-microcavity fiber,which offers a way to cancel the optical common mode noises.Driven by a single 980 nm pump,orthogonally polarized laser lines are generated in a pair of degeneracy breaking modes.The two laser lines produce a heterodyne beat note at 118.96 MHz,with frequency noise down to 200 Hz~2∕Hz at 1 MHz offset,demonstrating a linewidth of 930 Hz in vacuum.This compact device enables on-line and label-free NH_(3) gas detection with high resolution,realizing a detection limit on a single pmol/L level,and a capability to quantitatively trace gas–graphene interactions.Such a combination of graphene optoelectronics and microcavity photonics demonstrates a novel physical paradigm for microlaser control and offers a new scheme for in situ chemical sensing.YUCHEN WANG YIWEI LI YICHENG LI HAO ZHANG ZIHAN LIU YANHONG GUO ZEPING WANG JUN HE XUHAN GUO YIPING WANG BAICHENG YAO 2023Photonics Research2023,11,8:0
9Monolithic integration of embedded Ⅲ-Ⅴ lasers on SOI显示文摘Silicon photonic integration has gained great success in many application fields owing to the excellent optical device properties and complementary metal-oxide semiconductor(CMOS)compatibility.Realizing monolithic integration of Ⅲ-Ⅴ lasers and silicon photonic components on single silicon wafer is recognized as a long-standing obstacle for ultradense photonic integration,which can provide considerable economical,energy-efficient and foundry-scalable onchip light sources,that has not been reported yet.Here,we demonstrate embedded InAs/GaAs quantum dot(QD)lasers directly grown on trenched silicon-on-insulator(SOI)substrate,enabling monolithic integration with buttcoupled silicon waveguides.By utilizing the patterned grating structures inside pre-defined SOI trenches and unique epitaxial method via hybrid molecular beam epitaxy(MBE),high-performance embedded InAs QD lasers with monolithically out-coupled silicon waveguide are achieved on such template.By resolving the epitaxy and fabrication challenges in such monolithic integrated architecture,embedded Ⅲ-Ⅴ lasers on SOI with continuous-wave lasing up to 85°C are obtained.The maximum output power of 6.8mW can be measured from the end tip of the butt-coupled silicon waveguides,with estimated coupling efficiency of approximately-6.7 dB.The results presented here provide a scalable and low-cost epitaxial method for the realization of on-chip light sources directly coupling to the silicon photonic components for future high-density photonic integration.Wen-Qi Wei An He Bo Yang Zi-Hao Wang Jing-Zhi Huang Dong Han Ming Ming Xuhan Guo Yikai Su Jian-Jun Zhang Ting Wang 2023Light(Science & Applications)2023,12,5:0

11PRKG1 mutation identified by whole-exome sequencing:a potential genetic etiology for He-Zhao deficiency显示文摘Objective:He-Zhao deficiency was originally described as a severe type of nonsyndromic hypodontia,and the causative gene locus was mapped to chromosome 10q11.2.The aim of this study was to identify potential genetic mutations that could cause He-Zhao deficiency.Methods:Patients with He-Zhao deficiency and their unaffected relatives of the large pedigree were investigated.The whole-exome sequencing using next-generation sequencing was employed to identify genetic variants.The data generated from the whole-exome sequencing using the Illumina Novaseq 6000 system were further analyzed by Burrows-Wheeler Aligner software,Sequence Alignment/Map tools and ANNOVAR tool.In vitro luciferase assay was used to investigate the effect of the detected mutation on gene expression.R environment was used to conduct t-tests.The study protocol was approved by the Research Ethics Committee of Bio-X Institutes,Shanghai Jiao Tong University(M2011004).Results:The exomes of five patients with He-Zhao deficiency and two of their unaffected relatives identified a mutation in PRKG1αas the molecular etiology of the disease.The variant c.-144 C>A of PRKG1 isoform 1 cosegregated with permanent tooth agenesis in 93 family members who were older than 12,at which time the primary teeth should have been replaced with permanent teeth.Functional studies suggested that the mutant allele promotes gene transcription by increasing its promoter activity.Conclusion:c.-144 C>A variant of PRKG1αinvolving odontoclast-associated root resorption is responsible for He-Zhao deficiency,unlike other forms of hypodontia,which typically involve odontoblast dysfunction.Xiaowen Hu Dandan Wang Xuhan Yang Zhongchen Song Zuolin Wang Juan Zhang Chunling Wan Lin He 2022Journal of Bio-X Research2022,5,3:0
