| 2 | Expression of human FUS/TLS in yeast leads to protein aggregation and cytotoxicity,recapitulating key features of FUS proteinopathy显示文摘Mutations in the fused in sarcoma/translocated in liposarcoma(FUS/TLS)gene have been associated with amyotrophic lateral sclerosis(ALS).FUS-positive neuropathology is reported in a range of neurodegenerative diseases,including ALS and fronto-temporal lobar degeneration with ubiquitin-positive pathology(FTLDU).To examine protein aggregation and cytotoxicity,we expressed human FUS protein in yeast.Expression of either wild type or ALS-associated R524S or P525L mutant FUS in yeast cells led to formation of aggregates and cytotoxicity,with the two ALS mutants showing increased cytotoxicity.Therefore,yeast cells expressing human FUS protein recapitulate key features of FUSpositive neurodegenerative diseases.Interestingly,a significant fraction of FUS expressing yeast cells stained by propidium iodide were without detectable protein aggregates,suggesting that membrane impairment and cellular damage caused by FUS expression may occur before protein aggregates become microscopically detectable and that aggregate formation might protect cells from FUS-mediated cytotoxicity.The N-terminus of FUS,containing the QGSY and G rich regions,is sufficient for the formation of aggregates but not cytotoxicity.The C-terminal domain,which contains a cluster of mutations,did not show aggregation or cytotoxicity.Similar to TDP-43 when expressed in yeast,FUS protein has the intrinsic property of forming aggregates in the absence of other human proteins.On the other hand,the aggregates formed by FUS are thioflavin T-positive and resistant to 0.5%sarkosyl,unlike TDP-43 when expressed in yeast cells.Furthermore,TDP-43 and FUS display distinct domain requirements in aggregate formation and cytotoxicity. | Kazuo Fushimi Charles Long Neha Jayaram Xiaoping Chen Liming Li Jane Y.Wu | 2011 | Protein & Cell2011,2,2: | 4 |
| 3 | Evolution of microstructure and texture in copper during repetitive extrusion-upsetting and subsequent annealing显示文摘The evolution of the microstructure and texture in copper has been studied during repetitive extrusionupsetting(REU) to a total von Mises strain of 4.7 and during subsequent annealing at different temperatures. It is found that the texture is significantly altered by each deformation pass. A duplex 001 + 111 fiber texture with an increased 111 component is observed after each extrusion pass,whereas the 110 fiber component dominates the texture after each upsetting pass. During REU, the microstructure is refined by deformation-induced boundaries. The average cell size after a total strain of 4.7 is measured to be ~0.3 μm. This refined microstructure is unstable at room temperature as is evident from the presence of a small number of recrystallized grains in the deformed matrix. Pronounced recrystallization took place during annealing at 200?C for 1 h with recrystallized grains developing predominantly in high misorientation regions. At 350?C the microstructure is fully recrystallized with an average grain size of only 2.3 μm and a very weak crystallographic texture. | 4 | Update on Sphincter of Oddi Dysfunction: A Review显示文摘Sphincter of Oddi dysfunction(SOD)encompasses a spectrum of clinical syndromes that are not fully understood,and various diagnostic and therapeutic methods have had varying results depending on the type of dysfunction.This review explored various mechanisms that might play a role in SOD and methods of diagnosis and management.It is important to rule out other causes of abdominal pain with laboratory testing,imaging studies,and endoscopic procedures.Medications that affect sphincter motility should be identified as well.Manometry is the gold standard for diagnosis but it is not always required.For example,patients with type I SOD may have symptomatic improvement with sphincterotomy without need for a diagnostic manometry.Hepatobiliary scintigraphy and fatty meal sonography may also have diagnostic utility.Sphincterotomy is not always effective for symptomatic improvement in type II and III SOD.Alternate therapies with calcium channel blockers and botulinum toxin have been studied and might be considered as options after discussing the risks and benefits with the patients. | Jaimy Villavicencio Kim George Y.Wu | 2022 | Journal of Clinical and Translational Hepatology2022,10,3: | 4 |

