1Screening test for anti-Helicobacter pylori activity of traditional Chinese herbal medicines显示文摘AIM:To evaluate the anti-Helicobacter pylori (H.pylori) activity of 50 traditional Chinese herbal medicines in order to provide the primary evidence for their use in clinical practice.METHODS:A susceptibility test of water extract from 50 selected traditional Chinese herbal medicines for in vitro H.pylori Sydney strain 1 was performed with broth dilution method.Anti-H.pylori activity of the selected Chinese herbal medicines was evaluated according to their minimum inhibitory concentration (MIC).RESULTS:The water extract from Rhizoma Coptidis,Radix Scutellariae and Radix isatidis could significantly inhibit the H.pylori activity with their MIC less than 7.8 mg/mL,suggesting that traditional Chinese herbal medicines have anti-inflammatory and antibacterial effects and can thus be used in treatment of H.pylori infection.CONCLUSION:Rhizoma Coptidis,Radix Scutellariae and Radix isatidis are the potential sources for the synthesis of new drugs against H.pylori.Feng Ma Ye Chen Jing Li He-Ping Qing Ji-De Wang YaLi Zhang Yang Bai Bei-Guo Long 2010World Journal of Gastroenterology2010,16,44:34
2Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
3单周多西他赛加顺铂联合三维适形放疗治疗局部晚期食管癌的临床研究(英文)显示文摘Objective: This randomized controlled clinical study was to assess and compare the efficacy and safety of two chemoradiotherapy regimens [cisplatin+5-fluorouracil+3 dimensional conformal radiation therapy (3DCRT) and cisplatin+weekly docetaxel+3DCRT] in patients with locally advanced esophageal squamous cell carcinoma. Methods: A total of seventy-four patients with clinical stages IIB to IIIB esophageal squamous cell carcinoma were enrolled. Chemotherapy for PF group comprised 5-fluorouracil at days 1-5 (250 mg/m2/d) and cisplatin (20 mg/m2) at days 1-3 of every 28-day cycle; full treatment course included 2 cycles. Chemotherapy for DP group comprised docetaxel (20 mg/m2) and cisplatin (20 mg/m2) at days 1, 8, 15, 22, 29, and 36. Both groups treated with concurrent 60 Gy 3DCRT at 200 cGy/d. Results: Seventy-four patients were enrolled and 71 completed the planned treatment, with a follow-up rate of 95.94%. Short-term curative effect was not statistically significant between the two groups (P=0.471). The 2-year survival rates were 65.7% and 61.1%, respectively (P=0.806), 5 years survival rates were 34.29% and 27.78%, respectively (P=0.221), and there was no significant difference by Fisher test (P=0.734). As common side effects, incidence rates of radioactive esophagitis and hematological toxicity were lower in DP group. Conclusion: For locally advanced esophageal cancer patients, current chemoradiotherapy with chemotherapy regimen of weekly docetaxel plus cisplatin has equal curative effect with 5-fluorouracil plus cisplatin, but well-tolerated by reducing side effects such as radioactive esophagitis and bone marrow suppression.Minghua Bai Baofeng Wang Xijing Wang Hongbing Ma Yali Wang Zhongwei Wang 2013The Chinese-German Journal of Clinical Oncology2013,12,8:3
4Diagnostic value of negative enrichment and immune fluorescence in situ hybridization for intraperitoneal free cancer cells of gastric cancer显示文摘Objective:To explore the intraperitoneal free cancer cell(IFCC)detection value of negative enrichment and immune fluorescence in situ hybridization(NEimFISH)on chromosomes(CEN)8/17.Methods:To verify the reliability of NEimFISH,29 gastric cancer tumors,their adjacent tissues and greater omental tissues were tested.Our study then included 105 gastric cancer patients for IFCC.We defined patients as IFCC-positive if a signal was detected,regardless of the detailed cancer cell numbers.A comparison of clinicopathological features was conducted among IFCC groups.We also compared the diagnosis value and peritoneal recurrence predictive value among different detection methods.The comparison of IFCC number was also conducted among different groups.Results:A cutoff of 2.5 positive cells could