3Mycoplasma infections and different human carcinomas显示文摘AIM To explore relationships between human carcinomas and mycoplasma infection.METHODS Monoclonal antibody PD4, which specifically recognizes a distinct protein from mycoplasma hyorhinis, was used to detect mycoplasma infection in different paraffinembedded carcinoma tissues with immunohistochemistry. PCR was applied to amplify the mycoplasma DNA from the positive samples for confirming immunohistochemistry.RESULTS Fifty of 90 cases (56%) of gastric carcinoma were positive for mycoplasma hyorhinis. In other gastric diseases, the mycoplasma infection ratio was 28% (18/49) in chronic superficial gastritis, 30% (14/ 46) in gastric ulcer and 37% (18/ 49) in intestinal metaplasia. The difference is significant with gastric cancer (X2=12.06, P<0.05). In colon carcinoma, the mycoplasma infection ratio was 55.1% (32/58), but it was 20.9% (10/49) in adenomarous polyp (X2=13.46, P<0.005).Gastric and colon cancers with high differentiation had a higher mycoplasma infection ratio than those with low differentiation (P< 0.05). Mycoplasma infection in esophageal cancer, lung cancer, breast cancer and glioma was 50.9% (27/53), 52.6% (31/ 59), 39.7%(25/63) and 41% (38/91), respectively. The mycoplasma DNA was successfully amplified with the DNA extracted from the cancer tissues that were positive for mycoplasma infection (detected with antibody PD4).CONCLUSION There was high correlation between mycoplasma infection and different cancers, which suggests the possibility of an association between the two. 4Single-cell transcriptomic analysis defines the interplay between tumor cells,viral infection,and the microenvironment in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an Epstein-Barr virus(EBV)-associated malignancy with a complex tumor ecosystem.How the interplay between tumor cells,EBV,and the microenvironment contributes to NPC progression and immune evasion remains unclear.Here we performed single-cell RNA sequencing on^104,000 cells from 19 EBV^+NPCs and 7 nonmalignant nasopharyngeal biopsies,simultaneously profiling the transcriptomes of malignant cells,EBV,stromal and immune cells.Overall,we identified global upregulation of interferon responses in the multicellular ecosystem of NPC.Notably,an epithelial–immune dual feature of malignant cells was discovered and associated with poor prognosis.Functional experiments revealed that tumor cells with this dual feature exhibited a higher capacity for tumorigenesis.Further characterization of the cellular components of the tumor microenvironment(TME)and their interactions with tumor cells revealed that the dual feature of tumor cells was positively correlated with the expression of co-inhibitory receptors on CD8^+tumor-infiltrating T cells.In addition,tumor cells with the dual feature were found to repress IFN-γproduction by T cells,demonstrating their capacity for immune suppression.Our results provide new insights into the multicellular ecosystem of NPC and offer important clinical implications.Shanzhao Jin Ruoyan Li Ming-Yuan Chen Chao Yu Lin-Quan Tang Yan-Min Liu Jiang-Ping Li Yi-Na Liu Yi-Ling Luo Yifan Zhao Yu Zhang Tian-Liang Xia Shang-Xin Liu Qi Liu Guan-Nan Wang Rui You Jing-Yun Peng Jiang Li Feng Han Jianwei Wang Qiu-Yan Chen Li Zhang Hai-Qiang Mai Benjamin E.Gewurz Bo Zhao Lawrence S.Young Qian Zhong Fan Bai Mu-Sheng Zeng 2020Cell Research2020,30,11:16

8Ultrasensitive broadband phototransistors based on perovskite/organic-semiconductor vertical heterojunctions显示文摘Organolead halide perovskites have emerged as the most promising materials for various optoelectronic devices,especially solar cells,because of their excellent optoelectronic properties.Here,we present the first report of low-voltage high-gain phototransistors based on perovskite/organic-semiconductor vertical heterojunctions,which show ultrahigh responsivities of~10^(9)A W^(–1) and specific detectivities of~10^(14) Jones in a broadband region from the ultraviolet to the near infrared.The high sensitivity of the devices is attributed to a pronounced photogating effect that is mainly due to the long carrier lifetimes and strong light absorption in the perovskite material.In addition,flexible perovskite photodetectors have been successfully prepared via a solution process and show high sensitivity as well as excellent flexibility and bending durability.The high performance and facile solution-based fabrication of the perovskite/organic-semiconductor phototransistors indicate their promise for potential application for ultrasensitive broadband photodetection.Chao Xie Peng You Zhike Liu Li Li Feng Yan 2017Light(Science & Applications)2017,6,1:12
