3Opposing intraspecific vs.interspecific diversity effects on herbivory and growth in subtropical experimental tree assemblages显示文摘Aims Positive plant diversity-ecosystem function relations are ultimately driven by variation in functional traits among individuals that form a community.To date,research has largely focused on the role of species diversity for ecosystem functioning.However,substantial intraspecific trait variation is common and a significant part of this variation caused by genetic differences among individuals.Here,we studied the relative importance of species diversity and seed family(SF)diversity within species for growth and herbivory in experimental subtropical tree assemblages.Methods In 2010,we set up a field experiment in subtropical China,using four species from the local species pool.Trees were raised from seeds,with seeds from the same mother tree forming an SF.We established 23 plots containing one or four species(species diversity treatment)and one or four SFs per species(SF diversity treatment).Tree growth(stem diameter,plant height and crown expansion)and herbivory(percentage leaf loss due to leaf chewers)were monitored annually from 2011 to 2013.Important findings Tree species richness promoted growth but had no effect on herbivory.In contrast,SF diversity reduced growth and increased herbivory but only so in species mixtures.Most of the observed effects were time dependent,with the largest effect found in 2013.Our results suggest that biodiversity can affect plant performance directly via tree species-species interactions,or context dependent,via potential effects on inter-trophic interactions.Two important conclusions should be drawn from our findings.Firstly,in future studies regarding biodiversity and ecosystem functioning(BEF)relationships,intraspecific genetic diversity should be given similar weight as species diversity as it has often been neglected and its effects are not well understood.Secondly,we demonstrate opposite effects of biodiversity among and within species,stressing the importance to consider the effects of multiple levels of biodiversity simultaneously.Christoph Z.Hahn Pascal A.Niklaus Helge Bruelheide Stefan G.Michalski Miaomiao Shi Xuefei Yang Xueqin Zeng Markus Fischer Walter Durka 2017Journal of Plant Ecology2017,10,1:11
4Evaluating efficacy of screening for upper gastrointestinal cancer in China:a study protocol for a randomized controlled trial显示文摘Objective: To evaluate the efficacy and feasibility of screening procedure for upper gastrointestinal cancer in both high-risk and non-high-risk areas in China.Setting: Seven cities/counties, representing three economical-geographical regions(Eastern, Central and Western) in China, were selected as screening centers: three in high-risk areas and four in non-high-risk areas.Participants: Villages/communities in these seven centers regarded as clusters were randomly assigned to either intervention group(screening by endoscopic examination) or control group(with normal community care) in a 1:1ratio stratified by each center. Eligible participants are local residents aged 40–69 years in the selected villages/communities with no history of cancer or endoscopic examination in the latest 3 years who are mentally and physically competent. Those who are not willing to take endoscopic examination or are unwilling to sign the consent form are excluded from the study. Totally 140,000 participants will be enrolled.Interventions: In high-risk areas of upper gastrointestinal cancer, all subjects in screening group will be screened by endoscopy. In non-high-risk areas, 30% of the subjects in screening group, identified through a survey,will be screened by endoscopy.Primary and secondary outcome measures: The primary outcome is the mortality caused by upper gastrointestinal cancer. The secondary outcomes include detection rate, incidence rate, survival rate, and clinical stage distribution. Additional data on quality of life and cost-effectiveness will also be collected to answer important questions regarding screening effects.Conclusions: Screening strategy evaluated in those areas with positive findings may be promoted nationally and applied to the majority of Chinese people. 