distinguish all benign tissue samples and 97%of malignant tissue samples in our study.Compared to intestinal gastric cancer,patients with diffuse gastric cancer tended to have more IFCCs(6 vs.4,P=0.002).The IFCC counts were often higher in the lymphovascular invasion positive group than negative group(3 vs.1,P=0.022).All IFCC samples that were considered positive using conventional cytology were also found to be positive using NEimFISH.When compared to conventional cytology and paraffin pathology,NEimFISH had a higher IFCC positive rate(68.9%)and higher one-year peritoneal recurrence predictive value with area under the curve(AUC)of 0.922.Conclusions:Gastric cancer could be effectively diagnosed by NEimFISH.The IFCC number found using NEimFISH on CEN8/17 is closely associated with Lauren type and vascular invasion of cancer.NEimFISH is a reliable detection modality with a higher positive detection rate,higher one-year peritoneal recurrence predictive value and quantitative features for IFCC of gastric cancer.Anqiang Wang Zhongwu Li Qian Wang Yali Bai Xin Ji Tao Fu Ke Ji Yanwen Xue Tingxu Han Xiaojiang Wu Ji Zhang Yingjie Yang Guobin Xu Zhaode Bu Jiafu Ji 2019Chinese Journal of Cancer Research2019,31,6:3

8Regulation of angiotensin Ⅱ on Gαq/11 protein of vascular smooth muscle cell and its underlying mechanism显示文摘To study the regulation of angiotensin Ⅱ (Ang Ⅱ) on Gαq/11 protein of vascular smooth muscle cell (VSMC) and its underlying mechanism, the protein synthesis was detected by [3H]-leucine incorporation. Gαq/11 expression was measured by Western blot in cultured VSMC of rat aorta. The results showed that the level of Gαq/11 was down-regulated after stimulated by Ang Ⅱ for 1-6 h, while it was upregulated significantly by 12-24 h stimulation (P < 0.01) in VSMC. The [3H]-leucine incorporation of VSMC was increased after 24 h Ang Ⅱ stimulation. The biphase regulation of Ang Ⅱ on Gαq/11 protein was blocked by the Ang Ⅱ type I receptor (AT1) specific antagnist losartan or PLC inhibitor U73122, while PD98059 did not have this effect. 9Presence of organophosphate flame retardants(OPEs)in different functional areas in residential homes in Beijing,China显示文摘The production and application of organophosphate esters(OPEs)have dramatically increased in recent years due to their use as a replacement for brominated flame retardants.In this study,13 OPEs(∑_(13)OPEs)were analyzed in indoor air samples from kitchens and living rooms in 14 residential homes in Beijing,China.The concentrations of ∑_(13)OPEs in kitchen air samples(mean:13 ng/m^(3))were significantly(p<0.05)higher than in living room air samples(5.0 ng/m^(3)).In addition,paired window surface organic film samples were collected and analyzed to investigate film-air partitioning,exhibiting a mean concentration of ∑_(13)OPEs of 4100 ng/m^(2).The congener profiles showed that tris(2-chloroisopropyl)phosphate(TCPP)was the predominant compound in both window film samples(48%)and the corresponding indoor air sample(56%).The estimated daily intakes(EDI)of OPEs via indoor air inhalation were 2.8 and 1.4 ng/kg/day for infants and adults,respectively,both of which are below the reference dose values(RfDs).Overall,these findings indicate that OPEs in the indoor air environment of residential homes in Beijing are not likely to pose a health risk to the general population.Kun Lv Lu Bai Boyu Song Xindong Ma Minmin Hou Jie Fu Yali Shi Yawei Wang Guibin Jiang 2022Journal of Environmental Sciences2022,34,5:1