9Adsorption behavior of carbon dioxide and methane in bituminous coal:A molecular simulation study显示文摘The adsorption behavior of CO_2, CH_4 and their mixtures in bituminous coal was investigated in this study. First, a bituminous coal model was built through molecular dynamic(MD) simulations, and it was confirmed to be reasonable by comparing the simulated results with the experimental data. Grand Canonical Monte Carlo(GCMC)simulations were then carried out to investigate the single and binary component adsorption of CO_2 and CH_4with the built bituminous coal model. For the single component adsorption, the isosteric heat of CO_2 adsorption is greater than that of CH_4 adsorption. CO_2 also exhibits stronger electrostatic interactions with the heteroatom groups in the bituminous coal model compared with CH_4, which can account for the larger adsorption capacity of CO_2 in the bituminous coal model. In the case of binary adsorption of CO_2 and CH_4mixtures, CO_2 exhibits the preferential adsorption compared with CH_4 under the studied conditions. The adsorption selectivity of CO_2 exhibited obvious change with increasing pressure. At lower pressure, the adsorption selectivity of CO_2 shows a rapid decrease with increasing the temperature, whereas it becomes insensitive to temperature at higher pressure. 14Minimally-invasive treatment of communicating hydrocephalus using a percutaneous lumboperitoneal shunt显示文摘Objective:To investigate the clinical value of a minimally-invasive treatment of communicating hydrocephalus using a percutaneous lumboperitoneal (LP) shunt.Method:The clinical and long-term follow-up data of 256 patients suffering from communicating hydrocephalus and undergoing percutaneous LP shunt during 1998 to 2008 were retrospectively analyzed.Results:After the follow-up,which lasted 6 months to 10 years,219 cases of communicating hydrocephalus recovered well (ventricular size returned to normal and symptoms completely disappeared),25 cases were brought under control (ventricle size reduced by 50% and symptoms partially abated),and 12 cases showed no obvious changes.Fifteen obese subjects needed modifications of the shunt due to the obstruction of the abdominal end following wrapping,and one subject underwent extubation as the subject was unable to tolerate stimulation of the cauda equina.The effectiveness of shunting was 91.40% and the probability of shunt-tube obstruction,which occurs predominantly in the abdominal end,was only 5.85%,far lower than that of ventriculoperitoneal (VP) shunt.Three subjects had a history of infection following VP shunting.Conclusion:LP shunting is minimally invasive and effective in treating communicating hydrocephalus,with fewer complications.Lu JIA Zhong-xin ZHAO Chao YOU Jia-gang LIU Si-qing HUANG Min HE Pei-gang JI Jie DUAN Yi-jun ZENG Guo-ping LI 2011Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,4:6

16中国东北条子河中微生物群落变化与锌砷污染的关系研究(英文)显示文摘研究目的:通过研究河流微生物与污染物的互作关系,为河流污染治理等相关研究提供新途径。创新要点:研究选择的目标河流是报道较少的辽河二级支流,该研究对于了解支流污染特别是东北地区河流污染有重要的意义。将分子生态学技术应用于分析不同污染程度的河流底泥微生物变化,建立微生物种群变化和污染物的互作关系。研究方法:选取沿条子河的8个代表性区段,分析河水和底泥中重金属和污染物的变化情况。通过变性梯度凝胶电泳(DGGE)技术分析不同河流区段内真核和原核微生物种群变化,解析河流中污染物对底泥微生物的影响,建立微生物种群变化和重金属污染的互作关系。重要结论:重金属污染物的存在能抑制底泥中微生物生长,仅少量具有较强抗性的微生物存活(见图4);有机污染物的增加使对重金属有较强抗性的Limnodrilus hoffmeisteri 大量繁殖(见图2~4),从而大量消耗溶解氧,导致好氧微生物死亡和河水自净能力退化,最终引起水质恶化。由此可见,微生物对环境污染物的变化有很敏感的响应,通过对环境中微生物的监测有助于进一步分析环境污染的变化。Jun ZHAO Xin ZHAO Lei CHAO Wei ZHANG Tao YOU Jie ZHANG 2014Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2014,15,7:5
17Structural changes in the gray matter of unmedicated patients with obsessive-compulsive disorder: a voxel-based morphometric study显示文摘The aim of the current study was to use whole brain voxel-based morphometry(VBM)to assess the gray matter(GM)changes in unmedicated patients with obsessive-compulsive disorder(OCD)compared with normal controls.We compared the GM volumes in28 patients with 22 matched healthy controls using a1.5T MRI.Three-dimensional T1-weighted magnetic resonance images were obtained from all participants.VBM was performed to detect GM volume differences between the two groups.We detected increased regional GM volumes in the bilateral middle temporal gyri,bilateral middle occipital gyri,bilateral globus pallidus,right inferior parietal gyrus,left superior parietal gyrus,right parahippocampus,right supramarginal gyrus,right medial superior frontal gyrus,and left inferior frontal opercular cortex in the OCD patients relative to controls(P<0.001,uncorrected,cluster size>100 voxels).No decreased GM volume was found in the OCD group compared with normal controls.Our findings suggest that structural changes in the GM are not limited to fronto-striato-thalamic circuits in the pathogenesis of OCD.Temporo-parietal cortex may also play an important role.Ling Tan Qing Fan Chao You Jijun Wang Zhao Dong Xuemei Wang Kemin Chen Zeping Xiao Kaida Jiang 2013Neuroscience Bulletin2013,29,5:5

Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 19Wave Attenuation Performance and the Influencing Factors of A Lower Arc-Plate Breakwater显示文摘Comprehensive experimental and numerical studies have been undertaken to investigate wave energy dissipation performance and main influencing factors of a lower arc-plate breakwater. The numerical model, which considers nonlinear interactions between waves and the arc-plate breakwater, has been constructed by using the velocity wave- generating method, the volume of fluid (VOF) method and the finite volume method. The results show that the relative width, relative height and relative submergence of the breakwater are three main influencing factors and have significant influence on wave energy dissipation of the lower arc-plate open breakwater. The transmission coefficient is found to decrease with the increasing relative width, and the minimum transmission coefficient is 0.15 when the relative width is 0.45. The reflection coefficient is found to vary slightly with the relative width, and the maximum reflection coefficient is 0.53 when the relative width is 0.45. The transmission and reflection coefficients are shown to increase with the relative wave height for approximately 85% of the experimental tests when the relative width is 0.19 0.45. The transmission coefficients at relative submergences of 0.04, 0.02 and 0 are clearly shown to be greater than those at relative submergences of 0.02 and 0.04, while the reflection coefficient exhibits the opposite relationship. After the wave interacts with the lower arc-plate breakwater, the wave energy is mainly converted into transmission, reflection and dissipation energies. 20Screening of differentially expressed genes related to differentiation and proliferation by gene expression profiling of different grade astrocytoma cell lines显示文摘BACKGROUND: The detection of differential gene expression in brain is possible by cDNA microarray technology, and the screening of differentially expressed genes might provide a biological basis for gene-targeted therapy for tumors. OBJECTIVE: To detect the differential expression of genes among astrocytoma SHG-44 (WHO grade IV), CHG-5 (WHO grade II), and ATRA-treated SHG-44 cell lines by cDNA microarray. DESIGN: Laboratory experiments in vitro. SETTING: Department of Neurobiology, the Third Military Medical University. MATERIALS: The experiment was performed at the Department of Neurobiology in the Third Military Medical University of the Chinese PLA from January to October 2007. The SHG-44 cell line (WHO grade Ⅳ) was established by Prof. Ziwei Du, and the CHG-5 cell line (WHO grade II) was set up by Prof. Xiuwu Bian from the Third Military Medical University of the Chinese PLA. The cDNA microarray containing 9182 known genes was prepared and provided by Dr. Yang Zhong at the City University of Hong Kong. METHODS: To screen differentially expressed genes from the gene expression profiles detected by cDNA microarray comparisons were made between CHG-5 and SHG-44 cells and between SHG-44 cells with or without treatment with 10 μmol/L ATRA. Some differentially expressed genes were selected randomly for Northern Blot analysis to confirm the results of the microarray. The determination criteria for differential gene expression were as follows. ① The ratio of Cy5 signal to Cy3 was greater than 2.0 or less than 0.5. ② The results of the triplicate microarray hybridizations showed the same trend in three experiments. ③ A gene appeared at least two times on the triplicate microarray hybridizations, and the 3rd value did not show a contradictory trend. A normalized ratio of Cy5 intensity to Cy3 greater than 2.0 or less than 0.5 was considered to represent up-regulated or down-regulated gene expression, respectively. MAIN OUTCOME MEASURES: The identification of genes that were similarly regulated (overlapping) during tumor progression and differentiation, by comparison of gene expression profiles between CHG-5 and SHG-44 cells, and between SHG-44 cells with or without treatment with ATRA. RESULTS: Thirty-one overlapping genes were found to have similar regulatory effects on astrocytomas; among them, twenty genes were up-regulated and eleven were down-regulated in both comparisons between CHG-5 and SHG-44 cells, and between SHG-44 cells with or without treatment with ATRA. The four reported genes, SERPINF1, MAPK11, HIF1A and SOD2, were up-regulated in this study. CONCLUSION: The differentially expressed genes in different grade astrocytoma cell lines were revealed primarily by cDNA microarray; among them, five identified overlapping genes, SERPINF1, MAPK11, DCTN2, HIF1A and SOD2, were related to the malignant progression of astrocytoma cells.Yi Zeng Zhong Yang Yangyun Han Chao You 2008Neural Regeneration Research2008,3,3:5