| 6 | Methods for a blind analysis of isobar data collected by the STAR collaboration显示文摘In 2018,the STAR collaboration collected data from^(96)_(44)Ru+^(96)_(44)Ru and^(96)_(40)Zr+^(96)_(40)Zr at√^(S)NN=200 Ge V to search for the presence of the chiral magnetic effect in collisions of nuclei.The isobar collision species alternated frequently between 9644 Ru+^(96)_(44)Ru and^(96)_(40)Zr+^(96)_(40)Zr.In order to conduct blind analyses of studies related to the chiral magnetic effect in these isobar data,STAR developed a three-step blind analysis procedure.Analysts are initially provided a'reference sample'of data,comprised of a mix of events from the two species,the order of which respects time-dependent changes in run conditions.After tuning analysis codes and performing time-dependent quality assurance on the reference sample,analysts are provided a species-blind sample suitable for calculating efficiencies and corrections for individual≈30-min data-taking runs.For this sample,species-specific information is disguised,but individual output files contain data from a single isobar species.Only run-by-run corrections and code alteration subsequent to these corrections are allowed at this stage.Following these modifications,the'frozen'code is passed over the fully un-blind data,completing the blind analysis.As a check of the feasibility of the blind analysis procedure,analysts completed a'mock data challenge,'analyzing data from Au+Au collisions at√^(S)NN=27 Ge V,collected in 2018.The Au+Au data were prepared in the same manner intended for the | 7 | Designing Bulk Metallic Glass Composites with Enhanced Formability and Plasticity显示文摘To address the main stumbling-block of bulk metallic glasses(BMGs), i.e., room temperature brittleness,designing BMG matrix composites has been attracted extensive attention. Up to date, BMG composites in various alloy systems have been successfully developed by forming crystalline phases embedded in the amorphous matrix through either ex-situ or in-situ methods. In this paper, a brief review of our recent work in this topic will be presented and the novel approaches to improving composite formability and mechanical properties will also be highlighted. The main purpose of this manuscript is not to offer a comprehensive review of all the BMG composites, but instead focuses will be placed on illustrating recently developed advanced BMG composites including Fe-based BMG composite with no metalloids, Al-based BMG composite and BMG composites reinforced by the TRIP(transformation-induced plasticity) effects. | 12 | Enhancing strength and ductility of AlSi10Mg fabricated by selective laser melting by TiB_(2)nanoparticles显示文摘In the metallic components fabricated by the emerging selective laser melting(SLM)technology,most strategies used for strengthening the materials sacrifice the ductility,leading to the so-called strengthductility trade-off.In the present study,we report that the strength and ductility of materials can be enhanced simultaneously by introducing nanoparticles,which can break the trade-off of the metallic materials.In the case of in-situ nano-TiB_(2)decorated AlSi10Mg composites,the introduced nanoparticles lead to columnar-to-equiaxed transition,grain refinement and texture elimination.With increasing content of nanoparticles,the strength increases continually.Significantly,the ductility first increases and then decreases.Our results show that the ductility is controlled by the competition between the crack-induced catastrophic fracture and ductile fracture associated with dislocation activities.The first increase of ductility is mainly attributed to the suppression of crack-induced catastrophic fracture when TiB_(2)nanoparticles present.With the further increase of TiB_(2)nanoparticles,the subsequent decrease of ductility is mainly controlled by dislocation activities.Thus,the materials will exhibit the optimum strength and ductility combination in a certain range of TiB_(2)nanoparticles.This study clarifies the physical mechanism controlling ductility for nano-TiB_(2)decorated Al Si10Mg composites,which provides the insights for the design of structural materials. | Y.K.Xiao H.Chen Z.Y.Bian T.T.Sun H.Ding Q.Yang Y.Wu Q.Lian Z.Chen H.W.Wang | 2022 | Journal of Materials Science & Technology2022,,14: | 2 |