10OPTIMIZATION OF Ni BIOACCUMULATION BY SYNECHCOCCUS显示文摘Influencing factors on bioaccumulation of Ni by Synechcoccus were studied in this paper. The equilibration time of Ni bioaccumulation was about 80 min in aqueous solution. Bioaccumulation quantity reached maximum when mass ratio of Ni to dry weight concentration of Synechcoccus was 16?18%. Bioaccumulation quantity increased with in- creasing pH. The optimum pH was 9?10 and higher pH led to precipitation of Ni(OH)2. Bioaccumulation quantity was also influenced by temperature and light intensity reaching their optima at 35oC and 3 000 lx respectively. 12Single-cell transcriptome atlas reveals spatiotemporal developmental trajectories in the basal roots of moso bamboo (Phyllostachys edulis)显示文摘Roots are essential for plant growth and development.Bamboo is a large Poaceae perennial with 1642 species worldwide.However,little is known about the transcriptional atlas that underpins root cell-type differentiation.Here,we set up a modified protocol for protoplast preparation and report single-cell transcriptomes of 14279 filtered single cells derived from the basal root tips of moso bamboo.We identified four cell types and defined new cell-type-specific marker genes for the basal root.We reconstructed the developmental trajectories of the root cap,epidermis,and ground tissues and elucidated critical factors regulating cell fate determination.According to in situ hybridization and pseudotime trajectory analysis,the root cap and epidermis originated from a common initial cell lineage,revealing the particularity of bamboo basal root development.We further identified key regulatory factors for the differentiation of these cells and indicated divergent root developmental pathways between moso bamboo and rice.Additionally,PheWOX13a and PheWOX13b ectopically expressed in Arabidopsis inhibited primary root and lateral root growth and regulated the growth and development of the root cap,which was different from WOX13 orthologs in Arabidopsis.Taken together,our results offer an important resource for investigating the mechanism of root cell differentiation and root system architecture in perennial woody species of Bambusoideae.Zhanchao Cheng Changhong Mu Xiangyu Li Wenlong Cheng Miaomiao Cai Chongyang Wu Jutang Jiang Hui Fang Yucong Bai Huifang Zheng Ruiman Geng Junlei Xu Yali Xie Yuping Dou Juan Li Shaohua Mu Jian Gao 2023Horticulture Research2023,10,8:0
13Engineered human spinal cord-like tissues with dorsal and ventral neuronal progenitors for spinal cord injury repair in rats and monkeys显示文摘Transplanting human neural progenitor cells is a promising method of replenishing the lost neurons after spinal cord injury (SCI), but differentiating neural progenitor cells into the diverse types of mature functional spinal cord neurons in vivo is challenging. In this study, engineered human embryonic spinal cord-like tissues with dorsal and ventral neuronal characters (DV-SC) were generated by inducing human neural progenitor cells (hscNPCs) to differentiate into various types of dorsal and ventral neuronal cells on collagen scaffold in vitro. Transplantation of DV-SC into complete SCI models in rats and monkeys showed better therapeutic effects than undifferentiated hscNPCs, including pronounced cell survival and maturation. DV-SC formed a targeted connection with the host’s ascending and descending axons, partially restored interrupted neural circuits, and improved motor evoked potentials and the hindlimb function of animals with SCI. 14Plasma treated carbon paper electrode greatly improves the performance of iron-hydrogen battery for low-cost energy storage显示文摘A novel iron-hydrogen battery system, whose Fe^(3+)/Fe^(2+)cathode circumvents slowly dynamic oxygen reduction reaction and anode is fed with clean and cordial hydrogen, is systematically investigated. The maximum discharge power density of the iron-hydrogen battery reaches to 96.0 m W/cm^(2) under the room temperature. The capacity reaches to 17.2 Ah/L and the coulombic and energy efficiency are achieved to99% and 86%, respectively, during the galvanostatic charge-discharge test. Moreover, stable cycling test is observed for more than 240 h and 100 cycles with the iron sulfate in the sulfuric acid solutions. It is found that air plasma treatment onto the cathode carbon paper can generate the oxygen-containing groups and increase the hydrophilic pores proportion to ca. 40%, enlarging nearly 6-fold effective diffusion coefficient and improving the mass transfer in the battery performance.