5Epidermal growth factor improves intestinal morphology by stimulating proliferation and differentiation of enterocytes and mTOR signaling pathway in weaning piglets显示文摘Epidermal growth factor(EGF) has been shown to improve piglet intestinal morphology and epithelial recovery. In an attempt to further understand the mechanisms behind these improvements, this study tested the hypothesis that dietary EGF may affect intestinal morphology by stimulating the proliferation and differentiation of enterocytes in weaning piglets. In piglets receiving200 μg kg^-1 EGF, crypt depth and villus height increased(P<0.05). Adding 400 μg kg^-1 EGF increased villus height-to-crypt depth ratio(P<0.05), but reduced crypt depth(P<0.05). Dietary supplementation with 200 μg kg^-1 EGF significantly increased the number of Ki67-positive cells(P<0.01) and tended to increase the mRNA level of proliferating cell nuclear antigen(P<0.10).However, this supplementation decreased the expression level of intestinal fatty acid-binding protein(P<0.05). Piglets fed with400 μg kg^-1 EGF had an increased mRNA level of intestinal alkaline phosphatase(P<0.05). The phosphorylation of m TOR(mammalian target of rapamycin) was observed in the 200 μg kg^-1 EGF group. 6The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 7s-HBEGF/SIRT1 circuit-dictated crosstalk between vascular endothelial cells and keratinocytes mediates sorafenib-induced hand-foot skin reaction that can be reversed by nicotinamide显示文摘Hand-foot skin reaction(HFSR),among the most significant adverse effects of sorafenib,has been limiting the clinical benefits of this frontline drug in treating various malignant tumors.The mechanism underlying such toxicity remains poorly understood,hence the absence of effective intervention strategies.In the present study,we show that vascular endothelial cells are the primary cellular target of sorafenib-induced HFSR wherein soluble heparin-binding epidermal growth factor(s-HBEGF)mediates the crosstalk between vascular endothelial cells and keratinocytes.Mechanistically,s-HBEGF released from vascular endothelial cells activates the epidermal growth factor receptor(EGFR)on keratinocytes and promotes the phosphorylation of c-Jun N-terminal kinase 2(JNK2),which stabilizes sirtuin 1(SIRT1),an essential keratinization inducer,and ultimately gives rise to HFSR.The administration of s-HBEGF in vivo could sufficiently induce hyper-keratinization without sorafenib treatment.Furthermore,we report that HBEGF neutralization antibody,Sirtl knockdown,and a classic SIRTI inhibitor nicotinamide could all significantly reduce the sorafenib-induced HFSR in the mouse model.It is noteworthy that nicotinic acid,a prodrug of nicotinamide,could substantially reverse the sorafenib-induced HFSR in ten patients in a preliminary clinical study.Collectively,our findings reveal the mechanism of vascular endothelial cell-promoted keratinization in keratinocytes and provide a potentially promising therapeutic strategy for the treatment of sorafenib-induced HFSR.Peihua Luo Hao Yan Xueqin Chen Ying Zhang Ziying Zhao Ji Cao Yi Zhu Jiangxia Du Zhifei Xu Xiaochen Zhang Su Zeng Bo Yang Shenglin Ma Qiaojun He 2020Cell Research2020,30,9:5

9Pseudocapacitance boosted N-doped carbon coated Fe7S8 nanoaggregates as promising anode materials for lithium and sodium storage显示文摘Herein,the core-shell structured N-doped carbon coated Fe7S8 nano-aggregates(Fe7S8@NC)were controllably prepared via a simple three-step synthesis strategy.The appropriate thickness of N-doped carbon layer outside Fe7S8 nano-aggregates can not only inhibit the particle pulverization induced by the big volume changes of Fe7S8,but can increase the electron transfer efficiency.The hierarchical Fe7S8 nano-aggregates composed of some primary nanoparticles can accelerate the lithium or sodium diffusion kinetics.As anode materials for Li-ion batteries(LIBs),the well-designed Fe7S8@NC nanocomposites exhibit outstanding lithium storage performance,which is better than that of pure Fe7S8,Fe3O4@NC and Fe7S8@C.Among these nanocomposites,the N-doped carbon coated Fe7S8 with carbon content of 26.87 wt.%shows a high reversible specific capacity of 833 mAh·g^−1 after 1,000 cycles at a high current density of 2 A·g^−1.The above electrode also shows excellent high rate sodium storage performance.The experimental and theoretical analyses indicate that the outstanding electrochemical performance could be attributed to the synergistic effect of hierarchical Fe7S8 nanostructure and conductive N-doped carbon layer.The quantitative kinetic analysis indicates that the charge storage of Fe7S8@NC electrode is a combination of diffusion-controlled battery behavior and surface-induced capacitance behavior.Yanli Zhou Ming Zhang Qi Wang Jian Yang Xingyun Luo Yanlu Li Rong Du Xinsheng Yan Xueqin Sun Caifu Dong Xiaoyu Zhang Fuyi Jiang 2020Nano Research2020,13,3:4
