1Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 2Smartphone-powered iontophoresis-microneedle array patch for controlled transdermal delivery显示文摘The incidence rate of diabetes has been increasing every year in nearly all nations and regions.The traditional control of diabetes using transdermal insulin delivery by metal needles is generally associated with pain and potential infections.While microneedle arrays(MAs)have emerged as painless delivery techniques,the integration of MA systems with electronic devices to precisely control drug delivery has rarely been realized.In this study,we developed an iontophoresis-microneedle array patch(IMAP)powered by a portable smartphone for the active and controllable transdermal delivery of insulin.The IMAP in situ integrates iontophoresis and charged nanovesicles into one patch,achieving a one-step drug administration strategy of“penetration,diffusion and iontophoresis”.The MA of the IMAP is first pressed on the skin to create microholes and then is retracted,followed by the iontophoresis delivery of insulin-loaded nanovesicles through these microholes in an electrically controlled manner.This method has synergistically and remarkably enhanced controlled insulin delivery.The amount of insulin can be effectively regulated by the IMAP by applying different current intensities.This in vivo study has demonstrated that the IMAP effectively delivers insulin and produces robust hypoglycemic effects in a type-1 diabetic rat model,with more advanced controllability and efficiency than delivery by a pristine microneedle or iontophoresis.The IMAP system shows high potential for diabetes therapy and the capacity to provide active as well as long-term glycemic regulation without medical staff care.Jingbo Yang Yanjun Li Rui Ye Ying Zheng Xiangling Li Yuzhen Chen Xi Xie Lelun Jiang 2020Microsystems & Nanoengineering2020,6,1:3
3Down regulated expression of S_(2)-RNase attenuates self-incompatibility in “Guiyou No.1”pummelo显示文摘Self-incompatibility(SI)substantially restricts the yield and quality of citrus.Therefore,breeding and analyzing selfcompatible germplasm is of great theoretical and practical signi ficance for citrus.Here,we focus on the mechanism of a self-compatibility mutation in‘Guiyou No.1'pummelo(Citrus maxima),which is a spontaneous mutant of‘Shatian’pummelo(Citrus maxima,self-incompatibility).The rate of fruit set and the growth of pollen tubes in the pistil con firmed that a spontaneous mutation in the pistil is responsible for the self-compatibility of‘Guiyou No.1'.Segregation ratios of the S genotype in progeny,expression analysis,and western blotting validated that the reduced levels of S_(2)-RNase mRNA contribute to the loss of SI in‘Guiyou No.1'.Furthermore,we report a phased assembly of the‘Guiyou No.1'pummelo genome and obtained two complete and well-annotated S haplotypes.Coupled with an analysis of SV variations,methylation levels,and gene expression,we identi fied a candidate gene(CgHB40),that may in fluence the regulation of the S/^RNase promoter.Our data provide evidence that a mutation that affects the pistilled to the loss of SI in‘Guiyou No.1'by in fluencing a poorly understood mechanism that affects transcriptional regulation.This work signi ficantly advances our understanding of the genetic basis of the SI system in citrus and provides information on the regulation of S-RNase genes.ianbing Hu Qiang Xu Chenchen Liu Binghao Liu Chongling Deng Chuanwu Chen Zhuangmin Wei Muhammad Husnain Ahmad Kang Peng Hao Wen Xiangling Chen Peng Chen Robert MLarkin Junli Ye Xiuxin Deng Lijun Chai 2021Horticulture Research2021,8,1:2
4Differential gene expression in porcine SK6 cells infected with wild-type and SAP domain-mutant foot-and-mouth disease virus显示文摘Foot-and-mouth disease virus(FMDV) is the causative agent of a highly contagious disease in livestock. The viral proteinaseL^(rop) of FMDV is involved in pathogenicity, and mutation of theL^(rop) SAP domain reduces FMDV pathogenicity in pigs. To determine the gene expression profiles associated with decreased pathogenicity in porcine cells, we performed transcriptome analysis using next-generation sequencing technology and compared differentially expressed genes in SK6 cells infected with FMDV containingL^(rop) with either a wild-type or mutated version of the SAP domain. This analysis yielded 1,853 genes that exhibited a ≥ 2-fold change in expression and was validated by real-time quantitative PCR detection of several differentially expressed genes. Many of the differentially expressed genes correlated with antiviral responses corresponded to genes associated with transcription factors, immune regulation, cytokine production, inflammatory response, and apoptosis. Alterations in gene expression profiles may be responsible for the variations in pathogenicity observed between the two FMDV variants. 15Molecular phylogenetic analysis of sulfate-reducing bacteria from deep sediment layers of the tropical West Pacific warm pool显示文摘The diversity of sulfate-reducing bacteria (SRB) from deep layers of deep-sea sediments [more than 2 m bsf (below seafloor)] of two sites (W01-3 and WP01-4) in a tropical West Pacific warm pool region was characterized by using molecular phylogenetic analysis. The results of culture-independent samples demonstrated that the dominant clones from both sites were related to Gram-positive spore forming genus, Desulfotomaculum, which accounted for 36.8% of all the sequencing clones from Site WP01-3 and 62.8% from Site WP01-4. However, the other SRB group which was generally reported to be predominant in the deep-sea sediments of other regions,δ-subclass of the proteobacteria was found to be in very low percentages. Therefore, it could be speculated that there existed a unique chemical environment in the deep-sea sediment of this warm pool region. When comparing the Desulfotomaculum sp. related sequences from both sites, it was revealed that though the Desulfotomaculum-like sequences from Site WP01-3 were more diverse than those from Site WP01-4, all these sequences from both sites showed high similarity and formed a new phylogenetically homogeneous cluster in the Desulfotomaculum genus which had never been reported before. Successful enrichment of SRB was only achieved from samples of Site WP01-4 and the sequence analysis of culture-dependent samples further confirmed the dominance of Desulfotomaculum genus. But Desulfotomaculum-related sequences from culture-dependent and culture-independent samples belonged to two different clusters respectively.