| 15 | Persistently Rising Alpha-fetoprotein in the Diagnosis of Hepatocellular Carcinoma:A Review显示文摘Hepatocellular carcinoma(HCC),one of the most common malignant tumors worldwide,is known for its grim prognosis,with untreated life expectancy being only a matter of months after the diagnosis.The difficulty in making a diagnosis early is one of the main contributing factors to the poor prognosis.Alpha-fetoprotein(AFP)had long been used as a surveillance tool,but suboptimal specificity and sensitivity has prompted liver societies to abandon the recommendation for its universal use,even in combination with ultrasonography.Most studies have shown no obvious correlation between serum AFP level and HCC tumor size,stage,or survival post-diagnosis.However,some studies concluded that a gradual rise or persistent elevation in AFP were positive predictors for tumor development.Other studies reported a fall in AFP followed by a rise in patients with HCC as well as persistently rising AFP levels without development of HCC on follow up.Our calculation of the sensitivity and specificity of persistently rising AFP for HCC were both low,at 60%and 35.8%,respectively,indicating that the presence of persistently rising AFP per se did not offer diagnostic benefit.In addition,our calculated mean slopes of persistently rising AFP levels in HCC and non-HCC patients were numerically very different,but the difference was not statistically significant.We conclude that the published data do not support a role for rising AFP levels per se in the diagnosis of HCC. | Alla Turshudzhyan George Y.Wu | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 2 |
| 16 | Tailoring grain growth and solid solution strengthening of single-phase CrCoNi medium-entropy alloys by solute selection显示文摘In the present study,we selected solutes to be added to the Cr Co Ni medium-entropy alloy(MEA)based on the mismatch of self-diffusion activation energy(SDQ)between the alloying elements and constituent elements of the matrix,and then investigated their grain growth behavior and mechanical properties.Mo and Al were selected as the solutes for investigation primarily because they have higher and lower SDQ,respectively,than those of the matrix elements;a secondary factor was their higher and lower shear modulus.Their concentrations were fixed at 3 at.%each because previous work had shown these compositions to be single-phase solid solutions with the face-centered cubic structure.Three alloys were produced by arc melting,casting,homogenizing,cold rolling and annealing at various temperatures and times to produce samples with different grain sizes.They were(a)the base alloy Cr Co Ni,(b)the base alloy plus 3 at.%Mo,and(c)the base alloy plus 3 at.%Al.The activation energies for grain growth of the Cr Co Ni,Cr Co Ni-3Mo and CrCo Ni-3Al MEAs were found to be^251,~368 and^219 k J/mol,respectively,consistent with the notion that elements with higher SDQ(in this study Mo)retard grain growth(likely by a solute-drag effect),whereas those with lower values(Al)accelerate grain growth.The roomtemperature tensile properties show that Mo increases the yield strength by^40%but Al addition has a smaller strengthening effect consistent with their relative shear moduli.The yield strength as a function of grain size for the three single-phase MEAs follows the classical Hall-Petch relationship with much higher slopes(>600 MPaμm-0.5)than traditional solid solutions.This work shows that the grain growth kinetics and solid solution strengthening of the Cr Co Ni MEA can be tuned by selecting solute elements that have appropriate diffusion and physical properties. | G.W.Hu L.C.Zeng H.Du X.W.Liu Y.Wu P.Gong Z.T.Fan Q.Hu E.P.George | 2020 | Journal of Materials Science & Technology2020,54,19: | 2 |

| 20 | Spliceosomal genes in the D.discoideum genome:a comparison with those in H.sapiens,D.melanogaster,A.thaliana and S.cerevisiae显示文摘Little is known about pre-mRNA splicing in Dictyostelium discoideum although its genome has been completely sequenced.Our analysis suggests that pre-mRNA splicing plays an important role in D.discoideum gene expression as two thirds of its genes contain at least one intron.Ongoing curation of the genome to date has revealed 40 genes in D.discoideum with clear evidence of alternative splicing,supporting the existence of alternative splicing in this unicellular organism.We identified 160 candidate U2-type spliceosomal proteins and related factors in D.discoideum based on 264 known human genes involved in splicing.Spliceosomal small ribonucleoproteins(snRNPs),PRP19 complex proteins and late-acting proteins are highly conserved in D.discoideum and throughout the metazoa.In non-snRNP and hnRNP families,D.discoideum orthologs are closer to those in A.thaliana,D.melanogaster and H.sapiens than to their counterparts in S.cerevisiae.Several splicing regulators,including SR proteins and CUGbinding proteins,were found in D.discoideum,but not in yeast.Our comprehensive catalog of spliceosomal proteins provides useful information for future studies of splicing in D.discoideum where the efficient genetic and biochemical manipulation will also further our general understanding of pre-mRNA splicing. | Bing Yu Petra Fey Karen E.Kestin-Pilcher Alexei Fedorov Ashwin Prakash Rex L.Chisholm Jane Y.Wu | 2011 | Protein & Cell2011,2,5: | 1 |