10Intelligent lesion blood–brain barrier targeting nano-missiles for Alzheimer's disease treatment by anti-neuroinflammation and neuroprotection显示文摘The treatment of Alzheimer's disease(AD)is one of the most difficult challenges in neurodegenerative diseases due to the insufficient blood‒brain barrier(BBB)permeability and unsatisfactory intra-brain distribution of drugs.Therefore,we established an ibuprofen and FK506 encapsulated drug co-delivery system(Ibu&FK@RNPs),which can target the receptor of advanced glycation endproducts(RAGE)and response to the high level of reactive oxygen species(ROS)in AD.RAGE is highly and specifically expressed on the lesion neurovascular unit of AD,this property helps to improve targeting specificity of the system and reduce unselective distribution in normal brain.Meanwhile,these two drugs can be specifically released in astrocytes of AD lesion in response to high levels of ROS.As a result,the cognition of AD mice was significantly improved and the quantity of Aβplaques was decreased.Neurotoxicity was also alleviated with structural regeneration and functional recovery of neurons.Besides,the neuroinflammation dominated by NF-κB pathway was significantly inhibited with decreased NF-κB and IL-1βin the brain.Overall,Ibu&FK@RNPs can efficiently and successively target diseased BBB and astrocytes in AD lesion.Thus it significantly enhances intracephalic accumulation of drugs and efficiently treats AD by anti-neuroinflammation and neuroprotection.Xueqin He Xiaorong Wang Lianyi Yang Zhihang Yang Wenqi Yu Yazhen Wang Rui Liu Meiwan Chen Huile Gao 2022Acta Pharmaceutica Sinica B2022,12,4:3
11Essential role of Msx1 in regulating anterior-posterior patterning of the secondary palate in mice显示文摘Development of the secondary palate displays molecular heterogeneity along the anterior-posterior axis;however, the underlying molecular mechanism remains largely unknown. MSX1 is an anteriorly expressed transcription repressor required for palate development. Here, we investigate the role of Msx1 in regional patterning of the secondary palate. The Wnt1-Cre-mediated expression of Msx1(Rosa Msx1^(Wnt1-Cre))throughout the palatal mesenchyme leads to cleft palate in mice, associated with aberrant cell proliferation and cell death. Osteogenic patterning of the hard palate in Rosa Msx1^(Wnt1-Cre)mice is severely impaired, as revealed by a marked reduction in palatine bone formation and decreased expression of the osteogenic regulator Sp7. Overexpression and knockout of Msx1 in mice show that the transcription repressor promotes the expression of the anterior palate-specific Alx1 but represses the expression of the medialposterior palate genes Barx1, Meox2, and Tbx22. Furthermore, Tbx22 constitutes a direct Msx1 target gene in the secondary palate, suggesting that Msx1 can directly repress the expression of medial-posterior specific genes. Finally, we determine that Sp7 is downstream of Tbx22 in palatal mesenchymal cells,suggesting that a Msx1/Tbx22/Sp7 axis participates in the regulation of palate development. 15Metagenomic insights into the effects of submerged plants on functional potential of microbial communities in wetland sediments显示文摘Submerged plants in wetlands play important roles as ecosystem engineers to improve self-purification and promote elemental cycling.However,their effects on the functional capacity of microbial communities in wetland sediments remain poorly understood.Here,we provide detailed metagenomic insights into the biogeochemical potential of microbial communities in wetland sediments with and without submerged plants(i.e.,Vallisneria natans).A large number of functional genes involved in carbon(C),nitrogen(N)and sulfur(S)cycling were detected in the wetland sediments.However,most functional genes showed higher abundance in sediments with submerged plants than in those without plants.Based on the comparison of annotated functional genes in the N and S cycling databases(i.e.,NCycDB and SCycDB),we found that genes involved in nitrogen fixation(e.g.,nifD/H/K/W),assimilatory nitrate reduction(e.g.,nasA and nirA),denitrification(e.g.,nirK/S and nosZ),assimilatory sulfate reduction(e.g.,cysD/H/J/N/Q and sir),and sulfur oxidation(e.g.,glpE,soeA,sqr and sseA)were significantly higher(correctedp<0.05)in vegetated vs.unvegetated sediments.This could be mainly driven by environmental factors including total phosphorus,total nitrogen,and C:N ratio.The binning of metagenomes further revealed that some archaeal taxa could have the potential of methane metabolism including hydrogenotrophic,acetoclastic,and methylotrophic methanogenesis,which are crucial to the wetland methane budget and carbon cycling.This study opens a new avenue for linking submerged plants with microbial functions,and has further implications for understanding global carbon,nitrogen and sulfur cycling in wetland ecosystems.Binhao Wang Xiafei Zheng Hangjun Zhang Xiaoli Yu Yingli Lian Xueqin Yang Huang Yu Ruiwen Hu Zhili He Fanshu Xiao Qingyun Yan 2021Marine Life Science & Technology2021,3,4:1
16Simultaneously improving the EMI shielding performances and mechanical properties of CF/PEKK composites via MXene interfacial modification显示文摘In this study, two-dimensional MXene (Ti3 C2 Tx ) was employed to modify the interface of carbon fiber-reinforced polyetherketoneketone (CF/PEKK) composites, in order to simultaneously improve the electromagnetic interference (EMI) shielding performances and mechanical properties. The obtained CF/PEKK composites possessed outstanding EMI and mechanical performances, as anticipated. Specifically, the CF/PEKK composites modified with MXene at 1 mg mL–1 exhibited an excellent EMI shielding effectiveness of 65.2 dB in the X-band, a 103.1% enhancement compared with the unmodified CF/PEKK composites. The attractive EMI shielding performances of CF/PEKK composites originated from enhanced ohmic losses and multiple reflections of electromagnetic waves with the help of the MXene and CF layers. In addition, CF/PEKK composites achieved the best mechanical properties by optimizing the dispersion concentration of MXene to 0.1 mg mL–1 . The flexural strength, flexural modulus, and interlaminar shear strength of CF/PEKK composites reached 1127 MPa, 81 GPa, and 89 MPa, which were 28.5%, 9.5%, and 29.7% higher than that of the unmodified CF/PEKK composites, respectively. 20Geographic differentiation and phylogeographic relationships among world soybean populations显示文摘A fast-growing protein and oil crop,soybean was domesticated in ancient China and disseminated early in Asia and afterwards to other continents,in particular the Americas in recent centuries.After adaptation,locally developed landraces and cultivars formed a diversity of geographic-populations.In an investigation of their phylogeographic features,marker-derived traits were combined with geography-related photo-and temperaturesensitive traits to study 13 geographic-populations comprising 371 accessions.Extreme differentiation among geographic-populations was observed for flowering date(33–94 days),maturity date(79–181 days),and main stem node number(6–25 nodes).Restriction-site associated DNA sequencing revealed strong genetic differentiation among these geographic-populations,including genetic richness(alleles,35,242–44,986)and specific-present alleles(SPAs,0–67).More SPAs(28–67)emerged in some secondary and tertiary centers than in centers of origin(8–11).Phenotypic and genotypic clustering divided 11 of the 13 geographic-populations into the same five sets of sensitivity-similar geographic-populations and grouped the populations of northeast China and northern North America rather than center-of-origin populations as secondary centers,indicating the importance of geography-related traits in determining genetic differences among geographic-populations.A model of four soybean dissemination paths is presented:from the center of origin to the north,east,and south in Asia and from northeast China to Europe and the Americas.These findings provide a detailed phylogeographic understanding of worldwide soybeans.Xueqin Liu Jianbo He Yufeng Wang Guangnan Xing Yan Li Shouping Yang Tuanjie Zhao Junyi Gai 2020The Crop Journal2020,8,2:1
